Objective This study examined humanin expression in rat ovarian tissue,its cellular localization,and its correlation with rat age under physiological conditions.Methods A total of 40 Sprague-Dawley rats of various ages(2,12,30,and 60 days old and 1 year old)were grouped by age.Immunofluorescence and immunohistochemical techniques were used to observe humanin expression and cellular location in the ovarian tissues of rats from each age group.In addition,Western blotting and Real-time quantitative reverse transcription PCR(qRT-PCR)techniques were used to measure humanin expression level in the ovarian tissues of rats from each age group.Results The immunofluorescence and immunohistochemical results confirmed that humanin was expressed in rat ovarian tissues.In addition,cellular localization analysis showed that humanin was expressed in the cytoplasm of oocytes,interstitial cells,granulosa cells and theca cells in all levels of follicles after the primary follicles,and in the corpus luteum.The qRT-PCR results revealed that the level of humanin expression in the ovarian tissues of 12-day-old rats was non-significantly higher than that in the ovarian tissues of 2-day-old rats(P>0.05),whereas the levels of humanin expression in the ovarian tissues of 30-day-old,60-day-old,and 1-year-old rats were significantly lower than that in the ovarian tissues of 2-day-old rats(P<0.05).The Western blotting results demonstrated that the levels of humanin protein expression in the ovarian tissues of 60-day-old and 1-year-old rats were significantly lower than those of 2-day-old rats(P<0.01),whereas there was no significant difference in the level of humanin protein expression between the ovarian tissues of 12-day-old and 30-day-old rats.Conclusion This study confirmed that humanin is expressed in the cytoplasm of various cells in rat ovarian tissues.Moreover,the level of humanin expression was highest in the ovarian tissues of 12-day-old rats,and it subsequently decreased with age.The changes in the expression of humanin in the ovar

Molybdenum dinitrogen complexes supported by monodentate arylsilylamido ligand,[Ar(MeSi)N]MoNMg(THF)[N(SiMe)Ar](5) and [Ar(MeSi)N]MoNSiMe(6)(Ar=3,5-MeCH) were synthesized and structurally characterized,and proved to be effective catalysts for the disproportionation of cyclohexadienes and isomerization of terminal alkenes.The~1 H NMR spectrum suggested that the bridging nitrogen ligand remains intact during the catalytic reaction,indicating possible catalytic ability of the Mo-N=N motif.

Developing socio-hydrology:Research progress,opportunities and challenges

The development of industrialization and urbanization has intensified the coupling of human activities and hydrological processes and promoted the emergence of socio-hydrology.This paper addresses the issue of socio-hydrology due to new development and social demand for hydrological sciences and sustainable development.Four key scientific issues are identified through systematic analysis and summary of the relative research and international progress,i.e.,(1)the long-term dynamic process of socio-hydrological system evolution;(2)quantitative description and driving mechanism analysis of socio-hydrological coupling system;(3)prediction of the trajectories of socio-hydrological system co-evolution,and(4)integrated water resource management from the perspective of water systems.Moreover,opportunities and challenges for developing socio-hydrology are emphasized,including(1)strengthening the research of interdisciplinary theoretical systems;(2)improving and broadening socio-hydrological research technical methods,and(3)supporting integrated water resources management(IWRM)for sustainable utilization goals(SDGs).The review is expected to provide a reference for the future development of socio-hydrology discipline.

Predicted Mean Vote of Subway Car Environment Based on Machine Learning

The thermal comfort of passengers in the carriage cannot be ignored.Thus,this research aims to establish a prediction model for the thermal comfort of the internal environment of a subway car and find the optimal input combination in establishing the prediction model of the predicted mean vote(PMV)index.Data-driven modeling utilizes data from experiments and questionnaires conducted in Nanjing Metro.Support vector machine(SVM),decision tree(DT),random forest(RF),and logistic regression(LR)were used to build four models.This research aims to select the most appropriate input variables for the predictive model.All possible combinations of 11 input variables were used to determine the most accurate model,with variable selection for each model comprising 102350 iterations.In the PMV prediction,the RF model was the best when using the correlation coefficients square(R2)as the evaluation indicator(R^(2):0.7680,mean squared error(MSE):0.2868).The variables include clothing temperature(CT),convective heat transfer coefficient between the surface of the human body and the environment(CHTC),black bulb temperature(BBT),and thermal resistance of clothes(TROC).The RF model with MSE as the evaluation index also had the highest accuracy(R^(2):0.7676,MSE:0.2836).The variables include clothing surface area coefficient(CSAC),CT,BBT,and air velocity(AV).The results show that the RF model can efficiently predict the PMV of the subway car environment.

Metal distribution characteristic of MSWI bottom ash in view of metal recovery

Bottom ash is the major by-product of municipal solid waste incineration（MSWI）, and is often reused as an engineering material, such as road-base aggregate. However, some metals（especially aluminum） in bottom ash can react with water and generate gas that could cause expansion and failure of products containing the ash; these metals must be removed before the ash is utilized. The size distribution and the chemical speciation of metals in the bottom ash from two Chinese MSWI plants were examined in this study, and the recovery potential of metals from the ash was evaluated. The metal concentrations in these bottom ashes were lower than that generated in other developed countries. Specifically, the contents of Al,Fe, Cu and Zn were 18.9–29.2, 25.5–32.3, 0.7–1.0 and 1.6–2.5 g/kg, respectively. Moreover,44.9–57.0 wt.% of Al and 55.6–75.4 wt.% of Fe were distributed in bottom ash particles smaller than 5 mm. Similarly, 46.6–79.7 wt.% of Cu and 42.9–74.2 wt.% of Zn were concentrated in particles smaller than 3 mm. The Fe in the bottom ash mainly existed as hematite, and its chemical speciation was considered to limit the recovery efficiency of magnetic separation.

