Impact of the national hepatitis B immunization program in China:a modeling study

Background:Elimination of hepatitis B virus(HBV)is a striking challenge for countries with high or moderate disease burden.Therefore,using China as a practical case to share experiences for similar countries may accelerate the achievement of the WHO 2030 target of 90%reduction in HBV-related incidence.We aim to evaluate the impact of national HBV immunization strategies in China;and the feasibility to achieve WHO 2030 targets under diferent scenarios.Methods:We constructed an expanded Susceptible-Exposed-Infectious-Recovered(SEIR)model and decision treeMarkov model to estimate the epidemic of HBV in China,assess the feasibility of 2030 Elimination Goals through the projections and conduct the economic analysis.Least square method was used to calibrate the expanded SEIR model by yearly data of laboratory-confrmed HBV cases from 1990 to 2018.Two models were separately used to evaluate the impact and cost-efectiveness of HBV vaccine by comparing prevalence of chronic HBV infections,qualityadjusted life-years(QALYs),incremental cost efectiveness ratio and beneft–cost ratio(BCR)under various intervention options,providing a basis for exploring new containment strategies.Results:Between 1990 and 2020,the number of chronic HBV infections decreased by 33.9%.The current status quo would lead to 55.73 million infections(3.95%prevalence)in 2030,compared to 90.63 million(6.42%prevalence)of the“Without the NIP”scenario(NIP:National Immunization Program),114.78 million(8.13%prevalence)without any interventions.The prevention of mother to child transmission(PMTCT)strategy showed a net beneft as 12,283.50 dollars per person,with BCR as 12.66,which is higher than that of universal vaccination at 9.49.Compared with no screening and no vaccination,the PMTCT strategy could save 7726.03 dollars for each QALY increase.Conclusions:Our fndings proved the HBV vaccination has demonstrated a substantial positive impact on controlling the epidemic of HBV in terms of efectiveness and economy after about 30 years of implementation of the nati

In recent years,solid-state nanopore analysis technology has begun to show increasing intersections with nucleic acid self-assembly research.A series of breakthroughs have shown that solid-state nanopores are expected to locate accidental folded(knots) configurations in long-chain nucleic acids without label modification, as well as to distinguish precise nucleic acid assemblies with different 3D morphology. On this basis,our research group are focusing on the applications of solid-state nanopore in analysis and characterization of complicated nucleic acid assembly behaviors at a single molecular level(Fig.1).It is demonstrated that the solid-state nanopores can provide electrophoresis-like,but more sensitive characterization results for recognizing linear DNA assembly mixtures3,determining length-distribution of the mixtures,and even revealing the approximate information of folding structures. Further,those DNA self-assembly behaviors with amplification functions are used as sensitization/transduction probes to help the solid-state nanopores to perform separation-free and label-free detection for small molecule analytes that are originally lower than their spatial or temporal resolution.

Impact of immunization programs on 11 childhood vaccine-preventable diseases in China: 1950–2018

To evaluate the achievements of China’s immunization program between 1950 and 2018,we chose 11 vaccine-preventable diseases(VPDs)as representative notifiable diseases and used annual surveillance data obtained between 1950 and 2018 to derive disease incidence and mortality trends.Quasi-Poisson and polynomial regression models were used to estimate the impacts of specific vaccine programs,and life-table methods were used to calculate the loss of life expectancy,years of life lost,and loss of working years.The total notification number for the 11 VPDs was 211,866,000 from 1950 to 2018.The greatest number occurred in 1959,with a total incidence of 1,723 per million persons.From 1978 to 2018,a substantial decline was observed in the incidence of major infectious diseases.The incidence of pertussis fell 98%from 126.35 to 1.58 per million,and the incidences of measles,meningococcal meningitis,and Japanese encephalitis fell 99%,99%,and 98%,respectively.The regression models showed that most of the 11 diseases exhibited dramatic declines in morbidity after their integration into the Expanded Program on Immunization(EPI),while varicella and paratyphoid fever,which were not integrated into the EPI,showed increased morbidity.From 1978 to 2018,the total life expectancy for the 11 VPDs increased by 0.79 years,and similar results were obtained for different age groups.China has had great success in controlling VPDs in recent decades,and improving vaccination coverage is a key aspect of controlling VPDs in China.

