Effects of COL1A1 and SYTL2 on inflammatory cell infiltration and poor extracellular matrix remodeling of the vascular wall in thoracic aortic aneurysm

Background: Thoracic aortic aneurysm (TAA) is a fatal cardiovascular disease, the pathogenesis of which has not yet been clarified. This study aimed to identify and validate the diagnostic markers of TAA to provide a strong theoretical basis for developing new methods to prevent and treat this disease.Methods: Gene expression profiles of the GSE9106, GSE26155, and GSE155468 datasets were acquired from the Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) were identified using the "limma" package in R. Least absolute shrinkage and selection operator (LASSO), support vector machine-recursive feature elimination (SVM-RFE), random forest, and binary logistic regression analyses were used to screen the diagnostic marker genes. Single-sample gene set enrichment analysis (ssGSEA) was used to estimate immune cell infiltration in TAA.Results: A total of 16 DEGs were identified. The enrichment and functional correlation analyses showed that DEGs were mainly associated with inflammatory response pathways and collagen-related diseases. Collagen type I alpha 1 chain (COL1A1) and synaptotagmin like 2 (SYTL2) were identified as diagnostic marker genes with a high diagnostic value for TAA. The expression ofCOL1A1 andSYTL2 was considerably higher in TAA vascular wall tissues than in the corresponding normal tissues, and there were significant differences in the infiltration of immune cells between TAA and normal vascular wall tissues. Additionally,COL1A1 andSYTL2 expression were associated with the infiltration of immune cells in the vascular wall tissue. Single-cell analysis showed thatCOL1A1 in TAA was mainly derived from fibroblasts andSYTL2 mainly from cluster of differentiation (CD)8+ T cells. In addition, single-cell analysis indicated that fibroblasts and CD8+ T cells in TAA were significantly higher than those in normal arterial wall tissue.Conclusions: COL1A1 andSYTL2 may serve as diagnostic marker genes for TAA. Response of Freezing/Thawing Indexes to the Wetting Trend under Warming Climate Conditions over the Qinghai–Tibetan Plateau during 1961–2010:A Numerical Simulation

Since the 1990s,the Qinghai–Tibetan Plateau(QTP)has experienced a strikingly warming and wetter climate that alters the thermal and hydrological properties of frozen ground.A positive correlation between the warming and thermal degradation in permafrost or seasonally frozen ground(SFG)has long been recognized.Still,a predictive relationship between historical wetting under warming climate conditions and frozen ground has not yet been well demonstrated,despite the expectation that it will become even more important because precipitation over the QTP has been projected to increase continuously in the near future.This study investigates the response of the thermal regime to historical wetting in both permafrost and SFG areas and examines their relationships separately using the Community Land Surface Model version 4.5.Results show that wetting before the 1990s across the QTP mainly cooled the permafrost body in the arid and semiarid zones,with significant correlation coefficients of 0.60 and 0.48,respectively.Precipitation increased continually at the rate of 6.16 mm decade–1 in the arid zone after the 1990s but had a contrasting warming effect on permafrost through a significant shortening of the thawing duration within the active layer.However,diminished rainfall in the humid zone after the 1990s also significantly extended the thawing duration of SFG.The relationship between the ground thawing index and precipitation was significantly negatively correlated(−0.75).The dual effects of wetting on the thermal dynamics of the QTP are becoming critical because of the projected increases in future precipitation.

Distribution of bacterial communities across plateau freshwater lake and upslope soils

Microorganisms are involved in a variety of biogeochemical processes in natural environments.The differences between bacterial communities in freshwaters and upslope soils remain unclear. The present study investigated the bacterial distribution in a plateau freshwater lake,Erhai Lake（southwestern China）, and its upslope soils. Illumina MiS eq sequencing illustrated high bacterial diversity in lake sediments and soils. Sediment and soil bacterial communities were mainly composed of Proteobacteria, Acidobacteria, Actinobacteria,Bacteroidetes, Chloroflexi and Planctomycetes. However, a distinctive difference in bacterial community structure was found between soil and sediment ecosystems. Water content, nitrogen and p H affected the distribution of the bacterial community across Erhai Lake and its upslope soils. Moreover, the soil bacterial community might also be shaped by plant types. This work could provide some new insights into plateau aquatic and terrestrial microbial ecology.

Influences of 1-methylcyclopropene-containing papers on the metabolisms of membrane lipids in Anxi persimmons during storage

Objectives:The aim of this work was to analyse the effects of 1-methylcyclopropene(1-MCP)treatment on the metabolisms of membrane lipids in postharvest Anxi persimmons during storage.Materials and methods:Anxi persimmon(Diospyros kaki L.f.cv.Anxi)fruits were treated by paper containing 1-MCP with a concentration of 1.35μl/l.The cellular membrane permeability was analysed by the electric conductivity meter.The activities of lipoxygenase(LOX),phospholipase(PLD)and lipase were determined by spectrophotometry.The component and relative amounts of membrane fatty acids were determined using gas chromatograph(GC).Results:The 1-MCP-treated Anxi persimmons manifested a lower electrolyte leakage rate,lower LOX,PLD and lipase activities,higher levels of unsaturated fatty acids(USFAs),higher ratio of USFAs to saturated fatty acids(SFAs)(U/S),higher index of USFAs(IUFA),but lower levels of SFAs.Conclusions:The degradation and the metabolisms of membrane lipids could be suppressed by 1-MCP treatment,which might be accountable for the delaying softening of postharvest Anxi persimmons during storage.

