ELABELA-derived peptide ELA13 attenuates kidney fibrosis by inhibiting the Smad and ERK signaling pathways

Kidney fibrosis is an inevitable result of various chronic kidney diseases(CKDs)and significantly contributes to end-stage renal failure.Currently,there is no specific treatment available for renal fibrosis.ELA13(amino acid sequence:RRCMPLHSRVPFP)is a conserved region of ELABELA in all vertebrates;however,its biological activity has been very little studied.In the present study,we evaluated the therapeutic effect of ELA13 on transforming growth factor-β1(TGF-β1)-treated NRK-52E cells and unilateral ureteral occlusion(UUO)mice.Our results demonstrated that ELA13 could improve renal function by reducing creatinine and urea nitrogen content in serum,and reduce the expression of fibrosis biomarkers confirmed by Masson staining,immunohistochemistry,real-time polymerase chain reaction(RT-PCR),and western blot.Inflammation biomarkers were increased after UUO and decreased by administration of ELA13.Furthermore,we found that the levels of essential molecules in the mothers against decapentaplegic(Smad)and extracellular signal-regulated kinase(ERK)pathways were reduced by ELA13 treatment in vivo and in vitro.In conclusion,ELA13 protected against kidney fibrosis through inhibiting the Smad and ERK signaling pathways and could thus be a promising candidate for anti-renal fibrosis treatment.

Fabrication and luminescence of highly transparent C12A7:Tb^(3+)glass-ceramics via in-situ crystallization from aerodynamic levitation processed glasses

Rare earth doped 12CaO·7Al_(2)O_(3)(C12A7)glass-ceramics are one of attractive photonic materials due to their high transparency,high luminescent efficiency and eco-friendly nature.A series of C12A7 glasses with different Tb^(3+)doping concentrations was prepared by aerodynamic levitation method.The C12A7:Tb glass-ceramics were obtained via in-situ growth from the glasses with the identical chemical composition at 800℃.The microstructure observation demonstrates that the elemental distributions of Ca and Al are homogeneous in the glass-ceramics and the grain size of C12A7 is 4-70 nm.The in-line transmittance of glass-ceramics reaches as high as 82%at 1500 nm.The emission spectra of the C12A7:Tb glass-ceramics present characteristic Tb^(3+)emission peaks at 486 nm(^(5)D_(4)→^(7)F_6),541 nm(^(5)D_(4)→^(7)F_(5)),585 nm(^(5)D_(4)→^(7)F_(4))and 621 nm(^(5)D_(4)→^(7)F_(3)).The photolumine scence lifetime of C12A7:0.3%Tb glass-ceramic is 1.9 ms.Compared with the emission intensity of the glasses,that of the glass-ceramics increases due to the reducing in concentration of oxygen vacancy associated with network forming(NWF)cations.The highly transparent C12A7:Tb glass-ceramics may be a promising candidate for green light source in solid state lighting.

An attention-embedded GAN for SVBRDF recovery from a single image

Learning-based approaches have made substantial progress in capturing spatially-varying bidirectional reflectance distribution functions(SVBRDFs)from a single image with unknown lighting and geometry.However,most existing networks only consider per-pixel losses which limit their capability to recover local features such as smooth glossy regions.A few generative adversarial networks use multiple discriminators for different parameter maps,increasing network complexity.We present a novel end-to-end generative adversarial network(GAN)to recover appearance from a single picture of a nearly-flat surface lit by flash.We use a single unified adversarial framework for each parameter map.An attention module guides the network to focus on details of the maps.Furthermore,the SVBRDF map loss is combined to prevent paying excess attention to specular highlights.We demonstrate and evaluate our method on both public datasets and real data.Quantitative analysis and visual comparisons indicate that our method achieves better results than the state-of-the-art in most cases.