目的分析2004-2016年我国结核病登记病例的时空分布特征,探测聚集区域,为结核病防控提供理论依据。方法利用ArcGIS 10.0软件作为数据管理和呈现的平台,建立我国2004-2016年结核病空间分析数据库,对结核病疫情进行空间自相关分析,采用SaTScan 9.6软件进行时空扫描分析。结果2004-2016年全国共登记结核病病例13157794例,全国年均登记率为75.90/10万(27.95/10万~180.82/10万)。全局空间自相关结果显示结核病发病呈聚集性分布,局部Moran’s I自相关分析结果表明,新疆、西藏、贵州、广西和海南(省、自治区)为高-高聚集区域,北京、河北、天津、山东、江苏、上海(省、直辖市)为低-低聚集区域;局部G统计量热点分析结果显示,全国结核病疫情存在15个"热点"区域,其中3个"正热点"区域分别为新疆、西藏和海南(省、自治区),12个"负热点"区域分别为北京、天津、辽宁、内蒙古、河北、山东、江苏、安徽、上海、山西、河南和吉林(省、自治区、直辖市)。利用SaTScan 9.6软件进行分阶段时空扫描分析,3个阶段共探测出12个聚集区域,每个聚集区域差异均有统计学意义(均P<0.05)。结论2004-2016年我国结核病疫情呈现逐年下降的趋势,各省(自治区、直辖市)的年均登记率并非随机分布,呈明显的空间聚集性,分阶段时空扫描聚集区域逐渐减少,结核病防治工作取得一定进展,但高风险地区仍持续存在,需重点关注并采取针对性防控措施。

Oryza longistaminata is a close wild relative of the African cultivated rice and has several hallmark biological traits,such as rhizomatousness,self-incompatibility and high resistances to biotic and abiotic stresses. Deciphering genome of O. longistaminata has been hindered by the high heterozygosity of this species,which is a result of its inherent self-incompatibility. Here,we present the reference genome of O. longistaminata based on a large quantity of combined sequencing data(96× Illumina short reads and .9× Roche GS FLX+ long reads) and compared genomics revealed more resistance(R) genes/families. By comparing transcriptomes of 4 pairs of tissues related to the presence/absence of rhizomes or self-incompatibility,we identified candidate genes related to rhizomatousness and self-incompatibility. In aggregate,the draft genome and large quantity of transcriptome data of O. longistaminata provide a basic evidentiary foundation for targeted studies into the genes underlying its valuable phenotypic traits,future gene mining or breeding efforts,and further study into the evolution of African rice and the Oryza genus.

目的分析1990-2017年我国乙型肝炎(乙肝)发病和死亡的变化情况,为制定防控乙肝疫情的策略提供合理依据。方法利用中国公共卫生科学数据中心的乙肝发病和死亡数据及国家统计局的人口学资料,采用Excel 2016软件分别建立乙肝报告发病和死亡的数据库。利用Joinpoint 4.***.0.1软件进行Joinpoint回归模型分析,估计1990-2017年我国乙肝不同年龄组、不同省份年报告发病率和年报告死亡率的平均年度变化百分比,进一步分析乙肝疫情的变化趋势。采用R 3.6.2软件进行统计学分析和数据的可视化。结果从1990年1月至2017年12月共报告20793233例乙肝病例(年平均报告发病率为58.19/10万)。年平均报告发病率最高的年龄组是25~29岁(119.67/10万),≥45岁年龄组年报告发病率均呈上升趋势,≥85岁年龄组的报告病死率达最高值(2.26/1000)。有23个省份的报告发病率呈增长趋势(P0.05),2个省份的报告发病率呈下降趋势(P<0.05)。结论1990-2017年中国乙肝主要发病人群已后移至成年人群,且该人群的报告发病率呈上升趋势;东部地区的部分省份防控措施落实到位,治疗水平较高,乙肝疫情已趋于平稳;西部地区大部分省份仍处上升趋势。应进一步针对不同省份的疫情现况,因地制宜地制定合适的防治策略。

