Pathologically successful conversion hepatectomy for advanced giant hepatocellular carcinoma after multidisciplinary therapy:A case report and review of literature

BACKGROUND Hepatocellular carcinoma(HCC)is one of the leading causes of death due to its complexity,heterogeneity,rapid metastasis and easy recurrence after surgical resection.We demonstrated that combination therapy with transcatheter arterial chemoembolization(TACE),hepatic arterial infusion chemotherapy(HAIC),Epclusa,Lenvatinib and Sintilimab is useful for patients with advanced HCC.CASE SUMMARY A 69-year-old man who was infected with hepatitis C virus(HCV)30 years previously was admitted to the hospital with abdominal pain.Enhanced computed tomography(CT)revealed a low-density mass in the right lobe of the liver,with a volume of 12.9 cm×9.4 cm×15 cm,and the mass exhibited a“fast-in/fast-out”pattern,with extensive filling defect areas in the right branch of the portal vein and an alpha-fetoprotein level as high as 657 ng/mL.Therefore,he was judged to have advanced HCC.During treatment,the patient received three months of Epclusa,three TACE treatments,two HAIC treatments,three courses of sintilimab,and twenty-one months of lenvatinib.In the third month of treatment,the patient developed severe side effects and had to stop immunotherapy,and the Lenvatinib dose had to be halved.Postoperative pathological diagnosis indicated a complete response.The patient recovered well after the operation,and no tumor recurrence was found.CONCLUSION Multidisciplinary conversion therapy for advanced enormous HCC caused by HCV infection has a significant effect.Individualized drug adjustments should be made during any treatment according to the patient's tolerance to treatment.

Sequential extraction of RNA,DNA and protein from cultured cells of the same group

BACKGROUND Efficient extraction of nucleic acids and proteins(ENAP)from cells is a prerequisite for precise annotation of gene function,and has become laboratory routine for revealing the mysteries of life.However,cell samples are often from different culture dishes,resulting in inevitable experimental errors and sometimes poor repeatability.AIM To explore a method to improve the efficiency of ENAP,minimizing errors in ENAP processes,enhancing the reliability and repeatability of subsequent experimental results.METHODS A protocol for the sequential isolation of RNA,DNA,and proteins from the same cultured HepG2 cells using RNAzol reagent is presented here.The first step involves culturing HepG2 cells to the exponential phase,followed by the sequential isolation of RNA,DNA,and proteins from the same cultured cells in the second step.The yield of nucleic acids and proteins is detected in the third step,and their purity and integrity are verified in the last step.RESULTS The procedure takes as few as 3-4 d from the start to quality verification and is highly efficient.In contrast to the existing kits and reagents,which are primarily based on independent isolation,this RNAzol reagent-based method is characterized by the sequential isolation of RNA,DNA,and proteins from the same cells,and therefore saves time,and has low cost and high efficiency.CONCLUSION The RNA,DNA,and proteins isolated using this method can be used for reverse transcription-polymerase chain reaction,polymerase chain reaction,and western blotting,respectively.

Effects of Pinus massoniana bark extract on cell proliferation and apoptosis of human hepatoma BEL-7402 cells

AIM: To study the effects of Pinus massoniana bark extract (PMBE) on cell proliferation and apoptosis of human hepatoma BEL-7402 cells and to elucidate its molecular mechanism.METHODS: BEL-7402 cells were incubated with various concentrations (20-200 μg/mL) of PMBE for different periods of time. After 48 h, cell proliferation was determined by 3-(4,5-dimethyl-thiazolyl-2)-2,5-diphenyl tetrazolium bromide (MTT) assay. Apoptosis was evaluated by morphological observation, agarose gel electrophoresis,and flow cytometry analysis. Possible molecular mechanisms were primarily explored through immunohistochemical staining.RESULTS: PMBE (20-200 μg/mL) significantly suppressed BEL-7402 cell proliferation in a time- and dose-dependent manner. After treatment of BEL-7402 cells with 160 μg/mL PMBE for 24, 48, or 72 h, a typical apoptotic 'DNA ladder'was observed using agarose gel electrophoresis. Nuclear condensation and boundary aggregation or split, apoptotic bodies were seen by fluorescence and electron microscopy.Sub-G1 curves were displayed by flow cytometry analysis.PMBE decreased the expression levels of Bcl-2 protein in a time-dependent manner after treatment of cells with 160 μg/mL PMBE.CONCLUSION: PMBE suppresses proliferation of BEL-7402 cells in a time- and dose-dependent manner and induces cell apoptosis by possibly downregulating the expression of the bcl-2 gene.

Single-Ni-atoms on nitrogenated humic acid based porous carbon for CO_(2)electroreduction

tWe proposed a facile synthesis of single-Ni-atom catalysts on low-cost porous carbon using a calcina-tion method at the temperatures of 850-1000°C,which were used for CO_(2)electrochemical reduction to CO.The porous carbon was prepared by carbonizing cheap and abundant humic acid.The structural characterizations of the as-synthesized catalysts and their electrocatalytic performances were analyzed.The results showed that the single-Ni-atom catalyst activated at 950°C showed an optimum catalytic performance,and it reached a CO Faradaic efficiency of 91.9%with a CO partial current density of 6.9 mAcm^(-2)at-0.9 V vs.reversible hydrogen electrode(RHE).Additionally,the CO Faradaic efficiency and current density of the optimum catalyst changed slightly after 8 h of continuous operation,suggesting that it possessed an excellent stability.The structure-activity relations indicate that the variation in the CO_(2)electroche-mical reduction performance for the as-synthesized cataly-sts is ascribed to the combined effects of the increase in the content of pyrrolic N,the evaporation of Ni and N,the decrease in pore volume,and the change in graphitization degree.

