Circulating proteomic biomarkers for diagnosing sporadic amyotrophic lateral sclerosis:a cross-sectional study

Biomarke rs are required for the early detection,prognosis prediction,and monitoring of amyotrophic lateral sclerosis,a progressive disease.Proteomics is an unbiased and quantitative method that can be used to detect neurochemical signatures to aid in the identification of candidate biomarke rs.In this study,we used a label-free quantitative proteomics approach to screen for substantially differentially regulated proteins in ten patients with sporadic amyotrophic lateral scle rosis compared with five healthy controls.Su bstantial upregulation of serum proteins related to multiple functional clusters was observed in patients with spo radic amyotrophic lateral sclerosis.Potential biomarke rs were selected based on functionality and expression specificity.To validate the proteomics profiles,blood samples from an additional cohort comprising 100 patients with sporadic amyotrophic lateral sclerosis and 100 healthy controls were subjected to enzyme-linked immunosorbent assay.Eight substantially upregulated serum proteins in patients with spora dic amyotrophic lateral sclerosis were selected,of which the cathelicidin-related antimicrobial peptide demonstrated the best discriminative ability between patients with sporadic amyotrophic lateral sclerosis and healthy controls(area under the curve[AUC]=0.713,P<0.0001).To further enhance diagnostic accuracy,a multi-protein combined discriminant algorithm was developed incorporating five proteins(hemoglobin beta,cathelicidin-related antimicrobial peptide,talin-1,zyxin,and translationally-controlled tumor protein).The algo rithm achieved an AUC of 0.811 and a P-value of<0.0001,resulting in 79%sensitivity and 71%specificity for the diagnosis of sporadic amyotrophic lateral scle rosis.Subsequently,the ability of candidate biomarkers to discriminate between early-stage amyotrophic lateral sclerosis patients and controls,as well as patients with different disease severities,was examined.A two-protein panel comprising talin-1 and translationally-controlled tumor protein effectively d

Background:The Global Leadership Initiative on Malnutrition(GLIM)criteria were published to build a global consensus on nutritional diagnosis.Reduced muscle mass is a phenotypic criterion with strong evidence to support its inclusion in the GLIM consensus criteria.However,there is no consensus regarding how to accurately measure and define reduced muscle mass in clinical settings.This study aimed to investigate the optimal reference values of skeletal muscle mass index for diagnosing sarcopenia and GLIM-defined malnutrition,as well as the prevalence of GLIM-defined malnutrition in hospitalized cirrhotic patients.Methods:This retrospective study was conducted on 1002 adult patients with liver cirrhosis between January 1,2018,and February 28,2022,at Beijing You-An Hospital,Capital Medical University.Adult patients with a clinical diagnosis of liver cirrhosis and who underwent an abdominal computed tomography(CT)examination during hospitalization were included in the study.These patients were randomly divided into a modeling group(cohort 1,667 patients)and a validation group(cohort 2,335 patients).In cohort 1,optimal cut-off values of skeletal muscle index at the third lumbar skeletal muscle index(L3-SMI)were determined using receiver operating characteristic analyses against in-hospital mortality in different gender groups.Next,patients in cohort 2 were screened for nutritional risk using the Nutritional Risk Screening 2002(NRS-2002),and malnutrition was diagnosed by GLIM criteria.Additionally,the reference values of reduced muscle mass in GLIM criteria were derived from the L3-SMI values from cohort 1.Multivariate logistic regression analysis was used to analyze the association between GLIM-defined malnutrition and clinical outcomes.Results:The optimal cut-off values of L3-SMI were 39.50 cm 2/m 2 for male patients and 33.06 cm 2/m 2 for female patients.Based on the cut-off values,31.63%(68/215)of the male patients and 23.3%(28/120)of the female patients had CT-determined sarcopenia in cohort 2.The prevalence of GL

Expression and clinical significance of pattern recognition receptor-associated genes in hand, foot and mouth disease

