Molecular profiling of cell-surface proteins is a powerful strategy for precise cancer diagnosis.While mass cytometry(MC)enables synchronous detection of over 40 cellular parameters,its full potential in disease classification is challenged by the limited types of recognition probes currently available.In this work,we synthesize a panel of heavy isotopeconjugated aptamers to profile cancer-associated signatures on the surface of hematological malignancy(HM)cells.Based on 15 molecular signatures,we performed cell-surface profiling that allowed the precise classification of 8 HM cell lines.Combined with machine-learning technology,this aptamer-based MC platform also achieved multiclass identification of HM subtypes in clinical sampleswith 100%accuracy in the training cohort and 80%accuracy in the test cohort.Therefore,we report an effective and practical strategy for precise cancer classification at the singlecell level,paving the way for its clinical use in the near future.

How to deal with differences is a question that any political body must respond to.The individualist perspective emphasizes that actors will adopt measures to exclude or even eliminate divergent claims in order to gain control over interests power or statutory policy,but the rich experience of traditional Chinese scholar officials offers the possibility of going beyond the limitations of this perspective.The perspective of“tianxia qingshi(world circumstances)”as a new narrative reveals a unique practical logic and dynamic mechanism in Chinese political civilization that have been overlooked by the individualist perspective.The“Green Sprouts policy(qing miao fa)polemic”in the early years of the New Policies era of the Northern Song dynasty(960-1127)presents a historical situation that has not been given full attention by existing research.

BACKGROUND Synaptotagmins(SYTs)are a family of 17 membrane transporters that function as calcium ion sensors during the release of Ca2+-dependent neurotransmitters and hormones.However,few studies have reported whether members of the SYT family play a role in glucose uptake in diabetic retinopathy(DR)through Ca2+/glucose transporter-1(GLUT1)and the possible regulatory mechanism of SYTs.AIM To elucidate the role of the SYT family in the regulation of glucose transport in retinal pigment epithelial cells and explore its potential as a therapeutic target for the clinical management of DR.METHODS DR was induced by streptozotocin in C57BL/6J mice and by high glucose medium in human retinal pigment epithelial cells(ARPE-19).Bioinformatics analysis,reverse transcriptase-polymerase chain reaction,Western blot,flow cytometry,ELISA,HE staining,and TUNEL staining were used for analysis.RESULTS Six differentially expressed proteins(SYT2,SYT3,SYT4,SYT7,SYT11,and SYT13)were found between the DR and control groups,and SYT4 was highly expressed.Hyperglycemia induces SYT4 overexpression,manipulates Ca2+influx to induce GLUT1 fusion with the plasma membrane,promotes abnormal expression of the glucose transporter GLUT1 and excessive glucose uptake,induces ARPE-19 cell apoptosis,and promotes DR progression.Parkin deficiency inhibits the proteasomal degradation of SYT4 in DR,resulting in SYT4 accumulation and enhanced GLUT1 fusion with the plasma membrane,and these effects were blocked by oe-Parkin treatment.Moreover,dysregulation of the myelin transcription factor 1(Myt1)-induced transcription of SYT4 in DR further activated the SYT4-mediated stimulus-secretion coupling process,and this process was inhibited in the oe-MYT1-treated group.CONCLUSION Our study reveals the key role of SYT4 in regulating glucose transport in retinal pigment epithelial cells during the pathogenesis of DR and the underlying mechanism and suggests potential therapeutic targets for clinical DR.

Frequency-comb-linearized, widely tunable lasers for coherent ranging

Tunable lasers,with the ability to continuously vary their emission wavelengths,have found widespread applications across various fields such as biomedical imaging,coherent ranging,optical communications,and spectroscopy.In these applications,a wide chirp range is advantageous for large spectral coverage and high frequency resolution.Besides,the frequency accuracy and precision also depend critically on the chirp linearity of the laser.While extensive efforts have been made on the development of many kinds of frequency-agile,widely tunable,narrow-linewidth lasers,wideband yet precise methods to characterize and linearize laser chirp dynamics are also demanded.Here we present an approach to characterize laser chirp dynamics using an optical frequency comb.The instantaneous laser frequency is tracked over terahertz bandwidth at 1 MHz intervals.Using this approach we calibrate the chirp performance of 12 tunable lasers from Toptica,Santec,New Focus,EXFO,and NKT that are commonly used in fiber optics and integrated photonics.In addition,with acquired knowledge of laser chirp dynamics,we demonstrate a simple frequency-linearization scheme that enables coherent ranging without any optical or electronic linearization unit.Our approach not only presents novel wideband,highresolution laser spectroscopy,but is also critical for sensing applications with ever-increasing requirements on performance.

