Nitro-oleic acid ameliorates erectile dysfunction in a streptozotocin-induced rat model of diabetes by inhibiting oxidative stress and apoptosis and activating the NO/cGMP pathway

The major vascular complications associated with diabetes make the management of diabetic mellitus erectile dysfunction(DMED)a challenging endeavor.Notable factors contributing to DMED include oxidative stress,nitric oxide(NO)/cyclic guanosine monophosphate(cGMP)pathway activation,and apoptosis,while nitro-oleic acid(NO,-OA)has been shown to be beneficial in treating these aspects of this condition.We,herein,investigated the effects and possible mechanisms of NO,-OA on erectile function as assessed in a streptozotocin-induced rat model of diabetes.Our results revealed that the erectile function of DMED rats was significantly impaired compared with that of the control group.However,in response to 4 weeks of NO,-OA treatment,there was an improvement in erectile function.The expression of oxidative stress-related indicators was significantly increased and the NO/cGMP pathway was impaired in the DMED group.The expression of proapoptotic factors was increased,while that of antiapoptotic factors was decreased in the DMED group.Moreover,the cell morphology in the cavernous tissue of the DMED group also changed adversely.NO,-OA treatment significantly reversed all these changes observed in the DMED group.In conclusion,NO,-OA treatment partially improved erectile function in DMED rats through mechanisms that included inhibition of oxidative stress,activation of the NO/cGMP pathway,and a reduction in apoptosis.

Systemic immune monitoring is a crucial clinical tool for disease early diagnosis,prognosis and treatment planning by quantitative analysis of immune cells.However,conventional immune monitoring using flow cytometry faces huge challenges in large-scale sample testing,especially in mass health screenings,because of time-consuming,technical-sensitive and high-cost features.However,the lack of high-performance detection platforms hinders the development of high-throughput immune monitoring technology.To address this bottleneck,we constructed a generally applicable DNA framework signal amplification platform(DSAP)based on post-systematic evolution of ligands by exponential enrichment and DNA tetrahedral framework-structured probe design to achieve high-sensitive detection for diverse immune cells,including CD4+,CD8+T-lymphocytes,and monocytes(down to 1/100μl).Based on this advanced detection platform,we present a novel high-throughput immune-cell phenotyping system,DSAP,achieving 30-min one-step immune-cell phenotyping without cell washing and subset analysis and showing comparable accuracy with flow cytometry while significantly reducing detection time and cost.As a proof-of-concept,DSAP demonstrates excellent diagnostic accuracy in immunodeficiency staging for 107 HIV patients(AUC>0.97)within 30 min,which can be applied in HIV infection monitoring and screening.Therefore,we initially introduced promising DSAP to achieve high-throughput immune monitoring and open robust routes for point-of-care device development.

Pain management in patients with hepatocellular carcinoma after transcatheter arterial chemoembolisation: A retrospective study

BACKGROUND Pain after transcatheter arterial chemoembolisation(TACE)can seriously affect the prognosis of patients and the insertion of additional medical resources.AIM To develop an early warning model for predicting pain after TACE to enable the implementation of preventive analgesic measures.METHODS We retrospectively collected the clinical data of 857 patients(from January 2016 to January 2020)and prospectively enrolled 368 patients(from February 2020 to October 2022;as verification cohort)with hepatocellular carcinoma(HCC)who received TACE in the Hepatic Surgery Center of Tongji Hospital.Five predictive models were established using machine learning algorithms,namely,random forest model(RFM),support vector machine model,artificial neural network model,naive Bayes model and decision tree model.The efficacy of these models in predicting postoperative pain was evaluated through receiver operating characteristic curve analysis,decision curve analysis and clinical impact curve analysis.RESULTS A total of 24 candidate variables were included in the predictive models using the iterative algorithms.Age,preoperative pain,number of embolised tumours,distance from the liver capsule,dosage of iodised oil and preoperative prothrombin activity were closely associated with postoperative pain.The accuracy of the predictive model was compared between the training[area under the curve(AUC)=0.798;95%confidence interval(CI):0.745-0.851]and verification(AUC=0.871;95%CI:0.818-0.924)cohorts,with RFM having the best predictive efficiency(training cohort:AUC=0.869,95%CI:0.816-0.922;internal verification cohort:AUC=0.871;95%CI:0.818-0.924).CONCLUSION The five predictive models based on advanced machine learning algorithms,especially RFM,can accurately predict the risk of pain after TACE in patients with HCC.RFM can be used to assess the risk of pain for facilitating preventive treatment and improving the prognosis.

