Endoscopic ultrasound elastography for evaluation of lymph nodes and pancreatic masses:A multicenter study

AIM:To evaluate the ability of endoscopic ultrasound(EUS) elastography to distinguish benign from malignant pancreatic masses and lymph ***:A multicenter study was conducted and included 222 patients who underwent EUS examination with assessment of a pancreatic mass(n=121) or lymph node(n=101).The classification as benign or malignant,based on the real time elastography pattern,was compared with the classif ication based on the B-mode EUS images and with the fi nal diagnosis obtained by EUS-guided fi ne needle aspiration(EUS-FNA) and/or by surgical *** interobserver study was ***:The sensitivity and specificity of EUS elastography to differentiate benign from malignant pancreatic lesions are 92.3% and 80.0%,respectively,compared to 92.3% and 68.9%,respectively,for the conventional B-mode *** sensitivity and specificity of EUS elastography to differentiate benign from malignant lymph nodes was 91.8% and 82.5%,respectively,compared to 78.6% and 50.0%,respectively,for the B-mode *** kappa coefficient was 0.785 for the pancreatic masses and 0.657 for the lymph ***:EUS elastography is superior compared to conventional B-mode imaging and appears to be able to distinguish benign from malignant pancreatic masses and lymph nodes with a high sensitivity,specificity and *** might be reserved as a second line examination to help characterise pancreatic masses after negative EUS-FNA and might increase the yield of EUS-FNA for lymph nodes.

The medicinal mushroom Ganoderma lucidum(GL,Reishi or Lingzhi)exhibits an inhibitory effect on ***,the underlying mechanism of the antitumor activity of GL is not fully *** this study,we characterized the gene networks regulated by a commercial product of GL containing a mixture of spores and fruiting bodies namely“GLSF”,in colorectal *** found that in vitro co-administration of GLSF extract at non-toxic concentrations significantly potentiated growth inhibition and apoptosis induced by paclitaxel in CT26 and HCT-15 *** inhibited NF-κB promoter activity in HEK-293 cells but did not affect the function of P-glycoprotein in K562/DOX ***,we found that when mice were fed a modified diet containing GLSF for 1 month prior to the CT26 tumor cell inoculation,GLSF alone or combined with Nab-paclitaxel markedly suppressed tumor growth and induced ***-seq analysis of tumor tissues derived from GLSF-treated mice identified 53 differentially expressed genes compared to normal *** of the GLSFdown-regulated geneswere involved in NF-κB-regulated inflammation pathways,such as IL-1β,IL-11 and *** enrichment analysis suggested that several inflammatory pathways involving leukocytemigration and adhesion were most affected by the *** analysis predicted activation of multiple tumor suppressors such asα-catenin and TP53 and inhibition of critical inflammatory mediators.“Cancer”was the major significantly inhibited biological effect of GLSF *** results demonstrate that GLSF can improve the therapeutic outcome for colorectal cancer through a mechanism involving suppression of NF-κB-regulated inflammation and carcinogenesis.

Evaluation of HemogloBind<sup>TM</sup>treatment for preparation of samples for cholinesterase analysis

Acetylcholine is an essential neurotransmitter found throughout the nervous system. Its action on postsynaptic receptors is regulated through hydrolysis by various carboxylesterases, especially cholinesterases (ChEs). The acute toxicity of organophosphate (OP) compounds is directly linked to their action as inhibitors of ChE. One widely used assay for evaluating ChE activity is a spectrophotometric method developed by Ellman et al. When the enzyme source is from tissues or, in particular, blood, hemoglobin displays a spectrophotometric peak at the same wave-length used to analyze cholinergic activity. This creates a substantial background that interferes with the Ellman’s assay and must be overcome in order to accurately monitor cholinesterase activity. Herein, we directly compare blood processing methods: classical method (1.67 ± 0.30 U/mL) and HemogloBindTM treatment (1.51 ± 0.17 U/mL), and clearly demonstrate that pretreatment of blood samples with HemoglobindTM is both a sufficient and rapid sample preparation method for the assessment of ChE activity using the Ellman’s method.