目的探讨我国5种病毒性肝炎(肝炎)的时间序列特征,并通过有效的模型预测其发病率。方法按照甲型肝炎、乙型肝炎、丙型肝炎、戊型肝炎和未分型肝炎5种不同类型肝炎分类方式收集2009-2018年的月度发病数据,进行描述性和时间序列研究,采用趋势分解法以季节指数形式表示时间序列中的季节性,以线性回归模型表示其长期趋势,为每种肝炎建立差分自回归移动平均(ARIMA)模型。结果2009-2018年报告肝炎14856990例,5种肝炎季节指数的极差均<1,戊型肝炎的季节性特征较为显著,其发病呈单峰型,其余4种肝炎的季节性特征一般。甲型肝炎、戊型肝炎和未分型肝炎的发病基本趋于平稳,在一个较低的水平上呈缓慢下降趋势,乙型肝炎发病数在5种肝炎中占比最高(79.59%,11824262/14856990),但其下降趋势也为各型肝炎中最快(-0.01/10万)。丙型肝炎发病呈不断上升的趋势,上升速率一直保持稳定(0.005/10万)。ARIMA模型拟合的2009年1月至2018年12月的预测值与实际值较一致,平均绝对误差百分比范围为3.7568~8.0683。结论对于法定报告传染病监测数据的时间序列分析有助于更好地了解我国肝炎的发病特征,ARIMA模型可用于我国肝炎的短期预测,具有较好的应用价值。

为了从海量的移动目标轨迹数据中识别异常轨迹点数据,提出了一种基于自适应单类支持向量机的轨迹异常点检测方法。首先,提取轨迹点的运动特征构造特征向量作为模型输入;其次,基于粒子群算法构造最优单类支持向量机模型;最后,利用最优单类支持向量机模型识别异常轨迹点。实验结果表明,新提出的方法能够自适应地构造最优单类支持向量机模型,并有效识别轨迹数据中的异常点,具有很好的自适应性与准确性。

PITPNC1 promotes the thermogenesis of brown adipose tissue under acute cold exposure

Brown adipose tissue (BAT) plays an essential role in non-shivering thermogenesis. The phosphatidylinositol transfer protein, cytoplasmic 1 (PITPNC1) is identified as a lipid transporter that reciprocally transfers phospholipids between intracellular membrane structures. However, the physiological significance of PITPNC1 and its regulatory mechanism remain unclear. Here, we demonstrate that PITPNC1 is a key player in thermogenesis of BAT. While Pitpnc1^(−/−) mice do not differ with wildtype mice in body weight and insulin sensitivity on either chow or high-fat diet, they develop hypothermia when subjected to acute cold exposure at 4℃. The Pitpnc1^(−/−) brown adipocytes exhibit defective β-oxidation and abnormal thermogenesis-related metabolism pathways in mitochondria. The deficiency of lipid mobilization in Pitpnc1^(−/−) brown adipocytes might be the result of excessive accumulation of phosphatidylcholine and a reduction of phosphatidic acid. Our findings have uncovered significant roles of PITPNC1 in mitochondrial phospholipid homeostasis and BAT thermogenesis.

The genus Dalbergia contains approximately 250 species with many valuable trees being destroyed by targeted and illegal logging.DNA barcoding is a reliable method for the molecular identification of different species and resources conservation.In the present study,the specimen discrimination ability of internal transcribed spacer (ITS),matK,rbcL and psbA-trnH barcoding were tested on Dalbergia sequences,downloaded from the National Center for Biotechnology Information (NCBI),and the combined barcoding ITS+matK+rbcL was used to identify unknown specimens.It was found that ITS+matK+rbcL have good discrimination rates based on the analysis methods best match (BM) and best close match (BCM).These barcodes also have the best performance concerning barcode gap distribution,and are able to discriminate unknown specimens from South-China.Furthermore,it was demonstrated that D.tamarindifolia and D.rubiginosa are also relatively close to sister-species D.pinnata and D.candenatensis within the phylogenetic Dalbergia tree.Considering the overall performance of these barcodes,we suggest that the ITS+matK+rbcL region is a suitable barcode for identifying Dalbergia species.
