The border gateway protocol(BGP)has become the indispensible infrastructure of the Internet as a typical inter-domain routing protocol.However,it is vulnerable to misconfigurations and malicious attacks since BGP does not provide enough authentication mechanism to the route advertisement.As a result,it has brought about many security incidents with huge economic losses.Exiting solutions to the routing security problem such as S-BGP,So-BGP,Ps-BGP,and RPKI,are based on the Public Key Infrastructure and face a high security risk from the centralized structure.In this paper,we propose the decentralized blockchain-based route registration framework-decentralized route registration system based on blockchain(DRRS-BC).In DRRS-BC,we produce a global transaction ledge by the information of address prefixes and autonomous system numbers between multiple organizations and ASs,which is maintained by all blockchain nodes and further used for authentication.By applying blockchain,DRRS-BC perfectly solves the problems of identity authentication,behavior authentication as well as the promotion and deployment problem rather than depending on the authentication center.Moreover,it resists to prefix and subprefix hijacking attacks and meets the performance and security requirements of route registration.

SOX2 is a well-characterized pluripotent factor that is essential for stem cell self-renewal,reprogramming,and homeostasis.The cellular levels of SOX2 are precisely regulated by a complicated network at the levels of transcription,post-transcription,and posttranslation.In many types of human cancer,SOX2 is dysregulated due to gene amplification and protein overexpression.SOX2 overexpression is associated with poor survival of cancer patients.Mechanistically,SOX2 promotes proliferation,survival,invasion/metastasis,cancer stemness,and drug resistance.SOX2 is,therefore,an attractive anticancer target.However,little progress has been made in the efforts to discover SOX2 inhibitors,largely due to undruggable nature of SOX2 as a transcription factor.In this review,we first briefly introduced SOX2 as a transcription factor,its domain structure,normal physiological functions,and its involvement in human cancers.We next discussed its role in embryonic development and stem cell-renewal.We then mainly focused on three aspects of SOX2:(a)the regulatory mechanisms of SOX2,including how SOX2 level is regulated,and how SOX2 cross-talks with multiple signaling pathways to control growth and survival;(b)the role of SOX2 in tumorigenesis and drug resistance;and(c)current drug discovery efforts on targeting SOX2,and the future perspectives to discover specific SOX2 inhibitors for effective cancer therapy.

Many biological processes such as cell proliferation, differentiation, and cell death depend precisely on the timely synthesis and degradation of key regulatory proteins. While protein synthesis can be regulated at multiple levels, protein degradation is mainly controlled by the ubiquitin-proteasome system （UPS）, which consists of two distinct steps：（1） ubiquitylation of targeted protein by E1 ubiquitin-activating enzyme, E2 ubiquitin-conjugating enzyme and E3 ubiquitin ligase, and （2） subsequent degradation by the 26S proteasome. Among all E3 ubiquitin ligases, the SCF （SKP1-CUL1-F-box protein） E3 ligases are the largest family and are responsible for the turnover of many key regulatory proteins. Aberrant regulation of SCF E3 ligases is associated with various human diseases, such as cancers, including skin cancer. In this review, we provide a comprehensive overview of all currently published data to define a promoting role of SCF E3 ligases in the development of skin cancer. The future directions in this area of research are also discussed with an ultimate goal to develop small molecule inhibitors of SCF E3 ligases as a novel approach for the treatment of human skin cancer. Furthermore, altered components or substrates of SCF E3 ligases may also be developed as the biomarkers for early diagnosis or predicting prognosis.

耦合可再生能源生产的绿电为煤气化反应供热以消除燃烧供热实现自热平衡的影响,不仅可有效降低气化过程CO_(2)排放量,还能提高合成气氢碳比(H/C),从而降低以煤气化为源头生产化学品的水煤气变换工段中CO_(2)排放。对不同类型的常规气化反应器和相应的耦合绿电反应器进行建模与模拟,判明合成气组分的主要影响因素,分析了温度对合成气组分的影响,探讨合成气H/C随温度变化规律,并计算不同化学品生产过程的CO_(2)减排潜力。结果表明,耦合绿电的气化反应器合成气中CO_(2)排放较常规气化反应器分别减少了12.63%(固定床)、11.01%(气流床)、9.23%(输运床)和5.12%(流化床),且H/C呈上升趋势;温度对合成气组分影响较大,热解解耦和气化反应的反应器一定程度上影响合成气组分;温度低于1500 K时,H/C随温度升高而降低;温度高于1500 K时,H/C随温度升高而缓慢升高;耦合绿电煤基化学品生产过程CO_(2)排放大幅减少,操作在更低温度和压力的气化反应器具有更高的合成气H/C和CO_(2)减排潜力;耦合绿电的固定床系统效率优势明显;考虑到耦合绿电的高温反应器存在的技术困难,耦合绿电的固定床、流化床和输运床反应器更具可行性。

