Fragmentation stability and retention time-shift obtained by LC-MS/MS to distinguish sialylated N-glycan linkage isomers in therapeutic glycoproteins

Sialylated N-glycan isomers withα2-3 orα2-6 linkage(s)have distinctive roles in glycoproteins,but are difficult to ***-type(WT)and glycoengineered(mutant)therapeutic glycoproteins,cytotoxic T lymphocyte-associated antigen-4-immunoglobulin(CTLA4-Ig),were produced in Chinese hamster ovary cell lines;however,their linkage isomers have not been *** this study,N-glycans of CTLA4-Igs were released,labeled with procainamide,and analyzed by liquid chromatography-tandem mass spectrometry(MS/MS)to identify and quantify sialylated N-glycan linkage *** linkage isomers were distinguished by comparison of 1)intensity of the N-acetylglucosamine ion to the sialic acid ion(Ln/Nn)using different fragmentation stability in MS/MS spectra and 2)retention time-shift for a selective m/z value in the extracted ion *** isomer was distinctively identified,and each quantity(>0.1%)was obtained relative to the total N-glycans(100%)for all observed ionization *** sialylated N-glycan isomers with onlyα2-3 linkage(s)in WT were identified,and each isomer's sum of quantities was 50.4%.Furthermore,39 sialylated N-glycan isomers(58.8%)in mono-(3 N-glycans;0.9%),bi-(18;48.3%),tri-(14;8.9%),and tetra-(4;0.7%)antennary structures of mutant were obtained,which comprised mono-(15 N-glycans;25.4%),di-(15;28.4%),tri-(8;4.8%),and tetra-(1;0.2%)sialylation,respectively,with onlyα2-3(10 N-glycans;4.8%),bothα2-3 andα2-6(14;18.4%),and onlyα2-6(15;35.6%)linkage(s).These results are consistent with those forα2-3 neuraminidase-treated *** study generated a novel plot of Ln/Nn versus retention time to distinguish sialylated N-glycan linkage isomers in glycoprotein.

An Effective Threat Detection Framework for Advanced Persistent Cyberattacks

Recently,with the normalization of non-face-to-face online environments in response to the COVID-19 pandemic,the possibility of cyberattacks through endpoints has *** endpoint devices are managed meticulously to prevent cyberattacks and ensure timely responses to potential security *** particular,because telecommuting,telemedicine,and teleeducation are implemented in uncontrolled environments,attackers typically target vulnerable endpoints to acquire administrator rights or steal authentication information,and reports of endpoint attacks have been increasing *** persistent threats(APTs)using various novel variant malicious codes are a form of a sophisticated ***,conventional commercial antivirus and anti-malware systems that use signature-based attack detectionmethods cannot satisfactorily respond to such *** this paper,we propose a method that expands the detection coverage inAPT attack *** this model,an open-source threat detector and log collector are used synergistically to improve threat detection *** the scope of attack log collection through interworking between highly accessible open-source tools can efficiently increase the detection coverage of tactics and techniques used to deal with APT attacks,as defined by MITRE Adversarial Tactics,Techniques,and Common Knowledge(ATT&CK).We implemented an attack environment using an APT attack scenario emulator called Carbanak and analyzed the detection coverage of Google Rapid Response(GRR),an open-source threat detection tool,and Graylog,an open-source log *** proposed method expanded the detection coverage against MITRE ATT&CK by approximately 11%compared with that conventional methods.

How to improve patient satisfaction during midazolam sedation for gastrointestinal endoscopy?

AIM To determine the procedure-related factors that affect sedation satisfaction and to make a suggestion to improve *** We prospectively enrolled a total of 456 patients who underwent outpatient endoscopy procedures with midazolam sedation between March 2014 and August 2014. All patients completed both pre- and postendoscopy questionnaires about sedation expectations and satisfaction. RESULTS The study cohort included 167 (36.6%) patients whounderwent esophagogastroduodenoscopy (EGD), 167 (36.6%) who underwent colonoscopy, and 122(26.8%) who underwent a combined procedure (EGD and colonoscopy). Over 80% of all patients were satisfied with sedation using midazolam. In univariate and multivariate analyses, total procedure time in the EGD group, younger age (≤ 50 years), and longer colonoscopy withdrawal time in the colonoscopy group were related to decreased satisfaction with sedation. However, in active monitoring and intervention group, there was no decrease in grade of satisfaction despite longer procedure time due to more procedures during colonoscopy. Younger age (≤ 50 years), longer interprocedure time gap, and colonoscopy withdrawal time were related to decreased satisfaction in the combined EGD and colonoscopy group. CONCLUSION Midazolam is still a safe and effective sedative for gastrointestinal endoscopy. Satisfaction with sedation depends on several factors including age(≤ 50 years) and procedure time duration. To improve patient satisfaction with sedation, active monitoring of sedation status by the endoscopist should be considered for patients who require long procedure time.

