Lipid nanoparticles(LNPs) have gained clinical approval as carriers for both siRNA and mRNA. Among the crucial components of LNPs, ionizable lipids play a pivotal role in determining the efficiency of RNA delivery. In this study, we synthesized a series of ionizable lipids, denoted as HTO, with a higher count of hydroxyl groups compared to SM-102. Remarkably, LNPs based on HTO12.lipid demonstrated comparable mRNA delivery efficiency and biosafety to those based on SM-102. However, the former reduced the ratio of ionizable lipid/total lipids to mRNA in LNPs by 2.5 times compared to SM-102. The HTO12.LNP efficiently encapsulated adenine base editor mRNA and sgRNA targeting Pcsk9, leading to substantial gene editing within the liver of mice and effective reduction of the target protein. Our study underscores that ionizable lipids with multiple hydroxyl groups may facilitate an improved lipid-to-mRNA ratio to minimize the dosage of ionizable lipids for in vivo delivery.

Centrifuge modeling of unreinforced and multi-row stabilizing piles reinforced landslides subjected to reservoir water level fluctuation

With the construction of the Three Gorges Reservoir dam,frequent reservoir landslide events have been *** recent years,multi-row stabilizing piles(MRSPs)have been used to stabilize massive reservoir landslides in *** this study,two centrifuge model tests were carried out to study the unreinforced and MRSP-reinforced slopes subjected to reservoir water level(RWL)operation,using the Taping landslide as a *** results indicate that the RWL rising can provide lateral support within the submerged zone and then produce the inward seepage force,eventually strengthening the slope ***,a rapid RWL drawdown may induce outward seepage forces and a sudden debuttressing effect,consequently reducing the effective soil normal stress and triggering partial pre-failure within the RWL fluctuation ***,partial deformation and subsequent soil structure damage generate excess pore water pressures,ultimately leading to the overall failure of the reservoir *** study also reveals that a rapid increase in the downslope driving force due to RWL drawdown significantly intensifies the lateral earth pressures exerted on the ***,the MRSPs possess a considerable reinforcement effect on the reservoir landslide,transforming the overall failure into a partial deformation and failure situated above and in front of the *** mechanical transfer behavior observed in the MRSPs demonstrates a progressive alteration in relation to RWL *** the RWL rises,the mechanical states among MRSPs exhibit a growing *** shear force transfer factor(*** ratio of shear forces on pile of the n th row to that of the first row)increases significantly with the RWL *** indicates that the mechanical states among MRSPs tend toward an enhanced *** insights gained from this study contribute to a more comprehensive understanding of the failure mechanisms of reservoir landslides and the mechanical behavior of M

Seismic prediction of cracks is of great significance in many disciplines,for which the rock physics model is ***,up to now,multitudinous analytical models focus primarily on the cracked rock with the isotropic background,while the explicit model for the cracked rock with the anisotropic background is rarely investigated in spite of such case being often encountered in the ***,we first studied dependences of the crack opening displacement tensors on the crack dip angle in the coordinate systems formed by symmetry planes of the crack and the background anisotropy,respectively,by forty groups of numerical *** on the conclusion from the experiments,the analytical solution was derived for the effective elastic properties of the rock with the inclined penny-shaped cracks in the transversely isotropic ***,we comprehensively analyzed,according to the developed model,effects of the crack dip angle,background anisotropy,filling fluid and crack density on the effective elastic properties of the cracked *** analysis results indicate that the dip angle and background anisotropy can significantly either enhance or weaken the anisotropy degrees of the P-and SH-wave velocities,whereas they have relatively small effects on the SV-wave velocity ***,the filling fluid can increase the stiffness coefficients related to the compressional modulus by reducing crack compliance parameters,while its effects on shear coefficients depend on the crack dip *** increasing crack density reduces velocities of the dry rock,and decreasing rates of the velocities are affected by the crack dip *** comparing with exact numerical results and experimental data,it was demonstrated that the proposed model can achieve high-precision estimations of stiffness ***,the assumption of the weakly anisotropic background results in the consistency between the proposed model and Hudson's published theory for the or
