Treatment of condylar osteophyte in temporomandibular joint osteoarthritis with muscle balance occlusal splint and long-term follow-up:A case report

BACKGROUND Condylar osteophytes,a remodeling form of temporomandibular joint osteoarthritis(TMJ OA),mainly manifest as marginal angular outgrowths of the condyle.Previous researchers have advocated surgical removal of condylar osteophytes.Reports on the effect of occlusal splint on TMJ OA patients’joints have mostly focused on treatment with this splint,which can reduce the absorption of the affected condyle and promote repair and regeneration.However,the effect of the splint on the dissolution of condylar osteophytes has not yet been reported.CASE SUMMARY A 68-year-old female patient suffered from occlusal discomfort with left facial pain for 2 years.Cone beam computed tomography showed a rare osteophyte on top of her left condyle.She was finally diagnosed with TMJ OA.The patient refused surgical treatment and received conservative treatment with a muscle balance occlusal splint.The pain experienced by the patient on the left side of her face was relieved,and her chewing ability recovered after treatment.The osteophyte dissolved,and the condylar cortex remained stable during long-term follow-up observations.CONCLUSION The muscle balance occlusal splint could be a noninvasive means of treating condylar osteophytes in TMJ OA patients.

Yes-associated protein promotes endothelial-tomesenchymal transition of endothelial cells in choroidal neovascularization fibrosis

AIM:To reveal whether and how Yes-associated protein(YAP)promotes the occurrence of subretinal fibrosis in agerelated macular degeneration(AMD).METHODS:Cobalt chloride(Co Cl2)was used in primary human umbilical vein endothelial cells(HUVECs)to induce hypoxia in vitro.Eight-week-old male C57 BL/6 J mice weighing 19-25 g were used for a choroidal neovascularization(CNV)model induced by laser photocoagulation in vivo.Expression levels of YAP,phosphorylated YAP,mesenchymal markers[αsmooth muscle actin(α-SMA),vimentin,and Snail],and endothelial cell markers(CD31 and zonula occludens 1)were measured by Western blotting,quantitative real-time PCR,and immunofluorescence microscopy.Small molecules YC-1(Lificiguat,a specific inhibitor of hypoxia-inducible factor 1α),CA3(CIL56,an inhibitor of YAP),and XMU-MP-1(an inhibitor of Hippo kinase MST1/2,which activates YAP)were used to explore the underlying mechanism.RESULTS:Co Cl2 increased expression of mesenchymal markers,decreased expression of endothelial cell markers,and enhanced the ability of primary HUVECs to proliferate and migrate.YC-1 suppressed hypoxia-induced endothelialto-mesenchymal transition(End MT).Moreover,hypoxia promoted total expression,inhibited phosphorylation,and enhanced the transcriptional activity of YAP.XMU-MP-1 enhanced hypoxia-induced End MT,whereas CA3 elicited the opposite effect.Expression of YAP,α-SMA,and vimentin were upregulated in the laser-induced CNV model.However,silencing of YAP by vitreous injection of small interfering RNA targeting YAP could reverse these changes.CONCLUSION:The findings reveal a critical role of the hypoxia-inducible factor-1α(HIF-1α)/YAP signaling axis in End MT and provide a new therapeutic target for treatment of subretinal fibrosis in AMD.

Contrast visual acuity after multifocal intraocular lens implantation: aspheric versus spherical design

AIM: To evaluate contrast visual acuity(CVA) after implantation of an aspheric apodized diffractive intraocular lens(IOL) or a spherical apodized diffractive IOL in cataract surgery. ·METHODS: This prospective randomized controlled study with a 12-month follow-up compared the results of cataract surgery with implantation of an aspheric AcrySof ReSTOR SN6AD3 IOL(30 eyes) and a spherical AcrySof ReSTOR SN60D3 IOL(30 eyes). CVA with best distance correction was measured at 4 contrast levels(100%,25%,10% and 5%) under 3 levels of chart luminance [250,85 and 25 candelas per square meter(cd/m2)] using a multi-functional visual acuity tester(MFVA-100). ·RESULTS: At 12 months after surgery,there were no statistically significant differences in 100% CVA and 25% CVA under 250cd/m2(P100% =0.875 and P25% =0.057) and 85cd/m2(P100% =0.198 and P25% =0.193) between the aspheric group and the spherical group. However,the 10% CVA and 5% CVA were significant better in aspheric group than spherical group under 250cd/m2(P10% =0.042 and P5% = 0.007) and 85cd/m2(P10% =0.002 and P5%=0.039). Under the luminance level of 25cd/m 2,no significant differences was found in the 100% CVA between the 2 group(P100% = 0.245),while aspheric group had better visual acuity in the remaining 3 contracts(P25% =0.023,P10% =0.026 and P5% = 0.002,respectively). ·CONCULSION: The aspheric AcrySof ReSTOR SN6AD3 IOL provided patients with better low-contrast visual acuity than the spherical AcrySof ReSTOR SN60D3 IOL.