Application of mesenchymal stem cell therapy for premature ovarian insufficiency: Recent advances from mechanisms to therapeutics

The incidence of premature ovarian insufficiency(POI)is increasing worldwide,particularly among younger women,posing a significant challenge to fertility.In addition to menopausal symptoms,POI leads to several complications that profoundly affect female reproductive function and overall health.Unfortunately,current clinical treatment strategies for this condition are limited and often yield unsatisfactory outcomes.These approaches typically involve hormone repla-cement therapy combined with psychological support.Recently,mesenchymal stem cell(MSC)therapies for POI have garnered considerable attention in global research.MSCs can restore ovarian reproductive and endocrine functions through diverse mechanisms,including controlling differentiation,promoting angiogenesis,regulating ovarian fibrosis,inhibiting apoptosis,enhancing autocrine and paracrine effects,suppressing inflammation,modulating the immune system,and genetic regulation.This editorial offers a succinct summary of the application of MSC therapy in the context of POI,providing evidence for groundbreaking medical approaches that have potential to enhance reproductive health and overall well-being for women.

A partition of unity level set method with moving knot CS-RBFs for optimizing variable stiffness composites

A partition of unity level set method with moving knot Compactly Supported Radial Basis Functions(CS-RBFs)is proposed for optimizing variable stiffness composite structures.The iso-contours of a level set function are utilized to represent the curved fiber paths,and the tan-gent vector of the iso-contour defines the orientation of fiber.The level set function of the full design domain is constructed according to the Partition of Unity(POU)method by a set of local level set functions defined on an array of overlapping subdomains,and they are constructed by using the CS-RBFs.The positions of knots are iteratively changed during the optimization to improve the performance of composite structures.Several examples of compliance minimization are presented.

Boron Nitride Quantum Dots/Ti_(3)C_(2)T_(x)-MXene Heterostructure For Efficient Electrocatalytic Nitrogen Fixation

Electrocatalytic N_(2) fixation through N_(2) reduction reaction(NRR)has been regarded as a promising route for sustainable NH_(3) synthesis,while exploring high-performing NRR catalysts is pivotal yet challenging.Herein,BN quantum dots/Ti_(3)C_(2)T_(x)-MXene(BNQDs/Ti_(3)C_(2)T_(x))heterostructure is demonstrated as an efficient and durable NRR catalyst,exhibiting a high NH_(3) yield of 52.8±3.3μg h^(-1) mg^(-1) with an FE of 19.1±1.6%at0.4 V(vs.RHE),which stand at the high level among all reported BN-and MXene-based NRR catalysts.Theoretical computations reveal that the electronic interactions between BNQDs and Ti_(3)C_(2)T_(x) enrich the electron density of B atoms at the heterointerface and endow them with enhanced electron-donating capability for N_(2) activation and protonation.Meanwhile,the decorated BNQDs can block the active sites of Ti_(3)C_(2)T_(x) for hydrogen evolution,rendering a high N_(2)-to-NH_(3) selectivity.

Responses of radial growth, wood density and fiber traits to planting space in poplar plantations at a lowland site

Poplar is raw material for various panel, paper and fiber products. The 12 sample trees of clone Nanlin-895 from four spacings were destructively harvested after thirteen growing seasons to assess the influence of spacing on radial growth and wood properties. Spacing significantly affected tree-ring width and wood basic density (p < 0.05) but not fiber traits. The highest diameter and wood basic density at breast height (1.3 m) was in 6 m × 6 m and 3 m × 8 m spacings, respectively. However, no significant differences in tree-ring width, wood basic density and fiber traits were observed among the four sampling directions in discs taken at 1.3 m for each spacing. Growth rings from the pith and tree heights had significant effects on wood basic density and fiber anatomical characteristics, highlighting obvious temporal-spatial variations. Pearson correlation analysis showed a significantly negative relationship of tree-ring width to wood basic density, fiber length and fiber width, but a significantly positive relationship to hemicellulose. There was no relationship with cellulose and lignin contents. Based on a comprehensive assessment by the TOPSIS method, the 6 m × 6 m spacing is recommended for producing wood fiber at similar sites in the future.