CMIP5 Simulated Change in the Intensity of the Hadley and Walker Circulations from the Perspective of Velocity Potential

Based on the simulations of 31 global models in CMIP5, the performance of the models in simulating the Hadley and Walker circulations is evaluated. In addition, their change in intensity by the end of the 21st century （2080-2099） under the RCP4.5 and RCP8.5 scenarios, relative to 1986-2005, is analyzed from the perspective of 200 hPa velocity potential. Validation shows good performance of the individual CMIP5 models and the multi-model ensemble mean （MME） in re- producing the meridional （zonal） structure and magnitude of Hadley （Walker） circulation. The MME can also capture the observed strengthening tendency of the winter Hadley circulation and weakening tendency of the Walker circulation. Such secular trends can be simulated by 39% and 74% of the models, respectively. The MME projection indicates that the winter Hadley circulation and the Walker circulation will weaken under both scenarios by the end of the 21st century. The weak- ening amplitude is larger under RCP8.5 than RCP4.5, due to stronger external forcing. The majority of the CMIP5 models show the same projection as the MME. However, for the summer Hadley circulation, the MME shows little change under RCP4.5 and large intermodel spread is apparent. Around half of the models project an increase, and the other half project a decrease. Under the RCP8.5 scenario, the MME and 65% of the models project a weakening of the summer southern Hadley circulation.

Projection of the Future Changes in Tropical Cyclone Activity Affecting East Asia over the Western North Pacific Based on Multi-RegCM4 Simulations

Future changes in tropical cyclone(TC)activity over the western North Pacific(WNP)under the representative concentration pathway RCP4.5 are investigated based on a set of 21 st century climate change simulations over East Asia with the regional climate model RegCM4 driven by five global models.The RegCM4 reproduces the major features of the observed TC activity over the region in the present-day period of 1986-2005,although with the underestimation of the number of TC genesis and intensity.A low number of TCs making landfall over China is also simulated.By the end of the 21st century(2079-98),the annual mean frequency of TC genesis and occurrence is projected to increase over the WNP by16%and 10%,respectively.The increase in frequency of TC occurrence is in good agreement among the simulations,with the largest increase over the ocean surrounding Taiwan Island and to the south of Japan.The TCs tend to be stronger in the future compared to the present-day period of 1986-2005,with a large increase in the frequency of strong TCs.In addition,more TCs landings are projected over most of the China coast,with an increase of~18%over the whole Chinese territory.

Inhibition of Eukaryotic Initiating Factor eIF4E Overcomes Abemaciclib Resistance in Gastric Cancer

Objective Aberrant activating mutations in cyclin-dependent kinases 4 and 6(CDK4/6)are common in various cancers,including gastroesophageal malignancies.Although CDK4/6 inhibitors,such as abemaciclib and palbociclib,have been approved for breast cancer treatment,their effectiveness as a monotherapy remains limited for gastroesophageal tumors.The present study explored the underlying mechanism of abemaciclib resistance.Methods Abemaciclib-resistant gastric cancer cell lines were generated,and the phospho-eukaryotic translation initiation factor 4E(p-eIF4E)and eIF4E expression was compared between resistant and parental cell lines.In order to analyze the role of eIF4E in cell resistance,siRNA knockdown was employed.The effectiveness of ribavirin alone and its combination with abemaciclib was evaluated in the gastric cancer xenograft mouse model.Results The upregulation of eIF4E was a common feature in gastric cancer cells exposed to prolonged abemaciclib treatment.Gastric cancer cells with increased eIF4E levels exhibited a better response to eIF4E inhibition,especially those that were resistant to abemaciclib.Ribavirin,which is an approved anti-viral drug,significantly improved the efficacy of abemaciclib,both in vitro and in vivo,by inhibiting eIF4E.Importantly,ribavirin effectively suppressed the abemaciclib-resistant gastric cancer growth in mice without causing toxicity.Conclusion These findings suggest that targeting eIF4E can enhance the abemaciclib treatment for gastric cancer,proposing the potential combination therapy of CDK4/6 inhibitors with ribavirin for advanced gastric cancer.

