牛病毒性腹泻病毒(Bovine viral diarrhea virus, BVDV)是引起犊牛(Bos sp.)病毒性腹泻的主要病原之一，导致犊牛免疫力降低，患病率和死亡率升高。目前对BVDV常用的检测方法存在灵敏度较低、容易出现假阳性等状况。本研究基于微滴式数字PCR(droplet digital PCR, ddPCR)技术，建立BVDV精准检测技术。首先针对BVDV 5'UTR基因保守序列区域设计引物和探针，合成BVDV 5'UTR基因并构建质粒标准品。使用BVDV质粒标准品分别建立实时荧光定量PCR(real-time quantitative PCR, qPCR)和ddPCR检测方法，并优化其反应参数和条件，分析其灵敏度、特异性和重复性，并进行了临床应用。结果显示，本研究建立的qPCR方法中，最佳引物和探针浓度分别为300和500 nmol/L，标准曲线呈良好的线性关系，最低检测下限为1×10～4 copies/μL，具有良好的特异性和重复性。建立的ddPCR方法中，最低检测下限为0.64 copies/μL，比qPCR灵敏性高15 625倍，该方法具有良好的特异性和重复性。临床检测了82份样品，ddPCR检出率为15.58%,qPCR检出率为14.63%。本研究建立的ddPCR方法比qPCR方法更加灵敏、高效、特异且重复性良好，为BVDV鉴别诊断提供了技术支持。

AIM:To investigate the effect of transfected survivin antisense oligonucleotide (ASODN) on proliferation and apoptosis of gastric cancer ceils. METHODS:The authors designed ASODNs targeting different regions of survMn mRNA,including surviving ASODN1,ASODN2 and *** were transfected into gastric cancer cell line SGC 7901,cell growth was detected by MTT *** exposed to the potent oligonucleotide were also examined for apoptosis induction by FCM and fluorescence *** RT-PCR and Western blot examinations were carried for expression of survivin mRNA and protein. RESULTS:ASODN3 caused a statistically significant reduction of cell viability to 60.6% (±2.9%) (P0.05).The cell growth was also significantly inhibited by ASODN3, compared with reversal and scrambled sequence.A significant loss of survivin mRNA was presented in ASODN3 treated ceils and this was not seen in treatment with sense ODN or scramble *** level was significantly decreased 48 h after survivin ASODN trasfected by approximately 2-fold decrease comparedwith untreated ***. ASODN3 did not induce significant apoptosis response until 48 h after transfection (P>0.05). CONCLUSION:ASODN3,which targets translation initiation part,can be identified as a most potent antisense compound. Srvivin ASODN3 may provide a novel approach to therapy of gastric cancer.

We propose a novel face image representation – Local gabor dominant direction pattern(LGDDP)for face recognition. The face image is convolved with the Gabor filters, resulting in multiple response images of different orientations and scales. The response images' each pixel is encoded by the LGDDP descriptor from the pixel's dominant neighboring one or two pixels. The image formed by the LGDDP descriptor is partitioned into multiple regions and the histogram is extracted from each region. All the histograms are concatenated into the spatial *** nearest neighbor classifier and the weighted intersection histogram similarity measure are used for face image classification. The advantage of the proposed LGDDP method lies in the high recognition rate performance and the low computational complexity. Extensive experiments are performed on FERET face image database and the experimental results verify the LGDDP's effectiveness by comparing LGDDP with other well-known published face recognition methods.

In this paper, we extend the classical compound binomial risk model to the case where the premium income process is based on a Poisson process, and is no longer a linear function. For this more realistic risk model, Lundberg type limiting results for the finite time ruin probabilities are derived. Asymptotic behavior of the tail probabilities of the claim surplus process is also investigated.

