Oxidative stress(OS) plays a major role in the gastrointestinal disorders.Although probiotics were reported to repress OS,few researches compared the antioxidant ability of different Bacillus strains and decipher the mechanisms.To select a Bacillus strain with higher antioxidant capacity,we used HO to induce intestinal porcine epithelial cell 1(IPEC-1) OS model.The most suitable HO concentration and incubation time were determined by the half lethal dose and Methyl Thiazolyl Tetrazolium.Correlation analysis was performed to choose a sensitive indicator for OS.As for the comparison of Bacillus,cells were divided into:control,Bacillus treatment,HO treatment and Bacillus pre-protection+HO treatment.Bacillus were co-cultured with IPEC-1 for 3h in Bacillus and Bacillus pre-protection+HO treatments.Then,based on OS model,300 μmol/L HO were added into medium of HO and Bacillus pre-protection+HO treatments for another 12 h.Antioxidant and apoptosis gene expressions were detected to screen the target strain.Nuclear factor erythroid-derived 2 related factor 2(Nrf2)/Kelch-like ECH-associated protein 1(Keap1) pathway,reactive oxygen species(ROS) production,mitochondrial membrane potential(△ψm),apoptosis and necrosis were analyzed.Results revealed that heme oxygenase-1(HO-1) gene expression had a positive correlation with HO induction.Moreover,Bacillus amyloliquefaciens SC06(SC06)-meditated IPEC-1 showed the best antioxidant capacity though modulating Nrf2 phosphorylation.△ψm was elevated while ROS generation was reduced with SC06 pre-protection,resulting in decreased apoptosis and necrosis.Altogether,HO-1 expression could be regarded as an OS indicator.The regulation of Nrf2/Keap1 pathway and ROS production by SC06 are involved in alleviating OS of IPEC-1.

Objective :Pulmonary sarcomatous neoplasms are a rare group of tumors occur in the lung[1]. We received a pair of monozygotic twins discordant for sarcomatoid neoplasms of the right principal bronchus in their early age. Identical twins were grown in the same environment and the genetic similarity was highly consistent, so we thought the very small number of genetic differences between the sisters may be involved in the occurrence of the disaster. As we all know, most human pathogenic genes are caused by mutations in exons. In order to find out novel genes related to the patient’s tumor, exome sequencing which has unique advantages in identifying the causative genes of rare diseases was applied.Methods : Exome sequencing was used in the twins mentioned above. Through comparison analysis among the twins and the general population, 11 genes were first screened out. After further siRNA screening of these genes, we identified H4K20-specific histone methyltransferase SETD8 as the most possible participant in tumor occurrence. Two individual siRNAs of SETD8 were used in U2OS and human bronchial epithelium(HBE) cells, cell proliferation, apoptosis, ageing, cell cycle were analyzed. To determine the effect of single nucleotide variation(SNV, T904C) in patient’s setd8, we overexpressed wildtype and mutant SETD8 respectively in cells, immunoprecipitation and mass spectrometry were followed to analysis the differences in interaction with other proteins, we also amplified wildtype and mutant SETD8 protein in vitro to assess their methyltransferase activity.Results : : The silencing of SETD8 inhibited cell proliferation and increased apoptosis in both two cell lines. Cell cycle arrest and DNA damage also occurred without setd8. DNA fiber assay showed that compared with control, cells transfected with setd8 siRNA exhibited slower replication fork speed. Overexpress exogenous wildtype and mutant SETD8, protein interaction analysis revealed that the mutant SETD8 enhanced histone H4 binding and H4K20me1 levels. Methyltransferase

The current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)remains a threat to pregnant women.However,the impact of early pregnancy SARS-CoV-2 infection on the maternal-fetal interface remains poorly understood.Here,we present a comprehensive analysis of single-cell transcriptomics and metabolomics in placental samples infected with SARS-CoV-2 during early pregnancy.Compared to control placentas,SARS-CoV-2 infection elicited immune responses at the maternal-fetal interface and induced metabolic alterations in amino acid and phospholipid profiles during the initial weeks post-infection.However,subsequent immune cell activation and heightened immune tolerance in trophoblast cells established a novel dynamic equilibrium that mitigated the impact on the maternal-fetal interface.Notably,the immune response and metabolic alterations at the maternal-fetal interface exhibited a gradual decline during the second trimester.Our study underscores the adaptive immune tolerance mechanisms and establishment of immunological balance during the first two trimesters following maternal SARS-CoV-2 infection.