Objective:To explore which pattern recognition receptors(PRRs)play a key role in the development of hand,foot,and mouth disease(HFMD)by analyzing PRR-associated genes.Methods:We conducted a comparative analysis of PRR-associated gene expression in human peripheral blood mononuclear cells(PBMCs)infected with enterovirus 71(EV-A71)which were derived from patients with HFMD of different severities and at different stages.A total of 30 PRR-associated genes were identified as significantly upregulated both over time and across different EV-A71 isolates.Subsequently,ELISA was employed to quantify the expression of the six most prominent genes among these 30 identified genes,specifically,BST2,IRF7,IFI16,TRIM21,MX1,and DDX58.Results:Compared with those at the recovery stage,the expression levels of BST2(P=0.027),IFI16(P=0.016),MX1(P=0.046)and DDX58(P=0.008)in the acute stage of infection were significantly upregulated,while no significant difference in the expression levels of IRF7(P=0.495)and TRIM21(P=0.071)was found between different stages of the disease.The expression levels of BST2,IRF7,IFI16 and MX1 were significantly higher in children infected with single pathogen than those infected with mixed pathogens,and BST2,IRF7,IFI16 and MX1 expression levels were significantly lower in coxsackie B virus(COXB)positive patients than the negative patients.Expression levels of one or more of BST2,IRF7,IFI16,TRIM21,MX1 and DDX58 genes were correlated with PCT levels,various white blood cell counts,and serum antibody levels that reflect disease course of HFMD.Aspartate aminotransferase was correlated with BST2,MX1 and DDX58 expression levels.Conclusions:PRR-associated genes likely initiate the immune response in patients at the acute stage of HFMD.

We propose and experimentally realize a class of quasi-one-dimensional topological lattices whose unit cells are constructed by coupled multiple identical resonators,with uniform hopping and inversion symmetry.In the presence of coupling-path-induced effective zero hopping within the unit cells,the systems are characterized by complete multimerization with degenerate-1 energy edge states for open boundary condition.

CFD-DEM simulation of fluorination reaction in fluidized beds with local grid and time refinement method

The gas-solid reaction process with wide particle size distribution is extensively used in the chemical engineering field,especially the particle reacts with the gas gradually,such as fluorination reactions in fluidized beds.When the computational fluid dynamics-discrete element method(CFD-DEM)is used for the coupling simulation of multiphase and polydisperse particle reaction system,the grid size directly affects the accuracy of flow field information and simulation of chemical reaction.Furthermore,particle calculation time step will directly affect the efficiency of coupling calculation.In this work,a local grid and time step refinement method is proposed to simulate multiphase and polydisperse particle fluid-ization reaction system.In this method,the refined DEM grids are automatically generated in the computational domain around the fine particles,and the detailed fluid phase information is obtained with the interpolation algorithm.In the two-phase coupling process,particles are divided into different groups based on physical properties,each group has its own independent time step.The multistage conical-cylindrical spouted bed is proposed for the fluorination reaction process;the operating gas ve-locity range of the polydisperse particle system is extended by the new design while the particle size distribution changes with the gas-solid reaction process.It is demonstrated that the local grid and time step refinement method can improve the accuracy and efficiency of the traditional CFD-DEM method in the reaction process simulation,which describes a polydisperse particle system with wide particle size distribution.Aimed at improving the simulation accuracy and efficiency,this paper will be helpful for simulating the particle reaction process in the gas-solid fluidized bed and beneficial for the development of the CFD-DEM method.

A model for aircraft cabin evacuation considering passenger type

Pedestrian safety evacuation in aircraft cabins has been a challenging problem because of the aircraft’s unique characteristics,such as the diversity of passengers and the restricted evacuation environment.It is difficult to reproduce evacuation activities in aircraft cabin due to safety concerns and cost constraints.To fill this gap,an improved cellular automaton model of crowd evacuation for aircraft cabin is established by incorporating the characteristics of cabin space structures and passenger attributes.Passengers are divided into healthy individual passengers and disabled-healthy group passengers,whose movement mechanisms are quantified.Based on the constructed model,simulation experiments are conducted using the configuration cabin layout of B737-800 as an example.The results show that the evacuation time is prolonged with increased passenger density and the number of disabled passengers.Moreover,the overall evacuation time is insignificantly affected by whether disabled-healthy group passengers’seats are close to the aisle or window,and the evacuation efficiency is best when their seats are evenly distributed in the cabin.The evacuation time is the shortest when all cabin doors are open,and pedestrians are evacuated the slowest when the central emergency doors are closed.This study pro-vides valuable insights into effective strategies for pedestrian evacuation and crowd emergency management of civil aircraft.