PARAMETER OPTIMIZATION AND VIBRATION REDUCTION ANALYSIS OF DOUBLE ELECTROMAGNETIC SHUNT TUNED MASS DAMPER

A novel electromagnetic shunt tuned mass damper (EMS-TMD) is proposed for structural vibration control. A new feature of the damper is the electromagnetic damping element that replaces the viscous damping of the traditional double tuned mass dampers. To further improve the damping performance and robustness of the damper, a double electromagnetic shunt tuned mass damper (DEMS-TMD) is designed. According to the Alembert’s theorem, the dynamic model of the coupled system DEMS-TMD and a single degree of freedom structure under seismic excitation is established. Based on the Monte Carlo pattern search method with the target of minimizing the value of the maximum dynamic amplification coefficient of the main structure displacement, the optimal parameters of DEMS-TMD, the structure frequency ratio, the electrical frequency ratio, the electromagnetic damping ratio and the electromechanical coupling coefficient are obtained, which provides the theoretical guidance for damping parameter design. The frequency and time domain simulations of the DEMS-TMD are conducted to analyze the performance of the vibration suppression of the DEMS-TMD. The results show that the reduction of the peak and RMS values of main structure displacement of the DEMS-TMD is superior to traditional parallel double tuned mass dampers (DTMD) in the frequency domain simulations. The reduction of the peak and RMS values of displacement and acceleration of the DEMS-TMD is superior to traditional DTMD in the time domain simulations, which shows the better damping effect of the DEMS-TMD on structures. © 2024 Tsinghua University. An image mosaic technique with non-overlapping regions based on microscopic vision in precision assembly

Microscopic vision has been widely applied in precision assembly.To achieve sufficiently high resolution in measurements for precision assembly when the sizes of the parts involved exceed the field of view of the vision system,an image mosaic technique must be used.In this paper,a method for constructing an image mosaic with non-overlapping areas with enhanced efficiency is proposed.First,an image mosaic model for the part is created using a geometric model of the measurement system installed on a X-Y-Z precision stages with high repeatability,and a path for image acquisition is established.Second,images are captured along the same path for a specified calibration plate,and an entire image is formed based on the given model.The measurement results obtained from the specified calibration plate are utilized to identify mosaic errors and apply compensation for the part requiring measurement.Experimental results show that the maximum error is less than 4μm for a camera with pixel equivalent 2.46μm,thereby demonstrating the accuracy of the proposed method.This image mosaic technique with non-overlapping regions can simplify image acquisition and reduce the workload involved in constructing an image mosaic.

Estimation and testing of linkages between forest structure and rainfall interception characteristics of a Robinia pseudoacacia plantation on China’s Loess Plateau

