Exhausted T cells(T) is a hallmark of cancer and chronic viral infection,and a barrier to cancer *** underlying molecular mechanism in regulation of T in cancer microenvironments is still largely ***,we firstly observed a T phenotype,including increased frequency of effector memory(CD44CD62 Land CD45 ROCD45 RA)cell subset and reduced frequency of central memory(CD44CD62 L) and na?ve(CD44"CD62 L or CD45 ROCD45 RA)cell subsets,the highly expressed suppressive molecules including CD39,PD-1 and TIM-3,and decreased secretion of cytokines IFN-gama and Granzyme B(GrB) in peripheral blood and tumor-infiltrating T cells from NPC ***,we found that the exosomal miR-24,which was identified to enrich in exosomes from nasopharyngeal carcinoma(NPC) cells and resulting to T cell dysfunction in our previous study,induced T cell progressing to T and the reprograming the energy metabolism including Oxidative phosphorylation and extra-mitochondrial glycolysis during the process of T cell exhaustion in ***,we found the morphology of mitochondria became large and tubular,and the expression of mitochondrial constituent and dynamic related proteins including Mfn1 and 2,P-DRP1,TOMM20 was increased in T cells once blocking endogenous *** oligomycin,a inhibitor of mitochondria,promoted T cell progressing into T in ***,we identified that the direct target genes of miR-24 including FGF11 and MYC was involved in the regulation of miR-24-mediated T and energy metabolism ***,these finding demonstrate a newly molecular link between exosomal miR-24,target genes(FGF11 and MYC) and energy metabolism in the development of T,which will help to break the immunosuppression in NPC microenvironments and improve the effect of immunotherapy in NPC.

Type 1 iodothyronine deiodinase activity and mRna expression in rat thyroid tissue with different iodine intakes

Background Type 1 deiodinase （D1） plays an important role in the metabolism of thyroid hormone and has close relationship with thyroid function. In this study we explore the effects of iodine intake on D1 activity and its mRna expression and its possible mechanism. Methods Forty-eight Wistar rats were randomly divided into six groups with 8 in each： low iodine （LI）, normal iodine （NI）, five-fold iodine （HI5）, ten-fold iodine （HI10）, fifty-fold iodine （HI50）, one hundred-fold iodine （HI100） group. Three months, six months and twelve months after admistration of potassium iodate, they were sacrificed and thyroids were excised. The expression of D1 mRna in the thyroid tissue was determined by RT-PCR and D1 activity was analyzed by ^125I-rT3 as substrate. The thyroid hormone was measured with radioimmunoassay method. Results Compared with NI group, D1 mRna expression in LI groups slightly decreased, and D1 activity greatly increased. Both T3 and T4 in thyroid tissue significantly decreased, but the T3/T4 ratio increased. D1 mRna expression decreased in all HI groups, and D1 activity was significantly lower in HI groups. There was a tendency of decrease in D 1 activity with increased doses of iodine intakes. There was no significant difference in T4 in thyroid tissue between HI groups and NI group, but a tendency of decrease in T3 level was found in all HI groups. Conclusions In the case of iodine deficiency, D 1 activity increased greatly in order to convert more T4 to T3. Excess iodine can inhibit both D1 mRna expression and its activity to protect organism from being injured by excessive T3.

为评估本课题组研发的新型可动叶片车轮ALW(Active Lugged Wheel)与土壤交互作用的力学特性,搭建了ALW-土壤交互作用力学测量平台,测量了采用不同叶片运动轨迹的ALW机构与砂土间相互作用所产生的牵引力和垂直升力,将其与光滑车轮和单个叶片产生的土壤反作用力分别进行比较,辨识了车轮表面和叶片与砂土的耦合作用影响.根据测量结果进一步分析了叶片倾斜角度和沉陷长度对土壤反作用力的影响,并通过最小二乘法构建了叶片沉陷长度与土壤反作用力相关性的数学模型.结果表明:当车轮转角处于一定范围内时,ALW产生的牵引力和垂直升力比光滑车轮和单个叶片产生的作用力的总和更大,因此推断车轮表面和叶片与砂土的耦合作用会影响砂土的变形形式,ALW可以利用砂土的这种变形特性增大土壤反作用力;ALW受到的牵引力和竖直升力与叶片的倾斜角度没有明显关系,而与叶片的沉陷长度呈二次函数关系,因此可以通过主动调节叶片的沉陷长度来提高ALW机构在软土上的运动性能.

