The virulence regulator AbsR in avian pathogenic Escherichia coli has pleiotropic effects on bacterial physiology

Avian pathogenic Escherichia coli(APEC)belonging to extraintestinal pathogenic E.coli(ExPEC)can cause severe infections in extraintestinal tissues in birds and humans,such as the lungs and blood.MprA(microcin production regulation,locus A,herein renamed AbsR,a blood survival regulator),a member of the MarR(multiple antibiotic resistance regulator)transcriptional regulator family,governs the expression of capsule biosynthetic genes in human ExPEC and represents a promising druggable target for antimicrobials.However,a deep understanding of the AbsR regulatory mechanism as well as its regulon is lacking.In this study,we present a systems-level analysis of the APEC AbsR regulon using ChIP-Seq(chromatin immunoprecipitation sequencing)and RNA-Seq(RNA sequencing)methods.We found that AbsR directly regulates 99 genes and indirectly regulates 667 genes.Furthermore,we showed that:1)AbsR contributes to antiphagocytotic effects by macrophages and virulence in a mouse model for systemic infection by directly activating the capsular gene cluster;2)AbsR positively impacts biofilm formation via direct regulation of the T2SS(type II secretion system)but plays a marginal role in virulence;and 3)AbsR directly upregulates the acid tolerance signaling system EvgAS to withstand acid stress but is dispensable in ExPEC virulence.Finally,our data indicate that the role of AbsR in virulence gene regulation is relatively conserved in ExPEC strains.Altogether,this study provides a comprehensive analysis of the AbsR regulon and regulatory mechanism,and our data suggest that AbsR likely influences virulence primarily through the control of capsule production.Interestingly,we found that AbsR severely represses the expression of the type I-F CRISPR(clustered regularly interspaced short palindromic repeats)-Cas(CRISPR associated)systems,which could have implications in CRISPR biology and application.

Predesigned covalent organic framework with sulfur coordination: Anchoring Au nanoparticles for sensitive colorimetric detection of Hg(Ⅱ)

Targeted construction of new covalent organic frameworks(COFs)with specific purposes and rationalities to build colorimetric assay platform for environmental pollutant monitoring have attracted increasing interest.However,it is still challenging due to lack of available coordination sites inside COFs pores and only a slight bonding ability for anchoring metal.In this work,a two-dimensional(2D)COFs(termed as Tz-COF)with high crystallinity,excellent chemical stability,and abundant sulfur coordination in its skeletons was synthesized and used for the confined growth of Au NPs.It was found that the Au NPs showed significant dispersibility for the support of Tz-COF.The proposed Tz-COF@Au NPs possessed outstanding Hg^(2+)-activated peroxidase-like activity benefited from physicochemical properties of gold amalgam and synergistic effect between COFs and Au NPs to oxidize chromogenic substrate.Based on highly efficient activity and distinctive color evolution,the strategy for detecting Hg^(2+)was developed and successfully applied to determine the content of Hg^(2+)in real environmental samples.This work manifests that a potential strategy to establish a colorimetric assay platform for environmental pollutant monitoring based on the targeted manufacturing of novel COFs with specific functions.

We previously showed that hydrogen sulfide(H2S)has a neuroprotective effect in the context of hypoxic ischemic brain injury in neonatal mice.However,the precise mechanism underlying the role of H2S in this situation remains unclear.In this study,we used a neonatal mouse model of hypoxic ischemic brain injury and a lipopolysaccharide-stimulated BV2 cell model and found that treatment with L-cysteine,a H2S precursor,attenuated the cerebral infarction and cerebral atrophy induced by hypoxia and ischemia and increased the expression of miR-9-5p and cystathionineβsynthase(a major H2S synthetase in the brain)in the prefrontal cortex.We also found that an miR-9-5p inhibitor blocked the expression of cystathionineβsynthase in the prefrontal cortex in mice with brain injury caused by hypoxia and ischemia.Furthermore,miR-9-5p overexpression increased cystathionine-β-synthase and H2S expression in the injured prefrontal cortex of mice with hypoxic ischemic brain injury.L-cysteine decreased the expression of CXCL11,an miR-9-5p target gene,in the prefrontal cortex of the mouse model and in lipopolysaccharide-stimulated BV-2 cells and increased the levels of proinflammatory cytokines BNIP3,FSTL1,SOCS2 and SOCS5,while treatment with an miR-9-5p inhibitor reversed these changes.These findings suggest that H2S can reduce neuroinflammation in a neonatal mouse model of hypoxic ischemic brain injury through regulating the miR-9-5p/CXCL11 axis and restoringβ-synthase expression,thereby playing a role in reducing neuroinflammation in hypoxic ischemic brain injury.

