Molecular mechanism underlying the functional loss of cyclindependent kinase inhibitors p16 and p27 in hepatocellular carcinoma

Hepatocellular carcinoma （HCC） is one of the most common human cancers, and its incidence is still increasing in many countries. The prognosis of HCC patients remains poor, and identification of useful molecular prognostic markers is required. Many recent studies have shown that functional alterations of cellcycle regulators can be observed in HCC. Among the various types of cell-cycle regulators, p16 and p27 are frequently inactivated in HCC and are considered to be potent tumor suppressors, p16, a G1-specific cell-cycle inhibitor that prevents the association of cyclindependent kinase （CDK） 4 and CDK6 with cyclin DI, is frequently inactivated in HCC via CpG methylation of its promoter region, p16 may be involved in the early steps of hepatocarcinogenesis, since p16 gene methylation has been detected in subsets of pre-neoplastic liver cirrhosis patients, p27, a negative regulator of the G1-S phase transition through inhibition of the kinase activities of Cdk2/cyclin A and Cdk2/cyclin E complexes, is now considered to be an adverse prognostic factor in HCC. In some cases of HCC with increased cell proliferation, p27 is overexpressed but inactivated by sequestration into cyclin D1-CDK4-containing complexes. Since loss of p16 is closely related to functional inactivation of p27 in HCC, investigating both p16 and p27 may be useful for precise prognostic predictions in individuals with HCC.

Hepatocellular carcinoma (HCC) is a common malignant disease with poor prognosis. To improve the clinical outcome, early diagnosis of HCC arising from nonviral agents and hepatitis virus is important. Among several etiological factors, mycotoxins defined as carcinogens by the International Agency for Research in Cancer (IARC) might be one of the critical risk factors for nonviral HCC. Aflatoxin B1 is the most well-known carcinogenic mycotoxin for HCC, but the role of the other types of mycotoxin remains unclear. Several studies have reported that a chromatographic separation technique based on high-performance liquid chromatography can successfully detect the concentration of mycotoxins in plasma. Recently, serum level of ochratoxin A (OTA), a widely distributed mycotoxin classified as Group 2B by IARC, was evaluated in HCC patients in Egypt. The results suggested that serum OTA levels might be a good biomarker for HCC. In this article, we review recent studies of OTA, and discuss its possible significance as a biomarker of HCC.

AIM:To reveal the manner of hepatocellular carcinoma (HCC) development in patients with nonalcoholic steatohepatitis(NASH) focusing on multicentric occurrence (MO) of ***:We compared clinicopathological characteristics between patients with and without MO of HCC arising from NASH *** clinical features were implicated with reference to the literature ***:MO of HCC was identified with histological proof in 4 out of 12 patients with NASH-related HCC(2 males and 2 females).One patient had synchronous MO;an advanced HCC,two well-differentiated HCCs and a dysplastic nodule,followed by the development of metachronous MO of *** other three patients had multiple advanced HCCs accompanied by a well-differentiated HCC or a dysplastic *** these three patients,one had synchronous MO,one had metachronous MO and the other had both synchronous and metachronous *** were no obvious differences between the patients with or without MO in terms of liver function tests,tumor markers and anatomical extent of *** the other hand,all four patients with MO of HCC were older than 70 years old and had the comorbidities of obesity,type 2 diabetes mellitus(T2DM),hypertension and *** these conditions were not limited to MO of HCC,all the conditions were met in only one of eight patients without MO of ***,concurrence of these conditions may be a predisposing situation to synchronous MO of *** particular,old age,T2DM and cirrhosis were suggested to be prerequisite for MO because these factors were depicted in common among two other cases with MO of HCC under NASH in the ***:The putative predisposing factors and necessary preconditions for synchronous MO of HCC in NASH were suggested in this *** investigations are required to clarify the accurate prevalence and predictors of MO to establish better strategies for treatment and prevention leading to the prognostic improvement in NASH.

Miniaturization of Bubbles by Shock Waves in Gas-Liquid Two-phase Flow in the Venturi Tube

Fine bubbles have been widely applied in many fields such as industry,medical engineering and ***,many attentions have been paid to the study of fine bubble generations in order to increase the yield while decrease the ***,the generation process of fine bubbles is a quite complicated process in which multiple hydrodynamic forces are interacted in the gas-liquid two-phase *** studies focus on the techniques of the converging-diverging nozzle(venturi tube)generator,which is famous for its simple and cheap features,and generates fine bubbles by using the miniaturization phenomenon of bubbles occurring in the venturi ***,the impact conditions on the amount and size of bubbles such as nozzle geometry and bleed air haven’t been investigated *** this work,we implement many experiments on the venturi tube fine bubble generators with different geometries and generating conditions,and evaluate different factors impacting the production components such as the volume and the bubble *** experimental results show that the supersonic flow filed in the venturi tube promotes the miniaturization of the bubbles,and the convergent angle of the nozzle and air bleed have a great impact on the size and volume of bubbles.

