Voluntary running delays primary degeneration in rat retinas after partial optic nerve transection

Running is believed to be beneficial for human health. Many studies have focused on the neuroprotective effects of voluntary running on animal models. There were both primary and secondary degeneration in neurodegenerative diseases, including glaucoma. However, whether running can delay primary or secondary degeneration or both of them was not clear. Partial optic nerve transection model is a valuable glaucoma model for studying both primary and secondary degeneration because it can separate primary(mainly in the superior retina) from secondary(mainly in the inferior retina) degeneration. Therefore, we compared the survival of retinal ganglion cells between Sprague-Dawley rat runners and non-runners both in the superior and inferior retinas. Excitotoxicity, oxidative stress, and apoptosis are involved in the degeneration of retinal ganglion cells in glaucoma. So we also used western immunoblotting to compare the expression of some proteins involved in apoptosis(phospho-c-Jun N-terminal kinases, p-JNKs), oxidative stress(manganese superoxide dismutase, MnSOD) and excitotoxicity(glutamine synthetase) between runners and non-runners after partial optic nerve transection. Results showed that voluntary running delayed the death of retinal ganglion cells vulnerable to primary degeneration but not those to secondary degeneration. In addition, voluntary running decreased the expression of glutamine synthetase, but not the expression of p-JNKs and MnSOD in the superior retina after partial optic nerve transection. These results illustrated that primary degeneration of retinal ganglion cells might be mainly related with excitotoxicity rather than oxidative stress; and the voluntary running could down-regulate excitotoxicity to delay the primary degeneration of retinal ganglion cells after partial optic nerve transection.

System Size Resonance Associated with Canard Phenomenon in a Biological Cell System

The influence of internal noise on the calcium oscillations is studied. It is found that stochastic calcium oscillations occur when the internal noise is considered, while the corresponding deterministic dynamics only yields a steady state. Also,. the performance of such oscillations shows two maxima with the variation of the system size, indicating the occurrence of system size resonance. This behavior is found to be intimately connected with the canard phenomenon. Interestingly, it is also found that one of the optimal system sizes matches well with the real cell size, and such a match is robust to the variation of the control parameters.

Topological analysis of plasma flow control on corner separation in a highly loaded compressor cascade

In this paper, flow behavior and topology structure in a highly loaded compressor cascade with and without plasma aerodynamic actuation （PAA） are investigated. Streamline pattern, total pressure loss coefficient, outlet flow angle and topological analysis are considered to study the effect and mechanism of the plasma flow control on corner separation. Results presented include the boundary layer flow behavior, effects of three types of PAA on separated flows and performance parameters, topology structures and sequences of singular points with and without PAA. Two separation lines, reversed flow and backflow exist on the suction surface. The cross flow on the endwall is an important element for the comer separation. PAA can reduce the undertuming and overturning as well as the total pressure loss, leading to an overall increase of flow turning and enhancement of aerodynamic performance. PAA can change the topology structure, sequences of singular points and their corresponding separation lines. Types II and III PAA are much more efficient in controlling comer separation and enhancing aerodynamic performances than type I.

Grain structure and tensile property of Al-li alloy sheet caused by different cold rolling reduction

The effect of cold rolling reduction(50%-90%)on the grain structures of solutionized 1445 Al-li alloy sheet at525-575 ℃ was investigated through electron backscatter diffraction(EBSD).Although the solutionization temperature is elevated to 575 ℃,the sheet is not completely *** main recrystallization model is subgrain coalescence and growth,and the non-recrystallization is due to the formed nano-sized Al3(Sc,Zr)dispersoids,which pin the grain boundaries,subgrain boundaries and *** increasing the cold rolling reduction,the fraction and size of the recrystallized grains in the sheet solutionized at525 ℃ are decreased,but the fraction of the subgrains is increased,leading to a decrease in the fraction of the deformed ***,the number fraction of high-angle boundaries(HABs)is *** to the decreased fraction of the deformed structures and increased fraction of the HABs,the T8-aged 1445 Al-li alloy sheet displays a decrease trend in the strength and heterogeneity with increasing the cold rolling *** higher solutionization temperature of 575 ℃,the fraction of the recrystallized grains and their size are obviously increased.

Selection of Parents for Breeding Edible Varieties of Sweetpotato with High Carotene Content

Sweetpotato varieties with high carotene content have high value for human health. This work aims to improve the breeding efficiency of special varieties with high carotene content by combining molecular markers and agronomic traits to evaluate and screen the parents. The cluster analysis was carried out to identify and screen promising parents by utilizing phenotypic results of the agronomic and characteristic traits, and RAPD and ISSR markers of 15 parents and their offspring by group crossing. Among different parental materials, greater variations were observed in two important traits, the carotene content and the storage root yield. Negative correlation was found between the carotene content and both fresh and dried root yields. The most significant positive correlation was between the carotene content of parents and that of their offspring, with the coefficient of correlation value of 0.7932＊＊. The relationship based on the agronomic characters of the trial materials was not in agreement with the known genealogy, while that based on the molecular marker data showed better result. Each primer amplified 13.8 bands oflSSR markers on average, in which the rate of polymorphic loci was 89.6%. 9.4 bands of RAPD markers were arnplified per primer, in which the rate of polymorphic bands was 74.46%. Large genetic variation of carotene content was found among the parents. For analyzing the genetic diversity of sweetpotato, the molecular marker methods were better than morphological traits; ISSR markers were more efficient than RAPD markers. The combination of molecular markers and agronomic trait analysis may provide valuable theoretic basis for selection of parents to breed new varieties with high carotene content.

