Type 2 diabetes mellitus(T2DM)is a complex metabolic disease threatening human health.We investigated the effects of Tegillarca granosa polysaccharide(TGP)and determined its potential mechanisms in a mouse model of T2DM established through a high-fat diet and streptozotocin.TGP(5.1×10^(3) Da)was composed of mannose,glucosamine,rhamnose,glucuronic acid,galactosamine,glucose,galactose,xylose,and fucose.It could significantly alleviate weight loss,reduce fasting blood glucose levels,reverse dyslipidemia,reduce liver damage from oxidative stress,and improve insulin sensitivity.RT-PCR and Western blotting indicated that TGP could activate the phosphatidylinositol-3-kinase/protein kinase B signaling pathway to regulate disorders in glucolipid metabolism and improve insulin resistance.TGP increased the abundance of Allobaculum,Akkermansia,and Bifidobacterium,restored the microbiota abundance in the intestinal tracts of mice with T2DM,and promoted short-chain fatty acid production.This study provides new insights into the antidiabetic effects of TGP and highlights its potential as a natural hypoglycemic nutraceutical.

As coronavirus disease 2019(COVID-19)public health restrictions are relaxed,the circulation of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)alongside other respiratory viruses may lead to an increased likelihood of coinfection(1).Older patients face a higher risk of severe outcomes,when infected with multiple respiratory viruses(2).This study highlights the successful recovery of the oldest older adult(≥80 years)from pneumonia caused by the dual infection of human respiratory syncytial virus(HRSV)and SARS-CoV-2.

Identification and characterization of camptothecin tailoring enzymes in Nothapodytes tomentosa

Camptothecin is a complex monoterpenoid indole alkaloid with remarkable antitumor activity.Given that two C-10 modified camptothecin derivatives,topotecan and irinotecan,have been approved as potent anticancer agents,there is a critical need for methods to access other aromatic ringfunctionalized congeners(e.g.,C-9,C-10,etc.).However,contemporary methods for chemical oxidation are generally harsh and low-yielding when applied to the camptothecin scaffold,thereby limiting the development of modified derivatives.Reported herein,we have identified four tailoring enzymes responsible for C-9 modifications of camptothecin from Nothapodytes tomentosa,via metabolomic and transcriptomic analysis.These consist of a cytochrome P450(Nt CPT9H)which catalyzes the regioselective oxidation of camptothecin to 9-hydroxycamptothecin,as well as two methyltransferases(Nt OMT1/2,converting 9-hydroxycamptothecin to 9-methoxycamptothecin),and a uridine diphosphate-glycosyltransferase(Nt UGT5,decorating 9-hydroxycamptothecin to9-β-D-glucosyloxycamptothecin).Importantly,the critical residues that contribute to the specific catalytic activity of Nt CPT9H have been elucidated through molecular docking and mutagenesis experiments.This work provides a genetic basis for producing camptothecin derivatives through metabolic engineering.This will hasten the discovery of novel C-9 modified camptothecin derivatives,with profound implications for pharmaceutical manufacture.

We exhibit an explicit formula for the cardinality of solutions to a class of quadratic matrix equations over finite fields.We prove that the orbits of these solutions under the natural conjugation action of the general linear groups can be separated by classical conjugation invariants defined by characteristic polynomials.We also find a generating set for the vanishing ideal of these orbits.

Atorvastatin, etanercept and the nephrogenic cardiac sympathetic remodeling in chronic renal failure rats

