Hyperhomocysteinemia induces injury in olfactory bulb neurons by downregulating Hes1 and Hes5 expression

Hyperhomocysteinemia has been shown to be associated with neurodegenerative diseases; however, lesions or histological changes and mechanisms underlying homocysteine-induced injury in olfactory bulb neurons remain unclear. In this study, hyperhomocysteinemia was induced in apolipoprotein E-deficient mice with 1.7% methionine. Pathological changes in the olfactory bulb were observed through hematoxylin-eosin and Pischingert staining. Cell apoptosis in the olfactory bulb was determined through terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling （TUNEL） staining. Transmission electron microscopy revealed an abnormal ultrastructure of neurons. Furthermore, immunoreactivity and expression of the hairy enhancer of the split 1 （Hesl） and Hess were measured using immunohistochemistry, immunofluorescence, and western blot assay. Our results revealed no significant structural abnormality in the ol- factory bulb of hyperhomocysteinemic mice. However, the number of TUNEL-positive cells increased in the olfactory bulb, lipofuscin and vacuolization were visible in mitochondria, and the expression of Hes1 and Hes5 decreased. These findings confirm that hyperhomocyste- inemia induces injury in olfactory bulb neurons by downregulating Hes1 and Hes5 expression.

Dynamic changes in neuronal autophagy and apoptosis in the ischemic penumbra following permanent ischemic stroke

The temporal dynamics of neuronal autophagy and apoptosis in the ischemic penumbra following stroke remains ***,in this study,we investigated the dynamic changes in autophagy and apoptosis in the penumbra to provide insight into potential therapeutic targets for *** adult Sprague-Dawley rat model of permanent ischemic stroke was prepared by middle cerebral artery *** autophagy and apoptosis in the penumbra post-ischemia were evaluated by western blot assay and immunofluorescence staining with antibodies against LC3-Ⅱ and cleaved caspase-3,*** of both LC3-Ⅱ and cleaved caspase-3 in the penumbra gradually increased within 5 hours ***,levels of both proteins declined,especially LC3-Ⅱ.The cerebral infarct volume increased slowly 1–4 hours after ischemia,but subsequently increased rapidly until 5 hours after *** severity of the neurological deficit was positively correlated with infarct ***3-Ⅱ and cleaved caspase-3 levels were high in the penumbra within 5 hours after ischemia,and after that,levels of these proteins decreased at different ***3-Ⅱ levels were reduced to a very low level,but cleaved caspase-3 levels remained high 72 hours after *** results indicate that there are temporal differences in the activation status of the autophagic and apoptotic *** suggests that therapeutic targeting of these pathways should take into consideration their unique temporal dynamics.

目的利用CiteSpace知识图谱分析甘草抗肿瘤的中英文文献,探索其研究现状、热点及趋势。方法运用CiteSpace 6.2.R4软件对CNKI和Web of Science中收录的2002年1月至2023年9月甘草抗肿瘤的中英文相关文献,从发文量、作者、机构、期刊、基金资助、关键词等研究主题进行可视化分析。结果共纳入397篇中文文献,172篇英文文献。中英文文献作者分析表明郑秋生和Jung分别为甘草抗肿瘤中文和英文发文量最多的作者;北京中医药大学和翰林大学分别为中文和英文发文量最多的机构,英文文献中研究机构之间的合作相对较多;中英文关键词分析显示甘草抗肿瘤研究主要集中在乳腺癌、甘草及其活性成分、分子机制等方面。聚类分析显示,研究集中在甘草活性成分及临床应用,中医治法理论及复方常用药物,甘草抗肿瘤的实验研究及作用机制,甘草防治化疗药物引起的肾毒性等毒副作用的作用机制研究上。结论甘草抗肿瘤的机制探讨以及甘草与天然产物联合用药可能为今后的研究热点。

目的总结近年来甘草及其复方调节肠道微生物改善药源性肝损伤(DILI)的作用机制。方法在CNKI和PubMed数据库中以“甘草or甘草复方”and“肝损伤”and“肠道菌群”为关键词,对近年来甘草及复方缓解药源性肝损伤的文献进行筛选,总结肠道菌群与DILI的关系、甘草和复方通过调节肠道微生物缓解DILI的机制。结果药源性肝损伤可导致肠道菌群构成异常、肠道屏障损坏以及细菌及其代谢产物易位,而异常的肠道菌群可能通过多途径加重肝脏损伤。甘草中的成分和复方可以通过调节肠道菌群构成、恢复肠道屏障、抗炎以及增强免疫作用机制改善DILI。结论甘草及其复方可以通过调节肠道菌群从而缓解药DILI,为扩大药源性肝损伤的中医药研究和临床治疗提供参考。