Polysaccharides from Fuzhuan brick tea(FBTPS),one of most important bioactive components in tea,showed various health-promoting functions.Our previous work demonstrated that the crude FBTPS(CFBTPS)could modulate the gut microbiota.However,which purified fraction in CFBTPS contributing to the modulation of gut microbiota remains unclear.Thus,the fermentation characteristics and probiotic activity of a purified fraction(FBTPS-2-1)of CFBTPS were evaluated in this work.The results showed that gut microbiota could utilize FBTPS-2-1 to produce short-chain fatty acids including acetic,propionic,n-butyric and n-valeric acids.FBTPS-2-1 could modulate the structure and metabolic pathways of gut microbiota.FBTPS-2-1 could increase the health-promoting gut microbiota such as Prevotellaceae and Bifidobacteriaceae,and decreased the harmful bacteria such as Enterobacteriaceae and Fusobacteriaceae.The results of metagenomics showed that Prevotella copri and Megamonas funiformis were the dominant bacteria after fermentation of FBTPS-2-1.Furthermore,FBTPS-2-1 could regulate the biosynthesis and metabolism pathways of gut microbiota.Thus,the enrichment of food with FBTPS-2-1 is expected as a potential strategy for promoting human health due to modulation of gut microbiota.

Multiscale simulations of the tilted flat-ended nanoindentation with different tilted angles （from 5° ～ 30°） on the （-1 1 0） surface of nickel crystal were studied using the QC method. The model of the indentation is a flat-end indenter inclined by an angle ε driven into a half- plane vertically. Load-displacement responses, initiM stages of the plasticity deformations and dislocation emissions for nickel film at different inclined angles were obtained and analyzed as well. An energy criterion was successfully proposed to analyze the critical load for the first dislocation emission beneath the edge of the indenter. Simulation results agree well with analytical ones. An elastic model based on the contact theory and the Peierls-Nabarro dislocation model were combined to analyze when and where the dislocation will be emitted beneath the lower surface of an inclined indenter. Results indicate that the key parameter is the ratio of the contact half- width to the position of the slip plane. This parameter shows the range in which a dislocation will probably be emitted. This mechanism explains the simulation results well. This work is of value for understanding the mechanism of dislocation emissions of FCC crystals under tilted flat- ended nanoindentation while providing approaches to predicting when the first dislocation will be emitted and where subsequent dislocations will probably be emitted.

