FERONIA-mediated TIR1/AFB2 oxidation stimulates auxin signaling in Arabidopsis

The phytohormone auxin plays a pivotal role in governing plant growth and development.Although the TRANSPORT INHIBITOR RESPONSE1/AUXIN SIGNAliNG F-BOX(TIR1/AFB)receptors function in both the nucleus and cytoplasm,the mechanism governing the distribution of TIR1/AFBs between these cellular compartments remains unknown.In this study,we demonstrate that auxin-mediated oxidation of TIR1/AFB2 is essential for their targeting to the nucleus.We showed that small active molecules,reactive oxygen species(ROS)and nitric oxide(NO),are indispensable for the nucleo-cytoplasmic distribution of TIR1/AFB2 in trichoblasts and root hairs.Further studies revealed that this process is regulated by the FERONIA receptor kinase–NADPH oxidase signaling pathway.Interestingly,ROS and NO initiate oxidative modifications in TIR1C140/516 and AFB2C135/511,facilitating their subsequent nuclear import.The oxidized forms of TIR1C140/516 and AFB2C135/511 play a crucial role in enhancing the function of TIR1 and AFB2 in transcriptional auxin responses.Collectively,our study reveals a novel mechanism by which auxin stimulates the transport of TIR1/AFB2 from the cytoplasm to the nucleus,orchestrated by the FERONIA–ROS signaling pathway.

Coordinated responses of leaf and nodule traits contribute to the accumulation of N in relay intercropped soybean

Maize(Zea mays L.)-soybean(Glycine max L.Merr.)relay intercropping provides a way to enhance land productivity.However,the late-planted soybean suffers from shading by the maize.After maize harvest,how the recovery growth influences the leaf and nodule traits remains unclear.A three-year field experiment was conducted to evaluate the effects of genotypes,i.e.,supernodulating(nts1007),Nandou 12(ND12),and Guixia 3(GX3),and crop configurations,i.e.,the interspecific row spacing of 45(I45),60(I60),75 cm(I75),and sole soybean(SS),on soybean recovery growth and N fixation.The results showed that intercropping reduced the soybean total leaf area(LA)by reducing both the leaf number(LN)and unit leaflet area(LUA),and it reduced the nodule dry weight(NW)by reducing both the nodule number(NN)and nodule diameter(ND)compared with the SS.The correlation and principal component analysis(PCA)indicated a co-variability of the leaf and nodule traits in response to the genotype and crop configuration interactions.During the recovery growth stages,the compensatory growth promoted soybean growth to reduce the gaps of leaf and nodule traits between intercropping and SS.The relative growth rates of ureide(RGR_U)and nitrogen(RGR_N)accumulation were higher in intercropping than in SS.Intercropping achieved more significant sucrose and starch contents compared with SS.ND12 and GX3 showed more robust compensatory growth than nts1007 in intercropping.Although the recovery growth of relay intercropping soybean improved biomass and nitrogen accumulation,ND12 gained a more significant partial land equivalent ratio(pLER)than GX3.The I60 treatment achieved more robust compensation effects on biomass and N accumulation than the other configurations.Meanwhile,I60 showed a higher nodule sucrose content and greater shoot ureide and N accumulation than SS.Finally,intercropping ND12 with maize using an interspecific row spacing of 60 cm was optimal for both yield advantage and N accumulation.

For sessile plants, gene expression plays a pivotal role in responding to salinity stress by activating or suppressing specific genes. However, our knowledge of genetic variations governing gene expression in response to salt stress remains limited in natural germplasm. Through transcriptome analysis of the Global Mini-Core Rice Collection consisting of a panel of 202 accessions, we identified 22 345 and 27 610expression quantitative trait loci associated with the expression of 7787 and 9361 e Genes under normal and salt-stress conditions, respectively, leveraging the super pan-genome map. Notably, combined with genome-wide association studies, we swiftly pinpointed the potential candidate gene STG5—a major salt-tolerant locus known as q STS5. Intriguingly, STG5 is required for maintaining Na+/K+homeostasis by directly regulating the transcription of multiple members of the Os HKT gene family. Our study sheds light on how genetic variants influence the dynamic changes in gene expression responding to salinity stress and provides a valuable resource for the mining of salt-tolerant genes in the future.

According to the boot process of modern computer systems,whoever boots first will gain control first.Taking advantage of this feature,a malicious code called bootkit can hijack the control before the OS bootloader and bypass security mechanisms in boot process.That makes bootkits difficult to detect or clean up thoroughly.With the improvement of security mechanisms and the emergence of UEFI,the attack and defense techniques for bootkits have constantly been evolving.We first introduce two boot modes of modern computer systems and present an attack model of bootkits by some sophistical samples.Then we discuss some classic attack techniques used by bootkits from their initial appearance to the present on two axes,including boot mode axis and attack phase axis.Next,we evaluate the race to the bottom of the system and the evolution process between bootkits and security mechanisms.At last,we present the possible future direction for bootkits in the context of continuous improvement of OS and firmware security mechanisms.

