Bioinformatics analysis of human kallikrein 5 (KLK5) expression in metaplastic triple‐negative breast cancer

Background:Metaplastic breast carcinoma(MBC)is a rare breast cancer subtype;most cases are triple‐negative breast cancers(TNBCs)and are poorly responsive to conventional systemic therapy.Few potential diagnostic and prognostic markers for distinguishing between metaplastic TNBC and nonmetaplastic TNBC have been discovered.We performed bioinformatic analysis to explore the underlying mechanism by which metaplastic TNBC differs from nonmetaplastic TNBC and provides potential pathogenic genes of metaplastic TNBC.Methods:Differentially expressed genes(DEGs)in metaplastic tumors and nonmetaplastic tumors from TNBC patients were screened using GSE165407.The GSE76275 data set and The Cancer Genome Atlas(TCGA)database were used to screen DEGs in TNBC and non‐TNBC.Metascape and DAVID were used for the Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis and Gene Ontology(GO)analysis of DEGs.Online databases,including UALCAN,GEPIA,HPA,Breast Cancer Gene‐Expression Miner,and quantitative PCR and western blot,were used to examine KLK5 messenger RNA and protein expression in breast cancer.Analysis of KLK5‑associated genes was performed with TCGA data,and the LinkedOmics database was used to detect the genes co‐expressed with KLK5.STRING(Search Tool for the Retrieval of Interacting Genes)and Cytoscape were used to screen for hub genes.Kaplan‑Meier plotter was used for survival analysis.Results:KLK5 was identified among the DEGs in nonmetaplastic TNBC and metaplastic TNBC.The KLK5 gene was overexpressed in nonmetaplastic TNBC but downregulated in metaplastic TNBC.KEGG and GO analyses revealed that epithelial‐to‐mesenchymal transition was a pathogenic mechanism in metaplastic TNBC and an important pathway by which KLK5 and its associated genes DSG1and DSG3 influence metaplastic TNBC progression. Faster structured illumination microscopy using complementary encoding-based compressive imaging

Structured illumination microscopy (SIM) has been widely applied to investigate intricate biological dynamics due to its outstanding super-resolution imaging speed. Incorporating compressive sensing into SIM brings the possibility to further improve the super-resolution imaging speed. Nevertheless, the recovery of the superresolution information from the compressed measurement remains challenging in experiments. Here, we report structured illumination microscopy with complementary encoding-based compressive imaging (CECI-SIM) to realize faster super-resolution imaging. Compared to the nine measurements to obtain a super-resolution image in a conventional SIM, CECI-SIM can achieve a super-resolution image by three measurements;therefore, a threefold improvement in the imaging speed can be achieved. This faster imaging ability in CECI-SIM is experimentally verified by observing tubulin and actin in mouse embryonic fibroblast cells. This work provides a feasible solution for high-speed super-resolution imaging, which would bring significant applications in biomedical research.

进入和转换古代文学资源是文学现代性的题中应有之义。在20世纪，中国现代文学大致有四种进入古代文学资源的范式：在马克思主义批判地继承文化遗产的思想指引下，中国现代文学出现了以“精华糟粕”说为代表的社会政治化文学传统观范式；在西方阐释学等思潮激活下，产生了以“新文学源流”说为代表的精神启蒙型文学传统观范式；以科学释古与现代审美相结合为特征的“释古立美”类文学传统观范式，则是着重从传统内部进行解释和转化、更新和创造；此外还有一种以日常生活叙事为特征的民间常态化文学传统观范式，显示出古今文学贯通的文化整体性。在多种范式的共生互补、竞相发展中，重要的是在“互动”、“原创”中凸显问题意识，通过对古代文学资源的进入与激活，实现现实生命价值的重构。

Deterministic and Robust Volt-var Control Methods of Power System Based on Convex Deep Learning

Volt-var control(VVC) is essentially a non-convex optimization problem due to the non-convexity of power flow(PF) constraints, resulting in the difficulty in obtaining the optimum without convexity conversion. The existing second-order cone method for the convexity conversion often leads to a sharp increase in PF constraints and optimization variables, which in turn increases the optimization difficulty or even leads to optimization failure. This paper first proposes a deterministic VVC method based on convex deep learning power flow(DLPF). This method uses the input convex neural network(ICNN) to establish a single convex mapping between state parameters and node voltage to complete the convexity conversion while the optimization variables only correspond to reactive power equipment, which can ensure the global optimum with extremely fast computation speed. To cope with the impact brought by the uncertainty of distributed energy and omit the additional worst scenario search of traditional robust VVC, this paper proposes robust VVC method based on convex deep learning interval power flow(DLIPF), which continues to adopt ICNN to establish another convex mapping between state parameters and node voltage interval. Combining DLIPF with DLPF, this method decreases the modeling and optimization difficulty of robust VVC significantly. Test results on 30-bus, 118-bus, and 200-bus systems prove the correctness and rapidity of the proposed methods.