BACKGROUND Chronic renal failure(CRF) patients are predisposed to arrhythmias, while the detailed mechanisms are unclear. We hypothesized the chronic inflammatory state of CRF patients may lead to cardiac sympathetic remodeling, increasing the incidence of ventricular arrhythmia(VA) and sudden cardiac death. And explored the role of atorvastatin and etanercept in this process.METHODS A total of 48 rats were randomly divided into sham operation group(Sham group), CRF group, CRF + atorvastatin group(CRF + statin group), and CRF + etanercept group(CRF + rhTNFR-Fcgroup). Sympathetic nerve remodeling was assessed by immunofluorescence of growth-associated protein 43(GAP-43) and tyrosine hydroxylase positive area fraction. Electrophysiological testing was performed to assess the incidence of VA by assessing the ventricular effective refractory period and ventricular fibrillation threshold. The levels of tumor necrosis factor-alpha(TNF-α) and interleukin-1beta were determined by Western blotting and enzyme-linked immunosorbent assay.RESULTS Echocardiogram showed that compared with the Sham group, left ventricular end-systolic diameter and ventricular weight/body weight ratio were significantly higher in the CRF group. Hematoxylin-eosin and Masson staining indicated that myocardial fibers were broken, disordered, and fibrotic in the CRF group. Western blotting, enzyme-linked immunosorbent assay,immunofluorescence and electrophysiological examination suggested that compared with the Sham group, GAP-43 and TNF-α proteins were significantly upregulated, GAP-43 and tyrosine hydroxylase positive nerve fiber area was increased, and ventricular fibrillation threshold was significantly decreased in the CRF group. The above effects were inhibited in the CRF + statin group and the CRF + rhTNFR-Fcgroup.CONCLUSIONS In CRF rats, TNF-α was upregulated, cardiac sympathetic remodeling was more severe, and the nephrogenic cardiac sympathetic remodeling existed. Determination of dynamic capillary effect on two-phase flow in porous media: A perspective from various methods

The relationship between capillary pressure and saturation plays a critical role in the characterization of two-phase flow and transport in aquifers and oil reservoirs. This relationship is usually determined under the static condition, where capillary pressure is the only function of saturation. However,considerable experiments have suggested that the dependence of capillary pressure on desaturation rate is under the dynamic condition. Thus, a more general description of capillary pressure that includes dynamic capillary effect has been approved widely. A comprehensive understanding of the dynamic capillary effect is needed for the investigation of the two-phase flow in porous media by various methods. In general, dynamic capillary effect in porous media can be studied through the laboratory experiment, pore-to macro-scale modeling, and artificial neural network. Here, main principle and research procedures of each method are reviewed in detail. Then, research progress, disadvantages and advantages are discussed, respectively. In addition, upscaling study from pore-to macro-scale are introduced, which explains the difference between laboratory experiment and pore-scale modeling. At last, several future perspectives and recommendations for optimal solution of dynamic capillary effect are presented.

Association of Sandstone-Type Uranium Mineralization in the Northern China with Tectonic Movements and Hydrocarbons

In the continental basins of Northern China(NC),a series of energy resources commonly co-exist in the same basin.As the three typical superimposed basins of different genesis in the NC,the Junggar,Ordos,and Songliao basins were chosen as the research objects.The favorable uraniumbearing structures are generally shown as a basin-margin slope or transition belt of uplifts with the development of faults,which are conducive to a fluid circulation system.The Hercynian,Indosinian,and Yanshanian movements resulted in the development of uranium-rich intrusions which acted as the significant uranium sources.The main hydrocarbon source rocks are developed in the Carboniferous,Permian,Jurassic and Cretaceous.The mature stage of source rocks is concentrated in the Jurassic–Cretaceous,followed by the multi-stage expulsion events.Influenced by the India-Eurasian collision and the subduction of the Pacific Plate,the tectonic transformation in the Late Yanshanian and Himalayan periods significantly influenced the sandstone-type uranium mineralization.The hydrocarbon reservoirs are spatially consistent with sandstone-type uranium deposits,while the hydrocarbon expulsion events occur in sequence with sandstone-type uranium mineralization.In the periphery of the faults or the uplifts,both fluids met and formed uranium concentration.The regional tectonic movements motivate the migration of hydrocarbon fluids and uranium mineralization,especially the Himalayan movement.

Characteristics of Altered Ilmenite in Uranium-Bearing Sandstone and Its Relationship with Uranium Minerals in the Northeastern Ordos Basin