The critical role of the endolysosomal system in cerebral ischemia

Cerebral ischemia is a serious disease that triggers sequential pathological mechanisms, leading to significant morbidity and mortality. Although most studies to date have typically focused on the lysosome, a single organelle, current evidence supports that the function of lysosomes cannot be separated from that of the endolysosomal system as a whole. The associated membrane fusion functions of this system play a crucial role in the biodegradation of cerebral ischemia-related products. Here, we review the regulation of and the changes that occur in the endolysosomal system after cerebral ischemia, focusing on the latest research progress on membrane fusion function. Numerous proteins, including N-ethylmaleimide-sensitive factor and lysosomal potassium channel transmembrane protein 175, regulate the function of this system. However, these proteins are abnormally expressed after cerebral ischemic injury, which disrupts the normal fusion function of membranes within the endolysosomal system and that between autophagosomes and lysosomes. This results in impaired “maturation” of the endolysosomal system and the collapse of energy metabolism balance and protein homeostasis maintained by the autophagy-lysosomal pathway. Autophagy is the final step in the endolysosomal pathway and contributes to maintaining the dynamic balance of the system. The process of autophagosome-lysosome fusion is a necessary part of autophagy and plays a crucial role in maintaining energy homeostasis and clearing aging proteins. We believe that, in cerebral ischemic injury, the endolysosomal system should be considered as a whole rather than focusing on the lysosome. Understanding how this dynamic system is regulated will provide new ideas for the treatment of cerebral ischemia.

Skin‑Inspired Ultra‑Tough Supramolecular Multifunctional Hydrogel Electronic Skin for Human–Machine Interaction

Multifunctional supramolecular ultra-tough bionic e-skin with unique durability for human–machine interaction in complex scenarios still remains challenging.Herein,we develop a skininspired ultra-tough e-skin with tunable mechanical properties by a physical cross-linking salting-freezing-thawing method.The gelling agent(β-Glycerophosphate sodium:Gp)induces the aggregation and binding of PVA molecular chains and thereby toughens them(stress up to 5.79 MPa,toughness up to 13.96 MJ m^(−3)).Notably,due to molecular self-assembly,hydrogels can be fully recycled and reprocessed by direct heating(100°C for a few seconds),and the tensile strength can still be maintained at about 100%after six recoveries.The hydrogel integrates transparency(>60%),super toughness(up to 13.96 MJ m^(−3),bearing 1500 times of its own tensile weight),good antibacterial properties(E.coli and S.aureus),UV protection(Filtration:80%–90%),high electrical conductivity(4.72 S m^(−1)),anti-swelling and recyclability.The hydrogel can not only monitor daily physiological activities,but also be used for complex activities underwater and message encryption/decryption.We also used it to create a complete finger joint rehabilitation system with an interactive interface that dynamically presents the user’s health status.Our multifunctional electronic skin will have a profound impact on the future of new rehabilitation medical,human–machine interaction,VR/AR and the metaverse fields.

Analysis and Characterization of the GABA Transaminase and Succinate Semialdehyde Dehydrogenase Genes in the Microalga Isochrysis zhanjiangensis in Response to Abiotic Stresses

Gamma-aminobutyric acid(GABA),widely existing in different organisms,is rapidly accumulated in plants in response to environmental stresses.The main biosynthesis and degradation pathways of GABA constitute the GABA shunt,which is tied to the tricarboxylic acid(TCA)cycle.GABA transaminase(GABA-T)and succinate semialdehyde dehydrogenase(SSADH)are two essential enzymes for the GABA degradation pathway.While there are abundant studies on GABA shunt in higher plants at the physiological and genetic levels,research on its role in microalgae remains limited.This study aimed at exploring the function of GABA-T and SSADH genes in Isochrysis zhanjiangensis,an important diet microalga,under different stresses.We cloned two GABA-T genes,IzGABA-T1 and IzGABA-T2,and one SSADH gene IzSSADH from Isochrysis zhanjiangensis and conducted heterologous expression experiments.The results showed that the overexpression of IzGABA-T1 or IzGABA-T2 enhanced the survival rates of yeast transformants under heat or NaCl stress,while the overexpression of IzSSADH improved yeast tolerance to NaCl stress but had no obvious effect on heat stress.Additionally,the results of quantitative real-time polymerase chain reaction(qPCR)showed that IzGABA-T1 transcription increased in the HT(salinity 25,35℃)and LS(salinity 15,25℃)groups.At 24 h,the IzGABA-T2 transcriptions increased in the HT,LS,and HS(salinity 35,25℃)groups,but their transcription levels decreased in all groups at 48 h.IzSSADH transcription increased in the LS group.These results suggest that IzGABA-T1,IzGABA-T2,and IzSSADH are associated with temperature and salinity stresses and possess a certain preference for different stresses.
