Antibody-drug conjugates(ADCs)are a new type of targeting antibodies that conjugate with highly toxic anticancer drugs via chemical linkers to exert high specificity and efficient killing of tumor cells,thereby attracting considerable attention in precise oncology ***(Cet)is a typical antibody that offers the benefits of good targeting and safety for individuals with advanced and inoperable cutaneous squamous cell carcinoma(cSCC);however,its anti-tumor activity is limited to a single ***(CisPt)shows good curative effects;however,its adverse effects and non-tumor-targeting ability are major *** this study,we designed and developed a new ADC based on a new cytotoxic platinum(IV)prodrug(C8Pt(IV))and *** so-called antibody-platinum(IV)prodrugs conjugates,named Cet-C8Pt(IV),showed excellent tumor targeting in ***,it accurately delivered C8Pt(IV)into tumor cells to exert the combined anti-tumor effect of Cet and ***,metabolomic analysis showed that Cet-C8Pt(IV)promoted cellular apoptosis and increased DNA damage in cSCC cells by affecting the vitamin B6 metabolic pathway in tumor cells,thereby further enhancing the tumor-killing ability and providing a new strategy for clinical cancer treatment using antibody-platinum(IV)prodrugs conjugates.

Supercoiled DNA Minicircles under Double-strand Breaks

Understanding how supercoiled DNA releases intramolecular stress is essential for its functional ***,the molecular mechanism underlying the relaxation process remains insufficiently *** we employed md simulations based on the oxDNA2 model to investigate the relaxation process of a 336-base pair supercoiled minicircular DNA under double-strand breaks with two fixed *** simulations show that the conformational changes in the DNA occur continuously,with intramolecular stress release happening abruptly only when the DNA chain traverses the breakage *** relaxation process is influenced not only by the separation distance between the fixed ends but also their ***,we observe an inhibitory effect on the relaxation characterized by small angles,where short terminal loops impede DNA conformational adjustments,preserving the supercoiled *** findings elucidate the intricate interplay between DNA conformational change,DNA motion and intramolecular stress release,shedding light on the mechanisms governing the relaxation of supercoiled DNA at the molecular level.

在细粒度情感分析和情绪分析数据集中,标签之间的相关性和标签分布的不均匀性非常突出。类别标签分布不均匀,标签之间存在相关性容易影响学习模型的性能。针对这一问题,该文受计算机视觉领域中的Circle loss方法的启发,将梯度衰减、成对优化、添加余量引入损失函数来优化深度学习模型的性能。该方法可以很好地与预训练模型相结合,不需要修改骨干网络。与当前最新的经典方法相比,该方法在SemEval18数据集上Jaccard系数、micro-F1、macro-F1分别提升了1.9%、2%、1.9%;在GoeEmotions数据集上Jaccard系数、micro-F1、macro-F1分别提升了2.6%、1.9%、3.6%。实验表明,该文提出的损失函数对情感分析和情绪分析问题具有显著的提升作用。

Exploring pore-scale production characteristics of oil shale after CO_(2) huff‘n’puff in fractured shale with varied permeability

Recent studies have indicated that the injection of carbon dioxide(CO_(2))can lead to increased oil recovery in fractured shale reservoirs following natural *** advancements in understanding mass exchange processes in subsurface formations,there remains a knowledge gap concerning the disparities in these processes between the matrix and fractures at the pore scale in formations with varying *** study aims to experimentally investigate the CO_(2) diffusion behaviors and in situ oil recovery through a CO_(2) huff‘n’puff process in the Jimsar shale oil *** achieve this,we designed three matrix-fracture models with different permeabilities(0.074 md,0.170 md,and 0.466 md)and experimented at 30 MPa and 91℃.The oil concentration in both the matrix and fracture was monitored using a low-field nuclear magnetic resonance(LF-NMR)technique to quantify in situ oil recovery and elucidate mass-exchange *** results showed that after three cycles of CO_(2) huff‘n’puff,the total recovery degree increased from 30.28%to 34.95%as the matrix permeability of the core samples increased from 0.074 to 0.466 md,indicating a positive correlation between CO_(2) extraction efficiency and matrix *** similar fracture conditions,the increase in matrix permeability further promoted CO_(2) extraction efficiency during CO_(2) huff‘n’***,the increase in matrix permeability of the core had the greatest effect on the extraction of the first-cycle injection in large pores,which increased from 16.42%to 36.64%.The findings from our research provide valuable insights into the CO_(2) huff‘n’puff effects in different pore sizes following fracturing under varying permeability conditions,shedding light on the mechanisms of CO_(2)-enhanced oil recovery in fractured shale oil reservoirs.