单元主题教学法在全科住培教学中的应用

目的探索单元主题教学法在全科住培教学活动中的应用效果。方法选取2021年6月—2022年8月在西部战区总医院全科医学科进行住培轮转的43名住院医师为对象,按照教学活动安排的不同分为试验组和对照组,其中试验组24名,对照组19名。试验组采用单元主题教学法设计教学活动,对照组沿用常规教学活动安排,比较2组住院医师出科时的迷你临床演练评价(mini-clinical evaluation exercise,Mini-CEX)成绩、理论测试成绩、综合成绩及教学满意度。结果试验组的Mini-CEX评分、理论成绩及综合成绩均高于对照组[Mini-CEX评分(54.6±4.7)分vs.(50.5±6.6)分;理论成绩(121.2±8.7)分vs.(111.7±10.8)分;综合成绩(83.7±5.3)分vs.(77.4±6.9)分,P <0.05],差异有统计学意义。教学满意度中教学方式及能力提升两项平均分也高于对照组[教学方式(4.5±0.7)分vs.(3.8±1.2)分;能力提升(4.6±0.7)分vs.(3.7±1.2)分,P <0.05],差异有统计学意义。结论采用单元主题教学法设计全科住培教学活动能提高住院医师综合成绩及满意度,有助于提升全科住培教学效果。

Mettl3通过PI3K/AKT信号通路调控Ang Ⅱ诱导肾脏纤维化的机制研究

目的:探讨甲基转移酶3(Mettl3)在血管紧张素Ⅱ(Ang Ⅱ)诱导周细胞向肌成纤维细胞转分化及肾脏纤维化过程中的作用及相关机制。方法:使用C57BL/6J小鼠,(1)细胞实验中,采用磁珠分选法培养纯化小鼠肾脏周细胞,并予以1×10 6 mmol/L的Ang Ⅱ诱导周细胞-肌成纤维细胞转分化,为Ang Ⅱ组;正常培养的周细胞为对照组。采用免疫荧光染色、蛋白质印迹(Western blot)及实时反转录PCR(RT-qPCR)检测α平滑肌肌动蛋白(α-SMA)以验证转分化成功。采用斑点印迹、RT-qPCR、Western blot检测N6-甲基腺苷(m6A)修饰水平及相关酶[Mettl3、Mettl14、Wilms肿瘤蛋白1相关蛋白(WTAP)、肥胖相关蛋白(FTO)、ALKB同源蛋白5(ALKBH5)、YTH结构域家族蛋白(YTHDF)1、YTHDF2、YTHDC1、YTHDC2、YTHDC3]的表达水平。采用慢病毒转染Mettl3 shRNA的方法抑制细胞中的Mettl3表达,为sh-Mettl3组、Ang Ⅱ+sh-Mettl3组;以转染慢病毒空载体作为阴性对照,为Ang Ⅱ+sh-NC组,观察Ang Ⅱ对周细胞转分化的影响,并检测下游磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(AKT)信号通路蛋白的表达,包括PI3K、AKT(丝氨酸磷酸化位点473)[p-AKT(S473)]、AKT(苏氨酸磷酸化位点308)[p-AKT(T308)]等。加入放线菌素D与各组周细胞共培养以抑制PI3K基因的转录,通过检测不同共培养时间残余PI3K mRNA表达量以计算PI3K mRNA半衰期。在Ang Ⅱ+sh-Mettl3组周细胞中加入AKT激动剂SC79,观察是否可逆转Mettl3抑制对Ang Ⅱ诱导周细胞-肌成纤维细胞转分化的作用。(2)动物实验中,分为假手术组(仅给予0.9%无菌生理盐水)、Ang Ⅱ组(泵入Ang Ⅱ溶液)、sh-Mettl3组(注射Mettl3 shRNA慢病毒溶液)、Ang Ⅱ+sh-Mettl3组(泵入Ang Ⅱ溶液+注射Mettl3 shRNA慢病毒溶液)、Ang Ⅱ+sh-Mettl3+SC79组(Ang Ⅱ+sh-Mettl3组处理基础上注射SC79),每组6只。手术前后采用尾夹法测定血压,术后4周测定血清肌酐、尿素含量及尿液白蛋白含量。28 d后取肾脏组织,采用苏木精-伊红(HE)及Masson三色法对组织切片进行染色,检测肾脏纤维化程度。结果:(1)磁珠分选法可获得原代肾脏周细胞,以1×10 6 mmol/L的Ang Ⅱ处理周细胞48 h可成功诱导其向肌成纤维细胞转分化。斑点印迹结果显示,Ang Ⅱ组总RNA的m6A修饰水平高于对照组( P<0.05),RT-qPCR及Western blot结果显示,与对照组相比,Ang Ⅱ组中Mettl3 mRNA及蛋白表达水平上调( P均<0.05)。Ang Ⅱ+sh-Mettl3组细胞中的Mettl3蛋白表达水平低于Ang Ⅱ组,α-SMA、波形蛋白、肌间线蛋白、成纤维细胞激活蛋白a(FAPa)以及Ⅰ型胶原蛋白的表达水平亦低于Ang Ⅱ组( P均<0.05)。与对照组相比,Ang Ⅱ组的PI3K mRNA表达水平上调,且p-AKT(S473)和p-AKT(T308)蛋白高表达;Ang Ⅱ+sh-Mettl3组的PI3K mRNA表达水平低于Ang Ⅱ组,p-AKT(S473)和p-AKT(T308)蛋白表达下调( P均<0.05)。Ang Ⅱ+sh-Mettl3组的半衰期短于Ang Ⅱ+sh-NC组(2.34 h比3.42 h)。而Mettl3抑制对Ang Ⅱ诱导周细胞-肌成纤维细胞转分化的改善作用可被SC79逆转。(2)动物实验结果显示,与假手术组比较,Ang Ⅱ组小鼠的血压更高,血清肌酐、尿素及24 h尿蛋白测量值更高,纤维化面积更大( P均<0.05);而Ang Ⅱ+sh-Mettle3组的纤维化面积小于Ang Ⅱ组( P<0.05),但在加入SC79后肾脏纤维化又加重。结论:Mettl3介导的RNA m6A表观遗传调控参与Ang Ⅱ诱导的肾脏周细胞-肌成纤维细胞转分化及肾脏纤维化,Mettl3可能通过影响PI3K的稳定性,进而影响PI3K/AKT信号通路发挥调控作用。

