Mitochondrial components transferred by MSC-derived exosomes promoted bone regeneration under high salt microenvironment via DRP1/Wnt signaling

Bone homeostasis relies on the dynamic balance of osteoblast mediated bone construction and osteoclast-based bone resorption processes,which has been reported to be controlled by various mineral ***,there is no direct evidence of the effect and the underlying mechanism of high salt stimulation on bone *** this study,we demonstrated that high salt stimulation promoted excessive mitochondrial fission mediated by dynamin-related protein 1 in mesenchymal stem cells,which resulted in impaired mitochondrial morphology and ***,this impairment hindered the bone formation of mesenchymal stem cells,resulting in osteopenia in ***,the impaired property of mesenchymal stem cells which was caused by high salt was controlled by dynamin-related protein 1 mediated mitochondrial fission,which inhibited the classical Wnt signaling ***,the osteogenic property of mesenchymal stem cells decreased by high salt could be restored by exosomes to transfer the mitochondrial DNA into the impaired mesenchymal stem *** study provides not only new strategies for promoting bone regeneration but also new insights into the effect and mechanism of exosome-mediated delivery.

The terahertz(THz)wave is at the intersection between photonics and electronics in the electromagnetic *** the vibration mode of many biomedical molecules and the weak interaction mode inside the molecules fall in the THz regime,utilizing THz radiation as a signal source to operate substance information sensing has its unique ***,the metamaterial sensor(metasensor)has greatly enhanced the interaction between signal and substances and spectral selectivity on the subwavelength ***,most past review articles have demonstrated the THz metasensor in terms of their structures,applications,or *** recently,with the rapid development of metasensing technologies,the molecular information has paid much more attention to the platform of THz *** this review,we comprehensively introduce the THz metasensor for detecting not only the featureless refractive index but also the vibrational/chiral molecular information of *** objectives of this review are to improve metasensing specificity either by chemical material-assisted analyte capture or by physical molecular ***,to boost THz absorption features in a certain frequency,the resonant responses of metasensors can be tuned to the molecular vibrational modes of target molecules,while frequency multiplexing techniques are reviewed to enhance broadband THz spectroscopic *** chiral metasensors are also summarized to specific identification chiral ***,the potential prospects of next generation THz metasensors are *** to featureless refractive index metasensing,the specific metasensor platforms accelerated by material modification and molecular information will lead to greater impact in the advancement of trace detection of conformational dynamics of biomolecules in practical applications.

Rechargeable neutral aqueous zinc-air batteries(ZABs)are a promising type of energy storage device with longer operating life and less corrosiveness compared with conventional alkaline ***,the neutral ZABs normally possess poor oxygen evolution reactions(OERs)and oxygen reduction reactions performance,resulting in a large charge–discharge voltage gap and low round-trip ***,we demonstrate a sunlight-assisted strategy for achieving an ultralow voltage gap of 0.05 V in neutral ZABs by using the FeOOH-decorated BiVO4(Fe-BiVO4)as an oxygen *** sunlight,the electrons move from the valence band(VB)of Fe-BiVO_(4) to the conduction band producing holes in VB to promote the OER process and hence reduce the ***,the photopotential generated by the Fe-BiVO_(4) compensates a part of the charging potential of neutral ***,the energy loss of the battery could be compensated via solar energy,leading to a record-low gap of 0.05 V between the charge and discharge voltage with a high round-trip efficiency of 94%.This work offers a simple but efficient pathway for solar-energy utilization in storage devices,further guiding the design of high energy efficiency of neutral aqueous ZABs.

In artificial photosynthesis systems,synthetic diiron complexes are popular[FeFe]-hydrogenase mimics,which are attractive for the fabrication of photocatalyst-protein hybrid structures to amplify hydrogen(H2)generation ***,constructing a highly bionic and efficient catalytic hybrid system is a major ***,we designed an ideal hybrid nanofibrils system that incorporates the crucial components:(1)a[FeFe]-H2ase mimic,which has a three-arm architecture(named triFeFe)for more interaction sites and higher catalytic activity and(2)uniform hybrid nanofibrils as the biological environment in which cysteine-catalyst coordination and the hydrogen-bonding network play a vital role in both catalyst binding and hydrogen evolution reaction *** assembled hybrid nanofibrils achieve efficient H2 generation with a turnover number of 2.3×103,outperforming previously reported diiron catalyst-protein hybrid ***,the hybrid nanofibrils work with photosynthetic thylakoids to produce H2,without extra photosensitizers or electron shuttle proteins,which advances the bioengineering of living systems for solar-driven biofuel production.

