Development of small intestinal barrier function and underlying mechanism in Chinese indigenous and Duroc piglets during suckling and weaning periods

This study explored the developmental changes in small intestinal barrier function and the potential regulatory roles of intestinal microbiota and metabolites in different breeds of piglets during suckling and weaning periods. Taoyuan black(TB), Xiangcun black(XB), and Duroc(DR) piglets(10 litters per breed; half male and half female) were selected for sampling to evaluate the intestinal barrier-related indexes and intestinal microbiota and metabolites at 1, 10, 21(weaned), and 24(3 d after weaning)d old. The results showed that weaning led to severe shedding of small intestinal microvilli and sparse microvilli arrangement. D-lactate level in the ileum of TB and XB piglets during suckling and weaning periods was lower(P < 0.01) than that of DR piglets, as well as the ileal diamine oxidase level at 1 d old.The expression level of mucin 1 was higher(P < 0.05) in the ileum of TB and XB piglets than that of DR piglets, and it was the highest in the ileum of TB piglets at 21 d old. The expression levels of mucin 2 and mucin 13 were higher(P < 0.10) in TB and XB piglets than those of DR piglets at 21 d old, whereas mucin2 and mucin 13 in the ileum of TB and XB piglets were higher(P < 0.05) than those of DR piglets at 24 d old. TB and XB piglets had a lower relative abundance of Escherichia＿Shigella at 21 and 24 d old, but they had higher Streptococcus at 1 and 24 d old than DR piglets(P < 0.01). Differential metabolites between the three breeds of piglets were mainly related to oxidative phosphorylation, steroid biosynthesis, and bile acid synthesis. Collectively, these findings suggest that different pig breeds present differences in the development of the small intestinal barrier function. Compared with DR piglets, TB and XB piglets had higher intestinal permeability during the suckling period and a stronger intestinal mechanical barrier after weaning. Moreover, intestinal microbiota and metabolites are the key factors for developing small intestinal barrier functions in different breeds of piglets.

Assessment of Wet Season Precipitation in the Central United States by the Regional Climate Simulation of the WRFG Member in NARCCAP and Its Relationship with Large-Scale Circulation Biases

Assessment of past-climate simulations of regional climate models(RCMs)is important for understanding the reliability of RCMs when used to project future regional climate.Here,we assess the performance and discuss possible causes of biases in a WRF-based RCM with a grid spacing of 50 km,named WRFG,from the North American Regional Climate Change Assessment Program(NARCCAP)in simulating wet season precipitation over the Central United States for a period when observational data are available.The RCM reproduces key features of the precipitation distribution characteristics during late spring to early summer,although it tends to underestimate the magnitude of precipitation.This dry bias is partially due to the model’s lack of skill in simulating nocturnal precipitation related to the lack of eastward propagating convective systems in the simulation.Inaccuracy in reproducing large-scale circulation and environmental conditions is another contributing factor.The too weak simulated pressure gradient between the Rocky Mountains and the Gulf of Mexico results in weaker southerly winds in between,leading to a reduction of warm moist air transport from the Gulf to the Central Great Plains.The simulated low-level horizontal convergence fields are less favorable for upward motion than in the NARR and hence,for the development of moist convection as well.Therefore,a careful examination of an RCM’s deficiencies and the identification of the source of errors are important when using the RCM to project precipitation changes in future climate scenarios.

Transient receptor potential vanilloid 1 involved in the analgesic effects of total flavonoids extracted from Longxuejie(Resina Dracaenae Cochinchinensis)

OBJECTIVE: To evaluate the analgesic effects of total flavonoids of Longxuejie(Resina Dracaenae Cochinchinensis)(TFDB) and explore the possible analgesic mechanism associated with transient receptor potential vanilloid 1(TRPV1).METHODS: Whole-cell patch clamp technique was used to observe the effects of TFDB on capsaicin-induced TRPV1 currents. Rat experiments in vivo were used to observe the analgesic effects of TFDB. Western blot and immunofluorescence experiments were used to test the change of TRPV1 expression in DRG neurons induced by TFDB.RESULTS: Results showed that TFDB inhibited capsaicin-induced TRPV1 receptor currents in acutely isolated dorsal root ganglion(DRG) neurons of rats and the half inhibitory concentration was(16.7 ± 1.6) mg/L.TFDB(2-20 mg/kg) showed analgesic activity in the phase Ⅱ of formalin test and(0.02-2 mg per paw)reduced capsaicin-induced licking times of rats. TFDB(20 mg/kg) was fully efficacious on complete Freund's adjuvant(CFA)-induced inflammatory thermal hyperalgesia and capsaicin could weaken the analgesic effects. The level of TRPV1 expressions of DRG neurons was also decreased in TFDB-treated CFA-inflammatory pain rats.CONCLUSION: All these results indicated that the analgesic effect of TFDB may contribute to their modulations on both function and expression of TRPV1 channels in DRG neurons.