Residential relocation in response to light rail transit investment:case study of the Hudson–Bergen Light Rail system

It is widely acknowledged that the improved accessibility enabled by investment in public transport services can, under favorable market conditions, impact the local real estate market within the zone of influence of the service＇s stations. The motivation for this study is to establish the nature of two such impacts, specifically the spatial and socio-economic patterns of residential relocations that are driven by the new light rail transit （LRT） service. Using empirical data （n = 1,023） from the Hudson-Bergen Light Rail system in New Jersey （US）, we report findings regarding the impacts of the introduction of the new LRT service. We investigate two linked dimensions; the first is the distinctive socio-economic profile of LRT passengers who self-report having relocated to the new transit corridor due, at least in part, to the new transit service. The second is their proximity （following their resi- dential relocation） to the new LRT line＇ s stations. We present a novel analysis that accounts for endogeneity between these two dimensions of residential relocation. Of light rail passengers who engaged in a residential relocation in the 5 years prior to the survey, two-thirds （69 %） indicate that proximity to the light rail service was a ＇somewhat＇ or ＇very＇ important consideration. Via the multivariate analysis, we demonstrate that small household size, low income, youth （as opposed to older age）, and low car ownership are each positively linked, ceteris paribus, with having engaged in a residential reloca- tion motivated by the new transit service. Finally, higher household income is found to be associated with distance （after relocation） to the nearest transit station, which is consistent with bid-rent theory.

To the Editor:Anterior cervical discectomy and fusion(ACDF)is a"gold standard"for patients with spondylotic myelopathy when conservative treatments fail.[1]A zeroprofile,standalone device(Zero P;Synthes GmbH,Zuchwil,Switzerland)is more popular than the traditional plate and cage for ACDF.[2]Yun et al[3]showed that the insertion point of Zero-P in the intervertebral disc space might be an important factor for maintenance of anterior disc ***,no consensus exists regarding the position related to better clinical and radiological results.
