Synthesis of platelet-activating factor and its receptor expression in Kupffer cells in rat carbon tetrachloride-induced cirrhosis

AIM:To determine the platelet-activating factor (PAF) synthesis and its receptor expression in Kupffer cells in rat carbon tetrachloride-induced cirrhosis. METHODS:Kupffer cells, isolated from the livers of control and CCl4-induced cirrhotic rats, were placed in serum-free medium overnight. PAF saturation binding, ET-1 saturation and competition binding were assayed. ET-1 induced PAF synthesis, mRNA expression of PAF, preproendothelin-1, endothelin A (ETA) and endothelin B (ETB) receptors were also determined. RESULTS:A two-fold increase of PAF synthesis (1.42 ± 0.14 vs 0.66 ± 0.04 pg/μg DNA) and a 1.48-fold increase of membrane-bound PAF (1.02 ± 0.06 vs 0.69 ± 0.07 pg/μg DNA) were observed in activated Kupffer cells of cirrhotic rats. The application of ET-1 to Kupffer cells induced PAF synthesis in a concentration-dependent manner in both cirrhotic and normal rats via ETB receptor, but PAF synthesis in the activated Kupffer cells was more effective than that in the normal Kupffer cells. In activated Kupffer cells, PAF receptor expression and PAF binding capacity were markedly enhanced. Activated Kupffer cells raised the [125I]-ET-1 binding capacity, but changed neither the affinity of the receptors, nor the expression of ETA receptor. CONCLUSION:Kupffer cells in the course of CCl4-induced cirrhosis are the main source of increased PAF. ET-1 is involved endogenously in stimulating the PAF synthesis in activated Kupffer cells via ETB receptor by paracrine. ETA receptor did not appear in activated Kupffer cells, which may exacerbate the hepatic and extrahepatic complications of cirrhosis.

This paper presents an algorithm to evaluate estimated and exact system reliabilities for a computer network in the cloud computing *** the quality of service（QOS） viewpoint,the computer network should be maintained when falling to a specific state such that it cannot afford enough capacity to satisfy ***,the transmission time should be concerned as ***,the data can be sent through several disjoint minimal paths simultaneously to shorten the transmission *** the maintenance budget B and time constraint T,we evaluate the system reliability that d units of data can be sent from the cloud to the client through multiple *** procedures are integrated in the proposed algorithm-an estimation procedure for estimated system reliability and an adjusting procedure utilizing the branch-and-bound approach for exact system ***,the estimated system reliability with lower bound and upper bound,and exact system reliability are computed by applying the recursive sum of disjoint products（RSDP） algorithm.

BiAlO_(3)-modified BiFeO_(3)-BaTiO_(3) high Curie temperature lead-free piezoelectric ceramics with enhanced performance

BiFeO_(3)-BaTiO_(3)(BF-BT)lead-free piezoelectric ceramics have high piezoelectricity and high Curie temperature(T_(C)),but the mixed-valence Fe ions and Bi^(3+)volatilization would promote the formation of Bi_(25)FeO_(40)/Bi_(2)Fe_(4)O_9 and oxygen vacancy,which greatly degrade the insulation properties required for *** this study,it was found that the modification of BiAlO_(3)(BA)in BF-BT ceramics could effectively solve these problems,reducing the leakage current to 1×10^(-9)A·cm^(-2)and transiting the space charge-limited conduction to ohmic *** of the enhanced insulation properties and appropriate rhombohedral-pseudocubic phase ratio(C_R/C_(PC)),BF-BT-xBA ceramics in an optimized composition obtain enhanced piezoelectric performance:piezoelectric charge coefficient(d_(33))=196 pC·N^(-1),planar electromechanical coupling coefficient(k_(p))=31.1%,T_(C)=487℃and depolarization temperature(T_d)=250°C;unipolar strain(S_(uni))=0.17%and piezoelectric strain coefficient(d_(33)^(*))=335 pm·V^(-1)at 100℃.Especially,d_(33)exceeds 283 pC·N^(-1)at 233℃and d_(33)^(*)is 335 pm·V^(-1)at100℃,showing an excellent high-temperature piezoelectricity and high depolarization *** results are attributed to the domain structure of rhombohedral-pseudocubic phase coexistence and its high-temperature switching *** work provides a feasible and effective approach to improve the high temperature piezoelectric properties of BF-BT-xBA ceramics,making them more suitable for high temperature applications.