余甘子提取物调控LINC01772/miR-153对肺癌细胞A549生物学行为的影响

目的:探讨余甘子提取物对肺癌细胞A549增殖、迁移和侵袭的影响及机制。方法:体外培养A549细胞,分为对照组、不同剂量(低、中、高剂量)余甘子提取物组、si-NC组、si-LINC01772组、高剂量余甘子提取物+pcDNA组和高剂量余甘子提取物+pcDNA-LINC01772组,细胞计数试剂盒(CCK-8)法和克隆形成实验检测细胞增殖,划痕实验检测细胞迁移,嵌入式细胞共培养法(Transwell)检测细胞侵袭,免疫印迹法(Western Blot)检测细胞中上皮型钙黏蛋白(E-cadherin)和神经型钙黏蛋白(N-cadherin)蛋白表达水平,实时荧光定量PCR(RT-qPCR)检测LINC01772和miR-153表达水平。双荧光素酶报告基因实验验证LINC01772和miR-153调控关系。结果:与对照组相比,不同剂量余甘子提取物组A549细胞中LINC01772表达降低,且光密度值(OD值)、克隆形成数、迁移以及侵袭细胞数减少(P<0.05),而miR-153含量与E-cadherin蛋白表达升高(P<0.05),且呈剂量依赖性(P<0.05)。LINC01772在A549细胞中负调控miR-153表达。与si-NC组相比,si-LINC01772组A549细胞增殖,侵袭及迁移能力受到抑制(P<0.05)。与高剂量余甘子提取物+pcDNA组相比,高剂量余甘子提取物+pcDNA-LINC01772组A549细胞增殖,侵袭及迁移能力增强(P<0.05)。结论:余甘子提取物可能通过调控LINC01772/miR-153轴抑制肺癌细胞A549增殖、迁移和侵袭,其可能通过下调LINC01772进而上调miR-153表达发挥作用,具有开发为治疗肺癌药物的潜在价值。

Towards accurate mapping of loess waterworn gully by integrating google earth imagery and DEM using deep learning

Accurate mapping of loess waterworn gully(LWG)is essential to further study gully erosion and geomorphological evolution for the Chinese Loess Plateau(CLP).Due to the vertical joint and collapsibility of loess,LWGs have the characteristics of zigzag and unique slope abruptness under synthetic action of hydraulic force and gravity.This forces existing LWG mapping methods to either focus on the improvement of mapping accuracy or center on the increase of mapping efficiency.However,simultaneously achieving accurate and efficient mapping of LWG is still in its infancy under complex topographic conditions.Here,we proposed a method that innovatively integrates the loess slope abruptness feature into an improved deep learning semantic segmentation framework for LWG mapping using 0.6 m Google imagery and 5 m DEM data.We selected four study areas representing typical loess landforms to test the performance of our method.The proposed method can achieve satisfactory mapping results,with the F1 score,mean Intersection-over-Union(mIoU),and overall accuracy of 90.5%,85.3%,and 92.3%,respectively.In addition,the proposed model also showed significant accuracy improvement by inputting additional topographic information(especially the slope of slope).Compared with existing algorithms(Random forests,original DeepLabV3+,and Unet),the proposed approach in this study achieved a better accuracy-efficiency trade-off.Overall,the method can ensure high accuracy and efficiency of the LWG mapping for different loess landform types and can be extended to study various loess gully mapping and water and soil conservation.

