AZD1775 and anti-PD-1 antibody synergistically sensitize hepatoma to radiotherapy

Background:Radiation(IR)-induced DNA damage triggers cell cycle arrest and has a suppressive effect on the tumor microenvironment(TME).Wee1,a cell cycle regulator,can eliminate G2/M arrest by phosphorylating cyclin-dependent kinase 1(CDK1).Meanwhile,programed death-1/programed death ligand-1(PD-1/PDL-1)blockade is closely related to TME.This study aims to investigate the effects and mechanisms of Wee1 inhibitor AZD1775 and anti-PD-1 antibody(anti-PD-1 Ab)on radiosensitization of hepatoma.Methods:The anti-tumor activity of AZD1775 and IR was determined by 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazolium bromide(MTT)assay on human and mouse hepatoma cells HepG2,Hepa1-6,and H22.The anti-hepatoma mechanism of AZD1775 and IR revealed by flow cytometry and Western blot in vitro.A hepatoma subcutaneous xenograft mice model was constructed on Balb/c mice,which were divided into control group,IR group,AZD1775 group,IR+AZD1775 group,IR+anti-PD-1 Ab group,and the IR+AZD1775+anti-PD-1 Ab group.Cytotoxic CD8^(+)T cells in TME were analyzed by flow cytometry.Results:Combining IR with AZD1775 synergistically reduced the viability of hepatoma cells in vitro.AZD1775 exhibited antitumor effects by decreasing CDK1 phosphorylation to reverse the IR-induced G2/M arrest and increasing IR-induced DNA damage.AZD1775 treatment also reduced the proportion of PD-1^(+)/CD8^(+)T cells in the spleen of hepatoma subcutaneous xenograft mice.Further studies revealed that AZD1775 and anti-PD-1 Ab could enhance the radiosensitivity of hepatoma by enhancing the levels of interferonγ(IFNγ)^(+)or Ki67^(+)CD8 T cells and decreasing the levels of CD8^(+)Tregs cells in the tumor and spleen of the hepatoma mice model,indicating that the improvement of TME was manifested by increasing the cytotoxic factor IFNγexpression,enhancing CD8^(+)T cells proliferation,and weakening CD8^(+)T cells depletion.Conclusions:This work suggests that AZD1775 and anti-PD-1 Ab synergistically sensitize hepatoma to radiotherapy by enhancing IR-induced DNA damage and improv

Spiking neural network(SNN),widely known as the third-generation neural network,has been frequently investigated due to its excellent spatiotemporal information processing capability,high biological plausibility,and low energy consumption characteristics.Analogous to the working mechanism of human brain,the SNN system transmits information through the spiking action of neurons.Therefore,artificial neurons are critical building blocks for constructing SNN in hardware.Memristors are drawing growing attention due to low consumption,high speed,and nonlinearity characteristics,which are recently introduced to mimic the functions of biological neurons.Researchers have proposed multifarious memristive materials including organic materials,inorganic materials,or even two-dimensional materials.Taking advantage of the unique electrical behavior of these materials,several neuron models are successfully implemented,such as Hodgkin–Huxley model,leaky integrate-and-fire model and integrate-and-fire model.In this review,the recent reports of artificial neurons based on memristive devices are discussed.In addition,we highlight the models and applications through combining artificial neuronal devices with sensors or other electronic devices.Finally,the future challenges and outlooks of memristor-based artificial neurons are discussed,and the development of hardware implementation of brain-like intelligence system based on SNN is also prospected.

The integration of sensory information from different modalities,such as touch and vision,is essential for organisms to perform behavioral functions such as decision-making,learning,and memory.Artificial implementation of human multi-sensory perception using electronic supports is of great significance for achieving efficient human–machine interaction.Thanks to their structural and functional similarity with biological synapses,memristors are emerging as promising nanodevices for developing artificial neuromorphic perception.Memristive devices can sense multidimensional signals including light,pressure,and sound.Their in-sensor computing architecture represents an ideal platform for efficient multimodal perception.We review recent progress in multimodal memristive technology and its application to neuromorphic perception of complex stimuli carrying visual,olfactory,auditory,and tactile information.At the device level,the operation model and undergoing mechanism have also been introduced.Finally,we discuss the challenges and prospects associated with this rapidly progressing field of research.

耐久度是制约质子交换膜燃料电池大规模应用的主要障碍之一,性能退化预测技术可以有效提高质子交换膜燃料电池的耐久度。该文提出一种结合小波阈值去噪方法的正则化堆叠长短期记忆网络的性能退化预测方法。通过小波阈值去噪法,获得消除噪声和尖峰后的平滑数据。针对退化数据不确定性和高度非线性导致的特征难以提取问题,引入了正则化堆叠长短期记忆网络模型,该模型通过引入参数优化算法有效地避免了过拟合风险,提高了预测精度和可靠性。为验证该方法的有效性,采用两种不同工况下的质子交换膜燃料电池老化数据进行验证。结果表明,所提方法在稳态工况下的最大误差为0.0163V,误差区间在0.5%以内;动态工况下的最大误差为0.0064 V,误差区间在0.2%以内。

Conversion electrochemistry of copper selenides for robust and energetic aqueous batteries

The development of highly safe and low-cost aqueous batteries is of great significance in the background of carbon neutrality.However,the practical deployment of aqueous batteries has been plagued due to their relatively low capacity and poor cycling stability.Herein,we propose unique conversion electrochemistry of copper selenides for robust and energetic aqueous charge storage.In situ X-ray diffraction and operando Raman techniques reveal a reversible transformation from CuSe to Cu_(2)Se through the intermediates of Cu_(3)Se_(2) and Cu_(1.8)Se.Such a conversion process activates the redox carrier of Cu^(2+)ion and delivers a remarkable rate capability of 285 mAh g^(-1) at 20 A g^(-1) and electrochemical durability up to 30,000 cycles.Furthermore,Cu^(2+)and H+coinsertion chemistry is proposed to facilitate the conversion process.As a proof-of-concept,a hybrid aqueous pouch cell coupling CuSe//Zn is capable of affording maximum energy and power densities of 190 Wh kg^(-1) and 1366W kg^(-1),respectively.

