Effects of different calcium salts on the physicochemical properties of sliver carp myosin

To investigate the effects of different calcium salts on the physicochemical properties of myosin,the intermolecular interactions,particle size,surface hydrophobicity,reactive sulfhydryl group content and rheological properties of sliver carp myosin with the addition of four different kinds of calcium salts (CaCl_(2),calcium citrate (CC),nano-hydroxyapatite (HAP),tricalcium phosphate (TCP)) during gel processing were measured.The results showed that the addition of CaCl_(2) promoted the unwinding of α-helix,the increase in hydrophobic interactions,the formation of disulfide bonds of myosin,and increased the viscoelasticity of myosin.While,the addition of calcium citrate (CC) enhanced the hydrophobic interaction between myosin molecules,and the viscoelasticity of CC-added myosin was second only to the CaCl_(2)-added myosin.On the contrary,the addition of HAP and TCP interfered with the hydrophobic interaction between myosin molecules,and greatly reduced the viscoelasticity of myosin.In summary,calcium with high calcium release rate can improve the physicochemical properties of myosin,HAP and TCP with a very low calcium release rate interfered with the formation of myosin gelation.And it could be concluded that the calcium ions played a leading role in the physicochemical properties of myosin,the more calcium ion that the calcium salt can release,the better physicochemical properties the myosin were.Our work may provide an insight into the phenomenon why different kinds of calcium salts differ from each other on affecting the physicochemical properties of myosin during gel processing.

Excessive nitrogen(N)fertilizer application in crop production has resulted in a series of environmental issues in North China Plain(NCP).Breeding N-efficient maize varieties is one of the effective strategies to improve the N use efficiency of maize and reduce environmental pollution.This study designed an experiment for two maize varieties(‘Xian-Yu 335’[XY335]and‘Hua-Nong 138’[HN138])with three N-level treatments(0 kg/hm^(2),180 kg/hm^(2),300 kg/hm^(2),i.e.N0,N180,N300).The aim of the study was to determine the response characteristics of maize canopy structure,root morphology,and anatomical structure,as well as N uptake and utilization of N fertilizer.Results showed that grain yield,N use efficiency,and N harvest index of XY335 were higher 5.20%-13.68%,11.86%-19.11%,and 6.07%-3.33%than those of HN138,respectively.Compared with HN138,XY335 had a higher leaf area index and photosynthetic nitrogen-use efficiency.The root length,root length density,root absorption area,root absorption area,and root active absorption area of XY335 in each period were higher than that of HN138.The proportion of RCA of XY335 was significantly higher than that of HN138,which was 14.64%higher on average.In the future breeding process,the high N and low N double-high-efficiency varieties should pay more attention to the improvement of root cortex aeration tissue.

目的探讨SPARC(骨素)、cwcv和Kazal样结构域蛋白多糖1(Spock1)在肥大型与扩张型心肌病发生中的作用与分子机制。方法基于CNKI数据库明确Spock1在机体各组织中的表达情况,并获得Spock1参与信号通路的关联基因,对此关联基因进行GO和KEGG富集分析,对比同类基因与心脏疾病的关系,从而分析获得Spock1参与肥大型与扩张型心肌病的具体关联信息。结果Spock1在心脏中较高表达,并且其可能通过Adipogenesis信号通路参与肥大型心肌病(HCM)和扩张型心肌病(DCM)的发生与发展。结论Spock1可能与肥大型和扩张型心肌病等相关心血管疾病的发生密切相关,并可能成为心脏疾病发生发展的生物标记物,甚至可成为治疗心脏疾病的新靶点基因。

Design and experimental research on disc-type seeding device for single-bud sugarcane seeds

Mechanized sugarcane seeding is one of the effective technical measures for improving seeding uniformity and operation efficiency.However,the imperfection of supporting equipment limits the promotion and application of this technology.As there are some problems in domestic sugarcane planting machines currently,such as large auxiliary labor,high labor intensity,low seeding uniformity,and large amount of seeds,a disc-type single-bud sugarcane seed metering device was innovatively designed on the basis of the analysis of physical properties of single-bud sugarcane seeds and the combination with agronomic requirements for field planting in this paper.Solidworks was used to simulate and analyze the mechanism,check the theoretical parameters,and process the test prototype.Through single-factor experiments,the effective factors affecting seeding uniformity were determined.A multi-factor orthogonal rotation test was designed,data were collected and then SPSS was used to get the optimal parameters for the seeding uniformity:the advancement speed of machine is 0.22 m/s,the number of disc seeding grooves is 10,the rotating disc speed is 0.18 r/s,and the seeding uniformity is 86.2%;the seeding uniformity was verified by field trial,and the results showed that the average seeding uniformity of the field verification was 83%,with the error 3.2% relative to the optimization result.The relative error is within 10%,indicating that the optimization result is reliable and meets the requirements of seeding uniformity,thus providing theoretical basis for the research and development of the disc-type single-bud seeding device.