The back-streaming white-neutron beamline(Back-n)of the China Spallation Neutron Source is an essential neutronresearch platform built for the study of nuclear data,neutron physics,and neutron *** types of cross-sectional neutron-reaction measurements have been performed at Back-n since early *** measurements have shown that a significant number of gamma rays can be transmitted to the experimental stations of Back-n along with the neutron *** gamma rays,commonly referred to as in-beam gamma rays,can induce a non-negligible experimental background in neutron-reaction *** the characteristics of in-beam gamma rays is important for understanding the experimental ***,measuring in-beam gamma rays is challenging because most gamma-ray detectors are sensitive to neutrons;thus,discriminating between neutron-induced signals and those from in-beam gamma rays is *** this study,we propose the use of the black resonance filter method and a CeBr_(3) scintillation detector to measure the characteristics of the in-beam gamma rays of *** types of black resonance filters,^(181)Ta,^(59)Co,^(nat)Ag,and^(nat)Cd,were used in this *** time-of-flight(TOF)technique was used to select the detector signals remaining in the absorption region of the TOF spectra,which were mainly induced by in-beam gamma *** energy distribution and flux of the in-beam gamma rays of Back-n were determined by analyzing the deposited energy spectra of the CeBr_(3) scintillation detector and using Monte Carlo *** on the results of this study,the background contributions from in-beam gamma rays in neutron-reaction measurements at Back-n can be reasonably evaluated,which is beneficial for enhancing both the experimental methodology and data analysis.

Identifying the dynamic behaviors in complete reconstruction of Co-based complex precatalysts during electrocatalytic oxygen evolution

Transition metal-based nanomaterials have emerged as promising electrocatalysts for oxygen evolution reaction（OER）. Considerable research efforts have shown that self-reconstruction occurs on these nanomaterials under operating conditions of OER process. However, most of them undergo incomplete reconstruction with limited thickness of reconstruction layer, leading to low component utilization and arduous exploration of real catalytic mechanism. Herein, we identify the dynamic behaviors in complete reconstruction of Co-based complexes during OER. The hollow phytic acid（PA） cross-linked CoFe-based complex nanoboxes with porous nanowalls are designed because of their good electrolyte penetration and mass transport ability, in favor of the fast and complete reconstruction. A series of experiment characterizations demonstrate that the reconstruction process includes the fast substitution of PA by OH-to form Co（Fe）(OH)xand subsequent potential-driven oxidation to Co（Fe）OOH. The obtained CoFeOOH delivers a low overpotential of 290 mV at a current density of 10 mA cm-2and a long-term *** experiment results together with theory calculations reveal that the Fe incorporation can result in the electron rearrangement of reconstructed CoFeOOH and optimization of their electronic structure,accounting for the enhanced OER activity. The work provides new insights into complete reconstruction of metal-based complexes during OER and offers guidelines for rational design of high-performance electrocatalysts.

Maximizing biomass utilization: An integrated strategy for coproducing multiple chemicals

Lignocellulosic biomass is one of the viable solutions to alleviate the global warming. However, the limited utilization of biomass majorly focused on cellulose and hemicellulose restricts the economic and environmental feasibilities. To cope with this issue, we proposed an integrated process of co-producing 1,6-hexanediol（1,6-HDO） with tetrahydrofuran and adipic acid from biomass, referred to as Strategy A. To compare the impacts of lignin upgrading and feedstock, Strategy B, which co-produces tetrahydrofuran alone, and Strategy C, which is the traditional route to produce 1,6-HDO from fossil fuels, were used. Heat networks are also designed to reduce operating costs and indirect carbon emissions due to energy consumption, saving 87% and 83% of the heat and cooling requirements, respectively, in Strategy A. The market competitiveness of Strategy A was evaluated by determining the minimum selling price through techno-economic analysis, and sustainability was thoroughly investigated by quantifying the environmental impacts through both midpoint and endpoint life-cycle assessments（LCAs）.Strategy A was found to be the most favorable both economically（USRDSCHARDOLLAR3,402/ton） and environmentally（-26.9 kg CO2eq.）. This indicates that lignin valorization is not only economically but also environmentally preferred. Finally, changes in economic and environmental feasibilities depending on economic, process, and environmental parameters were investigated using sensitivity and uncertainty analyses. The results of these analyses provide valuable insight into bio-based chemical production.