When the underexpanded supersonic jet impinges on the obstacle, it is well known that the self-induced flow os- cillation occurs. This oscillation depends on the pressure ratio in the flowfield, the position of an obstacle and is related with the noise problems of aeronautical and other industrial engineering. The characteristic and the mechanism of self-induced flow oscillation, have to be clarified to control various noise problems. But, it seems that the characteristics of the oscillated flowfield and the mechanism of an oscillation have to be more cleared to control the oscillation. This paper aims to clarify the effect of the pressure ratio and the obstacle position and the mechanism of self-induced flow oscillation by numerical analysis and experiment, when the underexpanded su- personic jet impinges on the cylindrical body. From the result of this study, it is clear that occurrence of the self-induced flow osciUation depends on the pressure balance in the flowfield.

When the underexpanded supersonic jet impinges on the obstacle, it is well known that the self-induced flow oscillation occurs at the specific condition of the pressure ratio in the flowfield, the position of an obstacle and so on. This oscillation is related with the noise problems of aeronautical and other industrial engineering so that the characteristic and the mechanism of self-induced flow oscillation have to be cleared to control the various noise problems. But, it seems that the characteristics of the oscillated flowfield and the mechanism of oscillation have to be more clear to control the oscillation. This paper aims to clarify the effect of the plate position and the width for the self-induced flow oscillation of an underexpanded supersonic jet impinging on the perpendicular plate by the experiment and the numerical analysis. From the results, it is clear that the occurring domain of the self-induced flow oscillation and its dimension strongly depend on the plate position and the width.

When the high-pressure gas is exhausted to the vacuum chamber from the supersonic nozzle, the overexpanded supersonic jet is formed at specific condition. In two-dimensional supersonic jet, furthermore, it is known that the hysteresis phenomena for the reflection type of shock wave in the flow field is occurred under the quasi-steady flow and for instance, the transitional pressure ratio between the regular reflection （RR） and Mach reflection （MR） is affected by this phenomenon. Many papers have described the hysteresis phenomena for underexpanded supersonic jet, but this phenomenon under the overexpanded axisymmetric jet has not been detailed in the past papers. The purpose of this study is to clear the hysteresis phenomena for the reflection type of shock wave at the overexpanded axisymmetric jet using the TVD method and to discuss the characteristic of hysteresis phenomena.

Performance and Internal Flow of Sirocco Fan Using Contra-Rotating Rotors

A sirocco fan using contra-rotating rotors in which an inner rotor is settled inside the sirocco fan rotor and each rotor rotates in an opposite direction was proposed for the purpose of getting the higher pressure and making the structure of a sirocco fan more compact. If the high discharge pressure is obtained with the adoption of the contra-rotating rotors, it could be used for various purposes. Pressure coefficient of a sirocco fan with contra-rotating rotors is 2.5 times as high as the conventional sirocco fan and the maximum efficiency point of contra-rotating rotors shifts to larger flow rate than a conventional sirocco fan. On the other hand, it was clarified from the flow measurement results that circumferential velocity component at the outlet of the outer rotor of contra-rotating ro- tors becomes larger than a conventional one. In the present paper, the performance of a conventional sirocco fan and a sirocco fan with contra-rotating rotors are shown and the internal flow field at the outlet of outer rotor of both cases is clarified. Then, the effect of different kind of contra-rotating rotors on the performance and internal flow field is investigated and the rotor design with higher performance would be discussed.

When the shock wave propagating in the straight circular tube reaches at the open end, the impulsive wave is generated by the emission of a shock wave from an open end, and unsteady pulse jet is formed near the open end behind the impulsive wave under the specific condition. The pulse jet transits to spherical shock wave with the increase in the strength of shock wave. The strength is dependent on the Mach number of shock wave, which attenuates by propagation distance from the open end. In this study, the mechanism of generating the unsteady pulse jet, the characteristics of the pressure distribution in the flow field and the emission of shock wave from straight circular tube which has the infinite flange at open end are analyzed numerically by the TVD method. Strength of spherical shock wave, relation of shock wave Mach number,distance decay of spherical shock wave and directional characteristics are clarified.