Three-dimensional(3D)bioprinting,specifically direct ink writing(DIW)capable of printing biologically active substances such as growth factors or drugs under low-temperature conditions,is an emerging di-rection in bone tissue engineering.However,limited by the bio-ink mobility and the poor resolution of this printing technology,the lateral pores of current crisscross-stacked scaffolds printed through DIW tend to clog and are inimical to bone growth.Therefore,it is critical to develop DIW printed biologi-cal scaffold structure with high mechanical strength,porosity,and biocompatibility performance.Herein,patterned polylactic acid(PLA)/polycaprolactone(PCL)/nano-hydroxyapatite(n-HA)based scaffold was printed through DIW technological and rolled-up for properties characterization,cytocompatibility test,and bone repair experiment.The result not only shows that the hexagonal patterned scaffolds are me-chanically strong with porosity,but also demonstrated that the hierarchical pore structure formed during rolled-up has the potential to address the clogging problem and stimulates bone growth and repair.

Could social media reflect acquisitive crime patterns in London?

Embraced within the framework of crime opportunities integrated with Social Disorganization theory and Broken Windows theory,this paper intends to explore the patterns of four types of acquisitive crimes,using social media data,i.e.,Twitter,Foursquare and cross-sectional data acquired through text analysis technique.With Greater London as the study area,models like negative binominal regression(NBR)and geographically weighted regression(GWR)are performed to illustrate the aggregated relationships between acquisitive crimes and crime opportunities at London-wide and sub-regional MSOAs levels respectively.The results work towards to hypotheses that:the tweets sentiment could reflect property-related crime rates positively in light of Broken Windows Theory;more tweets with negative sentiment may incur increases in acquisitive crimes.It contributed to existing studies in(1)providing empirical evidence for integrating these three theories;(2)complementing current research on local discrepancies of acquisitive crimes by utilising both GWR and NBR models;(3)challenging the traditional stereotypes about racial disparities with the finding that ethnic heterogeneity and instrumental crimes have counterintuitive association,especially taking education factor into consideration;(4)implicating some localised acquisitive crime prevention strategies to policy makers in light of the reality that the relationship between local variations and different crime types may vary by place.

Biochemistry and transcriptome analysis reveal condensed tannins alleviate liver injury induced by regulating cholesterol metabolism pathway

Free cholesterol has been considered to be a critical risk factor of nonalcoholic fatty liver disease(NAFLD).It remains unknown whether dietary intake of condensed tannins(CTs)have distinguishable effects to alleviate liver damage caused by a high cholesterol diet.Male C57BL/6 mice were fed a high cholesterol diet for 6 weeks,and given CTs treatment at a dosage of 200 mg/(kg·day)at the same time.The results indicated that compared with mice fed a normal diet,a high cholesterol diet group resulted in significant weight loss,dysregulation of lipid metabolism in blood and liver,and oxidative stress in the liver,but CTs treatment dramatically reversed these negative effects.Hematoxylin and eosin(H&E)staining and frozen section observation manifested that CTs treatment could effectively reduce the deposition of liver cholesterol and tissue necrosis caused by high cholesterol intake.CTs alleviated liver injury mainly by regulating the expression of related genes in cholesterol metabolism pathway and AMPK phosphorylation.Our results confirmed that CTs have remarkable cholesterol lowering and anti-liver injury effects in vivo.