Evaluating refractive index and birefringence of nonlinear optical crystals:Classical methods and new developments

Nonlinear optical(NLO)materials play an increasingly important role in laser technology.Birefringence is one of the most important parameters for NLO materials to realize angle phase-matching conditions.In comparison with other desirable optical properties,the availability of birefringence and refractive index dispersion is especially problematic owing to the strict requirements for single crystals.In this review,we described how to obtain the refractive index and birefringence of NLO materials from crystals sub-millimeters to centimeters in size.Espe-cially,recently developed methods including the minimum deflection angle method,auto-collimation method,prism coupling method,oil immersion technique,interference color method,and theoretical calculation(DFT)for rapid assessment of birefringence are summarized,the contents of which are mainly focused on the principles and typical applications,together with the advantages and drawbacks.In addition,representative examples of bire-fringent measurements were presented.The purpose of this work is to provide a useful perspective on the characterization of birefringence for NLO materials.It is hoped that this review can give a clear description of the birefringence measurements and accelerate the discovery of new NLO crystals.

Length of stay and inpatient charges of total knee arthroplasty in China:analysis of a national database

Background:There are limited data on the resource utilization of total knee arthroplasty(TKA)in China.This study aimed to examine the length of stay(LOS)and inpatient charges of TKA in China,and to investigate their determinants.Methods:We included patients undergoing primary TKA in the Hospital Quality Monitoring System in China between 2013 and 2019.LOS and inpatient charges were obtained,and their associated factors were further assessed using multivariable linear regression.Results:A total of 184,363 TKAs were included.The LOS decreased from 10.8 days in 2013 to 9.3 days in 2019.The admission-to-surgery interval decreased from 4.6 to 4.2 days.The mean inpatient charges were 61,208.3 Chinese Yuan.Inpatient charges reached a peak in 2016,after which a gradual decrease was observed.Implant and material charges accounted for a dominating percentage,but they exhibited a downward trend,whereas labor-related charges gradually increased.Single marital status,non-osteoarthritis indication,and comorbidity were associated with longer LOS and higher inpatient charges.Female sex and younger age were associated with higher inpatient charges.There were apparent varieties of LOS and inpatient charges among provincial or non-provincial hospitals,hospitals with various TKA volume,or in different geographic regions.Conclusions:The LOS following TKA in China appeared to be long,but it was shortened during the time period of 2013 to 2019.The inpatient charges dominated by implant and material charges exhibited a downward trend.However,there were apparent sociodemographic and hospital-related discrepancies of resource utilization.The observed statistics can lead to more efficient resource utilization of TKA in China.

为实现高速动车组车轮踏面擦伤长度定量评估,提出了一种自适应连续小波模极大值算法,应用到基于轴箱振动加速度信号的车轮擦伤状态识别与损伤评估。利用连续小波变换系数模极大值对自适应滤波后的轴箱振动加速度信号突变点进行定位,从而建立擦伤车轮脱离钢轨腾空运行时间与擦伤长度及其影响参数的关系模型。仿真实例结果表明,该方法可在车辆高速运行时实现车轮擦伤程度的定量识别。与其它文献识别结果对比分析表明,该方法原理简单、计算结果精度高。

IT Investment and Innovation Performance of Industrial Enterprises:Empirical Evidence from Chinese Listed Companies

In this paper,a theoretical frame on how the IT investment influences the innovation performance of enterprise is set up,and the data of Chinese industrial listed companies is used to conduct an empirical test.The study shows that IT investment can improve the innovation performance of enterprises on the whole,but there is heterogeneity among different types of enterprises.Specifically,IT investment of low-innovation strategy-oriented enterprises has a stronger effect on innovation performance than high-innovation strategy-oriented enterprises;IT investment of large enterprises has a more significant effect on innovation performance;and IT investment of state-owned enterprises has a stronger effect on innovation performance than that of non-state-owned enterprises.These conclusions not only provide microscopic evidence for the promotion of innovation performance by IT investment of industrial enterprises,but also have important policy implications for the integration of informatization and industrialization and the construction of a digital China.
