To the Editor:Normal-tension glaucoma(NTG)is a progressive optic neuropathy characterized by irreversible blindness with statistically normal intraocular pressure(IOP)(≤21 mmHg),^([1,2])which makes early-stage diagnosis in the clinical practice difficult.Pathophysiological pathways underlying NTG include vascular dysregulation,toxic metabolite accumulation,and elevated translaminar cribrosa pressure difference(TLCPD).^([3])Although various small animal models have provided evidence of different pathophysiological mechanisms,the complicated clinical phenotype cannot be fully imitated by secondary disease models,and the gap between rodents and humans weakens their translational potential.Thus,the pathological changes associated with NTG remain unclear.

The hematopoietic system composed of hematopoietic stem and progenitor cells(HSPCs)and their differentiated lineages serves as an ideal model to uncover generic principles of cell fate transitions.From gastrulation onwards,there successively emerge primitive hematopoiesis(that produces specialized he-matopoietic cells),pro-definitive hematopoiesis(that produces lineage-restricted progenitor cells),and definitive hematopoiesis(that produces multipotent HSPCs).These nascent lineages develop in several transient hematopoietic sites and finally colonize into lifelong hematopoietic sites.The development and maintenance of hematopoietic lineages are orchestrated by cell-intrinsic gene regulatory networks and cell-extrinsic microenvironmental cues.Owing to the progressive methodology(e.g.,high-throughput lineage tracing and single-cell functional and omics analyses),our understanding of the developmental origin of hematopoietic lineages and functional properties of certain hematopoietic organs has been updated;meanwhile,new paradigms to characterize rare cell types,cell heterogeneity and its causes,and comprehensive regulatory landscapes have been provided.Here,we review the evolving views of HSPC biology during developmental and postnatal hematopoiesis.Moreover,we discuss recent advances in the in vitro induction and expansion of HSPCs,with a focus on the implications for clinical applications.

The frequent detection of pharmaceutical compounds in the environment has led to a growing awareness,which may pose a major threat to the aquatic environment.In this study,photodegradation(direct and indirect photolysis)of two different dissociation states of fluoxetine(FLU)was investigated in water,mainly including the determination of photolytic transition states and products,and the mechanisms of indirect photodegradation with·OH,CO_(3)^(*-)and NO_(3)^(*).The main direct photolysis pathways are defluorination and C–C bond cleavage.In addition,the indirect photodegradation of FLU in water is mainly through the reactions with·OH and NO_(3)^(*),and the photodegradation reaction with CO_(3)^(*-)is relatively difficult to occur in the water environment.Our results provide a theoretical basis for understanding the phototransformation process of FLU in the water environment and assessing its potential risk.

Phosphorus(P)is involved in various biochemical reactions in plant growth,so it is beneficial to plants growing in soils contaminated by metals,including cadmium(Cd).However,few studies have reported on the mechanistic roles of P in mitigating Cd toxicity to ryegrass root,and especially in alleviating the disruption of the mitochondrial function of living cells.In this study,the physiological and biochemical mechanisms associated with ryegrass growth under various Cd and P treatments were investigated using leaching and pot systems.The concentration of Cd in soil leachates showed a significantly positive relationship with redox potential(Pduced by 53.3%and 64.5%in the soil leachate with exogenous P application(20 mg/L and 80 mg/L),respectively.Notably,application of P decreased the Cd concentrations in the symplastic fractions and increased the Cd concentrations in the apoplastic fractions in root tips,which may help to alleviate Cd stress to the protoplast.Moreover,exogenous P was found to play a positive role in mitochondrial function and Ca^(2+)variation in root cells under Cd stress,which provides novel insights into the mechanisms of exogenous P in alleviating plant Cd injury.

Focusing on the low open circuit voltage(V_(OC))and fill factor(FF)in flexible Cu_(2)ZnSn(S,Se)_(4)(CZTSSe)solar cells,indium(In)ions are introduced into the CZTSSe absorbers near Mo foils to modify the back interface and passivate deep level defects in CZTSSe bulk concurrently for improving the performance of flexible device.The results show that In doping effectively inhibits the formation of secondary phase(Cu(S,Se)_(2))and VSndefects.Further studies demonstrate that the barrier height at the back interface is decreased and the deep level defects(Cu_(Sn)defects)in CZTSSe bulk are passivated.Moreover,the carrier concentration is increased and the V_(OC) deficit(V_(OC,def))is decreased significantly due to In doping.Finally,the flexible CZTSSe solar cell with 10.01%power conversion efficiency(PCE)has been obtained.The synergistic strategy of interface modification and bulk defects passivation through In incorporation provides a new thought for the fabrication of efficient flexible kesterite-based solar cells.

Insects show highly complicated adaptive and sophisticated behaviors, includ-ing spatial orientation skills, learning ability, and social interaction. These behaviors arecontrolled by the insect brain, the central part of the nervous system. The tiny insect brainconsists of millions of highly differentiated and interconnected cells forming a complexnetwork. Decades of research has gone into an understanding of which parts of the insectbrain possess particular behaviors, but exactly how they modulate these functional conse-quences needs to be clarified. Detailed description of the brain and behavior is required todecipher the complexity of cell types, as well as their connectivity and function. Single-cell RNA-sequencing (scRNA-seq) has emerged recently as a breakthrough technologyto understand the transcriptome at cellular resolution. With scRNA-seq, it is possible touncover the cellular heterogeneity of brain cells and elucidate their specific functions andstate. In this review, we first review the basic structure of insect brains and the links toinsect behaviors mainly focusing on learning and memory. Then the scRNA applicationson insect brains are introduced by representative studies. Single-cell RNA-seq has allowedresearchers to classify cell subpopulations within different insect brain regions, pinpointsingle-cell developmental trajectories, and identify gene regulatory networks. These devel-opments empower the advances in neuroscience and shed light on the intricate problemsin understanding insect brain functions and behaviors.

