The mechanisms for the production of hydrogen peroxide (H2O2) induced by abscisic acid (ABA) were investigated in suspension culture cells of tobacco BY-2 cells. The results showed that the immediate generation of H2O2, which was mainly derived from super- oxide dismutase-catalyzed dismutation of superoxide radical, was significantly induced by ABA. Furthermore, treatment of the cultured tobacco cells with ABA resulted in a time-dependent quick increase in plasma membrane (PM) NADPH oxidase activity, which coin- cided on time and magnitude with the elevation in ABA-induced accumulation of H2O2. Moreover, these enhanced effects were pro- nouncedly inhibited by two NADPH oxidase inhibitors, diphenylene iodonium and imidazole, suggesting that PM NADPH oxidase is involved in the rapid accumulation of H2O2 in cultured tobacco cells. In addition, analysis of the expression level of NtrbohD, a PM NADPH oxidase gene in tobacco, by RT-PCR and protein gel blot revealed that the gene at both mRNA and protein levels was upregulated by ABA, indicating that NtrbohD participates in the ABA-stimulated rapid production of H2O2 in tobacco culture cells. Taken together, these findings suggest that ABA induces the rapid accumulation of reactive oxygen species via NADPH oxidase in sus-pension culture cells of tobacco, and that NADPH oxidase and H2O2 appear to be important components in ABA signal transduction pathway in plants.

Walnut dreg protein hydrolysates(WDPHs)exhibit a variety of biological activities,however,the cyclooxygenase-2(COX-2)inhibitory peptide of WDPHs remain *** aim of this study was to rapidly screen for such peptides in WDPHs through a combination of in silico and in vitro *** total,1262 peptide sequences were observed by nano liquid chromatography/tandem mass spectrometry(nano LC-MS/MS)and 4 novel COX-2 inhibitory peptides(AGFP,FPGA,LFPD,and VGFP)were *** kinetic data indicated that AGFP,FPGA,and LFPD displayed mixed-type COX-2 inhibition,whereas VGFP was a non-competitive *** is mainly because the peptides form hydrogen bonds and hydrophobic interactions with residues in the COX-2 active *** results demonstrate that computer analysis combined with in vitro evaluation allows for rapid screening of COX-2 inhibitory peptides in walnut protein dregs.

In this paper, we will study the temporal-spatial landscape change from 1978 to 2001 in Daqing City, based on the mul- ti spectral scanner (MSS) and thematic mapper (TM) images of remote sensing (RS) of 1978, 1988, 1992, 1996 and 2001. In order to explain the effects of the component change on the whole landscape, we make a statistic analysis about the transfer probability matrix of the component types of the landscape by using the ARCGIS platform through the overlay analysis of different landscape components. Based on the transfer probability matrix, we calculate the in-transfer ratio, the out-transfer ratio and the contributor ratio of the special transfer process of the main landscape components. The results illustrate that the cultivated-land, grassland, swamp, and alkaline land are the main contributors resulting in the landscape change: the out-transfer ratios of grassland and swamp are much higher than the in-transfer ones; the in-transfer ratio of alkaline land is much higher than the out-transfer one; the in-transfer ratio of the land-lot is always higher than its out-transfer with the highest rate per year, although there is a small difference among the in-transfers and the out-transfers per year. The main landscape changing processes are the salination of cultivated-land, salination or desertification of grassland, salination and degeneration of swamp. Results in this paper can provide scientific basis for investigating the landscape change mechanism of resources-based city.

The Recognition of Glycolate Oxidase Apoprotein with Flavin Analogs in Higher Plants

The dependence of glycolate oxidase apoprotein (apoGO) activity on flavin analogs was surveyed in 9 higher plants from 7 families. Activities of all apoGOs depended not only on flavin mono- nucleotide (FMN) but also on flavin adenine dinucleotide (FAD), but not on riboflavin. The kinetic analysis showed that FMN was the optimum cofactor for apoGO from leaves of Brassica campestris. In plant kingdom, FMN, FAD and riboflavin are three flavin analogs with very similar structure, and they could coexist and be inter-converted from each other, so the question is how the apoprotein of glycolate oxidase (GO) recognized these flavin analogs. No inhibition effect of riboflavin on the activity of apoGO with FMN or FAD was found and no obvious quenching of riboflavin or apoGO protein fluorescence was detected with the addition of apoGO or riboflavin, respectively. These results indicated that riboflavin did not bind to apoGO tightly like FMN and FAD. Inorganic phosphate (Pi) did inhibit the activity of GO, and kinetic analysis revealed that this inhibition was caused by the competitive binding to apoGO between Pi and FMN. This competitive binding was further confirmed by the inhibition of Pi to the quenching of FMN and apoGO protein fluorescence with apoGO and FMN, respectively. It was suggested that the 5’-phosphate group of FMN or FAD may play a key role in the recognition and binding of riboflavin analog cofactors with apoGO.

Studies on the Relationship Between Grain-yield and Climatic Ecological Factors in Summer Corn Under Super-high-yielding Cultivation Conditions

Cultivation experiments on super-high-yield (^12000kg/ha) of summer corn (Zea mays L.) were conducted in Laizhou, Shangdong Province, from 1986 to 1997, and in Wenxian, henan Province, from 1996 to 1997. The results showed that requirements of accumulated temperature and hours of sunshine for super-high-yield of summer corn could be met in normal years in the areas of the Huanghuaihai Plain. Amount of precipitation influenced the yield most strongly in indirect way among all the meteorological factors, and the relationship between them displayed significant negative correlation (r= - 0.5418). The regression equation between yield and amount of precipitation at seedling stage and grain filling stage both reached significant level, and the partial regression coefficients were - 4.8735 and - 13.7415, respectively. Further research revealed that the key climatic-ecological factors influencing yield were as fellows: the average temperature in the third and the ninth Xun (note: a Xun indicates ten days and the accounting of Xun was from the corresponding sowing date), the hours of sunshine in the sixth and the eighth Xun, the amount of precipitation in the sixth, the seventh and the second Xun. Results obtained by analyzing yield components of summer corn showed that grain numbers per ear (GN/E) made greater contribution to super-high-yield than kernel weight (KW) and the numbers of ears did. The key factors influencing GN/E were the amount of precipitation in the sixth and the eighth Xun and the hours of sunshine in the sixth Xun, with the correlation coefficients of -0.6074, 0.5793 and 0.5854, respectively.

A pig BAC library was constructed with genomic DNA from a male Erhualian pig. After partial digestion with HindⅢor BamHⅠthe fragments obtained were cloned into the pBeloBAC11 vector. The library consists of 184320 clones which stored in 480 pieces 384-well plates (20 plates per superpool). A two-step 4-dimension PCR screening system was established to screen the positive clones. An average insert size of 128 kb was estimated from 105 randomly isolated clones, which indicates that the library is more than five times of genomic coverage. For the demonstration of the probability to pick out any unique genes or DNA markers from the library, 10 single-copy genes were screened out and the positive clones were yielded between 1 and 8 with an average of 3.6. Positive superpools were obtained for 32 microsatellite markers selected from different regions of pig genome.
