Altered metabolism has long been recognized as a central hallmark of cancer;however,in the fluorescence imaging field,few studies have been conducted to label tumors by exploiting metabolic differences between cancer cells and normal cells.In this work,we successfully developed a metabolic probe MB-C for specific imaging of glutathione(GSH)dynamic metabolic pathways.GSH was endogenously metabolized to produce SO_(2)via Na_(2)S_(2)O_(3) and thiosulfate sulfurtransferase,equilibrating with sulfites/bisulfites.MB-C was allowed to be activated by GSH along with multi-fluorescence emission increased in red and green channels and further sequence-response metabolites(SO_(2))of GSH in significant fluorescence ratio change of red and green channels.Furthermore,such evident fluorescence ratio changes could be used to distinguish cancer cells from normal cells and identify tumor and normal tissues.Therefore,GSH metabolic imaging was successfully applied to accurately label tumors,which provides a new idea and practical case for the precise visualization of malignant tumors.

Objective:The study aims to present a novel classification of tracheal defects and the corresponding reconstruction strategies.Methods:The retrospective study was designed to analyze patients with diagnosed primary or secondary tracheal tumors from 1991 to 2020.Surgical techniques,complications and prognosis were reviewed.Airway status and patient outcomes were the principal follow-up measures.Tracheal defects were classified into two plane sizes(vertical(V)and horizontal(H)planes).Vertical defects were further categorized into three groups based on their tracheal ring numbers(V 1,≤5 rings;V 2,6-10 rings;and V 3,>10 rings).Tracheal defects with horizontal plane size H 1 and H 2 represent defects less and more than one-half the circumference of trachea.Thus,suitable reconstruction strategies were planned primarily based on"V"and"H"classifications.The reconstruction strategies performed were sleeve resection followed by an end-to-end anastomosis,window resection with sternocleidomastoid myoperiosteal flap reconstruction,defects conversion with rotation anastomosis,and modified tracheostomy with secondary flap reconstruction.Results:A total of 106 patients diagnosed with tracheal defects were enrolled in the study,of whom 59 patients underwent sleeve resection followed by end-to-end anastomosis;40 patients received window resection alongside sternocleidomastoid(SCM)myoperiosteal flap reconstruction;five patients received converting defects with rotation anastomosis and two patients underwent modified tracheostomy with secondary stage flap reconstruction.Lumen stenosis occurred in three V 2H 1 defect cases and were treated by a second reconstruction surgery.Iatrogenic unilateral recurrent laryngeal nerve paralysis occurred in two patients with the V 3H 2 defect type,who were treated by temporary tracheotomy and partial vocal cord resection and extubated successfully during follow-up.All 106 patients achieved airway patency with adequate laryngeal function at the end of follow-up.No anastomotic dehiscence or bleeding

Mo–57Si–10B(at.%) coating was prepared by the spark plasma sintering(SPS) technique on a 2D woven Cf/Si C composite, and the oxidation behavior of the bare and the coated Cf/SiC samples at 1300°C were investigated.The coating was composed of a fine MoSi/Mo B/SiO/MoBout layer and the inner SiOinterface layer. The mass gain percentage was approximately 0.25% after oxidation at 1300°C for 100 h. The coated Cf/Si C substrate was effectively protected, owing to the rapid formation of the dense and protective borosilicate layer on the coating, the excellent microstructure stability of the coating and the inner SiOinterface layer.

Hydrogen peroxide(H2 O2), as important products of oxygen metabolism, plays an important role in many biological processes, such as immune responses and cellular signal transduction. However, abnormal production of H2 O2 can damage cellular biomolecules, which was closely associated with many diseases.Thus, it is urgent to monitor the level change of H2 O2 in living cells, particularly at subcellular levels.Toward this end, a wide variety of H2 O2 fluorescent probes have been designed, developed and applied for imaging of H2 O2 in subcellular levels. In this review, we highlight the representative cases of H2 O2 fluorescent probes with mitochondria, nuclei and lysosomes-targetable ability. The review contains organelle target strategies, structures, fluorescence behavior and biological applications of these probes.

Transformer architectures have facilitated the development of large-scale and general-purpose sequence models for prediction tasks in natural language processing and computer vision,e.g.,GPT-3 and Swin Transformer.Although originally designed for prediction problems,it is natural to inquire about their suitability for sequential decision-making and reinforcement learning problems,which are typically beset by long-standing issues involving sample efficiency,credit assignment,and partial observability.In recent years,sequence models,especially the Transformer,have attracted increasing interest in the RL communities,spawning numerous approaches with notable effectiveness and generalizability.This survey presents a comprehensive overview of recent works aimed at solving sequential decision-making tasks with sequence models such as the Transformer,by discussing the connection between sequential decision-making and sequence modeling,and categorizing them based on the way they utilize the Transformer.Moreover,this paper puts forth various potential avenues for future research intending to improve the effectiveness of large sequence models for sequential decision-making,encompassing theoretical foundations,network architectures,algorithms,and efficient training systems.

