The OsWRKY72-OsAAT30/OsGSTU26 module mediates reactive oxygen species scavenging to drive heterosis for salt tolerance in hybrid rice

Hybrid rice(Oryza sativa)generally outperforms its inbred parents in yield and stress tolerance,a phenomenon termed heterosis,but the underlying mechanism is not completely understood.Here,we combined transcriptome,proteome,physiological,and heterosis analyses to examine the salt response of super hybrid rice Chaoyou1000(CY1000).In addition to surpassing the mean values for its two parents(mid-parent heterosis),CY1000 exhibited a higher reactive oxygen species scavenging ability than both its parents(over-parent heterosis or heterobeltiosis).Nonadditive expression and allele-specific gene expression assays showed that the glutathione S-transferase gene OsGSTU26 and the amino acid transporter gene OsAAT30 may have major roles in heterosis for salt tolerance,acting in an overdominant fashion in CY1000.Furthermore,we identified OsWRKY72 as a common transcription factor that binds and regulates OsGSTU26 and OsAAT30.The salt-sensitive phenotypes were associated with the OsWRKY72^(paternal)genotype or the OsAAT30^(maternal)genotype in core rice germplasm varieties.OsWRKY72^(paternal)specifically repressed the expression of OsGSTU26 under salt stress,leading to salinity sensitivity,while OsWRKY72^(maternal)specifically repressed OsAAT30,resulting in salinity tolerance.These results suggest that the OsWRKY72-OsAAT30/OsGSTU26 module may play an important role in heterosis for salt tolerance in an overdominant fashion in CY1000 hybrid rice,providing valuable clues to elucidate the mechanism of heterosis for salinity tolerance in hybrid rice.

Influence of vibration on the lubrication effect of a splash-lubricated gearbox

In this study,we investigate the effect of rail vibrations on the lubrication and efficiency of a splash-lubricated gearbox;specifically,the gearbox of a rail-transit vehicle.A high-fidelity 3D numerical model of the gearbox in a moving reference system is described,as well as computational fluid dynamics(CFD)simulations of the gearbox with different vibration directions.The effects of rotational speed,oil-immersion depth,and different oil-injection volume rates on lubrication and efficiency are discussed.We propose a method of evaluating the internal lubrication condition of a splash-lubricated gearbox and quantitatively compare the effects of different operating parameters on lubrication and efficiency.Finally,our experiment to verify the feasibility of the simulation method is described.The results show that with vibration,the churning loss and oil supply for the bearings are significantly higher than those under static conditions.In addition,among different vibrational directions,lateral vibration has the greatest influence on the lubrication condition and efficiency of the gearbox.For the studied railway-vehicle gearbox,the best lubrication condition is achieved at a rotational speed of 1600 r/min and an oil-immersion depth of two times the tooth height(2.Oh).Rotational speed is the operating parameter that has the most significant effect on the lubrication and efficiencyofthegearbox.

Atroposelective Synthesis of 2-Arylindoles via Chiral Phosphoric Acid-Catalyzed Direct Amination of Indoles

Indole-based atropisomers are a very important class of axially chiral compounds.However,the atroposelective synthesis of axially chiral 2-arylindole remains largely unexplored.In this study,we report the successful synthesis of atropisomeric 2-arylindoles using direct amination of indoles with p-quinonediimines in the presence of chiral phosphoric acid as a catalyst.Quinonediimine acts as an aminating reagent through formal polarity inversion of imine.The malonate group on the 2-aryl of 2-indoles was found to be essen-tial for high enantioselectivity of the products.This could be due to the additional interaction between the ester group and the cata-lyst,as well as the intramolecular hydrogen bonding.Our findings provide a new strategy for the asymmetric construction of 2-arylindole atropisomers.

LncRNA pol-lnc78 as a ceRNA regulates antibacterial responses via suppression of pol-miR-n199-3p-mediated SARM down-regulation in Paralichthys olivaceus