In recent years,the close relationship between uranium and Ti-Fe oxides in the sandstonetype uranium deposits has been extensively recognized.However,the altered characteristics of ilmenite and its relationship with uranium enrichment still remain unclear.With this paper based on heavymineral sorting of uranium ore selected from the Tarangaole-Nalinggou deposit in the northeastern Ordos Basin,electron probe,backscattering image,energy spectrum and scanning electron microscopy were systematically performed.The ilmenite in the sandstone can be divided into four groups,including unaltered,weakly altered,moderately altered,and strongly altered ilmenite.The alteration of ilmenite in uranium ores is notably more intense than that of the surrounding rocks.In addition,weakly,moderately,and strongly altered ilmenite associated with uranium minerals in uranium ores demonstrate that the more intensity ilmenite altered,the closer its relationship with uranium minerals is.The ilmenite has likely been somewhat altered before mineralization,and the alteration intensifies by later exposure related to an oxygencontaining fluid.The alteration mechanism comprises a process of competitive diffusion between Fe^(2+)and O_(2)-ions.In the early stage,Fe ions was mainly diffused on the particle surface.Subsequently,diffusion of O ions into the particles began to be dominate.Most of the leached iron is stripped or carried away by fluid.In an alkaline and reductive environment,the remaining iron is reduced to form the surrounding pyrite,and TiO_(2)in a form of titanium sol recrystallizes(i.e.,anatase).Backscattering images show that uranium and altered ilmenite are close in space.Coffinite is often distributed along the edges of altered ilmenite as burrs in shape.Colloidal or knitted coffinite associated with anatase is formed in the voids of altered ilmenite.The chemical composition of altered ilmenite varies considerably from the core to edge,and the mineral assemblage sequence is from girdle with ilmenite,to leucosphenite,to anatase,and

Observation of therapeutic effect of lamp irradiation combined with purple gromwell oil gauze on alleviating intestinal colic in patients

BACKGROUND Intestinal colic is a common complication in patients who have undergone radical surgery for colorectal cancer.Traditional Chinese medicine has advantages,including safety and stability,for the treatment of intestinal colic.Lamp irra-diation for abdominal ironing has been applied in the treatment of many gas-trointestinal diseases.Purple gromwell oil has the effects of clearing heat,cooling blood,reducing swelling,and relieving pain.RESULTS The general effective rate in the observation group was 95.00%,which was significantly higher than that in the control group(86.67%,P0.05).After 1,2,3,and 4 d of treatment,the duration of symptoms in both groups were decreased,and the duration in the observation group was significantly lower than that in the control group(96.54±9.57 vs 110.45±11.23,87.26±12.07 vs 104.44±11.68,80.45±16.21 vs 99.44±14.95,73.18±15.58 vs 92.17±14.20;P0.05).CONCLUSION The use of lamp irradiation combined with purple gromwell oil gauze in patients with intestinal colic after radical surgery for colorectal cancer can reduce symptom duration,alleviate intestinal colic,and improve treatment efficacy,and this approach is safe.It is worth promoting the use of this treatment in clinical practice.

Restoration of formation processes of dolomite reservoirs based on laser U-Pb dating: A case study of Ordovician Majiagou Formation, Ordos Basin, NW China

To address the issue of non-unique interpretation of dolomite reservoir diagenetic and porosity evolution in the previous qualitative or semi-quantitative studies, we investigate two dolomite reservoir types, i.e. weathering-crust karstic reservoirs and mound-beach reservoirs, in the Ordovician Majiagou Formation, Ordos Basin by using in-situ laser ablation U-Pb dating as well as carbon and oxygen isotopic composition analysis. The results show that:(1) According to the dating of 8 reservoir samples, the Majiagou Formation experienced 5 diagenetic stages(Stage 1: deposition of matrix dolomite or penecontemporaneous dolomitization, in 444.0–494.0 Ma;Stage 2: dogtooth-or blade-shaped dolomite cementation, in 440.0–467.0 Ma;Stage 3: dolomitic silt filling, in 316.5–381.0 Ma;Stage 4: crystalline dolomite filling, in 354.0 Ma;Stage 5: crystalline calcite filling, in 292.7–319.0 Ma).(2) Supra-salt weathering-crust karstic dolomite reservoirs went through several diagenetic processes including penecontemporaneous dolomitization, compaction, weathering-crust karstification, packing, and rupturing in succession. Gypsum mold pores formed in the phase of hypergenic karstification and were filled with such minerals as dolomitic silts and calcites, and thus the porosity decreased from 10%–40% to 3%–8%.(3) Sub-salt mound-beach dolomite reservoirs went through the diagenetic processes including penecontemporaneous dolomitization, compaction, subsea cementation, penecontemporaneous corrosion, infiltration backflow dolomitization, packing, and rupturing. The porosity of reservoirs was originally 10%–30%, decreased to 0–6% due to seawater cementation, rose back to 5%–15% owing to penecontemporaneous corrosion, and finally declined to 2%–6% as a result of crystalline dolomites and calcites packing. The above methodology for the restoration of dolomitization and porosity evolution may be helpful for the restoration of porosity evolution in other basins or series of strata.