Superhydrophobic surface(SHS) has been well developed, as SHS renders the property of minimizing the water/solid contact interface. Water droplets deposited onto SHS with contact angles exceeding 150°, allow them to retain spherical shapes, and the low adhesion of SHS facilitates easy droplet collection when tilting the substrate. These characteristics make SHS suitable for a wide range of applications. One particularly promising application is the fabrication of microsphere and supraparticle materials. SHS offers a distinct advantage as a universal platform capable of providing customized services for a variety of microspheres and supraparticles. In this review, an overview of the strategies for fabricating microspheres and supraparticles with the aid of SHS, including cross-linking process, polymer melting,and droplet template evaporation methods, is first presented. Then, the applications of microspheres and supraparticles formed onto SHS are discussed in detail, for example, fabricating photonic devices with controllable structures and tunable structural colors, acting as catalysts with emerging or synergetic properties, being integrated into the biomedical field to construct the devices with different medicinal purposes, being utilized for inducing protein crystallization and detecting trace amounts of analytes. Finally,the perspective on future developments involved with this research field is given, along with some obstacles and opportunities.

Scattering of Water Waves by Dual Symmetric Inclined Floating Porous Barriers Using the DBEM

The scattering of normally incident water waves by two surface-piercing inclined perforated barriers in water with a uniform finite depth is investigated within the framework of linear water wave theory.Considering that thin barriers are zero-thickness,a novel numerical method involving the the coupling of the dual boundary element method(DBEM)with damping layers is applied.In order to effectively damp out the reflected waves,two damping layers,instead of pseudoboundaries are implemented near the two side boundaries of the computational domain.Thus,the modified linearized free surface boundary conditions are formulated and used for solving both the ordinary boundary integral equation as well as the hypersingular boundary integral equation for degenerate boundaries.The newly developed numerical method is validated against analytical methods using the matched eigenfunction expansion method for the special case of two vertical barriers or the inclined angle to the vertical being zero.The influence of the length of the two damping layers has been discussed.Moreover,these findings are also validated against previous results for several cases.After validation,the numerical results for the reflection coefficient,transmission coefficient and dissipation coefficient are obtained by varying the inclination angle and porosity-effect parameter.The effects of both the inclination angle and the porosity on the amplitudes of wave forces acting on both the front and rear barriers are also investigated.It is found that the effect of the inclination angle mainly shifts the location of the extremal values of the reflection and the transmission coefficients.Additionally,a moderate value of the porosity-parameter is quite effective at dissipating wave energy and mitigating the wave loads on dual barriers.

INVARIANTS-PRESERVING DU FORT-FRANKEL SCHEMES AND THEIR ANALYSES FOR NONLINEAR SCHRÖDINGER EQUATIONS WITH WAVE OPERATOR

Du Fort-Frankel finite difference method(FDM)was firstly proposed for linear diffusion equations with periodic boundary conditions by Du Fort and Frankel in 1953.It is an explicit and unconditionally von Neumann stable scheme.However,there has been no research work on numerical solutions of nonlinear Schrödinger equations with wave operator by using Du Fort-Frankel-type finite difference methods(FDMs).In this study,a class of invariants-preserving Du Fort-Frankel-type FDMs are firstly proposed for one-dimensional(1D)and two-dimensional(2D)nonlinear Schrödinger equations with wave operator.By using the discrete energy method,it is shown that their solutions possess the discrete energy and mass conservative laws,and conditionally converge to exact solutions with an order of for ofο(T^(2)+h_(x)^(2)+(T/h_(x))^(2))1D problem and an order ofο(T^(2)+h_(x)^(2)+h_(Y)^(2)+(T/h_(X))^(2)+(T/h_(y))^(2))for 2D problem in H1-norm.Here,τdenotes time-step size,while,hx and hy represent spatial meshsizes in x-and y-directions,respectively.Then,by introducing a stabilized term,a type of stabilized invariants-preserving Du Fort-Frankel-type FDMs are devised.They not only preserve the discrete energies and masses,but also own much better stability than original schemes.Finally,numerical results demonstrate the theoretical analyses.