AIM： To observe the therapeutic efficacy of Baicalin in rats with severe acute pancreatitis （SAP） and explore its therapeutic mechanisms. METHODS： The SAP rat models were randomly divided into the model control group, Baicalin treatment group, octreotide treatment group and sham operation group. All groups were randomly subdivided into 3 h, 6 h and 12 h groups with 15 rats in each group. The survival, ascites volume and pathological changes of pancreas in all rats were observed at different time points after operation. The plasma amylase content and serum TNF-α, IL-6, malonaldehyde （MDA） and PLA2 contents were also determined. RESULTS： The survival was not obviously different between the treated groups, and was significantly higher in treated groups at 12 h compared to the model control group （P 〈 0.05, 15 vs 10）. The ascites/body weight ratio at 3 h and 6 h was significantly lower in Baicalin treatment group compared to the model control group and octreotide treatment group （P 〈 0.05, 1.00 vs 2.02 and 1.43 and P 〈 0.001, 2.29 （1.21） vs 2.70 （0.80） and 2.08 （2.21）, respectively）. The contents of amylase, TNF-α, IL-6, MDA and PLA2 were significantly lower in the treated groups than in the model control group （P 〈 0.05, 4342 vs 5303, 5058 vs 6272 in amylase, P 〈 0.01, 21.90 vs 36.30, 23.80 vs 39.70, 36 vs 54.35 in MDA and 56.25 vs 76.10 in PIA2, or P 〈 0.001, 65.10 and 47.60 vs 92.15 in TNF-α, 3.03 vs 5.44, 2.88 vs 6.82, 2.83 vs 5.36 in IL-6, respectively）. The pathological scores of pancreas in the treated groups were significantly lower than that in the model control group （P 〈 0.05, 9.00 vs 10.05, 6.00 vs 9.00, 8.00 vs 10.05）, but no marked difference was found between the treated groups. CONCLUSION： The Baicalin injection has significant therapeutic effects on SAP rats, its effects are similar to those of octreotide. The Baicalin injection is also cheap and has a big application range, quite hopefully to be used in clinical treatment of SAP.

Validation and application of three-dimensional discontinuous deformation analysis with tetrahedron finite element meshed block

In the last decade, three dimensional discontin- uous deformation analyses （3D DDA） has attracted more and more attention of researchers and geotechnical engineers worldwide. The original DDA formulation utilizes a linear displacement function to describe the block movement and deformation, which would cause block expansion under rigid body rotation and thus limit its capability to model block de- formation. In this paper, 3D DDA is coupled with tetrahe- dron finite elements to tackle these two problems. Tetrahe- dron is the simplest in the 3D domain and makes it easy to implement automatic discretization, even for complex topol- ogy shape. Furthermore, element faces will remain planar and element edges will remain straight after deformation for tetrahedron finite elements and polyhedral contact detection schemes can be used directly. The matrices of equilibrium equations for this coupled method are given in detail and an effective contact searching algorithm is suggested. Valida- tion is conducted by comparing the results of the proposed coupled method with that of physical model tests using one of the most common failure modes, i.e., wedge failure. Most of the failure modes predicted by the coupled method agree with the physical model results except for 4 cases out of the total 65 cases. Finally, a complex rockslide example demon- strates the robustness and versatility of the coupled method.

