Evaluation of the Blood Pressure Lowering-Effect of the Lactotripeptides Valine-Proline-Proline and Isoleucine-Proline-Proline in Non-Hypertensive Japanese Subjects through a Meta-Analysis of Randomized-Controlled Studies

Several studies have reported the ability of the lactotripeptides valine-proline-proline (VPP) and isoleucine-proline-proline (IPP) to lower systolic blood pressure (SBP), including in Japanese populations. But the magnitude of the reported changes differs across trials. Conclusions from a previous meta-analysis in Japanese subjects suggest that this may be due at least partly to differences in subjects’ blood pressure (BP) status. Therefore, we decided to resume this analysis, focusing only on non-hypertensive subjects and including newly-published eligible studies, in order to further evaluate the SBP-lowering effect of VPP/IPP and study the influence of the ingested dose, type of ingredient (enzymatic or fermented) and food product (drink or supplement). The systematic search of four databases (including two in Japanese) allowed to identify 11 relevant randomized-controlled trials (581 subjects), which were included in the meta-analysis. Results reported a significant decrease in SBP following VPP/IPP intake in non-hypertensive Japanese individuals, with an estimated effect-size of -3.44 mm Hg (95% CI, -4.53 to -2.34, P < 0.0001) as compared to placebo. There was no indication of heterogeneity or publication bias. Furthermore, the type of food product and ingredient did not influence the SBP-lowering effect, which was significant and of same order of magnitude with either type of product and ingredient. Besides, the SBP-lowering effect remained significant when limiting to studies testing usual daily amounts of VPP/IPP (≤5 mg/d). This updated meta-analysis therefore confirms that VPP/IPP are effective in reducing SBP in non-hypertensive Japanese individuals, for amounts that may be ingested on a daily basis, and independently of the types of ingredient/food consumed. VPP/IPP-containing foods could therefore contribute to a better control of high-normal BP and/or to the maintenance of normal BP, and by such, may play a role in preventing high BP in individuals wit

Numerical Investigation for the Effect of Unsaturated Aliphatic Hydrocarbons Addition on Precursors on Carbon Black

To investigate the effect of unsaturated aliphatic hydrocarbons on PAHs （polycyclic aromatic hydrocarbons） formation, numerical analysis using detailed reaction mechanism is performed. For the carbon black produced by benzene feedstock, three kinds of unsaturated aliphatic hydrocarbons （acetylene, diacetylene, and vinylacetylene） are added. The detailed reaction mechanism which is proposed by Wang and Frenklach is consisted by 527 reactions and 99 chemical species. The formations of PAHs and nuclei are promoted by unsaturated aliphatic hydrocarbons addition. There exits optimal benzene/acetylene ratio （XA/XB = 0.50）, benzene/diacetylene mixture ratio （XD/XB = 0.60）, benzene/vinyl-acetylene mixture ratio （XV/XB = 0.25）. The formation of nuclei is most promoted by acetylene addition, and the benzene/acetylene mixture ratio is 0.50.

Change in Aggregate Shape of Heat Treated Carbon Nanoparticles

To better understand the fusion mechanism of heated carbon black, heat treatment is conducted for carbon black produced by benzene pyrolysis. The effects of （a） heating time, （b） heating temperature and （c） heating rate on the aggregate shape and mean primary particle diameter of the carbon black are investigated using TEM （transmission electron microscopy）. The mean primary particle diameter does not change significantly when carbon black is heat treated. For short heating times and low heating temperatures, the aggregate shapes become simple when compared with those of non-heated carbon black, and shapes become complex with an increase in the heating time. Also, for low heating rates, the aggregate shapes become significantly simple when compared with those of non-heated carbon black. The results of this study suggest that sintering between primary particles is promoted under relatively low heating temperatures, and Ostwald ripening among aggregates is promoted under relatively high heating temperatures.

Application of DEM modified with enlarged particle model to simulation of bead motion in a bead mill

We applied the discrete element method （DEM） of simulation modified by an enlarged particle model to simulate bead motion in a large bead mill. The stainless-steel bead mill has inner diameter of 102 mm and mill length of 198 mm. The bead diameter and filling ratio were fixed respectively at 0.5 mm and 85%. The agitator rotational speed was changed from 1863 to 3261 rpm. The bead motion was monitored experimentally using a high-speed video camera through a transparent mill body. For the simulation, enlarged particle sizes were set as 3-6 mm in diameter. With the DEM modified by the enlarged particle model, the motion of enlarged particles in a mill was *** velocity data of the simulated enlarged particles were compared with those obtained in the experiment. The simulated velocity of the enlarged particles depends on the virtual frictional coefficient in the DEM model. The optimized value of the virtual frictional coefficient can be determined by considering the accumulated mean value. Results show that the velocity of the enlarged particles simulated increases with an increase in the optimum virtual frictional coefficient, but the simulated velocity agrees well with that determined experimentally by optimizing the virtual frictional coefficient in the simulation. The computing time in the simulation decreases with increased particle size.