OBJECTIVE: To reveal the potential underlying mechanism of the Shenqi Tiaoshen formula(参芪调肾方, SQTS) in the treatment of chronic obstructive pulmonary disease(COPD) by utilizing network pharmacology, molecular docking, and experimental verification. METHODS: Multiple open-source databases and research related to Traditional Chinese Medicine or compounds were employed to screen active ingredients and corresponding potential targets of the SQTS. The protein-protein interaction network screened hub genes, the relevant molecular mechanism and gene regulation were initially identified through the Kyoto Encyclopedia of Genes and Genomes(KEGG) and Gene Ontology(GO) analysis, and molecular docking was used to confirm further the interaction of the main components bound to the core targets. In vivo experiments on the COPD combined Qi-deficiency syndrome rat model were performed to verify the intervention effects and predicted potential molecular mechanisms of the SQTS. RESULTS: This study selected 156 active compounds and 326 candidate targets for treating COPD. Quercetin, Nobiletin, Kaempferol, Luteolin, Ginsenoside Rh2 and Formononetin were probably the main active compounds of SQTS in COPD treatment as they affected the most COPD-related targets, and interleukin-1(IL-6), signal transducing activator of transcription 3(STAT3), matrix metalloproteinase-9(MMP9), vascular endothelial growth factor A(VEGFA), protein kinase B(AKT1), hypoxiainducible factor-1α(HIF-1α), and forkhead box O3(FoxO3) were identified as the hub genes associated with its therapeutic effect. KEGG analysis was mainly enriched in the signaling pathways closely related to inflammation, immunity and oxidative stress, such as HIF-1, tumor necrosis factor(TNF), phosphatidylinositol 3 kinase(PI3K)-AKT, FoxO, apoptosis, IL-17, and toll-like receptor. Molecular docking confirmed that the main active components shared a good affinity with the hub genes. Bacillus cereus,a spore-forming bacterium and frequent cause of food poisoning,poses a safety threat to dairy and rice industries due to its high contamination rates and ability to produce toxins such as cereulide.Because of widespread presence and thermal resistance of the spores,B.cereus cannot be eliminated from the environment and may survive in processing plants.Surviving spores can develop into vegetative cells,leading to a heightened risk of cereulide production in the processing environment.Both spores and vegetative cells have the ability to adhere to the surfaces of dairy plants and form biofilms,serving as the site for cereulide production and accumulation.Therefore,it is crucial for the food industry to address potential sources and pathways of B.cereus contamination and their connections to cereulide production in processing lines.In this review,sources of contamination of B.cereus,including spores,vegetative cells,and biofilms and their potential role in cereulide production at each stage of dairy and cooked rice processing were analyzed.In addition,control methods to prevent B.cereus contamination and cereulide production in processing lines were proposed,offering valuable insights for improving microbial risk management in the food industry.

Metal–ligand interfaces for well-defined gold nanoclusters

Ligand engineering for well-defined gold nanoclusters(Au NCs) is getting more extensive attention. Organizing the Au-ligand interfaces on gold NCs can achieve the structural and functional control. This review focuses on the Au-ligand interfaces including gold-phosphorus(Au-P), gold-sulfur(Au-S), gold-selenium(Au-Se), gold-carbon(Au-C), and gold-nitrogen(Au-N), derived from the bonding between Au atoms and the different ligands(e.g., organic phosphine, thiolate, selenolate, alkynyl,n-heterocyclic carbene and nitrogenous ligands). The formation mechanism of Au-ligand interfaces is well discussed. In addition, the effects of Au-ligand interfaces on the stability, optical property, and catalysis are also presented. We hope the advances in this research area can boost the development of Au NC sciences.

气相色谱-质谱法(GC-MS)的四极杆检测器具有扫描速率低、离子流失率高、浓度检测范围窄的特点,这些缺陷限制了该技术在代谢组学领域的广泛应用,因此亟需建立一种基于GC-MS的高覆盖率代谢组学分析方法。本文提出了一种基于GC-MS的广泛靶向代谢组学方法,广泛靶向代谢组学结合了靶向和非靶向的优点,可以实现对代谢物质的定性和半定量检测,该方法以The Fiehn library(FiehnLib)数据库中的代谢物质信息为基础,建立直链脂肪酸甲酯(FAMEs)的保留时间与FiehnLib数据库中的保留指数(RI)的关系,根据FiehnLib数据库中的保留指数计算数据库中代谢物质在具体实验条件下的保留时间;对比分析并确定保留时间的阈值为0.15 min,优化最佳扫描间隔为0.20 s;优化代谢物质的定量离子以避免出峰时间相近离子的干扰;最终构建了含有611种代谢物质的选择离子监测(SIM)方法表,这611种代谢物质覆盖了KEGG(Kyoto Encyclopedia of Genes and Genomes)中65%的代谢通路。与全扫描非靶向GC-MS方法相比,该广泛靶向GC-MS方法所检测的代谢物质数量增加20%~30%,信噪比提高15%~20%;稳定性试验结果表明,使用该方法分析样品时,84%的代谢物质保留时间的日内相对标准偏差(RSD)均小于2%,91%的代谢物质保留时间的日内RSD均小于3%;54%的代谢物质保留时间的日间RSD均小于2%,76%的代谢物质保留时间的日间RSD均小于3%;通过对常见生物样本的检测分析,证明该方法大大提升了被检测到的代谢物质的数量和信噪比,可扩展GC-MS在代谢组学中的应用范围。