Understanding the interaction between canopy structure and the parameters of interception loss is essential in predicting the variations in partitioning rainfall and water resources as affected by changes in canopy structure and in implementing water-based management in semiarid forest plantations.In this study,seasonal variations in rainfall interception loss and canopy storage capacity as driven by canopy structure were predicted and the linkages were tested using seasonal filed measurements.The study was conducted in nine 50 m×50 m Robinia pseudoacacia plots in the semiarid region of China’s Loess Plateau.Gross rain-fall,throughfall and stemflow were measured in seasons with and without leaves in 2015 and 2016.Results show that measured average interception loss for the nine plots were 17.9% and 9.4% of gross rainfall during periods with leaves (the growing season) and without leaves, respectively. Average canopy storage capacity estimated using an indirect method was 1.3 mm in the growing season and 0.2 mm in the leafless season. Correlations of relative interception loss and canopy storage capacity to canopy variables were highest for leaf/wood area index (LAI/WAI) and canopy cover, fol-lowed by bark area, basal area, tree height and stand density. Combined canopy cover, leaf/wood area index and bark area multiple regression models of interception loss and canopy storage capacity were established for the growing season and in the leafless season in 2015. It explained 97% and 96% of the variations in relative interception loss during seasons with and without leaves, respectively. It also explained 98% and 99% of the variations in canopy storage capacity during seasons with and without leaves, respectively. The empiri-cal regression models were validated using field data col-lected in 2016. The models satisfactorily predicted relative interception loss and canopy storage capacity during seasons with and without leaves. The analysis of optical spectra—emission or absorption—has been arguably the most powerful approach for discovering and understanding matter.The invention and development of many kinds of spectrometers have equipped us with versatile yet ultra-sensitive diagnostic tools for trace gas detection,isotope analysis,and resolving hyperfine structures of atoms and molecules.With proliferating data and information,urgent and demanding requirements have been placed today on spectrum analysis with ever-increasing spectral bandwidth and frequency resolution.These requirements are especially stringent for broadband laser sources that carry massive information and for dispersive devices used in information processing systems.In addition,spectrum analyzers are expected to probe the device’s phase response where extra information is encoded.Here we demonstrate a novel vector spectrum analyzer(VSA)that is capable of characterizing passive devices and active laser sources in one setup.Such a dual-mode VSA can measure loss,phase response,and dispersion properties of passive devices.It also can coherently map a broadband laser spectrum into the RF domain.The VSA features a bandwidth of 55.1 THz(1260–1640 nm),a frequency resolution of 471 kHz,and a dynamic range of 56 dB.Meanwhile,our fiber-based VSA is compact and robust.It requires neither high-speed modulators and photodetectors nor any active feedback control.Finally,we employ our VSA for applications including characterization of integrated dispersive waveguides,mapping frequency comb spectra,and coherent light detection and ranging(LiDAR).Our VSA presents an innovative approach for device analysis and laser spectroscopy,and can play a critical role in future photonic systems and applications for sensing,communication,imaging,and quantum information processing.

Coastal mudflats as reservoirs of extracellular antibiotic resistance genes: Studies in Eastern China

Despite coastal mudflats serving as essential ecological zones interconnecting terrestrial/freshwater and marine systems,little is known about the profiles of antibiotic resistance genes(ARGs)in this area.In this study,characteristics of typical ARGs,involving both intracellular(iARGs)and extracellular ARGs(eARGs)at different physical states,were explored in over 1000 km of coastal mudflats in Eastern China.Results indicated the presence of iARGs and eARGs at states of both freely present or attached by particles.The abundance of eARGs was significantly higher than that of iARGs(87.3%vs 12.7%),and their dominance was more significant than those in other habitats(52.7%-76.3%).ARG abundance,especially for eARGs,showed an increasing trend(p<0.05)from southern(Nantong)to northern(Lianyungang)coastalmudflats.Higher salinity facilitated the transformation from iARGs to eARGs,and smaller soil particle size was conducive to the persistence of eARGs in northern coastal mudflats.This study addresses the neglected function of coastal mudflats as eARGs reservoirs.

Vibration reduction control of a semisubmersible floating offshore wind turbine by the distributed tuned mass dampers under combined wind and wave excitations

To address the threat of harmful vibrations of semisubmersible floating offshore wind turbine（FOWT）in complex deep-sea environments to the safety and durability，a design of distributed tuned mass dampers（TMDs）is proposed to control the plat‑ form pitch motion under the randomly combined wind and wave excitations，in combination with the geometric structure of the 5MW prototype of NREL in the United States. The distributed TMDs are installed inside the platform to form an equilateral trian‑ gle arrangement. To better describe the performance of the distributed TMDs on the semisubmersible FOWT，a 9-degree-of-free‑ dom multi-body dynamics model is proposed and established for the coupled semisubmersible FOWT-TMDs system based on La‑ grange's equation and the modal superposition method. Based on the H∞ algorithm，whose optimization objective is the peak value of the frequency response function of platform pitch motion，the parameters of the distributed TMDs are optimally designed，where the coupling relationship between multiple TMDs is considered. The numerical simulation of the coupled FOWT-TMDs system under the combined wind and wave excitations is carried out to analyze the performance of the distributed TMDs on the platform pitch response of the wind turbine. The results show that the distributed TMDs with optimal design has good damping performance on the platform pitch motion of the semisubmersible FOWT. Under random wind and wave loads in three different working condi‑ tions，the peak and standard deviation vibration reduction rates of the power spectral density curve near the natural frequency of platform pitch can reach more than 39% and 52%，respectively. The research method and results can provide reference for dynam‑ ic analysis and vibration control design of large semisubmersible FOWT. © 2024 Nanjing University of Aeronautics an Astronautics.