MUC5AC/β-catenin expression and KRAS gene alteration in laterally spreading colorectal tumors

AIM： To clarify differences in mucin phenotype, prolif- erative activity and oncogenetic alteration among sub- types of colorectal laterally spreading tumor （LST）. METHODS： LSTs, defined as superficial elevated lesions greater than 10 mm in diameter with a low vertical axis, were macroscopically classified into two subtypes：（1） a granular type （Gr-LST） composed of superficially spread- ing aggregates of nodules forming a fiat-based lesion with a granulonodular and uneven surface; and （2） a non-granular type （NGr-LST） with a flat smooth surface and an absence of granulonodular formation. A total of 69 LSTs, comprising 36 Gr-LSTs and 33 NGr-LSTs, were immunohistochemically stained with MUC2, MUC5AC, MUC6, CD10 （markers of gastrointestinal cell lineage）, p53, 13-catenin and Ki-67 antibodies, and examined for alteration in exon 1 of v-Ki-ras2 Kirsten rat sarcoma viral oncogene homolog （KRAS） and exon 15 of v-raf murine sarcoma viral oncogene homologue B1 （BRAF） by poly- merase chain reaction followed by direct sequencing. RESULTS： Histologically, 15 Gr-LST samples were ad- enomas with low-grade dysplasia （LGD）, 12 were high- grade dysplasia （HGD） and 9 were adenocarcinomas invading the submucosa （INV）, while 12 NGr-LSTs demonstrated LGD, 14 HGD and 7 INV. In the proximal colon, MUC5AC expression was significantly higher in the Gr-type than the NGr-type. MUC6 was expressed only in NGr-LST. MUC2 or CD10 did not differ. P53 ex- pression demonstrated a significant stepwise increment in progression through LGD-HGD-INV with both types of LST. Nuclear β-catenin expression was significantly higher in the NGr-type. Ki-67 expression was signifi- cantly higher in the Gr-type in the lower one third zone of the tumor. In proximal, but not distal colon tumors, the incidence of KRAS provided mutation was signifi- cantly higher in the Gr-type harboring a specific muta- tional pattern （G12V）. New simulation model for bone formation markers in osteoporosis patients treated with once-weekly teriparatide

Daily 20-mg and once-weekly 56.5-mg teriparatide（parathyroid hormone 1–34） treatment regimens increase bone mineral density（BMD） and prevent fractures, but changes in bone turnover markers differ between the two regimens. The aim of the present study was to explain changes in bone turnover markers using once-weekly teriparatide with a simulation model. Temporary increases in bone formation markers and subsequent decreases were observed during once-weekly teriparatide treatment for 72 weeks. These observations support the hypothesis that repeated weekly teriparatide administration stimulates bone remodeling, replacing old bone with new bone and leading to a reduction in the active remodeling surface. A simulation model was developed based on the iterative remodeling cycle that occurs on residual old bone. An increase in bone formation and a subsequent decrease were observed in the preliminary simulation. For each fitted time point, the predicted value was compared to the absolute values of the bone formation and resorption markers and lumbar BMD. The simulation model strongly matched actual changes in bone turnover markers and BMD. This simulation model indicates increased bone formation marker levels in the early stage and a subsequent decrease. It is therefore concluded that remodeling-based bone formation persisted during the entire treatment period with once-weekly teriparatide.

A laterally-spreading tumor in a colonic interposition treated by endoscopic submucosal dissection

Herein we describe an early colonic carcinoma which developed in a colonic interposition 14 years after surgery for esophageal cancer, which was successfully treated by endoscopic submucosal dissection (ESD). An 80-year-old man underwent colonic interposition between the upper esophagus and stomach after surgery for an early esophageal squamous cell carcinoma in 1994. He received a surveillance endoscopy, and a laterally-spreading tumor of granular type, approximately 20 mm in size, was identified in the colonic interposition. An endoscopic biopsy revealed moderately differentiated adenocarcinoma histologically, however, we diagnosed the lesion as an intramucosal carcinoma based on the endoscopic findings. The lesion was safely and completely removed en bloc by ESD using a bipolar knife. Histologically, the lesion was an intramucosal moderately differentiated adenocarcinoma in a tubular adenoma.

Atypical lipomatous tumor in the ligamentum teres of liver:A case report and review of the literature

A 61-year-old male was referred to our hospital with a three-month history of persistent epigastralgia and right hypochondralgia. Initial examination revealed a fist-size mass at the epigastric fossa. Ultrasonography showed a hemangioma and a mosaic echoic lesion in the ventromedian with poor blood-flow signal and linear hyperechoic part inside, and a clear border to the surroundings. Dynamic computed tomography revealed a highly enhanced effect from the portal-venous phase continuing to the equilibrium phase. T1-weighted gadolinium-ethoxybenzyl-diethylenetriamine pentaacetic acid-enhanced image revealed a high intensity effect at the early phase that continued to the next phase. On the other hand, it contained a low intensity area by a fat suppression of that image. In addition, a T2-weighted image did not show a high intensity effect. Laparotomy was performed on the second day of hospitalization. The tumor had arisen from the ligamentum teres of the liver, and no metastasis or invasion of other organs was noted. It consisted of a lipid component of mature adipocytes and a fibrous component of deep dyeing pleomorphic or multinuclear atypical stromal cells. Immunohistochemical study of the atypical stromal cells demonstrated that they were positive for MDM2 and CDK4. A pathological diagnosis of atypical lipomatous tumor(ALT) was made, and the patient was discharged on the eighth day following the procedure. At the 6-mo follow-up dynamic CT, the patient was free of recurrence or metastasis. We experienced a patient with ALT in the ligamentum teres of the liver. This case suggests the need for a careful and detailed examination when encountering patients presenting with a mass; when neoplastic lesion is confirmed by image inspection, we should thoroughly investigate, including further image investigations and pathologic examination. The latter is the most important.
