Global,regional and national burdens of bipolar disorders in adolescents and young adults:a trend analysis from 1990 to 2019

Background Bipolar disorder is identified as a cause of severe damage to the physical,psychological and social functioning of adolescents and young adults.Aims The aim of this study is to ascertain the trends in the burden of bipolar disorder among individuals aged 10-24 years at global,regional and national levels from 1990 to 2019.Methods The data analysed in this study were from the Global Burden of Diseases 2019.The numbers,rates per 100000 population,average annual percentage changes(AAPCs)of incidence,prevalence and years lived with disability(YLDs)of bipolar disorder are reported at the global,regional and national levels among individuals aged 10-24 years.Global trends by age,sex and Social Development Index(SDI)were further analysed.Results Globally,the incidence of bipolar disorder among adolescents and young adults increased from 79.21 per 100000 population(95%uncertainty interval(Ul):58.13 to 105.15)in 1990 to 84.97 per 100000 population(95%Ul:61.73 to 113.46)in 2019,AAPC 0.24(95%confidence interval(Cl):0.22 to 0.26).In the past three decades,there has been an increase in incidence,prevalence and YLDs in both males and females.The largest increase in incidence between 1990 and 2019 was observed in those aged 20-24 years old from 51.76 per 100000 population(95%Ul:26.81 to 87.20)in 1990 to 58.37 per 100000 population(95%UI:30.39 to 98.55)in 2019;AAPC 0.42(95%Cl:0.38 to 0.47).By the SDI quintile,the largest increase in incidence was observed in the middle SDl;however,the high SDI countries had the highest incidence.Regionally,the largest increase in incidence was observed in southern Latin America.At the national level,the most pronounced increase in the incidence was in Greenland.Conclusions The global increase in incidence among adolescents and young adults between 1990 and 2019 indicates that strategies to improve their mental health still need to be emphasised.

Fine-tuning the activation behaviors of ternary modular cabazitaxel prodrugs for efficient and on-target oral anti-cancer therapy

The disulfide bond plays a crucial role in the design of anti-tumor prodrugs due to its exceptional tumor-specific redox responsiveness. However, premature breaking of disulfide bonds is triggered by small amounts of reducing substances (e.g., ascorbic acid, glutathione, uric acid and tea polyphenols) in the systemic circulation. This may lead to toxicity, particularly in oral prodrugs that require more frequent and high-dose treatments. Fine-tuning the activation kinetics of these prodrugs is a promising prospect for more efficient on-target cancer therapies. In this study, disulfide, steric disulfide, and ester bonds were used to bridge cabazitaxel (CTX) to an intestinal lymph vessel-directed triglyceride (TG) module. Then, synthetic prodrugs were efficiently incorporated into self-nanoemulsifying drug delivery system (corn oil and Maisine CC were used as the oil phase and Cremophor EL as the surfactant). All three prodrugs had excellent gastric stability and intestinal permeability. The oral bioavailability of the disulfide bond-based prodrugs (CTX-(C)S-(C)S-TG and CTX-S-S-TG) was 11.5- and 19.1-fold higher than that of the CTX solution, respectively, demonstrating good oral delivery efficiency. However, the excessive reduction sensitivity of the disulfide bond resulted in lower plasma stability and safety of CTX-S-S-TG than that of CTX-(C)S-(C)S-TG. Moreover, introducing steric hindrance into disulfide bonds could also modulate drug release and cytotoxicity, significantly improving the anti-tumor activity even compared to that of intravenous CTX solution at half dosage while minimizing off-target adverse effects. Our findings provide insights into the design and fine-tuning of different disulfide bond-based linkers, which may help identify oral prodrugs with more potent therapeutic efficacy and safety for cancer therapy.

Gut homeostasis is of importance to host health and imbalance of the gut usually leads to disorders or diseases for both human and animal.Postbiotics have been applied in manipulating of gut health,and utilization of postbiotics threads new lights into the host health.Compared with the application of probiotics,the characteristics such as stability and safety of postbiotics make it a potential alternative to probiotics.Studies have reported the beneficial effects of components derived from postbiotics,mainly through the mechanisms including inhibition of pathogens,strengthen gut barrier,and/or regulation of immunity of the host.In this review,we summarized the characteristics of postbiotics,main compounds of postbiotics,potential mechanisms in gut health,and their application in animal production.

L.reuteri ZJ617 inhibits inflammatory and autophagy signaling pathways in gut-liver axis in piglet induced by lipopolysaccharide

Background:This study investigated the protective effects of L.reuteri ZJ617 on intestinal and liver injury and the underlying mechanisms in modulating inflammatory,autophagy,and apoptosis signaling pathways in a piglet challenged with lipopolysaccharide(LPS).Methods:Duroc×Landrace×Large White piglets were assigned to 3 groups(n=6/group):control(CON)and LPS groups received oral phosphate-buffered saline for 2 weeks before intraperitoneal injection(i.p.)of physiological saline or LPS(25μg/kg body weight),respectively,while the ZJ617+LPS group was orally inoculated with ZJ617 for 2 weeks before i.***.of LPS.Piglets were sacrificed 4 h after LPS injection to determine intestinal integrity,serum biochemical parameters,inflammatory signaling involved in molecular and liver injury pathways.Results:Compared with controls,LPS stimulation significantly increased intestinal phosphorylated-p38 MAPK,phosphorylated-ERK and JNK protein levels and decreased IκBαprotein expression,while serum LPS,TNF-α,and IL-6 concentrations(P<0.05)increased.ZJ617 pretreatment significantly countered the effects induced by LPS alone,with the exception of p-JNK protein levels.Compared with controls,LPS stimulation significantly increased LC3,Atg5,and Beclin-1 protein expression(P<0.05)but decreased ZO-1,claudin-3,and occludin protein expression(P<0.05)and increased serum DAO and D-xylose levels,effects that were all countered by ZJ617 pretreatment.LPS induced significantly higher hepatic LC3,Atg5,Beclin-1,SOD-2,and Bax protein expression(P<0.05)and lower hepatic total bile acid(TBA)levels(P<0.05)compared with controls.ZJ617 pretreatment significantly decreased hepatic Beclin-1,SOD2,and Bax protein expression(P<0.05)and showed a tendency to decrease hepatic TBA(P=0.0743)induced by LPS treatment.Pretreatment of ZJ617 before LPS injection induced the production of 5 significant metabolites in the intestinal contents:capric acid,isoleucine 1TMS,glycerol-1-phosphate byproduct,linoleic acid,alanine-alanine(P<0.05).Conclusions:These results demonstrated