Long non-coding RNAs(lncRNAs)function as key modulators in mammalian immunity,particularly due to their involvement in lncRNA-mediated competitive endogenous RNA(ceRNA)crosstalk.Despite their recognized significance in mammals,research on lncRNAs in lower vertebrates remains limited.In the present study,we characterized the first immune-related lncRNA(pol-lnc78)in the teleost Japanese flounder(Paralichthys olivaceus).Results indicated that pol-lnc78 acted as a ceRNA for pol-miR-n199-3p to target the sterile alpha and armadillo motif-containing protein(SARM),the fifth discovered member of the Toll/interleukin 1(IL-1)receptor(TIR)adaptor family.This ceRNA network regulated the antibacterial responses of flounder via the Toll-like receptor(TLR)signaling pathway.Specifically,SARM acted as a negative regulator and exacerbated bacterial infection by inhibiting the expression of inflammatory cytokines IL-1βand tumor necrosis factor-α(TNF-α).Pol-miR-n199-3p reduced SARM expression by specifically interacting with the 3’untranslated region(UTR),thereby promoting SARM-dependent inflammatory cytokine expression and protecting the host against bacterial dissemination.Furthermore,pol-lnc78 sponged pol-miR-n199-3p to ameliorate the inhibition of SARM expression.During infection,the negative regulators pol-lnc78 and SARM were significantly down-regulated,while pol-miR-n199-3p was significantly up-regulated,thus favoring host antibacterial defense.These findings provide novel insights into the mechanisms underlying fish immunity and open new horizons to better understand ceRNA crosstalk in lower vertebrates.

Novel phenanthrene/bibenzyl trimers from the tubers of Bletilla striata attenuate neuroinﬂammation via inhibition of NF-κB signaling pathway

Five novel(9,10-dihydro)phenanthrene and bibenzyl trimers,as well as two previously identified biphenanthrenes and bibenzyls,were isolated from the tubers of Bletilla striata.Their structures were elucidated through comprehensive analyses of NMR and HRESIMS spectroscopic data.The absolute configurations of these compounds were determined by calculating rotational energy barriers and comparison of experimental and calculated ECD curves.Compounds 5b and 6 exhibited inhibitory effects on LPS-induced NO production in BV-2 cells,with IC_(50) values of 12.59±0.40 and 15.59±0.83μmol·L^(-1),respectively.A mechanistic study suggested that these compounds may attenuate neuroinflammation by reducing the activation of the AKT/IκB/NF-κB signaling pathway.Additionally,compounds 3a,6,and 7 demonstrated significant PTP1B inhibitory activities,with IC_(50) values of 1.52±0.34,1.39±0.11,and 1.78±0.01μmol·L^(-1),respectively.Further investigation revealed that compound 3a might inhibit LPS-induced PTP1B overexpression and NF-κB activation,thereby mitigating the neuroinflammatory response in BV-2 cells.

Serrated flow behavior of GH536 superalloy under different loading rates at room temperature

Serrated flow phenomenon has been widely observed among different metals due to the potential influence on their applications.Uniaxial tensile tests of nickel-based superalloy GH536 were carried out at loading rates of 0.06,0.60,3.60,and 36.00 mm·min-1at room temperature,respectively.The tensile stress–strain curves demonstrate repetitive and discontinuous yielding behavior,namely serrated flow.It is observed that the stress–strain curves were dominated by type B serration,and original annealing twin boundaries （TBs） were distorted to various degrees under comparatively different lower tensile rates.In contrast,the TBs almost disappear,and the type B serrations become smoother and regular under the higher loading rates.This phenomenon can be attributed to the interactions between TBs and dislocations during tensile deformation.Coupled effects of the mobile and immobile dislocations illustrate the unsteady amplitude of serrations observed in stress–strain curves.Transmission electron microscope images of tested pieces reveal the interaction of dislocation and TB,with dislocation tangling around the TBs.

Plant high-affinity K^(+) transporters(HKTs)mediate Na^(+) and K^(+) uptake,maintain Na^(+)/K^(+) homeostasis,and therefore play crucial roles in plant salt tolerance.In this study,we present cryoelectron microscopy structures of HKTs from two classes,classI HKT1;1 from Arabidopsis thaliana(AtHKT1;1)and class II HKT2;1 from Triticum aestivum(TaHKT2;1),in both Na^(+) -and K^(+) -bound states at 2.6-to 3.0-A resolutions.BothAtHKT1;1and TaHKT2;1function ashomodimers.Each HKT subunit consists of four tan-dem domain units(D1-D4)with a repeated K^(+) -channel-like M-P-M topology.In each subunit,D1-D4 assemble into an ion conduction pore with a pseudo-four-fold symmetry.Although both TaHKT2;1and AtHKT1;1 have only one putative Na^(+) ion bound in the selectivity filter with a similar coordination pattern,the two HKTs display different K^(+) binding modes in the filter.TaHKT2;1 has three K^(+) ions bound in the selec-tivity filter,but AtHKT1;1 has only two K^(+) ions bound in the filter,which has a narrowed external entrance due to the presence of a Ser residue in the first filter motif.These structures,along with compu-tational,mutational,and electrophysiological analyses,enable us to pinpoint key residues that are critical for the ion selectivity of HKTs.The findings provide new insights into the ion selectivity and ion transport mechanisms of plant HKTs and improve our understanding about how HKTs mediate plant salt tolerance and enhance crop growth.

