Gene expression profiling of gastric cancer by microarray combined with laser capture microdissection

AIM: To examine the gene expression profile of gastric cancer (GC) by combination of laser capture microdissection (LCM) and microarray and to correlate the profiling with histological subtypes. METHODS: Using LCM, pure cancer cells were procured from 45 cancerous tissues. After procurement of about 5 000 cells, total RNA was extracted and the quality of RNA was determined before further amplification and hybridization. One microgram of amplified RNA was converted to cDNA and hybridized to cDNA microarray. RESULTS: Among 45 cases, only 21 were qualified for their RNAs. A total of 62 arrays were performed. These included 42 arrays for cancer (21 cases with dyeswab duplication) and 20 arrays for non-tumorous cells (10 cases with dye-swab duplication) with universal reference. Analyzed data showed 504 genes were differentially expressed and could distinguish cancerous and non-cancerous groups with more than 99% accuracy. Of the 504 genes, trefoil factors 1, 2, and 3 were in the list and their expression patterns were consistent with previous reports. Immunohistochemical staining of trefoil factor 1 was also consistent with the array data. Analyses of the tumor group with these 504 genes showed that there were 3 subgroups of GC that did not correspond to any current classification system, including Lauren's classification. CONCLUSION: By using LCM, linear amplification of RNA, and cDNA microarray, we have identified a panel of genes that have the power to discriminate between GC and non-cancer groups. The new molecular classification and the identified novel genes in gastric carcinogenesis deserve further investigations to elucidate their dinicopathological significance.

2009年8月8日莫拉克台风重创南台湾,嘉义阿里山区72 h的累积雨量高达2 500 mm以上,百年历史的阿里山森林铁道因沿线路基严重流失而停驶。其中两处大崩塌路段的复建工程以顺应自然为原则,采取迂回隧道方式避开崩塌区,需克服沿线地质变化及地下水问题,评估结果是以凿岩机工法及钻爆工法进行开挖。两座隧道的西口段崩积层分别长达109.8、99 m,是隧道开挖阶段的关键地段。隧道开挖支撑设计系依据台湾岩体分类与支撑系统(PCCR系统)及新奥工法的理念,沿线采用先进支撑材料包括超前管棚钢管、桁型钢支护、钢纤维喷射混凝土及自钻式锚杆等,除了可确保施工安全外,亦可提高施工进度。目前两隧道均已贯通且变形量控制均符合安全,说明隧道采用先进支撑方式穿越破碎崩积层段的成效良好,其研究结果可为相关工程设计提供参考。

Antralization at the edge of proximal gastric ulcers:Does Helicobacter pylori infection play a role?

AIM: To determine the prevalence of antralization at the edge of proximal gastric ulcers, and the effect ofH. pylori eradication on the mucosal appearances.METHODS: Biopsies were taken from the antrum, body and the ulcer edge of patients with benign proximal gastric ulcers before and one year after treatment. Gastric mucosa was classified as antral, transitional or body type.H. pylori positive patients receivedeither triple therapy, or omeprazole.RESULTS: Patients with index ulcers in the incisura, body or fundus (n=116) were analyzed. Antral-type mucosa was more prevalent at the ulcer edge inH. pylori-positive patients ***-negative patients (93% vs 60%, OR=8.95,95%CI: 2.47-32.4, P=0.001). At one year, there was a significant reduction in the prevalence of antralization (from 93 % to 61%, P=0.004) at the ulcer edge in patients with H. pyloribeing eradicated. However, there was no difference in the prevalence of antralization at the ulcer edge in those with persistent infection.CONCLUSION: H. pylori infection is associated with antralization at the edge of proximal gastric ulcers, which may be reversible in some patients after eradication of the infection.

The objective of this study is to investigate the emission of polychlorinated dibenzo-p-dioxins and dibenzofurans from an industrial boiler fuelled by both woodchip and wastewater sludge and the output distributions of PCDD/F mass among the stack flue gas, bottom residue, fly ash of cyclone collected, and fly ash of bag house. In woodchip, the chlorine content was smaller than 1 mg kg-1, whereas in wastewater sludge, it was 2248 mg kg-1. The mean PCDD/F contents in dry basis in woodchip and wastewater sludge are 0.0718 and 0.831 ng-I-TEQ kg-1, respectively. The total PCDD/F contents in input fuel （90 wt% woodchip and 10 wt% sludge）, bottom residue, fly ash of cyclone, and fly ash of bag house were 0.148, 10.7, 3.51, and 74.1 ng I-TEQ kg-1, respectively. The total PCDD/F-I-TEQ concentration in the stack flue gas was 0.0274 ng I-TEQ Nm-3, and the most dominated PCDD/F congeners were OCDD and 1,2,3,4,6,7,8-HpCDD. The total PCDD/F output mass distribution in the stack flue gas, bottom residue, cyclone collected fly ash, and bag house collected fly ash were 93.3%, 0.736%, 0.148%, and 5.78%, respectively. The above results showed that in the co-combustion of both wood chip and wastewater sludge, the most dominant route for the PCDD/F emission from the industrial boiler is stack flue gas. Although, in this study, the mean PCDD/F concentration in the stack flue gas was only 0.0274 ng I-TEQ Nm-3 , which was far below the regulated standard （0.5 ng I-TEQ Nm-3）, the fact of high chlorine content and high PCDD/F content in the wastewater sludge will significantly elevate the PCDD/F emission from the combustion process. The combustible organic sludge being considered as an alternative fuel is a world trend. However, the emission of toxic compounds like PCDD/Fs still requires our great attention.
