Isolated axillary tumor deposit consistent with primary breast carcinoma:A case report

BACKGROUND We all know that lymph-node metastasis is an important factor for poor clinical outcome in breast cancer prognosis.Tumor deposit refers to a discrete collection of cancer cells that is found in the lymph nodes or other tissues adjacent to the primary tumor site.These tumor deposits are separate from the primary tumor and are often considered as a manifestation of lymph node metastasis.In gastric and colorectal cancer,tumor deposits in the lymph node drainage area have been included as independent prognostic factors.The question arises whether tumor deposits should also be considered as prognostic factors in breast cancer patients.This article aims to provoke some thoughts on this matter through a case study and literature review.CASE SUMMARY A 70-year-old female patient was found to have a right breast lump for over 2 years.On January 3,2023,a core needle biopsy of the right breast lump was performed,and the pathology report indicated invasive carcinoma.Subsequently,on January 17,2023,the patient underwent right breast-conserving surgery,sentinel lymph node biopsy,and right axillary lymph node dissection.The postoperative pathological staging was determined as stage IIB.The patient received chemotherapy,radiotherapy,and endocrine therapy.At present,nearly one year after the surgery,no obvious signs of metastasis have been observed in the follow-up examinations,but the long-term prognosis is still unknown.CONCLUSION There is a need for increased focus on the matter of tumor deposits in the lymph node drainage region,as well as a requirement for further clinical investigation to ascertain the relevance of tumor deposits in the prognosis of individuals with breast carcinoma.

Heat stress induced hepatocyte apoptosis in largemouth bass Micropterus salmoides via IRE1α/TRAF2/ASK1/JNK pathway

Heat stress(HS)has been shown to adversely affect fish livers and can lead to extensive apoptosis.To investigate the relationship between endoplasmic reticulum(ER)stress and HS-induced apoptosis in fish livers,we isolated and cultured primary hepatocytes of largemouth bass,Micropterus salmoides by trypsin method,then established an in-vitro model of liver cells under HS(35℃).The contents of lactic dehydrogenase(LDH)and hydrogen peroxide(H2O2)were determined to evaluate the effects of HS on hepatocyte injury and oxidative stress.RT-qPCR was performed to discover the key genes in unfolded protein response(UPR)pathways involved at different HS duration.ERS inhibitor 4-PBA and IRE1αinhibitor 4μ8C were used to further investigate the effects of HS on IRE1αapoptosis pathway in hepatocytes.Results show that HS led to significant increases in the release of LDH,the content of H2O2,and the expressions of oxidative protein folding genes(ero1αand pdi)under HS,suggesting severe hepatocyte injury and oxidative stress happened in heat-stressed largemouth bass hepatocytes.The continuous activation of IRE1αpathway genes(grp78,grp94,atf6,perk,eif2a,atf4,chop,ire1α,traf2,ask1,jnk1,and jnk2)indicated that HS led significantly to ER stress.In particular,the mRNA expression levels of ER stress-related genes(grp78,grp94,atf6,perk,ire1α,chop,jnk1,and jnk2)in the high temperature(HT)+4-PBA group and the HT+4μ8C group were significantly down-regulated under HS.After 4μ8C treatment,the expression levels of apoptosis-related genes(caspase-2,caspase-3,caspase-6,caspase-7,caspase-8,caspase-9,and caspase-10)and LDH content were significantly decreased,whereas the cell survival rate was significantly increased when given 4-PBA or 4μ8C treatment.These findings demonstrate that HS could induce liver apoptosis of largemouth bass through the IRE1αpathway,which may act as a key switch mediating liver apoptosis of largemouth bass under HS.