Licorice,a perennial herb of Leguminosa,is one of the oldest and most widely used herbal medicines worldwide.Its distinct sweet flavor and rich medicinal value make it an integral component of traditional Chinese medicine(TCM)formulations,which continue to be widely employed.The main chemical constituents of licorice include triterpenoid saponins,flavonoids,and polysaccharides.Experimental and clinical studies have demonstrated that various extracts and pure compounds derived from licorice exhibit a wide range of pharmacological properties including anti-inflammatory,antioxidant,antimicrobial,antiviral,antitumor,immune-regulatory,and neuroprotective activities.The bioactive constituents of licorice offer therapeutic benefits for cardiovascular and cerebrovascular diseases,diabetes mellitus,and liver disorders.This comprehensive review discusses the primary chemical constituents of licorice and their pharmacological activities,describes in vivo and in vitro models employed for studying licorice,and its potential targets and mechanisms of action.Furthermore,we discuss the toxicological profile,side effects,dosage recommendations,and clinical applications of licorice.This review aims to establish a foundation for further research on the safe and effective utilization of licorice while facilitating an in-depth exploration of its properties and fostering the development of novel therapeutic agents.

A hybrid WUDT-NAFnet for simultaneous source data deblending

Simultaneous source technology, which reduces seismic survey time and improves the quality of seismic data by firing more than one source with a narrow time interval, is compromised by the massive blended interference. Therefore, deblending algorithms have been developed to separate this interference.Recently, deep learning(DL) has been proved its great potential in suppressing the interference. The most popular DL method employs neural network as a filter to attenuate the blended noise in an iterative estimation and subtraction framework(IESF). However, there are still amplitude distortion and blended noise residual problems, especially when dealing with weak signal submerged in strong interference. To address these problems, we propose a hybrid WUDT-NAFnet, which contains two sub-networks. The first network is a wavelet based U-shape deblending transformer network(WUDTnet), incorporated into IESF as a robust regularization term to iteratively separate the blended interference. The second network is a nonlinear activate free network(NAFnet) designed to recover the event amplitude and further suppress the weak noise residual in IESF. With the hybrid network, the blended noise can be separated purposefully and accurately. Examples using synthetic and field seismic data demonstrate that the WUDTNAFnet outperforms traditional curvelet transform(CT) based method and the deblending transformer(DT) model in terms of deblending. Additionally, for field applications, the data augmentation method of bicubic interpolation is applied to mitigate the feature difference between synthetic and field data.Consequently, the trained network exhibits strong signal preservation ability in numerical field example without requiring additional training.

Late Triassic Intracontinental Deformation of South Tianshan,Central Asia:Evidence from Syn-tectonic Sedimentation and Detrital Zircon Provenances of the Kuqa Depression

The Tianshan range,a Paleozoic orogenic belt in Central Asia,has undergone multiple phases of tectonic activities characterized by the N-S compression after the early Mesozoic,including the far-field effects of the Cenozoic Indian-Asian collision.However,there are limited reports on the tectonic deformation and initiation of Triassic intracontinental deformation in the Tianshan range.Understanding this structural context is crucial for interpreting the early intracontinental deformation history of the Eurasian continent during the early Mesozoic.Growth strata and syn-tectonic sediments provide a rich source of information on tectonic activities and have been extensively used in the studies of orogenic belts.Based on detail fieldwork conducted in this study,the middle-late Triassic Kelamayi Formation of the northern Kuqa Depression in the southern Tianshan fold-thrust belt has been identified as the typical syn-tectonic growth strata.The youngest detrital zircon component in two lithic sandstone samples from the bottom and top of the Kelamayi growth strata yielded U-Pb ages of 223.4±3.1 and 215.5±2.9 Ma,respectively,indicating that the maximum depositional age of the bottom and top of the Kelamayi growth strata is 226-220 and 218-212 Ma.The geochronological distribution of detrital samples from the Early-Middle Triassic and Late Triassic revealed abrupt changes,suggesting a new source supply resulting from tectonic activation in the Tianshan range.The coupling relationship between the syn-tectonic sedimentation of the Kelamayi Formation and the South Tianshan fold-thrust system provides robust evidence that the Triassic intracontinental deformation of the South Tianshan range began at approximately 226-220 Ma(during the Late Triassic)and ended at approximately 218-212 Ma.These findings provide crucial constraints for understanding the intraplate deformation in the Tianshan range during the Triassic.