Background:Sarcopenia is an age-related progressive skeletal muscle disorder involving the loss of muscle mass or strength and physiological function.Efficient and precise AI algorithms may play a significant role in the diagnosis of sarcopenia.In this study,we aimed to develop a machine learning model for sarcopenia diagnosis using clinical characteristics and laboratory indicators of aging cohorts.Methods:We developed models of sarcopenia using the baseline data from the West China Health and Aging Trend(WCHAT)study.For external validation,we used the Xiamen Aging Trend(XMAT)cohort.We compared the support vector machine(SVM),random forest(RF),eXtreme Gradient Boosting(XGB),and Wide and Deep(W&D)models.The area under the receiver operating curve(AUC)and accuracy(ACC)were used to evaluate the diagnostic efficiency of the models.Results:The WCHAT cohort,which included a total of 4057 participants for the training and testing datasets,and the XMAT cohort,which consisted of 553 participants for the external validation dataset,were enrolled in this study.Among the four models,W&D had the best performance(AUC=0.916±0.006,ACC=0.882±0.006),followed by SVM(AUC=0.907±0.004,ACC=0.877±0.006),XGB(AUC=0.877±0.005,ACC=0.868±0.005),and RF(AUC=0.843±0.031,ACC=0.836±0.024)in the training dataset.Meanwhile,in the testing dataset,the diagnostic efficiency of the models from large to small was W&D(AUC=0.881,ACC=0.862),XGB(AUC=0.858,ACC=0.861),RF(AUC=0.843,ACC=0.836),and SVM(AUC=0.829,ACC=0.857).In the external validation dataset,the performance of W&D(AUC=0.970,ACC=0.911)was the best among the four models,followed by RF(AUC=0.830,ACC=0.769),SVM(AUC=0.766,ACC=0.738),and XGB(AUC=0.722,ACC=0.749).Conclusions:The W&D model not only had excellent diagnostic performance for sarcopenia but also showed good economic efficiency and timeliness.It could be widely used in primary health care institutions or developing areas with an aging population.Trial Registration:***,ChiCTR 1800018895.

采用Gleeble-1500D型热模拟实验机进行SA-765 Gr.Ⅱ钢等温热压缩实验,压缩变形温度分别为1000、1050、1100、1150和1200℃,应变速率分别为0.005、0.01和1 s^(-1),最大压缩变形量为50%。基于热压缩变形过程的真应力-真应变数据,采用加工硬化率法和三次多项式拟合,求得不同变形条件下SA-765 Gr.Ⅱ钢的临界应力与临界应变,建立了关联Zener-Hollomon参数的动态再结晶临界应变模型。结果表明:变形温度和应变速率会极大地影响SA-765 Gr.Ⅱ钢的动态再结晶机制的启动,动态再结晶临界应变与变形温度成反比,与应变速率成正比。动态再结晶临界应变模型为:ε_(c)=0.201035Z^(0.000402),依据模型计算得到的临界应变预测值与实验值之间的相关系数达到了0.9765。

为探究煤矸石在雨水冲刷作用下重金属元素的释放规律及特征,以新疆某采矿区煤矸石为研究对象,采用动态模拟淋滤实验,研究煤矸石中Fe、Mn、Zn和Cu元素的释放规律。结果表明,Fe、Mn、Zn和Cu元素在淋滤前期溶出速率较快,淋出浓度较高,之后随着淋滤时间的延长,各金属元素溶出速率降低,淋出浓度减小,最后趋于稳定,释放达到平衡,整体表现为快速-慢速-平稳释放特征。结合地下水质量标准的要求和各重金属元素在淋滤期内的最大淋滤浓度,该矸石所在地的地下水适用于农业和部分工业用水;金属元素在淋滤期内(8d)的淋出值大小表现为Fe>Mn>Zn>Cu的顺序,Fe元素有相对较强的淋出活性,有向土壤和地下水迁移的潜在风险;矸石中重金属元素的淋滤释放呈指数曲线规律衰减,根据衰减模型可定量预测矸石中重金属在动态淋滤时对土壤和地下水的污染强度。本次实验结果为建立煤矸石堆放条件提供了数据支撑,并为煤矸石堆放场周围水体和土壤的污染治理提供了重要的科学依据。