Reconfigurable metamaterial for asymmetric and symmetric elastic wave absorption based on exceptional point in resonant bandgap

Elastic wave absorption at subwavelength scale is of significance in many engineering applications.Non-Hermitian metamaterials show the ability in high-efficiency wave absorption.However,the single functionality of metamaterials is an important limitation on their practical applications for lack of tunability and reconfigurability.Here,we propose a tunable and reconfigurable non-Hermitian piezoelectric metamaterial bar,in which piezoelectric bars connect with resonant circuits,to achieve asymmetric unidirectional perfect absorption(UPA)and symmetric bidirectional perfect absorption(PA)at low frequencies.The two functions can be arbitrarily switched by rearranging shunted circuits.Based on the reverberation-ray matrix(RRM)method,an approach is provided to achieve UPA by setting an exceptional point(EP)in the coupled resonant bandgap.By using the transfer matrix method(TMM)and the finite element method(FEM),it is observed that a non-Hermitian pseudo-band forms between two resonant bandgaps,and the EP appears at the bottom of the pseudo-band.In addition,the genetic algorithm is used to accurately and efficiently design the shunted circuits for desired low-frequency UPA and PA.The present work may provide new strategies for vibration suppression and guided waves manipulation in wide potential applications.

Overexpression of c-Myc-dependent heterogeneous nuclear ribonucleoprotein A1 promotes proliferation and inhibits apoptosis in NOTCH1-mutated chronic lymphocytic leukemia cells

Background: NOTCH1 mutation is an essential molecular biologic aberration in chronic lymphocytic leukemia (CLL). CLL patients withNOTCH1 mutation have shown an unfavorable survival and a poor response to chemoimmunotherapy. This study aims to present the mechanisms of adverse prognosis caused byNOTCH1 mutation from the perspective of the splicing factor heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1).Methods: The microarray data in Gene Expression Omnibus datasets were analyzed by bioinformatics and the function of hnRNPA1 was checked by testing the proliferation and apoptosis of CLL-like cell lines. Afterward, quantitative reverse transcription-polymerase chain reaction and Western blotting were applied to explore the relationship among NOTCH1, c-Myc, and hnRNPA1.Results: RNA splicing was found to play a vital part inNOTCH1-mutated CLL cells;hence, hnRNPA1 was selected as the focus of this study. Higher expression of hnRNPA1 validated in primaryNOTCH1-mutated CLL samples could promote proliferation and inhibit apoptosis in CLL. The expression of hnRNPA1 increased when NOTCH1 signaling was activated by transfection with NOTCH1 intracellular domain (NICD)-overexpressed adenovirus vector and declined after NOTCH1 signaling was inhibited by NOTCH1-shRNA. Higher expression of c-Myc was observed in NICD-overexpressed cells and hnRNPA1 expression was downregulated after applying c-Myc inhibitor 10058-F4. Moreover, in NICD-overexpressed cells, hnRNPA1 expression decreased through c-Myc inhibition.Conclusion: Overexpression of c-Myc-dependent hnRNPA1 could promote proliferation and inhibit apoptosis inNOTCH1- mutated CLL cells, which might partly account for the poor prognosis of patients withNOTCH1 mutation.

COVID-19 and Alzheimer’s disease:how one crisis worsens the other

Alzheimer’s disease(AD)has emerged as a key comorbidity of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus-2(SARS-CoV-2).The morbidity and mortality of COVID-19 are elevated in AD due to multiple pathological changes in AD patients such as the excessive expression of viral receptor angiotensin converting enzyme 2 and pro-inflammatory molecules,various AD complications including diabetes,lifestyle alterations in AD,and drug-drug interactions.Meanwhile,COVID-19 has also been reported to cause various neurologic symptoms including cognitive impairment that may ultimately result in AD,probably through the invasion of SARS-CoV-2 into the central nervous system,COVID-19-induced inflammation,long-term hospitalization and delirium,and post-COVID-19 syndrome.In addition,the COVID-19 crisis also worsens behavioral symptoms in uninfected AD patients and poses new challenges for AD prevention.In this review,we first introduce the symptoms and pathogenesis of COVID-19 and AD.Next,we provide a comprehensive discussion on the aggravating effects of AD on COVID-19 and the underlying mechanisms from molecular to social levels.We also highlight the influence of COVID-19 on cognitive function,and propose possible routes of viral invasion into the brain and potential mechanisms underlying the COVID-19-induced cognitive impairment.Last,we summarize the negative impacts of COVID-19 pandemic on uninfected AD patients and dementia prevention.

Tuning heat transport via coherent structure manipulation:recent advances in thermal turbulence

Tuning transport properties through the manipulation of elementary structures has achieved great success in many areas,such as condensed matter physics.However,the ability to manipulate coherent structures in turbulent flows is much less explored.This article reviews a recently discovered mechanism of tuning turbulent heat transport via coherent structure manipulation.We first show how this me chanism can be realized by applying simple geometrical confinement to a classical thermally driven turbulence, which leads to the condensation of elementary coherent structures and significant heat-transport enhancement,despite the resultant slower flow.Some potential applications of this new paradigm in passive heat management are also discussed.We then explain how the heat transport behaviors in seemingly different turbulence systems can be understood by this unified framework of coherent structure manipulation.Several future directions in this research area are also outlined.