Based on a large sample of massive(M* 1010 M⊙) compact galaxies at 1.0 < z < 3.0 in five 3 D-HST/CANDELS fields, we quantify the fractional abundance and comoving number density of massive compact galaxies as a function of redshift. The samples of compact quiescent galaxies(cQGs) and compact star-forming galaxies(cSFGs) are constructed by various selection criteria of compact galaxies in the literature, and the effect of compactness definition on abundance estimate has proven to be remarkable,particularly for the cQGs and cSFGs at high redshifts. Regardless of the compactness criteria adopted,their overall redshift evolutions of fractional abundance and number density are found to be rather similar.Large samples of the cQGs exhibit a sustained increase in number density from z ~ 3 to 2 and a plateau at 1 < z < 2. For massive cSFGs, a plateau in the number density at 2 < z < 3 can be found, as well as a continuous drop from z ~ 2 to 1. The evolutionary trends of the cQG and cSFG abundances support the scenario that the cSFGs at z≥ 2 may have been rapidly quenched into quiescent phase via violent dissipational processes, such as major merger and disk instabilities. The rarity of the cSFGs at lower redshifts(z < 1) can be interpreted by the decrease of gas reservoirs in dark matter halos and the consequent low efficiency of gas-rich dissipation.

Objective Liraglutide is a commonly used hypoglycemic agent in clinical practice,and has been demonstrated to have protective effects against the development of cardiovascular disease.However,its potential role in myocardial fibrosis remains unexplored.The present study aims to assess the impact of liraglutide on the activation of cardiac fibroblasts.Methods Primary rat adult fibroblasts were isolated,cultured,and randomly allocated into 4 groups:control group,transforming growth factor beta1(TGFβ1)stimulation group,liraglutide group,and TGFβ1+liraglutide group.Fibroblast activation was induced by TGFβ1.Cell proliferation activity was assessed using the CKK-8 kit,and cellular activity was determined using the MTT kit.Reverse transcrition-quantitative polymerase chain reaction(RT-qPCR)was utilized to quantify the level of collagen transcription,immunofluorescence staining was performed to detect the expression level of type III collagen andα-smooth muscle protein(α-SMA),and immunoblotting was conducted to monitor alterations in signal pathways.Results The addition of 10,25,50 and 100 nmol/L of liraglutide did not induce any significant impact on the viability of fibroblasts(P>0.05).The rate of cellular proliferation was significantly higher in the TGFβl stimulation group than in the control group.However,the treatment with 50 and 100 nmol/L of liraglutide resulted in the reduction of TGFβl-induced cell proliferation(P<0.05).The RT-qPCR results revealed that the transcription levels of type I collagen,type III collagen,andα-SMA were significantly upregulated in the TGFβl stimulation group,when compared to the control group(P<0.05).However,the expression levels of these aforementioned factors significantly decreased in the TGFβl+liraglutide group(P<0.05).The immunofluorescence staining results revealed a significant increase in the expression levels of type III collagen andα-SMA in the TGFβl stimulation group,when compared to the control group(P<0.05).However,these expression levels significantly decreased in the TG

In the present research,the dynamic recrystallization(DRX) behavior of a newly-developed Mg-Al-Zn-RE alloy with abundant secondphase particles during hot extrusion is investigated by coupling finite element(FE) and cellular automaton(CA) models.A two-dimensional CA model is developed to quantitatively and topologically evaluate the DRX process during deformation with constant forming conditions.Considering the fact that second-phase particles with various sizes extensively exist in the studied Mg-Al-Zn-RE magnesium alloy,models of DRX nucleation and grain growth velocity are modified.The coefficients of the modified CA model are calibrated by isothermal compression experiments of the magnesium alloy.Subsequently,the CA model is coupled with FE analysis to investigate the DRX behavior during the hot extrusions of the Mg-Al-Zn-RE alloy.The DRX behavior of the magnesium alloy at different stages and positions of extruded plates is simulated by the established model.Finer grains near the edge than in the inner of the plates result from higher strain and strain rate.The influence of extrusion conditions on microstructural evolution is explored.Under the employed forming conditions,average grain size decreases 28-62 times from as-cast and solution-treated to as-extruded state due to grain refinement by DRX.With increasing initial billet temperature or extrusion speed,average grain size increases.The finest grains are obtained at the initial billet temperature of 623 K and the extrusion speed of 7.83 mm/s.Low initial billet temperature or high extrusion speed benefits homogeneous grain distribution.The simulated results are in good agreement with experimental ones.