Inhibitory effect of procyanidin B2 and tannin acid on cholesterol esterase and their synergistic effect with orlistat

This study was aimed to analyze the effect of procyanidin B2(PC)and tannin acid(TA)on the activities of cholesterol esterase(CEase)and the inhibitory mechanisms of enzymatic activity.The interaction mechanisms were investigated by enzymatic kinetics,multi-spectroscopy methods,thermodynamics analysis,molecular docking,and dynamic simulations.PC and TA could bind with CEase and inhibit the activity of enzyme in a mixed-competitive manner and non-competitive manner,which was verified by molecular docking simulations and dynamics simulations.Also,PC and TA showed the synergistic inhibition with orlistat.Fluorescence,UVvis and the thermodynamic analysis revealed that the complexes were formed from CEase and inhibitors by noncovalent interaction.As revealed by the circular dichroism results,both PC and TA decreased enzymatic activities by altering the conformations of CEase.The inhibition of PC and TA on CEase might be one mechanism for its cholesterol-lowering effect.

Solid-state zinc-ion capacitors are emerging as promising candidates for large-scale energy storage owing to improved safety,mechanical and thermal stability and easy-to-direct stacking.Hydrogel electrolytes are appealing solid-state electrolytes because of eco-friendliness,high conductivity and intrinsic flexibility.However,the electrolyte/electrode interfacial contact and anti-freezing properties of current hydrogel electrolytes are still challenging for practical applications of zinc-ion capacitors.Here,we report a class of hydrogel electrolytes that couple high interfacial adhesion and anti-freezing performance.The synergy of tough hydrogel matrix and chemical anchorage enables a well-adhered interface between hydrogel electrolyte and electrode.Meanwhile,the cooperative solvation of ZnCl2 and liCl hybrid salts renders the hydrogel electrolyte high ionic conductivity and mechanical elasticity simultaneously at low temperatures.More significantly,the Zn||carbon nanotubes hybrid capacitor based on this hydrogel electrolyte exhibits low-temperature capacitive performance,delivering high-energy density of 39 Wh kg^(-1)at-60°C with capacity retention of 98.7%over 10,000 cycles.With the benefits of the well-adhered electrolyte/electrode interface and the anti-freezing hydrogel electrolyte,the Zn/li hybrid capacitor is able to accommodate dynamic deformations and function well under 1000 tension cycles even at-60°C.This work provides a powerful strategy for enabling stable operation of low-temperature zinc-ion capacitors.

Industry-Oriented Detection Method of PCBA Defects Using Semantic Segmentation Models

Automated optical inspection(AOI)is a significant process in printed circuit board assembly(PCBA)production lines which aims to detect tiny defects in PCBAs.Existing AOI equipment has several deficiencies including low throughput,large computation cost,high latency,and poor flexibility,which limits the efficiency of online PCBA inspection.In this paper,a novel PCBA defect detection method based on a lightweight deep convolution neural network is proposed.In this method,the semantic segmentation model is combined with a rule-based defect recognition algorithm to build up a defect detection frame-work.To improve the performance of the model,extensive real PCBA images are collected from production lines as datasets.Some optimization methods have been applied in the model according to production demand and enable integration in lightweight computing devices.Experiment results show that the production line using our method realizes a throughput more than three times higher than traditional methods.Our method can be integrated into a lightweight inference system and pro-mote the flexibility of AOI.The proposed method builds up a general paradigm and excellent example for model design and optimization oriented towards industrial requirements.

Ecological selection of bacterial taxa with larger genome sizes in response to polycyclic aromatic hydrocarbons stress

Polycyclic aromatic hydrocarbons(PAHs)are ubiquitous priority pollutants that cause great damage to the natural environment and health.Average genome size in a community is critical for shedding light on microbiome's functional response to pollution stress within an environment.Here,microcosms under different concentrations were performed to evaluate the selection of PAHs stress on the average genome size in a community.We found the distinct communities of significantly larger genome size with the increase of PAHs concentration gradients in soils,and consistent trends were discovered in soils at different latitudes.The abundance of Proteobacteria and Deinococcus-Thermus with relatively larger genomes increased along with PAHs stress and well adapted to polluted environments.In contrast,the abundance of Patescibacteria with a highly streamlined and smaller genome decreased,implying complex interactions between environmental selection and functional fitness resulted in bacteria with larger genomes becoming more abundant.Moreover,we confirmed the increased capacity for horizontal transfer of degrading genes between communities by showing an increased connection number per node positively related to the nid A gene along the concentration gradients in the co-occurrence network.Our findings suggest PAHs tend to select bacterial taxa with larger genome sizes,with significant consequences for community stability and potential biodegradation strategies.