Robust Symmetry Prediction with Multi-Modal Feature Fusion for Partial Shapes

In geometry processing,symmetry research beneﬁts from global geo-metric features of complete shapes,but the shape of an object captured in real-world applications is often incomplete due to the limited sensor resolution,single viewpoint,and occlusion.Different from the existing works predicting symmetry from the complete shape,we propose a learning approach for symmetry predic-tion based on a single RGB-D image.Instead of directly predicting the symmetry from incomplete shapes,our method consists of two modules,i.e.,the multi-mod-al feature fusion module and the detection-by-reconstruction module.Firstly,we build a channel-transformer network(CTN)to extract cross-fusion features from the RGB-D as the multi-modal feature fusion module,which helps us aggregate features from the color and the depth separately.Then,our self-reconstruction net-work based on a 3D variational auto-encoder(3D-VAE)takes the global geo-metric features as input,followed by a prediction symmetry network to detect the symmetry.Our experiments are conducted on three public datasets:ShapeNet,YCB,and ScanNet,we demonstrate that our method can produce reliable and accurate results.

对内径为41~43μm,外径约为188μm、251μm和270μm的锗芯光纤进行激光退火实验,研究CO_(2)激光沿光纤轴向扫描速度对锗芯拉曼峰频率和光传输损耗特性的影响。研究发现,激光扫描速度是决定退火后光纤特性的重要参数。对不同外径的光纤,达到最优退火效果的激光扫描速度不同,188μm、251μm和270μm外径的锗芯光纤分别为10 mm·s^(-1)、14 mm·s^(-1)和16 mm·s^(-1),光传输损耗分别为3.435 dB·cm^(-1)、2.147 dB·cm^(-1)和3.578 dB·cm^(-1)。使用COMSOL软件对退火过程中纤芯表面固定点的温度变化进行了模拟仿真研究,仿真结果显示激光退火过程中温度呈脉冲形变化。相同外径条件下,激光扫描速度提高,温度脉冲的峰值升高、谷值降低、单脉冲持续时间缩短;相同扫描速度条件下,光纤外径减小,温度脉冲峰值提高,谷值降低。

Defect reduction to enhance the mechanical strength of nanocellulose carbon aerogel

Carbon aerogels prepared from renewable nano building blocks are rising-star materials and hold great promise in many fields.However,various defects formed during carbonization at high temperature disfavor the stress transfer and thus the fabrication of flexible carbon aerogel from renewable nano building blocks.Herein,a structural defect-reducing strategy is proposed by altering the pyrolysis route of cellulose nanofiber.Inorganic salt that inhibits the generation of tar volatilization during pyrolysis can prevent the formation of various structural defects.Microstructure with fewer defects can reduce stress concentration and remarkably enhance the compressibility of carbon aerogel,thus increasing the maximum stress retention of carbon aerogel.The carbon aerogel also has high stress sensor sensitivity and excellent temperature coefficient of resistance.The structural defect-reducing strategy will pave a new way to fabricate high-strength carbon materials for various fields.

Unc-51样自噬激活激酶1(ULK1)作为自噬启动的重要调控因子,是肿瘤治疗的关键靶点之一。首先,以已知ULK1抑制剂为基础构建药效团模型,通过药效团模型筛选、分子对接以及分子力学广义波恩表面积(MM/GBSA)结合自由能计算等方法,对含有52万多个类药性小分子的数据库进行虚拟筛选,得到具有较高理论亲和力的化合物。随后,50ns的分子动力学模拟验证了蛋白质-配体复合物结合的稳定性,最后10ns的平均结合自由能的计算研究进一步验证了配体的结合能力。结果表明,6个化合物(F5258-0159、F3407-0428、F0529-1100、F0696-3531、F3222-5280、F6525-5596)具有骨架新颖、分子对接分数和结合自由能数值优异及与ULK1的结合状态稳定等特点,可以作为新型潜在的ULK1抑制剂用于肿瘤治疗的研究,也为新型ULK1抑制剂的设计和研发提供新的研究思路。

神经病理性疼痛仍然缺乏有效的治疗方法,导致病人长期忍受慢性疼痛的折磨、生活质量受到严重影响。富血小板血浆(platelet-rich plasma, PRP)可以释放多种生物活性因子,具备神经修复和缓解疼痛的作用。本文通过对现有的研究和文献进行检索及归纳,发现PRP具有促进创伤愈合和抗炎的特性,不仅有助于修复受损的神经组织,而且能有效缓解多种神经病理性疼痛,为临床治疗提供了新的思路与方法。