Statistical characteristics and classification of ionospheric mid-latitude trough as revealed by the observations of DMSP-F18

Statistical characteristics and the classification of the topside ionospheric mid-latitude trough are systemically analyzed,using observations from the Defense Meteorological Satellite Program F18(DMSP-F18)*** data was obtained at an altitude of around 860 km in near polar orbit,throughout *** study identified the auroral boundary based on the in-situ electron density and electron spectrum,allowing us to precisely determine the location of the mid-latitude *** differs from most previous works,which only use Total Electron Content(TEC)or in-situ electron *** our study,the troughs exhibited a higher occurrence rate in local winter than in summer,and extended to lower latitudes with increasing geomagnetic *** was found that the ionospheric mid-latitude trough,which is associated with temperature changes or enhanced ion drift,exhibited distinct ***,the ionospheric mid-latitude troughs related to electron temperature(Te)peak were located more equatorward of auroral oval boundary in winter than in *** ionospheric mid-latitude troughs related to Te-maximum were less frequently observed at 60−70°S magnetic latitude and 90−240°E ***,the troughs related to ion temperature(Ti)maximums were observed at relatively higher latitudes,occurring more frequently in *** addition,the troughs related to ion velocity(Vi)maximums could be observed in all *** troughs with the maximum-Ti and maximum-Vi were located closer to the equatorward boundary of the auroral oval at the nightside,and in both *** implies that enhanced ion drift velocity contributes to increased collisional frictional heating and enhanced ion temperatures,resulting in a density depletion within the trough region.

It has been reported that the PI3K/AKT signaling pathway plays a key role in the pathogenesis of ischemic *** a result,the development of drugs targeting the PI3K/AKT signaling pathway has attracted increasing attention from *** article reviews the pathological mechanisms and advancements in research related to the signaling pathways in ischemic stroke,with a focus on the PI3K/AKT signaling *** key findings include the following:(1)The complex pathological mechanisms of ischemic stroke can be categorized into five major types:excitatory amino acid toxicity,Ca^(2+)overload,inflammatory response,oxidative stress,and apoptosis.(2)The PI3K/AKT-mediated signaling pathway is closely associated with the occurrence and progression of ischemic stroke,which primarily involves the NF-κB,NRF2,BCL-2,mTOR,and endothelial NOS signaling pathways.(3)natural products,including flavonoids,quinones,alkaloids,phenylpropanoids,phenols,terpenoids,and iridoids,show great potential as candidate substances for the development of innovative anti-stroke medications.(4)Recently,novel therapeutic techniques,such as electroacupuncture and mesenchymal stem cell therapy,have demonstrated the potential to improve stroke outcomes by activating the PI3K/AKT signaling pathway,providing new possibilities for the treatment and rehabilitation of patients with ischemic *** investigations should focus on the direct regulatory mechanisms of drugs targeting the PI3K/AKT signaling pathway and their clinical translation to develop innovative treatment strategies for ischemic stroke.

区域生态安全格局构建作为景观生态学的研究热点之一,为区域可持续发展和国土生态屏障建设提供了重要途径,尤其对于灾害频发、生态脆弱的滇中地区,其能够有效规避自然灾害、促进区域生态系统和社会经济系统协调发展.本文以玉溪市为例,针对区域生态环境基底特征,选取水源涵养、固碳释氧、土壤保持和生物多样性等生态系统服务评估自然生境重要性,综合考虑单一类型生态系统服务质量和多功能性识别生态源地;并采用地质灾害敏感性修正基于地类赋值的基本阻力面,运用最小累积阻力模型识别生态廊道,从而构建玉溪市生态安全格局.结果表明:玉溪市生态源地斑块数量为81个,占玉溪市土地总面积的38.4%,与各级自然保护区重合率达75.2%,主要分布在市域西部的山林地和东部的湖泊湿地区域;玉溪市生态廊道总长度1642.04 km,呈"一横三纵"的空间分布格局,沿河谷、断陷盆地等植被覆盖较好的区域延伸.本文针对滇中山地生态脆弱区地质灾害频发特征,构建山地生态安全格局,对于区域山地开发与生态保护能够提供决策指引.