Growth,optical dispersion and magnetic behavior of Dy^(3+) doped yttrium iron garnet crystals

Y_(3)Fe_(5)O_(12)(YIG)crystals are highly desirable for the fabrication of mid-infrared isolators with the rapid growth of optical communications,although it is rather challenging to grow large size and high-quality single crystals.Dy^(3+)doping is expected to improve the optical and magneto-optical prope rties.However,high quality Dy:YIG crystals and the adjustment of Dy^(3+)on the structure and optical behavior of YIG crystal remain unclear,impeding its practical applications.Herein,a series of Y_(3-x)Dy_(x)Fe_(5)O_(12)(x=0,0.5,1.0,1.5,3)solid solution crystals was grown by the flux-Bridgman method and single crystals up to 25 mm were obtained.With the introduction of Dy^(3+),lattice parameters are gradually enlarged from 1.2379 nm(YIG)to 1.2420 nm(DyIG).Typical Dy^(3+)absorption peaks are observed around 1070,1265 and 1670 nm.The refractive index decreases from 2.37(500 nm)to 2.10(2500 nm)for YIG crystal,and it reduces from 2.47(500 nm)to 2.16(2500 nm)for DyIG crystal.The optical bandgaps remain almost unchanged for Dy:YIG crystals.The optical dispersion of the refractive indices was finely fitted by the Wemple and DiDomenico(WDD)and the Sellmeier models,respectively.With the increase of Dy^(3+)content,the saturation magnetization(Ms)decreases significantly from 23.62 emu/g(YIG)to 5.33 emu/g(DyIG).Small coercive field is persisted for this system,endowing small external magnetic field.These results provide valuable references for the manipulation of rare earths on the properties of magnetooptical crystals,which is beneficial to the design of high-performance garnet crystals for the application of optical switching and non-reciprocal related devices.

建设高质量教育体系是新时期教育改革发展的方向。民办高等教育作为我国高等教育系统的重要组成部分,实现高质量发展有其必要性。该文首先分析民办高等教育高质量发展的核心要义,明确其内涵与阶段性特征。其次,基于民办高等教育发展现状,分别从宏观与微观层面指出当前民办高等教育高质量发展面临的现实困境,即存在融合发展程度不够、办学同质化现象严重、人才培养重结果轻过程、高层次人才缺乏、经费来源相对单一、优质教学资源不足等问题。最后,结合民办高校体制机制优势,提出实现高质量发展的路径:把握发展趋势,对接现代职业教育;坚持质量为本,构建人才培养质量管理模式;加大引进与培养,提升教师发展能力;发挥资源优势,提供优质教育服务;发挥多方力量,多渠道筹措办学经费。如此,以推动民办高等教育更高质量、更有特色地发展。

基于GA改进BP神经网络预测热变形流变应力模型的建立

应力-应变曲线对研究金属热变形过程中的加工硬化、动态再结晶和动态回复的变化具有重要的意义,而预测不同热变形参数下的应力-应变曲线有助于研究热加工过程中金属的可加工性和不稳定性。在应变速率为0.01~3 s^(-1)以及变形温度为1000~1200℃条件下,利用Gleeble-3500热模拟试验机对Nb-V-Ti微合金钢进行热压缩实验,研究了Nb-V-Ti微合金钢的热变形行为。建立BP神经网络模型和基于GA改进BP神经网络模型,分别预测在应变速率0.5 s^(-1)、变形温度1050℃和应变速率1 s^(-1)、变形温度1100℃条件下的流动应力行为并验证模型效果。研究结果表明:经GA改进后的BP神经网络模型对测试数据的应力-应变曲线与实验曲线具有很好的吻合,相关系数分别达0.99202和0.99734,误差仅为2.7816%和2.1703%,预测结果与实验结果相对误差在[-2,2]范围内,证明了模型的预测可靠性,且适用于较广的应变范围,为工业生产轧制工艺提供理论指导。