光响应非均匀噪声(photo response nonuniformity,PRNU)是光学成像传感器成像时引入的一种独特噪声,可有效识别压缩视频的来源。针对现有算法提取压缩视频的PRNU效果并不显著的问题,论文提出了一种改进PRNU提取算法。首先,去除视频编解码的环路滤波器,对视频帧使用双密度双树复小波变换进行分解;然后对高频子带使用基于贝叶斯阈值估计的双变量收缩算法进行估计,再使用自适应加窗维纳滤波进行二次估计,得到噪声残差;最后用基于量化参数值加权的最大似然估计法聚合噪声残差,再与视频帧估计得到PRNU。实验结果表明:该文提出的方法在20 s时WhatsApp视频的识别率为75%。

【目的】明确番石榴果实采后环斑病发生的致病病原菌及其对番石榴果实采后品质的影响,筛选能抑制该病原菌的有效抑制剂。【方法】病原菌分离于发生环斑病的番石榴果皮病健交界处,并对该致病菌进行形态学鉴定、分子鉴定与系统发育树分析。另外,初步研究番石榴环斑病菌的生物学特性、环斑病菌侵染对果实采后品质的影响和评价不同抑制剂(ε-聚赖氨酸、水杨酸和褪黑素)对环斑病菌的体外抑菌效果。【结果】根据环斑病菌的菌丝与分生孢子的形态特征及基于rDNA-ITS、TUB和TEF-1α测序结果构建的系统发育树,将福建番石榴环斑病菌鉴定为棒状新拟盘多毛孢(Neopestalotiopsis clavispora)。葡萄糖和D-果糖、蛋白胨、7、25℃分别作为番石榴N. clavispora菌丝生长的最适碳源、氮源、pH、温度。此外,与未接种N. clavispora番石榴果实相比,接种N. clavispora果实具有较高的病斑直径和细胞膜透性,较低的果实硬度和色调角h值。体外试验表明,适当浓度的ε-聚赖氨酸、水杨酸和褪黑素处理对番石榴N. clavispora的菌丝生长有明显的抑制作用,可作为抑制N. clavispora侵染所致番石榴果实采后环斑病的抗菌剂。【结论】引起福建番石榴果实环斑病的病原菌为棒状新拟盘多毛孢(N. clavispora),且4.000 mg·mL^(-1)ε-聚赖氨酸对病原菌具有很好的抑制效果,可为后续番石榴环斑病的防治研究提供科学依据。

大陈岛礁海域鱼类群落结构特征

基于2020年9月、2020年11月、2021年1月、2021年4月连续4个季度的大陈岛礁海域21个底拖网站位的鱼类样本,对局地尺度下大陈岛礁海域鱼类群落的时空格局进行了分析。结果表明:4个季度共采集鱼类99种,隶属15目48科81属,其中鲈形目占比最大(50.51%);优势鱼类中,龙头鱼(Harpadon nehereus)为全年优势种,六丝钝尾虾虎鱼(Amblychaeturichthys hexanema)是除夏季外其他季节的优势种,镰鲳(Pampus echinogaster)是夏秋季的优势种,而大头银姑鱼(Pennahia macrocephalus)、小黄鱼(Larimichthys polyactis)、灰海鳗(Muraenesox cinereus)和大黄鱼(Larimichthys crocea)等仅在单一季节成为优势种;聚类和排序分析显示,大陈岛礁海域的夏、秋和冬3季鱼类群落可明显分为岛礁近河口群落和开阔海域群落2种类型,而春季鱼类群落则可分为岛礁群落和非岛礁群落2类,且群落种类组成在各季节不同区域间均存在显著差异(P<0.05);冗余分析表明,深度、表层温度、底层盐度和底层温度为影响该海域鱼类群落结构的首要环境因子(P<0.05)。研究发现,大陈岛礁海域的鱼类群落存在局地尺度上的区域差异和季节动态,资源结构以小型低值鱼类为主。

水蜜桃具有特殊的生理特性,其贮藏保鲜技术一直是世界性难题。水蜜桃对低温敏感,低温会导致冷害(CI)发生,引起水蜜桃贮藏期变短。由于水蜜桃出色的颜值,口感,风味,具有相对较高的市场价值和很大的开发潜力。目前研究主要集中在对保鲜、贮运环节的桃的品质提升。保鲜方法主要有低温、冰温、1-甲基环丙烯(1-Methylcyclopropene,1-MCP)等。在加工方面则有果汁、果浆、果冻、果酒、果醋、冻干品、可溶性膳食纤维等方面的研究和应用。但总体而言,水蜜桃的产业化过程包括品种选育、保鲜技术、加工技术、设备配套、新产品开发、标准制定等方面仍存在衔接不足等诸多问题。水蜜桃全产业链发展还处于初级阶段。