Saliva in the diagnosis of diseases

Saliva is secreted from the salivary glands and has multiple functions, including mouth cleaning and protection, antibacterial effects and digestion. With the rapid advancement in salivaomics, saliva is well recognized as a pool of biological markers. Saliva, as a non-invasive and safe source, could be a substitute for blood in the diagnosis and prognosis of diseases. This review summarizes the latest advancements in saliva-related studies and addresses the potential value of saliva in the early diagnosis of oral diseases, such as dental caries and periodontal disease, as well as cancer, diabetes and other systemic disorders. Saliva biomarkers range from changes in the biochemical indices of DNA, RNA and proteins to the diversification of microbiota structures. This study integrates data reported in the recent literature and discusses the clinical significance and prospects for the application of saliva in the early diagnosis of diseases, translational medicine and precision medicine.

Effect of annealing temperature on the microstructure and optoelectrical properties of ZnO thin films and their application in self-powered accelerometers

This paper reports a piezoelectric nanogenerator(NG) with a thickness of approximately 80 μm for miniaturized self-powered acceleration sensors. To deposit the piezoelectric zinc oxide(ZnO) thin film, a magnetron sputtering machine was used. Polymethyl methacrylate(PMMA) and aluminum-doped zinc oxide(AZO) were used as the insulating layer and the top electrode of the NG, respectively. The experimental results show that the ZnO thin films annealed at 150℃ exhibited the highest crystallinity among the prepared films and an optical band gap of 3.24 eV. The NG fabricated with an AZO/PMMA/ZnO/stainless steel configuration exhibited a higher output voltage than the device with an AZO/ZnO/PMMA/stainless steel configuration. In addition, the annealing temperature affected the open-circuit voltage of the NGs;the output voltage reached 3.81 V when the annealing temperature was 150℃. The open-circuit voltage of the prepared self-powered accelerometer increased linearly with acceleration. In addition, the small NG-based accelerometer, which exhibited excellent fatigue resistance, can be used for acceleration measurements of small and lightweight devices.

Sodium tanshinone ⅡA sulfonate prevents radiation-induced damage in primary rat cardiac fibroblasts

This study investigated the effects of X-ray irradiation on primary rat cardiac fibroblasts(CFs) and its potential mechanism, as well as whether sodium tanshinone ⅡA sulfonate(STS) has protective effect on CFs and its possible mechanism. Our data demonstrated that X-rays inhibited cell growth and increased oxidative stress in CFs, and STS mitigated X-ray-induced injury. Enzyme-linked immuno-sorbent assay showed that X-rays increased the levels of secreted angiotensin Ⅱ(Ang Ⅱ) and brain natriuretic peptide(BNP). STS inhibited the X-ray-induced increases in Ang Ⅱ and BNP release. Apoptosis and cell cycle of CFs were analyzed using flow cytometry. X-rays induced apoptosis in CFs, whereas STS inhibited apoptosis in CFs after X-ray irradiation. X-rays induced S-phase cell cycle arrest in CFs, which could be reversed by STS. X-rays increased the expression of phosphorylated-P38/P38,cleaved caspase-3 and caspase-3 as well as decreased the expression of phosphorylated extracellular signal-regulated kinase 1/2(ERK1/2)/ERK 1/2 and B cell lymphoma 2(Bcl-2)/Bcl-2 associated X protein(BAX) in CFs, as shown by Western blotting. STS mitigated the X-ray radiation-induced expression changes of these proteins. In conclusion, our results demonstrated that STS may potentially be developed as a medical countermeasure to mitigate radiation-induced cardiac damage.

Bimagnetic metal constructed core-shell structure of Fe@Co nanowires for multi-band microwave absorption

Controlled fabrication of materials by surface modification techniques has been a hot research topic. In this paper,one-dimensional(1D) core-shell structure Fe@Co nanowires(NWs) were successfully prepared by anchoring 0-dimensional(0D) Co nanoparticles on 1D Fe NWs based on the in situ reduction method. The electromagnetic parameters of the specimens at a filler mass fraction of 25 wt% were tested and the microwave loss mechanism was deeply analyzed. Binary magnetic metals in a core-shell structure have excellent multi-band microwave absorption capability(S-band, X-band and Ku-band). This is due to the heterogeneous interface and magnetic coupling effects tuning the dielectric and magnetic loss. This study offers a workable plan for creating effective multi-band magnetic metal-based microwave absorbers.
