Activin receptor-like kinase 1(ALK1)is a transmembrane serine/threonine receptor kinase of the transforming growth factor beta(TGFβ)receptor ***1 is specifically expressed in vascular endothelial cells,and its dynamic changes are closely related to the proliferation of endothelial cells,the recruitment of pericytes to blood vessels,and functional differentiation during embryonic vascular *** pathophysiology of many cerebrovascular diseases is today understood as a disorder of endothelial cell function and an imbalance in the proportion of vascular ***,mutations in ALK1 and its co-receptor endoglin are major genetic risk factors for vascular arteriovenous *** studies have shown that ALK1 is closely related to the development of cerebral aneurysms,arteriovenous malformations,and cerebral *** this review,we describe the various roles of ALK1 in the regulation of angiogenesis and in the maintenance of cerebral vascular homeostasis,and we discuss its relationship to functional dysregulation in cerebrovascular *** review should provide new perspectives for basic research on cerebrovascular diseases and offer more effective targets and strategies for clinical diagnosis,treatment,and prevention.

Treatment of chronic heart failure in the 21st century: A new era of biomedical engineering has come

Chronic heart failure (CHF) is a challenging burden on public health. Therapeutic strategies for CHF have developed rapidly in the past decades from conventional medical therapy, which mainly includes administration of angiotensin-converting enzyme inhibitors, angiotensin receptor blockers, beta-blockers, and aldosterone antagonists, to biomedical engineering methods, which include interventional engineering, such as percutaneous balloon mitral valvotomy, percutaneous coronary intervention, catheter ablation, biventricular pacing or cardiac resynchronization therapy (CRT) and CRT-defibrillator use, and implantable cardioverter defibrillator use;mechanical engineering, such as left ventricular assistant device use, internal artery balloon counteq^ulsation, cardiac support device use, and total artificial heart implantation;surgical engineering, such as coronary artery bypass graft, valve replacement or repair of rheumatic or congenital heart diseases, and heart transplantation (HT);regenerate engineering, which includes gene therapy, stem cell transplantation, and tissue engineering;and rehabilitating engineering, which includes exercise training, low-salt diet, nursing, psychological interventions, health education, and external counterpulsation/enhanced external counterpulsation in the outpatient department. These biomedical engineering therapies have greatly improved the symptoms of CHF and life expectancy. To date, pharmacotherapy, which is based on evidence-based medicine, large-scale, multi-center, randomized controlled clinical trials, is still a major treatment option for CHF;the current interventional and mechanical device engineering treatment for advanced CHF is not enough owing to its individual status. In place of HT or the use of a total artificial heart, stem cell technology and gene therapy in regenerate engineering for CHF are very promising. As we all known,renin-angiotensin system(RAS) is a common risk factor and involved in several cardiovasular diseases,such as hypertension,chronic heart failure,AS,coronary heart disease(AMI and post-MI remodeling),cardiovasular hypertrophy and fibrosis,type I or type II diabetes mellitus(DM) and nephropathy,and OOH syndrome we named firstly,and other related diseases includes chronic renal disease,non-DM renal disease,IgA renal disease,etc.,and cardiovascular events and cerebrovascular ***,RAS is also a specific target for above cardiovasular *** mechanisms in RAS related cardiovascular diseases involved mainly in transforming growth factor beta 1(TGFbetal), peroxisome proliferator-activated receptor gamma(PPAR-gamma),microalbuminuria,urinary protein, haemodynamic effects and renoprotective role,*** is easy to understand that dual blockade of RAS is now a new strategy for cardiovasular ***,we introduced simply current angiotensin-converting enzyme inhibitor(ACEI),*** and angiotensin II receptor blockers(ARB),***, eprosartan,candesartan(*** evidences of dual blockade are from two aspects,that is to say, animal experiments and clinical ***,we compared the effect of dual blockade of RAS and monotherapy with ACEI on cardiovasular diseases,and both are generally safe and well tolerated.. Arguments focused on the benefial role of dual blockade of RAS to cardiovasular diseases,but we believed that prospects of dual blockade is great and more data on animal experiments and clinical trials are needed and should be ***,to develop novel drugs of dual blockade of RAS for cardiovasular diseases is promising and waits for our work in future.

