Erianin inhibits the proliferation of lung cancer cells by suppressing mTOR activation and disrupting pyrimidine metabolism

Objective: Erianin has potential anticancer activities, especially against lung cancer. The specific mechanisms underlying the anticancer effects, including the molecular targets and signaling pathways in lung cancer, remain poorly understood and necessitate further ***: Lung cancer cell viability was evaluated using the CCK-8 assay. Flow cytometry was used to examine the effects of erianin on apoptosis and cell cycle progression. m RNA sequencing and metabolomics analysis were utilized to explore erianin-induced biological changes. Potential targets were identified and validated through molecular docking and Western blot analysis. The roles of mammalian target of rapamycin(m TOR) and carbamoyl-phosphate synthetase/aspartate transcarbamylase/dihydroorotase(CAD) in erianin-induced growth inhibition were studied using gene overexpression/knockdown techniques with uridine and aspartate supplementation confirming pyrimidine metabolism involvement. Additionally, lung cancer-bearing nude mouse models were established to evaluate the anti-lung cancer effects of erianin in ***: Erianin significantly inhibits the proliferation of lung cancer cells, induces apoptosis, and causes G2/M phase cell cycle arrest. Integrative analysis of m RNA sequencing and metabolomics data demonstrated that erianin disrupts pyrimidine metabolism in lung cancer cells. Notably, uridine supplementation mitigated the inhibitory effects of erianin, establishing a connection between pyrimidine metabolism and anticancer activity. Network pharmacology analyses identified m TOR as a key target of erianin. Erianin inhibited m TOR phosphorylation, thereby blocking downstream effectors(S6K and CAD), which are essential regulators of pyrimidine ***: Erianin is a promising therapeutic candidate for lung cancer. Erianin likely inhibits lung cancer cell growth by disrupting pyrimidine metabolism by suppressing m TOR activation.

Multi-channel microelectrode arrays for detection of single-cell level neural information in the hippocampus CA1 under general anesthesia induced by low-dose isoflurane

Timely monitoring of anesthesia status during surgery is important to prevent an overdose of isoflurane anesthesia. Therefore, in-depth studies of the neural mechanisms of anesthetics are warranted. Hippocampal CA1 plays an important role during anesthesia. Currently, a high spatiotemporal resolution microdevice technology for the accurate detection of deep brain nuclei is lacking. In this research, four-shank 32-channel implantable microelectrode arrays (MEAs) were developed for the real-time recording of single-cell level neural information in rat hippocampal CA1. Platinum nanoparticles were modified onto the microelectrodes to substantially enhance the electrical properties of the microelectrode arrays. The modified MEAs exhibited low impedance (11.5 ± 1 kΩ) and small phase delay (-18.5° ± 2.54°), which enabled the MEAs to record single-cell level neural information with a high signal-to-noise ratio. The MEAs were implanted into the CA1 nuclei of the anesthetized rats, and the electrophysiological signals were recorded under different degrees of anesthesia mediated by low-dose concentrations of isoflurane. The recorded signals were analyzed in depth. Isoflurane caused an inhibition of spike firing rate in hippocampal CA1 neurons, while inducing low-frequency oscillations in CA1, thus enhancing the low-frequency power of local field potentials. In this manner, the spike firing rate and the power of local field potentials in CA1 could characterize the degree of isoflurane anesthesia. The present study provides a technical tool to study the neural mechanisms of isoflurane anesthesia and a research method for monitoring the depth of isoflurane anesthesia in clinical practice. © 2023

The reform stems from honesty and determination. Since 2005, organ donation and transplantation in China has undergone thorough reform, which complies with legislation requirements and ethical principles established by the World Health Organization(WHO). Reform in China has demonstrated the unwavering confidence and utmost determination of the Chinese government and the Chinese transplantation community. The year 2015 marked a historic turning point when voluntary donations from Chinese citizens became the sole legitimate source for organ transplantation. Since 2015, China has gradually established and refined the “Chinese Mode” and “China System” for organ donation and transplantation, fulfilling its political pledge of reform, and has garnered international recognition, and fostered a social culture which promotes organ donation. This article reviewed the history of reform on organ donation and transplantation in China, presented a new pattern of establishment of organ donation system in the new era of the country, and the direction of advances in the future.

Sympathetic Solar Eruption on 2024 February 9

In this paper,we perform a follow-up investigation of the solar eruption originating from active region 13575 on2024 February *** primary eruption of a hot channel generates an X3.4 class flare,a full-halo coronal mass ejection （CME）,and an extreme-ultraviolet （EUV） *** between the wave and a quiescent prominence（QP） leads to a large-amplitude,transverse oscillation of *** the transverse oscillation,QP loses equilibrium and rises *** ascending motion of the prominence is coherently detected and tracked up to～1.68 R⊙by the Solar Ultra Violet Imager onboard the GOES-16 spacecraft and up to～2.2 R⊙by the Solar Corona Imager（SCI＿UV） of the Lyα Solar Telescope onboard the ASO-S *** velocity increases linearly from 12.3 to68.5 km s-1at 18:30 *** sympathetic eruption of QP drives the second CME with a typical three-part *** bright core comes from the eruptive prominence,which could be further observed up to～3.3 R⊙by the Large Angle Spectroscopic Coronagraph onboard the Solar and Heliospheric Observatory *** leading edge of the second CME accelerates continuously from～120 to～277 km *** EUV wave plays an important role in linking the primary eruption with the sympathetic eruption.