Compared with electrical neural networks,optical neural networks(ONNs)have the potentials to break the limit of the bandwidth and reduce the consumption of energy,and therefore draw much attention in recent years.By far,several types of ONNs have been implemented.However,the current ONNs cannot realize the acceleration as powerful as that indicated by the models like quantum neural networks.How to construct and realize an ONN with the quantum speedup is a huge challenge.Here,we propose theoretically and demonstrate experimentally a new type of optical convolutional neural network by introducing the optical correlation.It is called the correlated optical convolutional neural network(COCNN).We show that the COCNN can exhibit“quantum speedup”in the training process.The character is verified from the two aspects.One is the direct illustration of the faster convergence by comparing the loss function curves of the COCNN with that of the traditional convolutional neural network(CNN).Such a result is compatible with the training performance of the recently proposed quantum convolutional neural network(QCNN).The other is the demonstration of the COCNN’s capability to perform the QCNN phase recognition circuit,validating the connection between the COCNN and the QCNN.Furthermore,we take the COCNN analog to the 3-qubit QCNN phase recognition circuit as an example and perform an experiment to show the soundness and the feasibility of it.The results perfectly match the theoretical calculations.Our proposal opens up a new avenue for realizing the ONNs with the quantum speedup,which will benefit the information processing in the era of big data.

Gastric cancer is among the most frequently occurring cancers and a leading cause of cancer-related deaths globally. Because gastric cancer is highly heterogenous and comprised of different subtypes with distinct molecular and clinical characteristics, the management of gastric cancer calls for better-defined, biomarker-guided, molecular-based treatment strategies. MET is a receptor tyrosine kinase mediating important physiologic processes, such as embryogenesis, tissue regeneration, and wound healing. However, mounting evidence suggests that aberrant MET pathway activation contributes to tumour proliferation and metastasis in multiple cancer types, including gastric cancer, and is associated with poor patient outcomes. As such, MET-targeting therapies are being actively developed and promising progress has been demonstrated, especially with MET tyrosine kinase inhibitors. This review aims to briefly introduce the role of MET alterations in gastric cancer and summarize in detail the current progress of MET tyrosine kinase inhibitors in this disease area with a focus on savolitinib, tepotinib, capmatinib, and crizotinib. Building on current knowledge, this review further discusses existing challenges in MET alterations testing, possible resistance mechanisms to MET inhibitors, and future directions of MET-targeting therapies.

State-Based Opacity Verification of Networked Discrete Event Systems Using Labeled Petri Nets

The opaque property plays an important role in the operation of a security-critical system,implying that pre-defined secret information of the system is not able to be inferred through partially observing its behavior.This paper addresses the verification of current-state,initial-state,infinite-step,and K-step opacity of networked discrete event systems modeled by labeled Petri nets,where communication losses and delays are considered.Based on the symbolic technique for the representation of states in Petri nets,an observer and an estimator are designed for the verification of current-state and initial-state opacity,respectively.Then,we propose a structure called an I-observer that is combined with secret states to verify whether a networked discrete event system is infinite-step opaque or K-step opaque.due to the utilization of symbolic approaches for the state-based opacity verification,the computation of the reachability graphs of labeled Petri nets is avoided,which dramatically reduces the computational overheads stemming from networked discrete event systems.

Numerical Simulation of the Settling Flux of Biodeposition in a Bay with Cage Culture Through Similarity Theory and a Simplified Pollution Source

The settling flux of biodeposition affects the environmental quality of cage culture areas and determines their environmental carrying capacity.Simple and effective simulation of the settling flux of biodeposition is extremely important for determining the spatial distribution of biodeposition.Theoretically,biodeposition in cage culture areas without specific emission rules can be simplified as point source pollution.Fluent is a fluid simulation software that can simulate the dispersion of particulate matter simply and efficiently.Based on the simplification of pollution sources and bays,the settling flux of biodeposition can be easily and effectively simulated by Fluent fluid software.In the present work,the feasibility of this method was evaluated by simulation of the settling flux of biodeposition in Maniao Bay,Hainan Province,China,and 20 sampling sites were selected for determining the settling fluxes.At sampling sites P1,P2,P3,P4,P5,Z1,Z2,Z3,Z4,A1,A2,A3,A4,B1,B2,C1,C2,C3 and C4,the measured settling fluxes of biodeposition were 26.02,15.78,10.77,58.16,6.57,72.17,12.37,12.11,106.64,150.96,22.59,11.41,18.03,7.90,19.23,7.06,11.84,5.19 and 2.57 g d^(−1)m^(−2),respectively.The simulated settling fluxes of biodeposition at the corresponding sites were 16.03,23.98,8.87,46.90,4.52,104.77,16.03,8.35,180.83,213.06,39.10,17.47,20.98,9.78,23.25,7.84,15.90,6.06 and 1.65 g d^(−1)m^(−2),respectively.There was a positive correlation between the simulated settling fluxes and measured ones(R=0.94,P=2.22×10^(−9)<0.05),which implies that the spatial differentiation of biodeposition flux was well simulated.Moreover,the posterior difference ratio of the simulation was 0.38,and the small error probability was 0.94,which means that the simulated results reached an acceptable level from the perspective of relative error.Thus,if nonpoint source pollution is simplified to point source pollution and open waters are simplified based on similarity theory,the setting flux of biodeposition in the open waters can be simply and effectively