Summary:Dexmedetomidine(DEX),a potent and highly selective agonist for a2-adrenergic receptors(a2AR),exerts neuroprotective effects by reducing apoptosis through decreased neuronal Ca^2+***,the exact action mechanism of DEX and its effects on oxygen-glucose deprivation-reoxygenation(OGD/R)injury in vitro are *** demonstrate that DEX pretreatment reduced OGD/R injury in PC12 cells,as evidenced by decreased oxidative stress,autophagy,and neuronal ***,DEX pretreatment decreased the expression levels of stromal interaction molecule 1(STIM1)and calcium release-activated calcium channel protein 1(Orail),and reduced the concentration of intracellular calcium *** addition,variations in cytosolic calcium concentration altered apoptosis rate of PC12 cells after exposure to hypoxic conditions,which were modulated through STIM 1/Orail ***,DEX pretreatment decreased the expression levels of Beclin-1 and microtubule-associated protein 1A/1B-light chain 3(LC3),hallmark markers of autophagy,and the formation of *** conclusion,these results suggested that DEX exerts neuroprotective effects against oxidative stress,autophagy,and neuronal apoptosis afier OGD/R injury via modulation of Caf-STIM1/Orai1 *** results offer insights into the molecular mechanisms of DEX in protecting against neuronal ischemia-reperfusion injury.

Novel strategies halt cardiovascular, diabetes, and cancer strips

In this article, we introduce briefly several strategies for preventing atherosclerotic cardiovascular disease and promoting healthcare for non-communicable diseases (NCDs). These novel strategies include four core elements of health-sleep, emotion, exercise, and diet-and consist of SEED intervention (SEEDi) and E(e)SEEDi due to supplementation of the environment as a core element, and hu's healthy lifestyles intervention (HHLi) which originates from E(e)SEED-BasED healthy lifestyles. They are suitable for the early evaluation of risk factors, and play a key role in the prevention and management of human NCDs when combined with the RT-ABCDEF strategy and the Grade 210 prevention, which include obesity-OSA-hypertension syndrome and C-type hypertension, especially in halting cardiovascular, diabetes and cancer (CDC) strips we first discovered. After successful clinical practice, we may expect our novel strategies for controlling these chronic diseases according to the conception of mass prevention and treatment.

The serum anion gap is associated with disease severity and all-cause mortality in coronary artery disease

Objective To evaluate the associations between the serum anion gap （AG） with the severity and prognosis of coronary artery disease （CAD）. Methods We measured serum electrolytes in 18,115 CAD patients indicated by coronary angiography. The serum AG was calculated according to the equation： AG = Na^＋ [（mmol/L）＋ K^＋（mmol/L）] - [Cl^- （mmol/L）＋ HCO3^- （mmol/L）]. Results A total of 4510 （24.9%） participants had their AG levels greater than 16 mmol/L. The serum AG was independently associated with measures of CAD severity, including more severe clinical types of CAD （P 〈 0.001） and worse cardiac function （P = 0.004）. Patients in the 4th quartile of serum AG （≥ 15.92 mmol/L） had a 5.171-fold increased risk of 30 days all-cause death （P 〈 0.001）. This association was robust, even after adjustment for age, sex, evaluated glomerular filtration rate [hazard ratio （HR）： 4.861, 95% confidence interval （CI）： 2.150–10.993, P 〈 0.001], clinical diagnosis, severity of coronary artery stenosis, cardiac function grades, and other confounders （HR： 3.318, 95% CI： 1.76–2.27, P = 0.009）. Conclusion In this large population-based study, our findings reveal a high percentage of increased serum AG in CAD. Higher AG is associated with more severe clinical types of CAD and worse cardiac function. Furthermore, the increased serum AG is an independent, significant, and strong predictor of all-cause mortality. These findings support a role for the serum AG in the risk-stratification of CAD.

The method can effectively deal with nonlinearities, constraints, and noises in the system, optimize the performance metric, and present an upper bound on the stable output of the system.

Forecasting step-like landslide displacement through diverse monitoring frequencies

The precision of landslide displacement prediction is crucial for effective landslide prevention and mitigation ***,the role of surface monitoring frequency in influencing prediction accuracy has been largely *** study examined the effect of varying monitoring frequencies on the accuracy of displacement predictions by using the Baijiabao landslide in the Three Gorges Reservoir Area(TGRA)as a case *** collected surface automatic monitoring data at different intervals,ranging from daily to *** Ensemble Empirical Mode Decomposition(EEMD)algorithm was utilized to dissect the accumulated displacements into periodic and trend components at each monitoring *** fitting was applied to forecast the trend component while the periodic component was predicted with two state-of-the-art neural network models:Long Short-Term Memory(LSTM)and Gated Recurrent Unit(GRU).The predictions from these models were integrated to derive cumulative displacement forecasts,enabling a comparative analysis of prediction accuracy across different monitoring *** results demonstrate that the proposed models achieve high accuracy in landslide displacement forecasting,with optimal performance observed at moderate monitoring ***,the daily mean average error(MAE)decreases sharply with increasing monitoring frequency,reaching a *** findings were corroborated by a parallel analysis of the Bazimen landslide,suggesting that moderate monitoring intervals of approximately 7 to 15 days are most conducive to achieving enhanced prediction accuracy compared to both daily and monthly intervals.