Modified NiFe-MOF with defects induced by-NH_(2)and-SH for enhanced adsorption and photocatalytic reduction of CO_(2)

In this study,a Ni-Fe metal-organic framework modified by-NH_(2)and-SH was synthesized using a sim-ple hydrothermal process for enhancing photocatalytic CO_(2)reduction.As expected,the N2S-NiFe(NH_(2)-BDC/H_(4)DSBDC,2:1)displayed excellent photocatalytic CO_(2)reduction activity and high CO selectivity(12,412.23μmol g^(−1)h^(−1))under visible light irradiation(λ≥420 nm),which is 5 times that of NiFe-MOF.Notably,the excellent photocatalytic performance of N2S-NiFe can benefit from the rich defect trap site caused by the introduction of amino and sulfhydryl groups,accelerating charge transfer and promoting space charge separation,as supported by the photo-electrochemical properties.To better understand the CO_(2)adsorption mechanism,density functional theory calculations were performed,which revealed that the Niov site has more negative adsorption energy compared with the Feov site.This study provides a simple strategy to establish efficient photocatalysts for CO_(2)reduction through the modification of organic ligands.

Deciphering the neuronal response to injury in the spinal cord is essential for exploring treatment strategies for spinal cord injury(SCI).However,this subject has been neglected in part because appropriate tools are lacking.Emerging in vivo imaging and labeling methods offer great potential for observing dynamic neural processes in the central nervous system in conditions of health and disease.This review first discusses in vivo imaging of the mouse spinal cord with a focus on the latest imaging techniques,and then analyzes the dynamic biological response of spinal cord sensory and motor neurons to SCI.We then summarize and compare the techniques behind these studies and clarify the advantages of in vivo imaging compared with traditional neuroscience examinations.Finally,we identify the challenges and possible solutions for spinal cord neuron imaging.

N-glycan biosignatures as a potential diagnostic biomarker for earlystage pancreatic cancer

BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)has a poor prognosis,with a 5-year survival rate of less than 10%,owing to its late-stage diagnosis.Early detection of pancreatic cancer(PC)can significantly increase survival rates.AIM To identify the serum biomarker signatures associated with early-stage PDAC by serum N-glycan analysis.METHODS An extensive patient cohort was used to determine a biomarker signature,in-cluding patients with PDAC that was well-defined at an early stage(stages I and II).The biomarker signature was derived from a case-control study using a case-cohort design consisting of 29 patients with stage I,22 with stage II,4 with stage III,16 with stage IV PDAC,and 88 controls.We used multiparametric analysis to identify early-stage PDAC N-glycan signatures and developed an N-glycan sig-nature-based diagnosis model called the“Glyco-model”.RESULTS The biomarker signature was created to discriminate samples derived from patients with PC from those of controls,with a receiver operating characteristic area under the curve of 0.86.In addition,the biomarker signature combined with cancer antigen 19-9 could discriminate patients with PDAC from controls,with a receiver operating characteristic area under the curve of 0.919.Glyco-model demonstrated favorable diagnostic performance in all stages of PC.The diagnostic sensitivity for stage I PDAC was 89.66%.Core Tip:This study employed a patient cohort to investigate the N-glycan signature of early-stage pancreatic cancer(PC).Serum N-glycans analysis was conducted to identify the serum biomarker signature associated with early-stage pancreatic ductal adenocarcinoma(PDAC),resulting in the identification of nine early-stage PDAC N-glycan signatures.Subsequently,utilizing these biosignatures,a diagnostic model named the“Glyco-model”was developed,demonstrating promising diagnostic performance across all stages of PC.The study revealed that the diagnostic sensitivity for stage I PDAC was determined to be 89.66%.Consequently,this diagnostic model exhibits potent

The development of new design strategies to create innovative structural materials,refine existing ones,and achieves compatible combinations of strength and plasticity remains a worldwide goal.Promising alloys,such as shape memory alloys(SMAs),bulk metallic glasses(BMGs),high entropy alloys(HEAs),and heterogeneous pure metals such as Cu,have excellent mechanical responses,but they still fall short of meeting all the requirements of structural materials due to specific flaws,such as lack of tensile de-formation for BMGs and low yielding strength for HEAs.To address these shortcomings,proposals such as integrating glassy matrices and crystallized alloys,such as HEAs/SMAs,have been suggested.However,these solutions have unresolved issues,such as the challenging control of B2 phase formation in BMG composites.Recently,glass-crystal(A/C)laminated alloys with alternating layers have been reported to exhibit improved mechanical properties and activated work-hardening behaviors,but they still face press-ing issues such as bonding interfaces and unknown deformation mechanisms.This review focuses on design routes such as the selection of alloy components and processing techniques,exploration of micro-structural evolution and deformation modes with an increase in strain,and future solutions to address pressing and unsolved issues.These prominent advantages include diversified deformation mechanisms,such as deformation twinning,martensitic phase transformation,and precipitation hardening,as well as tuned interactive reactions of shear bands(SBs)near the A/C interfaces.Thus,this review provides a promising pathway to design and develop structural materials in the materials field community.