The direct cleavage of C–NO_(2)bonds for reductive denitration of nitroarenes remains a challenging transformation in synthetic organic ***,we report a biocompatible palladium-deposited graphdiyne nanocatalyst(Pd@GDY/DSPE-PEG)that can catalyze reductive denitration of nitroarenes under ambient physiological *** studies support this transformation via the oxidative addition of nitroarenes with Pd(0)and subsequent ligand exchange to form arylpalladium *** one-step reductive denitration via Pd@GDY/DSPE-PEG successfully facilitates the repair of the nitrated proteins arising from endogenic ONOO−and restores their physiological function,including blocking the apoptosis pathway in living ***,Pd@GDY/DSPE-PEG was further successfully applied for catalytic denitration to reduce the level of 3-nitrotyrosine residues of proteins located in the mouse brain hippocampus in *** study provides an ideal strategy for designing highly active enzymatic mimicking synthetic catalysts for the regulation of the nitrated protein level and the detoxification of nitrative damage of living cells and tissues.

Mitochondrial dysfunction is a hallmark of Alzheimer’s *** previously showed that neural stem cell-derived extracellular vesicles improved mitochondrial function in the cortex of AP P/PS1 *** Alzheimer’s disease affects the entire brain,further research is needed to elucidate alterations in mitochondrial metabolism in the brain as a ***,we investigated the expression of several important mitochondrial biogenesis-related cytokines in multiple brain regions after treatment with neural stem cell-derived exosomes and used a combination of whole brain clearing,immunostaining,and lightsheet imaging to clarify their spatial ***,to clarify whether the sirtuin 1(SIRT1)-related pathway plays a regulatory role in neural stem cell-de rived exosomes interfering with mitochondrial functional changes,we generated a novel nervous system-SIRT1 conditional knoc kout AP P/PS1mouse *** findings demonstrate that neural stem cell-de rived exosomes significantly increase SIRT1 levels,enhance the production of mitochondrial biogenesis-related fa ctors,and inhibit astrocyte activation,but do not suppress amyloid-β***,neural stem cell-derived exosomes may be a useful therapeutic strategy for Alzheimer’s disease that activates the SIRT1-PGC1αsignaling pathway and increases NRF1 and COXIV synthesis to improve mitochondrial *** addition,we showed that the spatial distribution of mitochondrial biogenesis-related factors is disrupted in Alzheimer’s disease,and that neural stem cell-derived exosome treatment can reverse this effect,indicating that neural stem cell-derived exosomes promote mitochondrial biogenesis.

Single-atom catalysts were widely used to treat atmospheric pollution and alleviate energy crises through ***,how to prevent the aggregation of single atoms during the preparation and catalytic processes remained a great ***,a novel ultrathin two-dimensional porphyrin-based single-atom photocatalyst Ti-MOF(abbreviated as TMPd)obtained through a simple hydrothermal synthesis strategy was used for photocatalytic hydrogen evolution and NO removal,in which the singleatom Pd tightly anchored in the center of porphyrin to ensure single-atom Pd stable *** with most reported MOFs-based photocatalysts,the TMPd showed an excellent hydrogen evolution rate(1.32 mmol g^(-1)h^(-1))and the NO removal efficiency(62%)under visible light *** high-angle annular dark-field scanning transmission electron microscope(HAADF-STEM)and synchrotron-radiation-based X-ray absorption fine-structure spectroscopy(XAFS)proved that pd in TMPd existed in an isolated state,and the atomic force microscope(AFM)proved the ultrathin morphology of *** calculations had demonstrated that single-atom Pd could serve as the active center and more effectively achieve electron transfer,indicating that single-atom Pd played a vital role in photocatalytic hydrogen *** addition,a possible photocatalytic pathway of NO removal was proposed based on ESR and in-situ infrared spectra,in which the catalysts anchored with single-atom Pd could produce more active substances and more effectively oxidize NO to NO_(2)^(-)or NO_(3)^(-).The results suggested that coordinating single-atom metal species as the active site in the center of porphyrin could be a feasible strategy to obtain various ultrathin porphyrin-based single-atom photocatalysts to acquire excellent photocatalytic performance further.

The rapid advancement of sequencing technologies poses challenges in managing the large volume and exponential growth of sequence data efficiently and on *** address this issue,we present GenBase(https://***/genbase),an open-access data repository that follows the International Nucleotide Sequence Database Collaboration(INSDC)data standards and structures,for efficient nucleotide sequence archiving,searching,and *** a core resource within the National Genomics Data Center(NGDC)of the China National Center for Bioinformation(CNCB;https://***),GenBase offers bilingual submission pipeline and services,as well as local submission assistance in *** also provides a unique Excel format for metadata description and feature annotation of nucleotide sequences,along with a real-time data validation system to streamline sequence *** of April 23,2024,GenBase received 68,251 nucleotide sequences and 689,574 annotated protein sequences across 414 species from 2319 *** of these,63,614(93%)nucleotide sequences and 620,640(90%)annotated protein sequences have been released and are publicly accessible through GenBase’s web search system,File Transfer Protocol(FTP),and Application Programming Interface(API).Additionally,in collaboration with INSDC,GenBase has constructed an effective data exchange mechanism with GenBank and started sharing released nucleotide ***,GenBase integrates all sequences from GenBank with daily updates,demonstrating its commitment to actively contributing to global sequence data management and sharing.