Atomic arrays provide an important quantum optical platform with photon-mediated dipole–dipole interactions that can be engineered to realize key applications in quantum information processing.A major obstacle for such applications is the fast decay of the excited states.By controlling two-band Bloch oscillations of single excitation in an atomic array under an external magnetic field,here we show that exotic subradiance can be realized and maintained with orders of magnitude longer than the spontaneous decay time in atomic arrays with the finite size.The key finding is to show a way for preventing the wavepacket of excited states scattering into the dissipative zone inside the free space light cone,which therefore leads to the excitation staying at a subradiant state for an extremely long decay time.We show that such operation can be achieved by introducing a spatially linear potential from the external magnetic field in the atomic arrays and then manipulating interconnected two-band Bloch oscillations along opposite directions.Our results also point out the possibility of controllable switching between superradiant and subradiant states,which leads to potential applications in quantum storage.

User Preference Aware Hierarchical Edge-User Cooperative Caching Strategy

The emergence of various new services has posed a huge challenge to the existing network architecture.To improve the network delay and backhaul pressure,caching popular contents at the edge of network has been considered as a feasible scheme.However,how to efficiently utilize the limited caching resources to cache diverse contents has been confirmed as a tough problem in the past decade.In this paper,considering the time-varying user requests and the heterogeneous content sizes,a user preference aware hierarchical cooperative caching strategy in edge-user caching architecture is proposed.We divide the caching strategy into three phases,that is,the content placement,the content delivery and the content update.In the content placement phase,a cooperative content placement algorithm for local content popularity is designed to cache contents proactively.In the content delivery phase,a cooperative delivery algorithm is proposed to deliver the cached contents.In the content update phase,a content update algorithm is proposed according to the popularity of the contents.Finally,the proposed caching strategy is validated using the MovieLens dataset,and the results reveal that the proposed strategy improves the delay performance by at least 35.3%compared with the other three benchmark strategies.

Realizing high-rate capability and high-efficiency utilization of polyanionic cathode materials is of great importance for practical sodium-ion batteries(SIBs) since they usually suffer from extremely low electronic conductivity and limited ionic diffusion kinetics. Herein, taking Na_(3.5)V_(1.5)Mn_(0.5)(PO_(4))_(3)(NVMP) as an example, a reinforced concrete-like hierarchical and porous hybrid(NVMP@C@3DPG) built from 3D graphene(“rebar”) frameworks and in situ generated carbon coated NVMP(“concrete”) has been developed by a facile polymer assisted self-assembly and subsequent solid-state method. Such hybrids deliver superior rate capability(73.9 m Ah/g up to 20 C) and excellent cycling stability in a wide temperature range with a high specific capacity of 88.4 m Ah/g after 5000 cycles at 15 C at room temperature, and a high capacity retention of 97.1% after 500 cycles at 1 C(-20 ℃), and maintaining a high reversible capacity of 110.3 m Ah/g in full cell. This work offers a facile and efficient strategy to develop advanced polyanionic cathodes with high-efficiency utilization and 3D electron/ion transport systems.

目的了解全国中医院校护理学类本科专业招生现状,为中医院校培养具有中医特色的护理人才提供参考。方法采用目的抽样法,选取全国24所中医院校2021-2022年公布的护理学类本科专业招生情况作为研究对象,通过文本分析法进行资料分析。结果专业设置和专业布点在校际区域间差异较大,专业目标和课程设置对中医特色逐渐彰显,针对拔尖人才培养聚力不够,招生模式传统单一,生源质量整体偏弱。结论高校可通过优化专业结构、调整招生策略、深化教学改革、强化培养特色等,提升中医院校护理本科人才培养质量。