A comprehensive research on wear resistance of GH4169 superalloy in longitudinal-torsional ultrasonic vibration side milling with tool wear and surface quality

To improve the service and machining performance of the workpiece, the tool wear mechanisms, surface machining quality, and wear resistance in conventional side milling(CSM)and longitudinal-torsional ultrasonic vibration side milling(LTUVSM) of GH4169 superalloy at different cutting lengths are investigated systematically. Tool wear mechanisms are revealed and the correlation between machined surface quality with tool wear is analyzed correspondingly. Tool wear patterns mainly include adhesive wear, diffusion wear, abrasive wear, and chipping *** surface quality is achieved in LTUVSM due to a maximum reduction of flank wear bandwidth and wear rate by 71.9% and 71.5%, respectively, compared to CSM. The friction coefficient,initial wear stage time, and wear volume of dry sliding wear were measured to evaluate the workpiece wear resistance. The maximum reductions in friction coefficient and wear volume in LTUVSM are 18.2% and 15.8% compared to CSM. The regular ultrasonic vibration textures suppress the friction and the growth of contact nodes in the contact area, decreasing the degree of surface wear, which is demonstrated by a 38.8% increase in initial wear stage time compared to *** conclusion, the workpiece in LTUVSM exhibits higher wear resistance because of the improvement of tool wear and the guarantee of surface quality.

Central Asia(CA)is one of the most drought-prone regions in the world with complex climate regimes,it is extremely vulnerable to water *** studies on drought in CA,as a whole,have been carried out,whereas there is a lack of studies on the drying and wetting trends of different climatic zones within *** this study,CA was divided into three different climatic zones based on the Koppen climate classification method,precipitation climatology,and aridity *** were the temperate continental(Df),dry arid desert(BW),and Mediterranean continental(Ds)climatic *** regional drying and wetting trends during the years 1961—2015 were investigated using the monthly gridded Standardized Precipitation Evapotranspiration Index(SPEI).The empirical orthogonal function(EOF)was applied to analyze spatial and temporal variation *** mode 1(EOF1)analysis found increasingly wet conditions throughout CA over the duration of the study,and EOF mode 2(EOF2)analysis found a contrast between northern and southern CA:as Df became drier and BW and Ds became *** mode 3(EOF3)analysis found a western and eastern inverse phase *** SPEI displayed a decreasing trend from 1961 to 1974 for CA as a whole,before increasing from 1975 to 2015,with a particularly significant increase over the first seven years of that *** a regional scale,the BW and Ds zones experienced a wetting trend due to increased precipitation during 1961—2015,but the Df zone experienced a drying trend due to reduced evapotranspiration and an increasing temperature,particularly from 1961 to ***,the spatio-temporal patterns in dryness and wetness across CA can be categorized according to climatic regions.

Stabilized effects of L-S cement-mixed batter pile composite foundation for existed warm frozen soil subgrade

In permafrost regions with warm frozen soil,subgrade thaw-collapse phenomenon commonly occurs,facing thaw collapse problems of the existed frozen soil subgrade,thus it is difficult to use traditional methods such as active cooling and passive protection technology to stabilize the existed warm frozen soil *** study derives a novel stabilizer method,a long-short(L-S)cement-mixed batter pile composite foundation to stabilize the existed warm frozen soil *** solve the thawcollapse problems in warm frozen soil subgrade,high water content and large compressibility characteristics were compared between soft soil and warm frozen *** analysis of heat conduction and numerical simulation of finite element model were used to study the freeze–thaw process and evaluate the stabilized effects of the L-S cement-mixed batter piles on the warm frozen soil foundation of the Qinghai-Tibet ***,the thaw process and mechanical properties of foundation and piles were analyzed by introducing the hydration heat factor in the thermodynamic control *** results indicate that the thawing displacement of the existed warm frozen soil subgrade was reduced owing to the“support”and“grasp”effects of the L-S cement-mixed batter piles on the surrounding *** composite ground formed by strengthening the warm frozen ground with batter piles could considerably improve the bearing capacity of the existed warm frozen ground,effectively restrain the deformation of the upper embankment,and improve the strength of the *** analysis can provide method for the construction design of cement mixing batter pile foundation in cold regions.