Previous studies have shown that berberine has neuroprotective effects against Alzheimer’s disease,including antagonizing tau phosphorylation,and inhibiting acetylcholinesterase activity and neural cell ***,its low bioavailability and adverse reactions with conventional administration limit its clinical *** this study,we prepared berberine nanoliposomes using liposomes characterized by low toxicity,high entrapment efficiency,and biodegradability,and modified them with ***-modified berberine nanoliposomes had uniform particle size and high entrapment *** used the lactoferrin-modified berberine nanoliposomes to treat a mouse model of Alzheimer’s disease established by injection of amyloid-beta 1-42 into the lateral ***-modified berberine nanoliposomes inhibited acetylcholinesterase activity and apoptosis in the hippocampus,reduced tau over-phosphorylation in the cerebral cortex,and improved mouse *** findings suggest that modification with lactoferrin can enhance the neuroprotective effects of berberine nanoliposomes in Alzheimer’s disease.

Tomato Fruit Development and Ripening Are Altered by the Silencing of LeEIN2 Gene

Loss-of-function ethylene insensitive 2 （EIN2） mutations showed ethylene insensitivity in Arabidopsis, which indicated an essential role of EIN2 in ethylene signaling. However, the function of EIN2 in fruit ripening has not been investigated. To gain a better understanding of EIN2, the temporal regulation of LeEIN2 expres- sion during tomato fruit development was analyzed. The expression of LeEIN2 was constant at different stages of fruit development, and was not regulated by ethylene. Moreover, LeEIN2-silenced tomato fruits were developed using a virus-induced gene silencing fruit system to study the role of LeEIN2 in tomato fruit ripening. Silenced fruits had a delay in fruit development and ripening, related to greatly descended expression of ethylene-related and ripening-related genes in comparison with those of control fruits. These results suggested LeEIN2 positively mediated ethylene signals during tomato development. In addition, there were fewer seeds and Iocules in the silenced fruit than those in the control fruit, like the phenotype of parthenocarpic tomato fruit. The content of auxin and the expression of auxin-regulated gene were declined in silenced fruit, which indicated that EIN2 might be important for crosstalk between ethylene and auxin hormones.

A new series of 12-benzyl matrinic amide/ethanamide derivatives were synthesized from matrinine（1）and evaluated for their anti-HCV activity,taking compound 2 as the *** revealed that the introduction of a suitable substituent at the N’-end of matrinic amide might greatly enhance the *** them,matrinic acid 17 and N’-substituted matrinic amides 18a-d exhibited promising potency with low micromolar EC50 values ranging from 1.03μmol/L to 7.54 μmol/L,and better therapeutic window with SI from 66 to ***,compound 17 displayed an excellent PK and safety profile in vivo,demonstrating good drug-like ***,it has been selected for further investigation,with an advantage of decreased chances of inducing drug-resistance mutations.

Alpinia oxyphylla,a traditional herb,is widely used for its neuroprotective,antioxidant and memory-improving ***,the neuroprotective mechanisms of action of its active ingredients are *** this study,we investigated the neuroprotective effects of various organic extracts of Alpinia oxyphylla on PC12 cells exposed to hydrogen peroxide-induced oxidative injury in *** oxyphylla was extracted three times with 95%ethanol(representing extracts 1–3).The third 95%ethanol extract was dried and resuspended in water,and then extracted successively with petroleum ether,ethyl acetate and n-butanol(representing extracts 4–6).The cell counting kit-8 assay and microscopy were used to evaluate cell viability and observe the morphology of PC12 *** protective effect of the three ethanol extracts(at tested concentrations of 50,100 and 200μg/mL)against cytotoxicity to PC12 cells increased in a concentration-dependent *** ethyl acetate,petroleum ether and n-butanol extracts(each tested at 100,150 and 200μg/mL)had neuroprotective effects as *** optimum effective concentration ranged from 50–200μg/mL,and the protective effect of the ethyl acetate extract was comparatively *** results demonstrate that organic extracts of Alpinia oxyphylla protect PC12 cells against apoptosis induced by hydrogen *** findings should help identify the bioactive neuroprotective components in Alpinia oxyphylla.

Determination of Dopamine in the Presence of Ascorbic Acid on the IrO2-Ta2O5 Type DSA Electrode

IrO2/Ta2O5 (70%: 30%, molar ratio) electrodes (ITEs) were prepared by the thermal decomposition method. Typical “mud-cracked” surface and surface enrichment of Ta were observed. The determination of dopamine (DA) was carried out in the presence of ascorbic acid (AA). The detection limit of DA was 5×10-5 mol/L without the interference of AA.