The open-circuit fault is one of the most common faults of the automatic ramming drive system(ARDS),and it can be categorized into the open-phase faults of Permanent Magnet Synchronous Motor(PMSM)and the open-circuit faults of Voltage Source Inverter(VSI). The stator current serves as a common indicator for detecting open-circuit faults. Due to the identical changes of the stator current between the open-phase faults in the PMSM and failures of double switches within the same leg of the VSI, this paper utilizes the zero-sequence voltage component as an additional diagnostic criterion to differentiate them.Considering the variable conditions and substantial noise of the ARDS, a novel Multi-resolution Network(Mr Net) is proposed, which can extract multi-resolution perceptual information and enhance robustness to the noise. Meanwhile, a feature weighted layer is introduced to allocate higher weights to characteristics situated near the feature frequency. Both simulation and experiment results validate that the proposed fault diagnosis method can diagnose 25 types of open-circuit faults and achieve more than98.28% diagnostic accuracy. In addition, the experiment results also demonstrate that Mr Net has the capability of diagnosing the fault types accurately under the interference of noise signals(Laplace noise and Gaussian noise).

The ocean,with its highly variable and complex meteorological conditions,harbors enormous renewable resources.Triboelectric nanogenerators(TENGs),which possess unique advantages,show exciting prospects in water wave energy collection.How to design and optimize TENGs to cover all characteristic water wave energies and achieve efficient energy utilization is emergent.In this paper,we carefully designed and fabricated a columnar multi-layer sliding TENG(CMLS-TENG)that can harvest water wave energy independent of wave height and direction.Drive rods with a hollow acrylic spherical shell were introduced to deliver wave energy,ensuring that the CMLS-TENG can work in all directions from 0°to 360°.Based on the sliding structure,switching the optimized CMLS-TENG is independent of wave heights.The optimized CMLS-TENG can achieve a total power density of 730 mW/m^(3) at a wave height of only 4.8 cm regardless of wave direction,which can illuminate multiple light-emitting diodes(LEDs)to provide lighting and provide power to a watch and a hygrometer for temperature and humidity monitoring.This work provides new choices and hopes for the effective collection of full-range water wave energy.

Argatroban promotes recovery of spinal cord injury by inhibiting the PAR1/JAK2/STAT3 signaling pathway

Argatroban is a synthetic thrombin inhibitor approved by U.S.Food and Drug Administration for the treatment of thrombosis.However,whether it plays a role in the repair of spinal cord injury is unknown.In this study,we established a rat model of T10 moderate spinal cord injury using an NYU Impactor ModerⅢand performed intraperitoneal injection of argatroban for 3 consecutive days.Our results showed that argatroban effectively promoted neurological function recovery after spinal cord injury and decreased thrombin expression and activity in the local injured spinal cord.RNA sequencing transcriptomic analysis revealed that the differentially expressed genes in the argatroban-treated group were enriched in the JAK2/STAT3 pathway,which is involved in astrogliosis and glial scar formation.Western blotting and immunofluorescence results showed that argatroban downregulated the expression of the thrombin receptor PAR1 in the injured spinal cord and the JAK2/STAT3 signal pathway.Argatroban also inhibited the activation and proliferation of astrocytes and reduced glial scar formation in the spinal cord.Taken together,these findings suggest that argatroban may inhibit astrogliosis by inhibiting the thrombin-mediated PAR1/JAK2/STAT3 signal pathway,thereby promoting the recovery of neurological function after spinal cord injury.

Antibacterial mechanism of kojic acid and tea polyphenols against Escherichia coli O157:H7 through transcriptomic analysis

Escherichia coli O157:H7 is one of the major foodborne pathogenic bacterial that cause infectious diseases in humans.The previous found that a combination of kojic acid and tea polyphenols exhibited better activity against E.coli O157:H7 than using either alone.This study aimed to explore responses underlying the antibacterial mechanisms of kojic acid and tea polyphenols from the gene level.The functional enrichment analysis by comparing kojic acid and tea polyphenols individually or synergistically against E.coli O157:H7 found that acid resistance systems in kojic acid were activated,and the cell membrane and genomic DNA were destructed in the cells,resulting in“oxygen starvation”.The oxidative stress response triggered by tea polyphenols inhibited both sulfur uptake and the synthesis of ATP,which affected the bacteria's life metabolic process.Interestingly,we found that kojic acid combined with tea polyphenols hindered the uptake of iron that played an essential role in the synthesis of DNA,respiration,tricarboxylic acid cycle.The results suggested that the iron uptake pathways may represent a novel approach for kojic acid and tea polyphenols synergistically against E.coli O157:H7 and provided a theoretical basis for bacterial pathogen control in the food industry.

The energy revolution requires coordination in energy consumption, supply, storage and institutional systems.Renewable energy generation technologies, along with their associated costs, are already fully equipped for large-scale promotion.However, energy storage remains a bottleneck, and solutions areneeded through the use of electric vehicles, which traditionallyplay the role of energy consumption in power systems. Toclarify the key technologies and institutions that support EVsas terminals for energy use, storage, and feedback, the CSEEJPES forum assembled renowned experts and scholars in relevantfields to deliver keynote reports and engage in discussions ontopics such as vehicle–grid integration technology, advancedsolid-state battery technology, high-performance electric motortechnology, and institutional innovation in the industry chain.These experts also provided prospects for energy storage andutilization technologies capable of decarbonizing new powersystems.
