A new diatom genus Spargeria gen.nov.(Bacillariophyceae)from Xizang,China,and the description of two new species

During our investigation of diatom biodiversity in Xizang,two species exhibited unique morphological features discriminative from all previously known genera.Herein we describe these two species and describe as new the genus,Spargeria gen.nov.The new genus features narrow to wide rectangular valves,narrow valve mantles,filiform raphe branches that occur on the valve face only,terminal raphe fissures straight or slightly deflected to same side,bow-tie shaped central areas,chambered striae present on the valve face only,being absent from the mantle,wider striae near the axial area and very narrow near the margin,multiseriate striae with small and round areolae that are occluded externally.Comparatively,Spargeria zhuii sp.***.has larger and robust valves,radiate striae,with one divergent stria near the apices,while Spargeria chenia sp.***.is smaller,with narrow valves,striae slightly radiate in the middle,becoming convergent or parallel near apices.This new genus belongs to the family Pinnulariaceae,and it was compared and contrasted with other genera of this family.Our work suggests the need for continued studies to document the biodiversity of diatoms in Xizang.

Semi-transparent perovskite solar cells(ST-PSCs)have broad applications in building integrated photovoltaics.However,the stability of ST-PSCs needs to be improved,especially in n-i-p ST-PSCs since the doped 2,2',7,7'-tetrakis(N,N-di-p-methoxyphenyl-amine)-9,9'-spirobifluorene(Spiro-OMeTAD)is unstable at elevated temperatures and high humidity.In this work,aπ-conjugated polymer poly[(2,6-(4,8-bis(5-(2-ethylhexyl)thiophene-2-yl)-benzo[1,2-b:4,5-b']dithiophene))-alt-(5,5-(1',3'-di-2-thienyl-5',7'-bis(2-ethylhexyl)benzo[1',2'-c:4',5'-c']dithiophene-4,8-dione)](PBDB-T)is selected to form a polymer composite hole transport layer(HTL)with Spiro-OMeTAD.The sulfur atom of the thiophene unit and the carbonyl group of the polymer interact with the undercoordinated Pb2+at the perovskite surface,which stabilizes the perovskite/HTL interface and passivates the interfacial defects.The incorporation of the polymer also increases the glass transition temperature and the moisture resistance of Spiro-OMeTAD.As a result,we obtain ST-PSCs with a champion efficiency of 13.71%and an average visible light transmittance of 36.04%.Therefore,a high light utilization efficiency of 4.94%can be obtained.Moreover,the encapsulated device can maintain 84%of the initial efficiency after 751 h under continuous one-sun illumination(at 30%relative humidity)at the open circuit and the unencapsulated device can maintain 80%of the initial efficiency after maximum power tracking for more than 1250 h under continuous one-sun illumination.

Neural stem cell-derived exosomes promote mitochondrial biogenesis and restore abnormal protein distribution in a mouse model of Alzheimer's disease

Mitochondrial dysfunction is a hallmark of Alzheimer’s disease.We previously showed that neural stem cell-derived extracellular vesicles improved mitochondrial function in the cortex of AP P/PS1 mice.Because Alzheimer’s disease affects the entire brain,further research is needed to elucidate alterations in mitochondrial metabolism in the brain as a whole.Here,we investigated the expression of several important mitochondrial biogenesis-related cytokines in multiple brain regions after treatment with neural stem cell-derived exosomes and used a combination of whole brain clearing,immunostaining,and lightsheet imaging to clarify their spatial distribution.Additionally,to clarify whether the sirtuin 1(SIRT1)-related pathway plays a regulatory role in neural stem cell-de rived exosomes interfering with mitochondrial functional changes,we generated a novel nervous system-SIRT1 conditional knoc kout AP P/PS1mouse model.Our findings demonstrate that neural stem cell-de rived exosomes significantly increase SIRT1 levels,enhance the production of mitochondrial biogenesis-related fa ctors,and inhibit astrocyte activation,but do not suppress amyloid-βproduction.Thus,neural stem cell-derived exosomes may be a useful therapeutic strategy for Alzheimer’s disease that activates the SIRT1-PGC1αsignaling pathway and increases NRF1 and COXIV synthesis to improve mitochondrial biogenesis.In addition,we showed that the spatial distribution of mitochondrial biogenesis-related factors is disrupted in Alzheimer’s disease,and that neural stem cell-derived exosome treatment can reverse this effect,indicating that neural stem cell-derived exosomes promote mitochondrial biogenesis.

Generally shortened 3′UTR due to alternative polyadenylation(APA)is widely observed in cancer,but its regulation mechanisms for cancer are not well characterized.Here,with profiling of APA in colorectal cancer tissues and poly(A)signal editing,we firstly identified that the shortened 3′UTR of CTNNIBP1 in colorectal cancer promotes cell proliferation and migration.We found that liquid-liquid phase separation(LLPS)of PABPN1 is reduced albeit with higher expression in cancer,and the reduction of LLPS leads to the shortened 3′UTR of CTNNBIP1and promotes cell proliferation and migration.Notably,the splicing factor SNRPD2 upregulated in colorectal cancer,can interact with glutamic-proline(EP)domain of PABPN1,and then disrupt LLPS of PABPN1,which attenuates the repression effect of PABPN1 on the proximal poly(A)sites.Our results firstly reveal a new regulation mechanism of APA by disruption of LLPS of PABPN1,suggesting that regulation of APA by interfering LLPS of 3′end processing factor may have the potential as a new way for the treatment of cancer.