BACKGROUND Liver injury is common in severe acute pancreatitis(SAP).Excessive autophagy often leads to an imbalance of homeostasis in hepatocytes,which induces lipid peroxidation and mitochondrial iron deposition and ultimately leads to ferroptosis.Our previous study found that milk fat globule epidermal growth factor 8(MFG-E8)alleviates acinar cell damage during SAP via binding toαvβ3/5 integrins.MFG-E8 also seems to mitigate pancreatic fibrosis via inhibiting chaperone-mediated autophagy.AIM To speculate whether MFG-E8 could also alleviate SAP induced liver injury by restoring the abnormal autophagy flux.METHODS SAP was induced in mice by 2 hly intraperitoneal injections of 4.0 g/kg L-arginine or 7 hly injections of 50μg/kg cerulein plus lipopolysaccharide.mfge8-knockout mice were used to study the effect of MFG-E8 deficiency on SAPinduced liver injury.Cilengitide,a specificαvβ3/5 integrin inhibitor,was used to investigate the possible mechanism of MFG-E8.RESULTS The results showed that MFG-E8 deficiency aggravated SAP-induced liver injury in mice,enhanced autophagy flux in hepatocyte,and worsened the degree of ferroptosis.Exogenous MFG-E8 reduced SAP-induced liver injury in a dose-dependent manner.Mechanistically,MFG-E8 mitigated excessive autophagy and inhibited ferroptosis in liver cells.Cilengitide abolished MFG-E8’s beneficial effects in SAP-induced liver injury.CONCLUSION MFG-E8 acts as an endogenous protective mediator in SAP-induced liver injury.MFG-E8 alleviates the excessive autophagy and inhibits ferroptosis in hepatocytes by binding to integrinαVβ3/5.

Epilepsy can be defined as a dysfunction of the brain network,and each type of epilepsy involves different brain-network changes that are implicated diffe rently in the control and propagation of interictal or ictal discharges.Gaining more detailed information on brain network alterations can help us to further understand the mechanisms of epilepsy and pave the way for brain network-based precise therapeutic approaches in clinical practice.An increasing number of advanced neuroimaging techniques and electrophysiological techniques such as diffusion tensor imaging-based fiber tra ctography,diffusion kurtosis imaging-based fiber tractography,fiber ball imagingbased tra ctography,electroencephalography,functional magnetic resonance imaging,magnetoencephalography,positron emission tomography,molecular imaging,and functional ultrasound imaging have been extensively used to delineate epileptic networks.In this review,we summarize the relevant neuroimaging and neuroelectrophysiological techniques for assessing structural and functional brain networks in patients with epilepsy,and extensively analyze the imaging mechanisms,advantages,limitations,and clinical application ranges of each technique.A greater focus on emerging advanced technologies,new data analysis software,a combination of multiple techniques,and the construction of personalized virtual epilepsy models can provide a theoretical basis to better understand the brain network mechanisms of epilepsy and make surgical decisions.

AWells-Riley/based COVID-19 infectiousrisk assessment model combining both short rangeandroom scale effects

There is growing evidence of the high transmission potential of COVID-19 through virus-laden aerosols.Because aerosols are inhaled in various concentrations,an overall assessment of transmission risks at different indoor scales is crucial.However,a comprehensive risk assessment method that evaluates the direct link between short-range and room-scale zones is stl lacking.In this paper,a risk assessment model combining both short-range and room-scale effects is developed to obtain effective reproduction number in confined indoor environments,called Turbulent Jet Wells Riley(TJWR)model.Combined with the viral load data and aerosol generation data of different human respiratory activities,the Monte Carlo simulation method is applied to calculate the quanta emission rate,which further provides the input parameters of the TJWR model.Three known outbreaks(Hangzhou banquet hall X,Guangzhou restaurant Y,and Hong Kong restaurant Z,China)are chosen to validate the TJWR model.Results show that the TJWR model is more efficient than the original Wells-Riley model.The average relative error of the TJWR model ranges between 9%and 44%,while for the Wells-Riley model,it ranges between 57%and 78%.The TJWR model also proves that infection risk assessments using the well-mixed assumption can systematically underestimate the transmission risk for those close to the source.Additionally,there is a significant positive linear correlation between the total number of exposed individuals at the short-range and the effective reproduction number.This newly developed TJWR model has great potential for rapid and real-time overall airborne transmission risk assessment in buildings and cities.

Multiscale observation in wide-spatial radar surveillance based on coherent FDA design

Wide-spatial radar surveillance missions are challenging tasks, requiring an increased power budget and agility in transmission aimed at extracting information from multiple targets in different environments. These requirements necessitate high transmitting degrees of freedom(DOF) to achieve the objective of multiscale observation for specific tasks in specialized regions of interest. Herein, we exploit the multiscale observation ability in wide-spatial radar surveillance based on frequency diverse array(FDA)radar. The proposed method facilitates spatial anisotropic control of multiple radar resources, including the transmitting waveforms, beampattern, and bandwidth. By utilizing a coherent FDA radar, we offer principles for the selection of baseband waveforms, in addition to the quantitative design of the beampattern gain and optimal bandwidth from the perspective of detection. The feasibility of the proposed method is validated through numerical experiments, thus indicating the potential in wide-spatial radar surveillance. Moreover,this work can be regarded as a preliminary attempt to gauge the efficacy of the computational array, a novel academic concept.