The effectiveness of physical activity interventions on blood pressure in children and adolescents:A systematic review and network meta-analysis

背景鞘内注射小剂量吗啡可产生与静脉注射吗啡相似的心肌保护作用，但鞘内注射吗啡预处理（IT—MPC）与缺血预处理（IPC）作用强度的差别还未知，IT-MPC是否由阿片类受体所介导也未阐明。因此，本实验比较IT—MPC与IPC作用强度差别并探讨阿片类受体的作用。方法80只SD大鼠麻醉后开胸，成功置入鞘内导管后随机分为13组（n=6—7）。IPC组接受3次5分钟缺血-5分钟再灌注的心肌IPC（阻闭左冠状动脉），随之给予30分钟缺血-2小时再灌注诱导心肌缺血再灌注（IR）损伤。IT-MPC组按剂量不同分为4个亚组（分别是0．03、0．3、3及30μg／kg），以3次5分钟注射-5分钟间停的模式平均注入鞘内，随之诱导IR损伤。静脉注射吗啡预处理组（IV-MPC）组经静脉给予吗啡300μg／kg，对照组鞘内给予10μl生理盐水。受体阻滞剂组选择在IT—MPC（3μg/kg）前分别给予选择性δ、κ、μ受体阻滞剂NTD、nor-BNI及CTOP，以评价阿片类受体亚型的作用。通过2，3，5-tripheny ltetrazolium染色，计算心肌梗死面积（IS），即梗死心肌占缺血危险区（AAR）的百分比。结果与对照组相比，鞘内注射0．3～30μg／kg吗啡降低心肌IS，3组IS／AAR分别为33％±10％（0．3μg／kg）、29％±10％（3μg／kg）、29％±16％（30μg/kg），对照组为53％±8％（P〈0．01）。IT-MPC降低心肌IS／AAR程度与IV-MPC（33％±6％，P=0．84）及IPC（22％±4％，P=0．41）相近。注射选择性受体阻滞剂后，由IT-MPC（3μg/kg）产生的心肌预处理效果降低（NTD＋IT-MPC，50％±9％：nor-BNI＋IT-MPc，43％±6％；CTOP＋IT—MPC，53％±9％；与对照组相比，P=0．14）。结论IT—MPC产生与IPC及IV—MPC相近的心肌保护作用，其机制涉及到δ、κ、μ受体。

采用分子动力学(md)方法,预测了1,1-二氨基-2,2-二硝基乙烯(FOX-7)在真空条件下以及3种不同溶剂-非溶剂体系(DMSO-H_(2)O、DMSO-乙醇、DMAC-乙醇,溶剂与非溶剂体积比为1∶2)下的晶体形貌,确定真空条件下FOX-7的形态学重要生长面和附着能,计算溶剂与晶面之间的相互作用能、溶剂影响下的晶面附着能。结果表明:FOX-7在真空中的主要生长面有6个面,分别为(0 1 1)、(1 0-1)、(1 0 1)、(0 0 2)、(1 1 0)、(1 1-1),其中(0 1 1)晶面面积最大,占比达到49.38%,是FOX-7晶体形态的最重要晶面。在DMSO-H_(2)O体系中形貌为方形,在DMSO-乙醇中晶体较扁,趋于圆形,在DMAC-乙醇中晶体形貌更为立体,趋于球形。对预测效果较好的DMAC-乙醇体系进行试验,得到的晶体形貌规整,为类球形,与模拟结果相符,说明了预测的准确性。