Heterosis in root microbiota inhibits growth of soil-borne fungal pathogens in hybrid rice

In nature,plants are colonized by various microbes that play essential roles in their growth and health.Heterosis is a natural genetic phenomenon whereby first-generation hybrids exhibit superior phenotypic performance relative to their parents.It remains unclear whether this concept can be extended to the“hybridization”of microbiota from two parents in their descendants and what benefits the hybrid microbiota might convey.Here,we investigated the structure and function of the root microbiota from three hybrid rice varieties and their parents through amplicon sequencing analysis of bacterial 16S ribosomal DNA(rDNA)and fungal internal transcribed spacer(ITS)regions.We show that the bacterial and fungal root microbiota of the varieties are distinct from those of their parental lines and exhibit potential heterosis features in diversity and composition.Moreover,the root bacterial microbiota of hybrid variety LYP9 protects rice against soil-borne fungal pathogens.Systematic analysis of the protective capabilities of individual strains from a 30-member bacterial synthetic community derived from LYP9 roots indicated that community members have additive protective roles.Global transcription profiling analyses suggested that LYP9 root bacterial microbiota activate rice reactive oxygen species production and cell wall biogenesis,contributing to heterosis for protection.In addition,we demonstrate that the protection conferred by the LYP9 root microbiota is transferable to neighboring plants,potentially explaining the observed hybrid-mediated superior effects of mixed planting.Our findings suggest that some hybrids exhibit heterosis in their microbiota composition that promotes plant health,highlighting the potential for microbiota heterosis in breeding hybrid crops.

Producing lightweight,mechanically strong,ductile,and biocompatible materials remains a significant challenge in material engineering due to the conflict between structural and mechanical features.Inspired by the“brick-and-mortar”structure of nacre,a construction with a naturally optimized structure-performance-function relationship,this study developed silk fibroin(SF)nacre as a silk protein-based nacre by integrating ice-templating and thermoplastic molding techniques.SF nacres are similar to natural nacre in microstructure,and their strength and toughness are even superior to natural nacres.These mechani-cal properties permit machining by extreme processing techniques,such as ion beam lithography.Furthermore,SF nacre can be used to modulate the polarization of laser beams and generate bright structural colors.Biocompatibility,mechanical robustness,good processability,and tunable coloration allow SF nacres to be used as a plastic replacement for structural engineering and biomedical use,showing promising advancement of such implantable devices towards clinical translation.

Light and temperature signals are the most important environmental cues regulating plant growth and development. Plants have evolved various strategies to prepare for, and adapt to environmental changes. Plants integrate environmental cues with endogenous signals to regulate various physiological processes, including flowering time. There are at least five distinct pathways controlling flowering in the model plant Arabidopsis thaliana: the photoperiod pathway, the vernalization/thermosensory pathway, the autonomous floral initiation, the gibberellins pathway, and the age pathway. The photoperiod and temperature/vernalization pathways mainly perceive external signals from the environment, while the autonomous and age pathways transmit endogenous cues within plants. In many plant species, floral transition is precisely controlled by light signals(photoperiod) and temperature to optimize seed production in specific environments. The molecular mechanisms by which light and temperature control flowering responses have been revealed using forward and reverse genetic approaches. Here we focus on the recent advances in research on flowering responses to light and temperature.

Recursive Zero-COVID model and quantitation of control efforts of the Omicron epidemic in Jilin province

Since the beginning of March 2022,the epidemic due to the Omicron variant has developed rapidly in Jilin Province.To figure out the key controlling factors and validate the model to show the success of the Zero-COVID policy in the province,we constructed a Recursive Zero-COVID Model quantifying the strength of the control measures,and defined the control reproduction number as an index for describing the intensity of interventions.Parameter estimation and sensitivity analysis were employed to estimate and validate the impact of changes in the strength of different measures on the intensity of public health preventions qualitatively and quantitatively.The recursive Zero-COVID model predicted that the dates of elimination of cases at the community level of Changchun and Jilin Cities to be on April 8 and April 17,respectively,which are consistent with the real situation.Our results showed that the strict implementation of control measures and adherence of the public are crucial for controlling the epidemic.It is also essential to strengthen the control intensity even at the final stage to avoid the rebound of the epidemic.In addition,the control reproduction number we defined in the paper is a novel index to measure the intensity of the prevention and control measures of public health.

利用美国国家大气研究中心(NCAR)提供的过去千年(850~2005年)集合模拟试验(CESM-LME)资料,在与重建资料、现代观测/再分析资料进行对比验证的基础上,分析了过去千年不同世纪东亚与全球地表温度变化的同步性,并结合单因子敏感性试验结果,分析了造成东亚与全球地表温度各个世纪同步性及变化幅度差异的原因。结果表明:过去千年里,在20世纪以前,东亚地区与全球均经历了中世纪暖期与小冰期,温度变化在百年尺度上整体一致,10世纪、 11世纪、 13世纪、 18世纪和19世纪东亚与全球温度变化同步性较高,差异较小;但在12世纪、 14~17世纪中,其变化振幅存在显著差异,东亚地区温度变化幅度总体上小于全球,太阳辐射和火山活动等自然外强迫及内部变率对差异的形成可能起到了主导作用;而20世纪以来,两者变化显示出较大的不同步性,东亚地区温度增长没有全球温度增长剧烈,甚至出现了降温,人为外强迫在这一时期起主导作用。温室气体浓度变化对温度的影响在两极有明显的放大效应,是东亚与全球温度变化差异形成的主要原因,同时,人为气溶胶排放使得东亚地区温度降低也是东亚与全球温度变化不同步的原因之一。
