Effects of Psychological Stress from a National License Examination on the Urine 8-Hydroxy-Deoxyguanosine Levels in Young Female Students, Taking into Account the Menstrual Cycle

Objective: Urine 8-hydroxy-deoxyguanosine (U8-OHdG) was a marker of oxidative stress-induced DNA damage;it was increased by psychological stress. Since the menstrual cycle may confound or modify the association between psychological stress and U8-OHdG. The aim of this study was to verify the effect of psychological stress from a national license examination on levels of U8-OHdG, which is a biomarker of oxidative stress. And the effects of women’s menstrual cycles, which should be considered in mental and physical assessments, on U8-OHdG, were evaluated. Methods: The subjects were 18 female university students at a medical university in whom Self-rating Depression Scale (SDS) scores, State-Trait Anxiety Inventory (STAI) scores, and U8-OHdG levels were measured in 3 phases of the menstrual cycle, the follicular phase, ovulatory phase, and luteal phase. The mean values were taken for the group during a time of classroom learning. The same measurements were also made one week before and the day after a national license examination and the measurements were compared among the three periods. Results and Conclusion: State anxiety and U8-OHdG levels were significantly higher in those with a week before the national license examination than in those with classroom lecture (State anxiety: p = 0.002, U8-OHdG levels: p = 0.007). The menstrual cycle did not show a significant correlation with U8-OHdG levels. This study demonstrated that levels of the oxidative stress biomarker U8-OHdG are not affected by changes in the menstrual cycle. It also showed that U8-OHdG levels increased with the psychological stress of a national license examination.

Development of a Method to Produce Chromosome Lacking Lines (CLLs) in <i>Nicotiana tabacum</i>L. “Red Russian”

Monosomic lines of Nicotiana tabacum are helpful to confirm the location of genes on specific chromosomes. In the cross N. nudicaulis and N. tabacum, hybrid seedlings express lethal symptoms, which are controlled by the S subgenome of N. tabacum. To identify the responsible chromosome, we needed to produce chromosome lacking lines (CLLs) of N. tabacum L. “Red Russian” and use them to cross with N. nudicaulis. From a cross of (N. tabacum × N. tomentosiformis) × N. tabacum, 380 BC1 individuals were obtained. Using a Haplo-Q line (a monosomic line lacking the single linkage group 11) and N. tabacum, we found that qPCR is a simple and reliable screening method for CLLs of N. tabacum. The marker PT30342 is located on linkage group 11, and the -Ct value (Ct Actin - Ct PT30342) was 2.0 for a disomic line and was 1.097 for a Haplo-Q line. By the use of flow cytometry, qPCR and chromosome counting together as a screening method, we identified 6 CLLs lacking 2 to 6 chromosomes. Compared with conventional methods, our method is a rapid technique for making and screening CLLs ofthe S or S/T subgenome of N. tabacum. Further, these CLLs will be useful to identify the location of two or more factors on chromosomes controlling a variety of genetic problems affecting breeding. Here, we only made CLLs of the S or S/T subgenome of N. tabacum. We will use the method established in this study to produce CLLs of the T subgenome of N. tabacum, and gather a full set of CLLs of N. tabacum. qPCR could also be applied to the identification of chromosome aberrations in other plants.

Background: Health promotion for the elderly mainly targets improvements in physical function, and a lack of strengthening of subjective well-being as well as the creation of interaction-providing opportunities for activity and participation has been reported. Evaluations of positive and negative aspects for subjective well-being are needed. Elderly subjects were intervened through a 6-month complex-type program combining amusement and exercise and its influences on subjective well-being and physical and physiological functions were investigated. Methods: Subjects in this survey were 37 healthy elderly females living in M city, Hiroshima Prefecture (age: 69.7 ± 5.9 years old), and the survey was performed between September 2016 and March 2017. The complex-type program comprised amusement and exercise programs. The survey was performed at 4 time points during the intervention period: before the intervention and 1, 3, and 6 months during the intervention. Subjective well-being (Japanese version of the Subjective Well-being Inventory (SUBI): Mental health scores, Mental fatigue scores), salivary amylase levels, and the locomotive syndrome risk test (two-step test, stand-up test, and 25-question risk assessment) were surveyed at each time point. Regarding the duration of the intervention as a factor, changes in salivary amylase levels were analyzed using repeated measures one-way ANOVA and SUBI was tested using the Friedman test (EZR Ver 1.32). In addition, the relationship between the locomotive syndrome risk level and duration of the intervention was examined using Fisher’s test. Results: SUBI and the locomotive syndrome risk level significantly differed with time during the intervention. Slight changes were observed in salivary amylase levels during the intervention. Conclusion: The present results suggest that the complex-type program promoted interactions among community-dwelling elderly subjects and positively influenced the maintenance of and improvements in physical and physiological functions. Dear Editor,Prostate cancer is the most common cancer among men[1].Androgen deprivation therapy(ADT)has remained the primary treatment of metastatic-hormone-sensitive prostate cancer(mHSPC),providing a temporary disease control in the majority of patients.Despite initial ADT response,castration-resistance prostate cancer(CRPC)still develops.Previous study have attempted to determine possible biomarkers for poor prognosis in patients with CRPC[2].

AIM:To investigate the immunological repertoire in the peritoneal cavity of gastric cancer patients.METHODS:The peritoneal cavity is a compartment in which immunological host-tumor interactions can occur.However,the role of lymphocytes in the peritoneal cavity of gastric cancer patients is unclear.We observed 64 patients who underwent gastrectomy for gastric cancer and 11 patients who underwent laparoscopic cholecystectomy for gallstones and acted as controls.Lymphocytes isolated from both peripheral blood and peritoneal lavage were analyzed for surface markers of lymphocytes and their cytokine production by flow cytometry.CD4+CD25high T cells isolated from the patient's peripheral blood were co-cultivated for 4 d with the intra-peritoneal lymphocytes,and a cytokine assay was performed.RESULTS:At gastrectomy,CCR7 -CD45RA-CD8+ef-fector memory T cells were observed in the peritoneal cavity.The frequency of CD4+CD25 high T cells in both the peripheral blood and peritoneal cavity was elevated in patients at advanced stage[control vs stage Ⅳin the peripheral blood:6.89(3.39-10.4)vs 15.34(11.37-19.31),Pitoneal cavity:8.65(5.28-12.0)vs 19.56(14.81-24.32),Pitoneal cavity in patients with gastric cancer.Immune regulation at advanced stage is reversible at the point of gastrectomy.CONCLUSION:The immunological milieu in the peritoneal cavity of patients with advanced gastric cancer elicited a Th2 response even at gastrectomy,but this response was reversible.

We successfully demonstrated low power penalty operation of a cross-phase modulated (XPM) wavelength converter using a semiconductor optical amplifier (SOA) power equalizer. We also clarified the SOA equalizing level for more adaptive wavelength conversion and achieved a power penalty of less than 1 dB over the wide input dynamic range of 15 dB.
