Development and application of a multi-step porcine in vitro system to evaluate feedstuffs and feed additives for their efficacy in nutrient digestion, digesta characteristics, and intestinal immune responses

In vitro model provides alternatives to the use of live animals in research. In pig nutrition, there has been a tremendous increase in in vivo research over the decades. Proper utilization of in vitro models could provide a screening tool to reduce the needs of in vivo studies, research duration, cost, and the use of animals and feeds. This study aimed to develop a multi-step porcine in vitro system to simulate nutrient digestion and intestinal epithelial immune responses affected by feedstuffs and feed additives. Seven feedstuffs(corn, corn distillers dried grains with solubles [corn DDGS], barley, wheat, soybean meal, soy protein concentrates, and Corynebacterium glutamicum cell mass [CGCM]), feed enzymes(xylanase and phytase), and supplemental amino acids(arginine, methionine, and tryptophan), were used in this in vitro evaluation for their efficacy on digestibility, digesta characteristics, and intestinal health compared with the results from previously published in vivo studies. All in vitro evaluations were triplicated. Data were analyzed using Mixed procedure of SAS9.4. Evaluations included(1) nutrient digestibility of feedstuffs,(2) the effects of feed enzymes, xylanase and phytase, on digestibility of feedstuffs and specific substrates, and(3) the effects of amino acids, arginine, tryptophan, and methionine, on anti-inflammatory, anti-oxidative, and anti-heat stress statuses showing their effects(P hosphorus release from phytate. Supplementation of arginine, tryptophan, and methionine modulated(P < 0.05) cellular inflammatory and oxidative stress responses. Despite its low water activity (Aw), food poisoning accidents associated with beef jerky consumption and problems due to mold growth have been reported. The objects of this study were to investigate the effect of yuza powder and the mixture of sodium lactate (SL) and sodium acetate (SA) to prevent the growth of S. aureus and A. flavus, as well as aflatoxin production in beef jerky. Different concentrations of the SL and SA (0, 0.75, and 1%) mixture were added to sliced beef and the Aws were adjusted to 0.70, 0.75, and 0.80. The effect of yuza powder and the SL and SA mixture on S. aureus in beef jerky (Aw 0.80) was also investigated at 10°C and 25°C. A rapid decline is that S. aureus population was observed in beef jerky containing yuza powder and mixtures of SL and SA. Antifungal effects against A. flavus were observed in beef jerky containing the SL and SA mixture at Aw below 0.75. The growth of A. flavus and aflatoxin production were prevented in beef jerky containing the yuza powder and a 1% mixture of SL and SA at 10°C and 0.80 Aw. The addition of yuza powder increased the hardness score of the beef jerky. Under current Aw regulations (0.80), beef jerky may pose a public health risk due to the growth of A. flavus and the presence of aflatoxin.

To attain a circular carbon economy and resolve CO_(2)electroreduction technology obstacles,single‐atom catalysts(SACs)have emerged as a logical option for electrocatalysis because of their extraordinary catalytic *** SACs,metal–organic frameworks(MOFs)have been recognized as promising support materials because of their exceptional ability to prevent metal *** study shows that atomically dispersed Ni single atoms on a precisely engineered MOF nanosheet display a high Faradaic efficiency of approximately 100%for CO formation in H‐cell and three‐compartment microfluidic flow‐cell reactors and an excellent turnover frequency of 23,699 h−1,validating their intrinsic catalytic *** results suggest that crystallographic variations affect the abundant vacancy sites on the MOF nanosheets,which are linked to the evaporation of Zn‐containing organic linkers during ***,using X‐ray absorption spectroscopy and density functional theory calculations,a comprehensive investigation of the unsaturated atomic coordination environments and the underlying mechanism involving CO^(*)preadsorbed sites as initial states was possible and provided valuable insights.

A conventional electrode composite for rechargeable zinc-ion batteries(ZIBs)includes a binder for strong adhesion between the electrode material and the current ***,the introduction of a binder leads to electrochemical inactivity and low electrical conductivity,resulting in the decay of the capacity and a low rate *** present a binder-and conducting agent-free VO_(2)composite electrode using in situ polymerization of dopamine on a flexible current collector of pyroprotein-based *** as-fabricated composite electrode was used as a substrate for the direct growth of VO_(2)as a self-supported form on polydopamine-derived pyroprotein-based fibers(pp-fibers@VO_(2)(B)).It has a high conductivity and flexible nature as a current collector and moderate binding without conventional binders and conducting agents for the VO_(2)(B)*** addition,their electrochemical mechanism was *** energy storage is induced by Zn^(2+)/H^(+)coinsertion during discharging,which can be confirmed by the lattice expansion,the formation of by-products including Zn_(x)(OTf)_(y)(OH)_(2x−y)·nH_(2)O,and the reduction of V^(4+)to V^(3+).Furthermore,the assembled Zn//pp-fibers@VO_(2)(B)pouch cells have excellent flexibility and stable electrochemical performance under various bending states,showing application possibilities for portable and wearable power sources.

Sleep posture surveillance is crucial for patient comfort,yet current systems face difficulties in providing compre-hensive studies due to the obstruction caused by *** posture assessment remains challenging because of the complex nature of the human body and variations in sleep ***,this study introduces an innovative method utilizing RGB and thermal cameras for comprehensive posture classification,thereby enhancing the analysis of body position and *** method begins by capturing a dataset of sleep postures in the form of videos using RGB and thermal cameras,which depict six commonly adopted postures:supine,left log,right log,prone head,prone left,and prone *** study involves 10 participants under two conditions:with and without ***,the database is normalized into a video *** subsequent step entails training a fine-tuned,pretrained Visual Geometry Group(VGG16)and ResNet50 *** the third phase,the extracted features are utilized for *** fourth step of the proposed approach employs a serial fusion technique based on the normal distribution to merge the vectors derived from both the RGB and thermal ***,the fused vectors are passed to machine learning classifiers for final *** dataset,which includes human sleep postures used in this study’s experiments,achieved a 96.7%accuracy rate using the Quadratic Support Vector Machine(QSVM)without the ***,the Linear SVM,when utilized with a blanket,attained an accuracy of 96%.When normal distribution serial fusion was applied to the blanket features,it resulted in a remarkable average accuracy of 99%.

This study explores the impact of introducing vacancy in the transition metal layer of rationally designed Na_(0.6)[Ni_(0.3)Ru_(0.3)Mn_(0.4)]O_(2)(NRM)cathode *** incorporation of Ru,Ni,and vacancy enhances the structural stability during extensive cycling,increases the operation voltage,and induces a capacity increase while also activating oxygen redox,respectively,in Na_(0.7)[Ni_(0.2)V_(Ni0.1)Ru_(0.3)Mn_(0.4)]O_(2)(V-NRM)*** analytical techniques including transmission electron microscopy,X-ray absorption near edge spectroscopy,operando X-ray diffraction,and operando differential electrochemical mass spectrometry are employed to assess changes in the average oxidation states and structural *** results demonstrate that V-NRM exhibits higher capacity than NRM and maintains a moderate capacity retention of 81%after 100 ***,the formation of additional lone-pair electrons in the O 2p orbital enables V-NRM to utilize more capacity from the oxygen redox validated by density functional calculation,leading to a widened dominance of the OP4 phase without releasing O_(2) *** findings offer valuable insights for the design of advanced high-capacity cathode materials with improved performance and sustainability in sodium-ion batteries.