N-methyl-D-aspartate receptor expression in the spinal dorsal horn of a rat model of formalin-induced inflammatory pain following intrathecal injection of butorphanol

Clinical and animal experiments have proved that intrathecal injection of butorphanol has an analgesic effect. However, whether the analgesic effect is associated with activation of the N-methyI-D-aspartate （NMDA） receptor remains unclear. This study presumed that intrathecal injection of butorphanol has an analgesic effect on formalin-induced inflammatory pain in rats, and its analgesic effect is associated with inhibition of NMDA receptors. Concurrently, ketamine was injected into the intrathecal space, which is a non-competitive NMDA receptor antagonist, to determine the analgesic mechanism of butorphanol. The total reflection time in phase 1 and phase 2 of rat hind paws carding action was reduced when the butorphanol dose was increased to 25 μg, or a low dose of butorphanol was combined with ketamine. Intrathecal injection of a high dose of butorphanol alone or a low dose of butorphanol combined with ketamine can remarkably reduce NMDA receptor expression in the Ls spinal dorsal hom of formalin-induced pain rats. The results suggest that intrathecal injection of butorphanol has analgesic effects on formalin-induced inflammatory pain, and remarkably reduces NMDA receptor expression in the rat spinal dorsal horn Ketamine strengthens this analgesic effect. The analgesic mechanism of intrathecal injection of butorphanol is associated with inhibition of NMDA receptor activation.

Sodium-ion batteries are considered one of the most promising candidates for affordable and scalable energy storage as required in smart grid and renewable energy.One of the principal challenges sodium-ion batteries being faced is to search suitable anode materials that can accommodate and store large amounts of Nat ions reversibly and sustainably at reasonable galvanostatic rates.Molybdenum-based materials such as oxides and sulfides might meet this challenge as they afford a capacity much greater than those of the carbo-naceous materials and exhibit rich Na-reaction chemistry.However,these materials are facing several technical issues,such as multiple phase transfor-mation,particle pulverization as the result of volume swelling,and low surface activity during sodiation/desodiation.To tackle these issues,materials design and engineering are of indispensability.In this brief review,we present a state-of-the-art overview of the research progress of molybdenum-based materials for sodium storage,and highlight materials engineering strategies that are capable of addressing the mentioned challenges.We also offer valuable insights into their further development direction and discuss their potentiality in practical batteries.

Differences and similarities in radial growth of Betula species to climate change

Betula platyphylla and Betula costata are important species in mixed broadleaved-Korean pine(Pinus koraiensis)forests.However,the specific ways in which their growth is affected by warm temperatures and drought remain unclear.To address this issue,60 and 62 tree-ring cores of B.platyphylla and B.costata were collected in yichun,China.Using dendrochronological methods,the response and adaptation of these species to climate change were examined.A“hysteresis effect”was found in the rings of both species,linked to May–September moisture conditions of the previous year.Radial growth of B.costata was positively correlated with the standardized precipitation-evapotranspiration index(SPEI),the precipitation from September to October of the previous year,and the relative humidity in October of the previous year.Growth of B.costata is primarily restricted by moisture conditions from September to October.In contrast,B.platyphylla growth is mainly limited by minimum temperatures in May–June of both the previous and current years.After droughts,B.platyphylla had a faster recovery rate compared to B.costata.In the context of rising temperatures since 1980,the correlation between B.platyphylla growth and monthly SPEI became positive and strengthened over time,while the growth of B.costata showed no conspicuous change.Our findings suggest that the growth of B.platyphylla is already affected by warming temperatures,whereas B.costata may become limited if warming continues or intensifies.Climate change could disrupt the succession of these species,possibly accelerating the succession of pioneer species.The results of this research are of great significance for understanding how the growth changes of birch species under warming and drying conditions,and contribute to understanding the structural adaptation of mixed broadleaved-Korean pine(Pinus koraiensis)forests under climate change.

文章旨在探讨营造老旧小区公共空间高品质的光照场景与环境风貌的更新设计思路与应用策略。通过以天津本溪楼小区为例,运用问卷调研、访谈观察、需求卡片、行为注记等方法,剖析其公共空间的光环境资源分布、居民活动类型、行为习惯特征以及光照需求程度间的动态联系,提出遵循光照优先性、空间整体性、边界共享性、尺度适用性的设计原则。针对区内复合型街道、灵活型边界、开放型活动空间类型分别采取了功能融合与光照适配、场地互借与需求过渡、场景聚焦与群体共享的应用策略,有效提升了区内公共空间的光照资源的利用率与空间、设施功能适配的精准度,可使居民在日常活动中获得良好的光环境体验。
