Enhancing m6A modification in the motor cortex facilitates corticospinal tract remodeling after spinal cord injury

Spinal cord injury typically causes corticospinal tract disruption. Although the disrupted corticospinal tract can self-regenerate to a certain degree, the underlying mechanism of this process is still unclear. N6-methyladenosine（m6A） modifications are the most common form of epigenetic regulation at the RNA level and play an essential role in biological processes. However, whether m6A modifications participate in corticospinal tract regeneration after spinal cord injury remains unknown. We found that expression of methyltransferase 14 protein（METTL14） in the locomotor cortex was high after spinal cord injury and accompanied by elevated m6A levels. Knockdown of Mettl14 in the locomotor cortex was not favorable for corticospinal tract regeneration and neurological recovery after spinal cord injury. Through bioinformatics analysis and methylated RNA immunoprecipitation-quantitative polymerase chain reaction, we found that METTL14 regulated Trib2 expression in an m6A-regulated manner, thereby activating the mitogen-activated protein kinase pathway and promoting corticospinal tract regeneration. Finally, we administered syringin, a stabilizer of METTL14, using molecular docking. Results confirmed that syringin can promote corticospinal tract regeneration and facilitate neurological recovery by stabilizing METTL14. Findings from this study reveal that m6A modification is involved in the regulation of corticospinal tract regeneration after spinal cord injury.

Triple-path feature transform network for ring-array photoacoustic tomography image reconstruction

Photoacoustic imaging(PAI)is a noninvasive emerging imaging method based on the photoacoustic effect,which provides necessary assistance for medical diagnosis.It has the characteristics of large imaging depth and high contrast.However,limited by the equipment cost and reconstruction time requirements,the existing PAI systems distributed with annular array transducers are difficult to take into account both the image quality and the imaging speed.In this paper,a triple-path feature transform network(TFT-Net)for ring-array photoacoustic tomography is proposed to enhance the imaging quality from limited-view and sparse measurement data.Specifically,the network combines the raw photoacoustic pressure signals and conventional linear reconstruction images as input data,and takes the photoacoustic physical model as a prior information to guide the reconstruction process.In addition,to enhance the ability of extracting signal features,the residual block and squeeze and excitation block are introduced into the TFT-Net.For further efficient reconstruction,the final output of photoacoustic signals uses‘filter-then-upsample’operation with a pixel-shuffle multiplexer and a max out module.Experiment results on simulated and in-vivo data demonstrate that the constructed TFT-Net can restore the target boundary clearly,reduce background noise,and realize fast and high-quality photoacoustic image reconstruction of limited view with sparse sampling.

Atomization characteristics of pyrolysis oil derived from waste tires

The atomization characteristics play a key role in the highly efficient combustion of pyrolysis oil derived from waste tires.In this study,the fuel properties of tire pyrolysis oil(TPO)were initially studied,and then a high-speed camera and a phase Doppler particle analyzer were employed to characterize the atomization feature of TPO.The influence of pressure and nozzle orifice diameter on atomization characteristics such as spray angle,droplet velocity,and droplet size distribution was investigated.The results showed that TPO had a high calorific value of about 43.6 MJ/kg and a low viscosity of 3.84×10^(–6)m^(2)/s at 40℃,which made it have the potential to be used as an alternative fuel.Higher pressure expanded the spray angle and extended the spray in both the axial and radial directions.With increasing pressure,spray angle and droplet velocity raised,and the increase in crushing effect of air reduced the Sauter mean diameter(SMD)of the droplets.To obtain proper atomization quality for combustion,the pressure is expected to be higher than 1.25 MPa.With increasing nozzle orifice diameter,droplet velocity increased,and the SMD of the droplets increased as well due to weakened crushing effect of the orifice.Therefore,the pressure must be increased to maintain the atomization quality when using a nozzle with a larger orifice.Due to the lower viscosity,the velocity and particle size distribution of TPO droplets after atomization were smaller than those of diesel droplets.The extremely small carbon black contained in TPO also contributed to the breaking of droplets and played a certain role in the size reduction of the oil droplets,but it may cause the risk of nozzle blockage.In summary,TPO showed great atomization characteristics for alternative fuel applications.

A transfer zone in rift basins preserves important information on regional tectonic evolution and plays significant roles in hydrocarbon accumulation.Based on the systematic analysis of 3D seismic data and hydrocarbon accumulation conditions,the geometry,kinematics,and reservoir control of a large synthetic overlapping transfer zone in the south of the Wenchang A subsag in the Zhujiang(Pearl)River Mouth basin were investigated.Results indicate that the development and evolution of the transfer zone was controlled by the interaction between pre-existing faults and regional stress transformation.The intense rifting of the main faults of the transfer zone controlled the development of source rocks and faultcontrolled slope break paleogeomorphology.The strike-slip overprint since the Oligocene is conducive to the formation of a large-scale fault-anticline trap,and the secondary faults in the transfer zone contribute to the hydrocarbon transportation.The conjugate intersection area of the NE-and NW-trending faults offers more opportunity for hydrocarbon migration and accumulation.