目的探讨Ras相关结合蛋白23(RAB23)在食管鳞状细胞癌(简称食管鳞癌)细胞侵袭和迁移中的作用和机制。方法采用实时荧光定量聚合酶链反应检测16例配对食管鳞癌及癌旁正常组织中RAB23 mRNA的表达。比较基因表达综合(GEO)数据库的GSE20347数据集中食管鳞癌和配对癌旁正常组织RAB23 mRNA的表达水平。免疫组织化学检测106例配对食管鳞癌和癌旁正常组织、33例淋巴结阳性与10例淋巴结阴性患者原发灶与淋巴结组织中RAB23蛋白含量。在食管鳞癌KYSE30和KYSE150细胞中瞬时敲降RAB23表达(转染si-RAB23-1和si-RAB23-9)或者稳定敲降RAB23表达(转染sh-RAB23), 采用Western blot法验证RAB23敲降效率, 采用细胞计数试剂盒8法和裸鼠皮下成瘤实验检测食管鳞癌细胞的增殖能力, 采用Transwell实验和裸鼠尾静脉-肺转移实验检测食管鳞癌细胞的侵袭和迁移能力, 采用细胞黏附实验检测食管鳞癌细胞的黏附能力, 采用转录组测序技术分析RAB23敲降后对细胞转录谱的影响, 并通过Western blot检测验证相关信号通路。结果 16例食管鳞癌组织中RAB23 mRNA表达水平为0.009 7±0.008 9, 高于癌旁正常组织[0.003 2±0.003 7, P=0.006]。对GEO数据库GSE20347数据集中食管鳞癌和配对癌旁正常组织表达谱的生信分析显示, 食管鳞癌组织中RAB23 mRNA表达水平为4.30±0.25, 高于癌旁正常组织(4.10±0.17, P=0.037)。106例食管鳞癌组织中, 51例RAB23低表达, 55例RAB23高表达;而配对癌旁正常组织中, 82例RAB23低表达, 24例RAB23高表达, 食管鳞癌组织中RAB23表达水平高于配对癌旁正常组织(P<0.001)。33例淋巴结阳性食管鳞癌组织中, 1例RAB23低表达, 32例RAB23高表达;10例淋巴结阴性的食管鳞癌组织中, 3例RAB23低表达, 7例RAB23高表达。淋巴结阳性食管鳞癌组织中RAB23表达水平高于淋巴结阴性食管鳞癌组织(P=0.024)。33例阳性淋巴结组织中, 1例RAB23低表达, 32例RAB23高表达, 而10例阴性淋巴结组织中均为RAB23低表达。阳性淋巴结组织中RAB23表达水平高于阴性淋巴结组织(P<0.001)。瞬时或稳定敲降RAB23后, KYSE30细胞体外和体内的增殖能力无明显变化, KYSE30细胞的细胞侵袭和迁移能力下降, 而KYSE150细胞仅侵袭能力下降, 迁移能力无明显变化。sh-RAB23组KYSE30细胞黏附细胞的数量为(313.75±89.34)个, 少于sh-NC组[(1 030.75±134.29)个, P<0.001]。sh-RAB23组KYSE150细胞的黏附细胞数为(710.5±31.74)个, 也少于sh-NC组[(1 005.75±61.09)个, P<0.001]。转录组测序分析显示, 敲降RAB23后, si-RAB23-1组和si-RAB23-9组KYSE30细胞中黏着斑相关信号通路均被削弱, sh-RAB23组KYSE30和KYSE150细胞中p-FAK和p-paxillin表达水平降低。结论 RAB23在食管鳞癌组织中高表达, 与淋巴结转移有关。敲低RAB23表达后能够削弱黏着斑相关信号通路, 进而抑制食管鳞癌细胞的侵袭、迁移和黏附。

Multi-mode microcavity frequency engineering through a shifted grating in a photonic crystal ring

Frequency engineering of whispering-gallery resonances is essential in microcavity nonlinear optics.The key is to control the frequencies of the cavity modes involved in the underlying nonlinear optical process to satisfy its energy conservation criterion.Compared to the conventional method that tailors dispersion by cross-sectional geometry,thereby impacting all cavity mode frequencies,grating-assisted microring cavities,often termed as photonic crystal microrings,provide more enabling capabilities through mode-selective frequency control.For example,a simple single period grating added to a microring has been used for single frequency engineering in Kerr optical parametric oscillation(OPO)and frequency combs.Recently,this approach has been extended to multifrequency engineering by using multi-period grating functions,but at the cost of increasingly complex grating profiles that require challenging fabrication.Here,we demonstrate a simple approach,which we term as shifted grating multiple mode splitting(SGMMS),where spatial displacement of a single period grating imprinted on the inner boundary of the microring creates a rotational asymmetry that frequency splits multiple adjacent cavity modes.This approach is easy to implement and presents no additional fabrication challenges compared to an unshifted grating,and yet is very powerful in providing multi-frequency engineering functionality for nonlinear optics.We showcase an example where SGMMS enables OPO across a wide range of pump wavelengths in a normal-dispersion device that otherwise would not support OPO.

人类通过感官与世界互动获得对事物的直观把握。知觉意义以感官感知为基础，在感官直观感知的基础上，经由认知初加工，形成知觉意义。由感官感知到语言编码，经历了感官感知—知觉加工产生意义—认知加工编码产生语言等过程。知觉意义的记录和交换催生了知觉语言的编码。知觉语言的产生与各感官之间联系紧密，关系错综复杂，亟待建立一个以知觉意义的生成和编码问题为核心议题的语言学研究新平台。基于此，提出“知觉语言学”学科界域，并以五官模型、体认语言学和认知隐喻为理论切入点，阐释“知觉语言学”的学科理论渊源；从通感阐释、通感隐喻迁移次序的差异化阐释、感官灵敏度与词汇丰富度之间的关联阐释、知觉意义的多模态表征及失聪和失明个体的知觉语言研究等方面爬梳研究议题。知觉语言学对探测人类的感知世界，回答感官系统如何与词语建立连接，以及知觉意义如何在语言中编码等问题，具有重要的学科意义与价值。