A framework of force of information influence and application for C4KISR system

The subversive nature of information war lies not only in the information itself, but also in the circulation and application of information. It has always been a challenge to quantitatively analyze the function and effect of information flow through command, control, communications, computer, kill, intelligence,surveillance, reconnaissance (C4KISR) system. In this work, we propose a framework of force of information influence and the methods for calculating the force of information influence between C4KISR nodes of sensing, intelligence processing,decision making and fire attack. Specifically, the basic concept of force of information influence between nodes in C4KISR system is formally proposed and its mathematical definition is provided. Then, based on the information entropy theory, the model of force of information influence between C4KISR system nodes is constructed. Finally, the simulation experiments have been performed under an air defense and attack scenario. The experimental results show that, with the proposed force of information influence framework, we can effectively evaluate the contribution of information circulation through different C4KISR system nodes to the corresponding tasks. Our framework of force of information influence can also serve as an effective tool for the design and dynamic reconfiguration of C4KISR system architecture.

Originating but free from chromosomal DNA,extrachromosomal circular DNAs(eccDNAs)are organized in circular form and have long been found in unicellular and multicellular eukaryotes.Their biogenesis and function are poorly understood as they are characterized by sequence homology with linear DNA,for which few detection methods are available.Recent advances in high-throughput sequencing technologies have revealed that eccDNAs play crucial roles in tumor formation,evolution,and drug resistance as well as aging,genomic diversity,and other biological processes,bringing it back to the research hotspot.Several mechanisms of eccDNA formation have been proposed,including the breakage-fusion-bridge(BFB)and translocation-deletion-amplification models.Gynecologic tumors and disorders of embryonic and fetal development are major threats to human reproductive health.The roles of eccDNAs in these pathological processes have been partially elucidated since the first discovery of eccDNA in pig sperm and the double minutes in ovarian cancer ascites.The present review summarized the research history,biogenesis,and currently available detection and analytical methods for eccDNAs and clarified their functions in gynecologic tumors and reproduction.We also proposed the application of eccDNAs as drug targets and liquid biopsy markers for prenatal diagnosis and the early detection,prognosis,and treatment of gynecologic tumors.This review lays theoretical foundations for future investigations into the complex regulatory networks of eccDNAs in vital physiological and pathological processes.

Shape optimisation of basin insulator for DC gas insulated switchgear/gas insulated transmission lines based on artificial bee colony algorithm

The surface charge accumulation on the basin insulator is a particular problem in high-voltage direct current(HVDC)gas insulated switchgears(GIS)and gas insulated trans-mission lines(GIL),which restricts the development and application of extra/ultra high-voltage(EHV/UHV)technology.The surface charge distribution and anti-flashover performance are closely related to the insulator shape.To obtain the optimal basin insulator,a shape optimisation model based on artificial bee colony algorithm was established,and the minimisation objective functions based on capacitive(initial)electric field distribution and resistive(steady-state)electric field distribution were respectively constructed.The optimisation results of the two objective functions were calculated,and the electric field and charge distribution on the surface of the insulator before and after optimisation were evaluated and compared using the simulation model.The results indicated that the optimisation based on the capacitive electric field can significantly inhibit the surface charge accumulation,and the optimised insulator can significantly reduce the surface normal electric field and make the distribution of the tangential electric field more uniform,and the maximum tangential electric field can be reduced by 10%;the optimisation based on resistive electric field can achieve similar results,but requires more computational resources.Based on the results,the‘anti-charge’basin insulator structure suitable for the DC field was given.

Numerical study of submerged bending vegetation under unidirectional flow

Submerged vegetation commonly grows and plays a vital role in aquatic ecosystems,but it is also regarded as a barrier to the passing flow.Numerical simulations of flow through and over submerged vegetation were carried out to investigate the effect of vegetation density on flow field.Numerical simulations were computationally set up to replicate flume experiments,in which vegetation was mimicked with flexible plastic strips.The fluid-structure interaction between flow and flexible vegetation was solved by coupling the two modules of the COMSOL packages.Two cases with different vegetation densities were simulated,and the results were successfully validated against the experimental data.The contours of the simulated time-averaged streamwise velocity and Reynolds stress were extracted to highlight the differences in mean and turbulent flow statistics.The turbulence intensity was found to be more sensitive to vegetation density than the time-averaged velocity.The developing length increased with the spacing between plants.The snapshots of the bending vegetation under instantaneous velocity and vorticity revealed that flexible vegetation responded to the effects of eddies in the shear layer by swaying periodically.The first two rows of vegetation suffered stronger approaching flow and were prone to more streamlined postures.In addition,the origin of tip vortices was investigated via the distribution of vorticity.The results reveal the variation of flow properties with bending submerged vegetation and provide useful reference for optimizationofrestorationprojects.