遵义红茶是产自贵州省遵义市的名优工夫红茶,目前对其内含化学成分和生物活性的相关报道较少,影响其进一步发展。本文系统分析遵义红茶的特征化学成分和降脂活性,结果表明:遵义红茶中含量最丰富的矿物质元素是K元素(22 655.89 mg/kg),且有害重金属As、Cd、Cr和Pb等元素含量均低于相关食品标准限量;醇类挥发性成分相对丰度占比最高(56.49%),主要包括具有花果香的芳樟醇(36.58%)和香叶醇(10.91%);总氨基酸含量为10 396.67 mg/kg,其中含量较高的茶氨酸(3 443.33 mg/kg)是遵义红茶鲜爽口感的主要来源,其它鲜味氨基酸如谷氨酸、天冬氨酸和天冬酰胺的含量也较高。电子舌结果显示遵义红茶不具有苦味和涩味;通过斑马鱼高脂喂养模型验证遵义红茶的降脂效果,采用800μg/mL的遵义红茶水提物处理的高脂斑马鱼的脂质水平降低54.98%(P <0.05)。本研究为遵义红茶的理化品质特征和降脂活性研究提供数据支撑,为其开发和推广提供技术参考。

研究了中国石化济南分公司(简称济南炼化)A1820芳香基环保橡胶增塑剂的生产工艺、产品性质,对比研究了当期市场占有率较高的参比产品的应用性能。物理精制工艺生产的A1820热稳定性和光安定性优良;用作填充油时,济南炼化A1820和参比产品在丁苯橡胶中效果相当;作为操作油,前者赋予胶料相对更好的加工性能、动态性能和更优的抗老化性能;两种操作油的相同配方轮胎制品均性能优良,但采用A1820的轮胎湿地刹车性能相对更好。研究结果被市场广泛接受,济南炼化A1820在高端轮胎橡胶制造领域的占比逐步增长。

Echinacea purpurea is an indigenous North American purple cone flower used by North Americans for treatment of various infectious diseases and wounds.This study investigated the effect of polysaccharide enriched extract of Echinacea purpurea(EE) on the polarization of macrophages.The results showed that 100 μg/mL of EE could markedly activate the macrophage by increasing the expression of CD80,CD86,and MHCII molecules.Meanwhile,EE upregulated the markers of classically activated macrophages(Ml) such as CCR7 and the production of IL-1β,IL-6,IL-12 p70,TNF-αand NO.The functional tests showed that EE enhanced the phagocytic and intracellular bactericidal activity of macrophage against ST.Furthermore,we demonstrated that JNK are required for EE-induced NO and M1-related cytokines production.Together,these results demonstrated that Echinacea purpurea can polarize macrophages towards Ml phenotype,which is dependent on the JNK signaling pathways.

目的芽孢杆菌是目前畜牧养殖业中应用较为广泛的益生菌。本试验以实验室前期从土壤中分离得到一株解淀粉芽孢杆菌SC06为材料,研究其替代抗生素(金霉素)对仔猪生长性能、肠黏膜物理和免疫屏障功能及其相关信号通路的影响。方法选取体重相近的杜长大公仔猪90头,分成3个组,每组3个重复。对照组饲喂添加150 ppm金霉素的基础饲粮(Anti组),处理组Ⅰ饲粮添加300克芽孢杆菌Ba/吨饲料和75 ppm金霉素(Anti+Ba组),处理组Ⅱ饲粮添加600克芽孢杆菌/吨,不添加任何抗生素(Ba组),试验期28天。结果1)与对照组相比,Anti+Ba组和Ba组平均日增重(ADG)均显著提高(P<0.05);2)Ba组和Ba+抗生素组的绒毛高度、绒毛高度与隐窝深度比值均显著高于对照组(P<0.05),隐窝深度低于对照组(P<0.05),此外,Ba组和Ba+抗生素组杯状细胞数量和分布、微绒毛完整性和致密程度、肠上皮细胞之间的紧密连接和线粒体完整性均优于对照组,进而显著增加空肠上皮电阻值(P<0.01);3)与对照组相比,Ba+抗生素组Ba组黏蛋白基因MUC-1(P<0.01)以及Ba组紧密连接基因Claudin 3、Claudin 4和Occludin均显著下调(P<0.05),但绒毛蛋白Vill基因表达量显著提高(P<0.01);4)与Anti组相比,Ba组肠黏膜细胞因子TNF-α(P<0.01)、IL-1α(P<0.05)基因表达以及TLR信号通路相关基因TLR6(P<0.01)和NFκB-p50(P<0.01)mRNA表达量均显著降低;5)通过检测MAPKs信号通路关键蛋白发现,Anti+Ba组肠黏膜JNK磷酸化水平降低,Ba组Erk1/2、p38、磷酸化JNK和磷酸化p38的蛋白表达量也降低。结论上述结果表明,日粮中添加益生茵Ba可改善仔猪生长性能,并通过抑制MAKPs信号通路,调节TLR信号通路相关基因以及细胞因子的mRNA表达增强仔猪肠黏膜屏障功能。