To the Editor:Infertility affects 15%to 20%of couples worldwide,and assisted reproductive technology(ART)is the most effective treatment strategy.In addition to female age,many other factors,such as antral follicle count(AFC),basal follicle-stimulating hormone(FSH)level,and anti-Mullerian hormone(AMH)level,collectively known as biomarkers for ovarian reserve tests(ORTs),are widely used to predict ovarian responses and assisted-pregnancy outcomes.[1]Among them,AMH and FSH levels were discordant in 20%to 43%of women undergoing ART treatment,and AMH was proven to be superior in predicting live birth rate(LBR).[2]

Glioma is the most prevalent brain tumor,presenting with limited treatment options,while patients with malignant glioma and glioblastoma(GBM)have poor prognoses.The physical obstacle to drug delivery imposed by the blood‒brain barrier(BBB)and glioma stem cells(GSCs),which are widely recognized as crucial elements contributing to the unsatisfactory clinical outcomes.In this study,we found a small molecule,gambogic amide(GA-amide),exhibited the ability to effectively penetrate the blood-brain barrier(BBB)and displayed a notable enrichment within the tumor region.Moreover,GA-amide exhibited significant efficacy in inhibiting tumor growth across various in vivo glioma models,encompassing transgenic and primary patient-derived xenograft(PDX)models.We further performed a genome-wide clustered regularly interspaced short palindromic repeats(CRISPR)knockout screen to determine the druggable target of GA-amide.By the combination of the cellular thermal shift assay(CETSA),the drug affinity responsive target stability(DARTS)approach,molecular docking simulation and surface plasmon resonance(SPR)analysis,WD repeat domain 1(WDR1)was identified as the direct binding target of GA-amide.Through direct interaction with WDR1,GA-amide promoted the formation of a complex involving WDR1,MYH9 and Cofilin,which accelerate the depolymerization of F-actin to inhibit the invasion of patient-derived glioma cells(PDCs)and induce PDC apoptosis via the mitochondrial apoptotic pathway.In conclusion,our study not only identified GA-amide as an effective and safe agent for treating glioma but also shed light on the underlying mechanisms of GA-amide from the perspective of cytoskeletal homeostasis.

Studies have found a U-shaped relationship between sleep duration and chronic kidney disease(CKD)risk,but limited research evaluated the association of reallocating excessive sleep to other behavior with CKD.We included 104538 participants from the nationwide cohort of the Risk Evaluation of Cancers in Chinese Diabetic Individuals:A Longitudinal Study,with self-reported time of daily-life behavior.Using isotemporal substitution models,we found that substituting 1 h of sleeping with sitting,walking,or moderate-to-vigorous physical activity was associated with a lower CKD prevalence.Leisure-time physical activity displacement was associated with a greater prevalence reduction than occupational physical activity in working population.In stratified analysis,a lower CKD prevalence related to substitution toward physical activity was found in long sleepers.More pronounced correlations were observed in long sleepers with diabetes than in those with prediabetes,and they benefited from other behavior substitutions toward a more active way.The U-shaped association between sleep duration and CKD prevalence implied the potential effects of insufficient and excessive sleep on the kidneys,in which the pernicious link with oversleep could be reversed by time reallocation to physical activity.The divergence in the predicted effect on CKD following time reallocation to behavior of different domains and intensities and in subpopulations with diverse metabolic statuses underlined the importance of optimizing sleeping patterns and adjusting integral behavioral composition.

单一线路高速列车转向架缺少足量故障数据特征,导致故障诊断模型泛化能力有限,为实现诊断多条线路高速列车的转向架故障,提出一种基于联邦学习的转向架全局故障诊断方法.针对每条线路各自的转向架振动信号,在本地使用多尺度卷积融合算法,提取不同尺度下的故障特征并融合,在本地建立局部转向架故障诊断模型;在不泄露数据隐私的前提下,所有线路的故障诊断模型通过第三方聚合,调整模型参数权重,对故障诊断模型进行优化,最终实现多方联合训练转向架全局故障诊断模型.实验表明:在联邦学习框架下,转向架全局故障诊断模型不仅对参与联邦建模的线路转向架故障诊断准确率达到93%以上,而且对于未参与联邦建模的线路转向架故障诊断率也可达到75%以上,给轨道交通中的“数据孤岛”问题提供了一种切实可行的方案.

Wildfire smoke plays a crucial role in climate change,air quality,human health,and the ecosystem.With the warming and drying of the climate,the scope,frequency,and intensity of global wildfire events are increasing rapidly[1,2].Southeast Asia,South America,Africa,and Australia are the major regions in which wildfires occur[3,4];Southeast Asia is the hotspot of wildfires and neighbors the South China Sea(SCS)[5].Therefore,the air quality and climate in the SCS can be affected by the transport of wildfire smoke in Southeast Asia.Many studies have focused on the transport pathways from Southeast Asia to the SCS,especially studies based on BASE-ASIA(Biomass-burning Aerosols in South-East Asia)and 7-SEAS(7-South-East Asian Studies)experiments[5,6].