Copper-coated Porous Polyimide as Ultralight and Safe Current Collectors for Advanced LIBs

Metallic copper is widely used as current collector(CC) for graphite anode of lithium-ion batteries(LIBs) due to its high electrical conductivity and electrochemical stability. However, the large volume density of commercial copper foil(~8.9 g·cm^(-3)) limits the increase of energy density of battery. Here, copper-coated porous polyimide(Cu@PPI) was prepared by vacuum evaporation as collector for the graphite anode. The sandwich structure connects the copper metal on both sides of the collector with excellent electrical conductivity. Compared to commercial Cu foil, Cu@PPI has lighter mass(≤3.9 mg for disc of 12 mm diameter versus 9.9 mg of ~10 μm Cu foil) and lower volume density(≤3.3 g·cm^(-3)). In addition, the porous structure allows of better adhesion of reactive substances and electrochemical properties than pure Cu foils. It is estimated that the energy density of Cu@PPI should be much higher than that of Cu foil. This strategy should be applicable for other current collectors.

Differential expression of miRNAs and mRNAs in the livers of largemouth bass Micropterus salmoides under heat stress

Global warming threatens freshwater ecosystems and compromises fish survival.To elucidate the role of miRNAs in the livers of heat stressed largemouth bass,juvenile fish was subject to heat stress under 37°C.Both mRNA-seq and miRNA-seq were conducted on the liver tissues under control and heat stress conditions.Differential gene expression analysis and enrichment analysis were performed on mRNA and miRNA expression profiles.A total of 406 differentially expressed genes(DEGs)were discovered,of which 212 were up-regulated and 194 were down-regulated.Most of the DEGs were significantly implicated in the regulation of“protein processing in endoplasmic reticulum”,“proteasome”,“steroid biosynthesis”,and“ornithine decarboxylase inhibitor activity”pathways.In addition,47 differentially expressed miRNAs(DEMs)were identified in largemouth bass livers under heat stress,including 21 up-regulated and 25 down-regulated miRNAs.A negatively regulated miRNA-mRNA network including 12 miRNAs and 19 mRNAs was constructed with DEMs involved in“protein degradation”,“calcium ion regulation”,“cell apoptosis”,and“lipid metabolism”.Moreover,this study indicated novel-miR-144 activated the IRE1 signaling pathway by targeting txndc5 to induce liver apoptosis in largemouth bass under heat stress.This study revealed the involvement of miRNA regulation in largemouth bass in response to heat stress.

Aptasensing biosynthesized phosphatidylserine with a AuNPs nanozyme-based colorimetric aptasensor

Sensitive monitoring of the target products during the biosynthesis process is crucial,and facile analytical approaches are urgently needed.Herein,phosphatidylserine(PS)was chosen as the model target,a colorimetric aptasensor was developed for the rapid quantitation in biosynthesis samples.A chimeric aptamer was constructed with two homogeneous original PS aptamers.Specific recognition between the chimeric aptamer and PS results in the desorption of aptamer from the surface of the AuNPs nanozyme,and the peroxidase-like enzymatic activity of the AuNPs nanozyme was weakened in a relationship with the different concentrations.The developed aptasensor performed well when applied for analyzing PS in biosynthesis samples.The aptasensor offers good sensitivity and selectivity,under optimal conditions,achieving monitoring and quantitation of PS in the range of 2.5-80.0μmol/L,with a limit of detection at 536.2 nmol/L.Moreover,the aptasensor provides good accuracy,with comparison rates of 98.17%-106.40%,when compared with the HPLC-ELSD.This study provides a good reference for monitoring other biosynthesized products and promoting the development of aptamers and aptasensors in real-world applications.