Effects of dietary high dose DHA omega-3 supplement in dry eye with meibomian gland dysfunction

AIM:To evaluate the clinical efficacy of dietary supplement of high dose DHA omega-3 in dry eye with meibomian gland dysfunction(MGD).METHODS:Prospective randomized double-masked,placebo-controlled clinical trial was conducted in mild to moderate dry eye patients with *** have no history of taking any dietary omega-3 supplements before 3 *** were divided into two groups:24 patients in the omega-3 group and 26 patients in the placebo *** omega-3 group received two capsules of Easyeye Dry?,total containing 600 mg of EPA and 1640 mg of DHA,while the placebo group received two capsules containing 3000 mg of olive *** patients take two pills once a *** examination of MGD scores,tear break-up time(TBUT),corneal staining test(NEI),strip meniscometry(SM tube),and ocular surface disease index(OSDI)scores were performed at baseline,after 4 and 8 ***:A total of 50 patients were *** were no differences in baseline characteristics between the two groups,such as age,sex,and other ocular examination *** TBUT,NEI,and OSDI scores significantly improved after 4 and 8 wk in both *** after 8 wk TBUT(6.00±1.62 s vs 5.08±1.28 s,P=0.034)and MGD score(7.2±1.8 vs 8.1±2.6,P=0.033)in the omega-3 group was more significantly improved than that of the placebo ***:Dry eye with the MGD patient,a high dose of DHA omega-3 dietary supplement can improve TBUT and MGD score after 8 wk,effective in stabilizing the tear film.

Multi-hop relay-based maritime visible light communication

This Letter presents a multi-hop relay visible light communication （VLC） system for maritime applications. Maritime VLC systems suffer from limited coverage distance due inherently to the usage of light-emitting diodes and photodetectors. The proposed system employs a multiple of decode-and-forward relays to extend coverage distance in maritime environments. The multi-hop relay based maritime VLC is analyzed under a maritime channel modeled by the JONSWAP spectrum and gamma-gamma distribution. It is found that the use of relays in maritime environments can extend the coverage distance significantly and also improve the performance. In addition, the performance of the system is analyzed using various combining techniques at the receiver to enhance the performance. The maximal ratio combining technique is found to provide superior link quality in maritime environments.

Aluminum doping for optimization of ultrathin and high-k dielectric layer based on SrTiO3

An ultrathin SrTiO3 dielectric layer is optimized through Al doping to solve the problems existing in development of ultra-high-k oxide MOS *** post-deposition annealing,Al doping induces changes in the electronic structure of SrTiO3,thereby effectively reducing leakage current to 80) of ultrathin SrTMO3 MOS *** titanate(SrTiO3) is a high-k material but its bandgap is smaller than that of other oxide dielectrics(e.g.,SiO2,Al2 O3).Consequently,an ultrathin SrTiO3 film may have a high tunneling leakage current,which is not suitable for capacitor-based *** improve the performance of metal-oxide-semiconductor(MOS) capacitors using SrTiO3,an approach based on homogenous and uniform aluminum doping to SrTiO3 through co-sputtering is *** bandgap of a pristine SrTiO3 film showed an increase of 0.5 eV after Al ***,Al doping decreased the leakage current of SrTiO3/Si-based MOS capacitors by more than five orders of magnitude(at the level of nanoampere per square centimeter).Importantly,a dielectric constant of 81.3 and equivalent oxide thickness less than 5 A were achieved in an 8-nm-thick Al-doped SrTiO3 film owing to changes in its crystal structure and conduction band edge electronic ***,the obtained data show the effectiveness of the proposed approach for solving the problems existing in the development of ultra-high-k oxide MOS capacitors.

Air in the main pancreatic duct:A case of innocent air

Air in the main pancreatic duct has been reported only rarely and might be associated with either a spontaneous or a surgically induced alteration of the anatomy of the biliary *** report a case of "innocent" air found incidentally in the main pancreatic *** our knowledge,this is only the third such case reported.A 54-year-old woman presented with hemoptysis that had lasted for 3 *** underwent a chest computed tomography scan,which revealed not only focal bronchiectasis in the left lower lobe,but also air in the main pancreatic duct and dilatation of the common bile *** was managed conservatively for the hemoptysis and no further problems *** had no specific gastrointestinal symptoms and had no history of surgery or *** laboratory parameters were *** resonance cholangiopancreatography also demonstrated air in the main pancreatic duct and a dilated common bile duct(CBD).Duodenoscopy revealed separate biliary and pancreatic orifices with patulous openings and some air bubbles appearing in the pancreatic *** retrograde cholangiopancreatography(ERCP) showed the dilated CBD and pancreatic duct with some air bubbles,but no other abnormal *** was discharged with no further *** patients with air in the main pancreatic duct have had a pancreatobiliary disease,or a history of pancreatobiliary disease,pancreatobiliary surgery or *** the air is innocent,as in our case,ERCP should be performed to evaluate any altered sphincteric function or anatomy such as patulous openings.
