We employed a multiplex polymerase chain reaction （PCR） coupled with capillary electrophoresis （mPCR-CE） targeting six Clostridium difficile genes, including tPi, tcdA, tcdB, cdtA, cdtB, and a deletion in tcdC for simultaneous detection and characterization of toxigenic C. difficile directly from fecal specimens. The mPCR-CE had a limit of detection of 10 colony-forming units per reaction with no cross-reactions with other related bacterial genes. Clinical validation was performed on 354 consecutively collected stool specimens from patients with suspected C. difficile infection and 45 isolates. The results were compared with a reference standard combined with BD MAX Cdiff, real-time cell analysis assay （RTCA）, and mPCR-CE. The toxigenic C. difficile species were detected in 36 isolates and 45 stool specimens by the mPCR-CE, which provided a positive rate of 20.3% （81/399）. The mPCR-CE had a specificity of 97,2% and a sensitivity of 96.0%, which was higher than RTCA （χ^2 = 5.67, P = 0.017） but lower than BD MAX Cdiff （P = 0.245）. Among the 45 strains, 44 （97.8%） were determined as non- ribotype 027 by the mPCR-CE, which was fully agreed with PCR ribotyping. Even though ribotypes 017 （n = 8, 17.8%）, 001 （n = 6, 13.3%）, and 012 （n = 7, 15.6%） were predominant in this region, ribotype 027 was an important genotype monitored routinely. The mPCR-CE provided an alternative diagnosis tool for the simultaneous detection of toxigenic C. difficile in stool and potentially differentiated between RT027 and non-RT027.

In order to enhance the service life of refractories for melting and casting of titanium alloy,preparation of yttrium oxide coatings on the refractories surface is an effective *** improvement of thermal shock stability of the coating is beneficial to its spalling resistance during the high-temperature service,and the available slurry with good performance is the *** effects of the varieties and contents of dispersant(sodium tripolyphosphate,sodium hexametaphosphate,sodium pyrophosphate,citric acid and polycarboxylate ammonium salt)and binder(aluminum dihydrogen phosphate,zirconium acetate and yttria sol)on the stability,rheological and thixotropic properties of yttrium oxide waterbased slurry were investigated,and the effects of slurry composition design on the thermal shock resistance of the coating were *** results showed that the introduction of polycarboxylate ammonium salt as dispersant significantly improved the stability of the slurry,and the varieties of the binders had significant effects on the dispersibility of the *** aluminum dihydrogen phosphate and polycarboxylate ammonium salt were used as binder and dispersant,respectively,the slurry had better stability,suitable rheological and thixotropic *** using aluminum dihydrogen phosphate as binder,the coating had good thermal shock resistance and no obvious cracks were observed for the coating after thermal shock,which was attributed to the yttrium aluminate binding phase generated in the *** effective method was provided for preparing yttrium oxide coatings with enhanced thermal shock resistance applied in the process of titanium alloy melting and casting.

The emergence of nucleic acid-based molecular techniques has significantly enhanced laboratory diagnosis and monitoring of atypical pneumonia. These techniques have not only provided rapid and sensitive detection of fastidious microbial organisms but have also played critical roles in identifying and characterizing emerging patho- gens that cause atypical pneumonia. Other benefits that molecular techniques can bring to the field include organism differentiation, quantitation, typing, and antibiotic resistance profiles. Gradually becoming standardized and widely available, the future will see some promising molecular methods become a mainstay in clinical laboratories for recognition and diagnosis of atypical pneumonia pathogens.

Mitochondrial base editing tools hold great promise for the investigation and treatment of mitochondrial diseases. Mitochondrial DNA base editors(mitoBEs) integrate a programmable transcriptionactivator-like effector(TALE) protein with single-stranded DNA deaminase(TadA8e-V106W,APOBEC1, etc.) and nickase(MutH, ***6I(C), etc.) to achieve heightened precision and efficiency in mitochondrial base editing. This innovative mitochondrial base editing tool exhibits a number of advantages, including strand-selectivity for editing, high efficiency, and the capacity to perform diverse types of base editing on the mitochondrial genome by employing various deaminases. In this context,we provide a detailed experimental protocol for mitoBEs to assist others in achieving proficient mitochondrial base editing.
