Cell-cycle dependent nuclear gene delivery enhances the effects of E-cadherin against tumor invasion and metastasis

Gene delivery is the process by which foreign DNA is transferred to host cells,released from intracellular vesicles,and transported to the nuclei for *** process is frequently inefficient and difficult to control *** developed a gene delivery strategy that uses ultrasound to directly deliver plasmid DNA into nuclei via gas vesicles(GVs)-based intracellular ***-binding GVs can be taken up by cells and cause intracellular cavitation when exposed to acoustic irradiation and delivering their pDNA payloads into ***,GVs can remain stable in the cytoplasm in the absence of acoustic irradiation,allowing for temporally controlled nuclear gene *** were able to achieve spatiotemporal control of E-cadherin nuclear gene delivery in this manner,demonstrating its efficacy in tumor invasion and metastasis ***,we discovered that nuclear gene delivery of E-cadherin during the G2.M phase of the cell cycle in C6 tumor cells inhibited tumor invasion and metastasis more effectively than during the G1 and S *** gene delivery of E-cadherin at the G2.M phase resulted in significantly lower expression of Fam50a,which reduced Fam50a/runx2.interaction and led to reduced transactivation of MMp13,an important factor for epithelial-mesenchymal transition,as observed in a molecular mechanism ***,using remote acoustic control of intracellular cavitation of pDNA-GVs,we developed a high spatiotemporally controllable gene delivery strategy and achieved stronger tumor invasion and metastasis inhibition effects by delivering the E-cadherin gene at the G2.M phase.

The South china Sea(SCS)is a large marginal sea connecting the Indian and pacific *** the factors of monsoons,strait transport,and varied bathymetry,the SCS presents a three-layer structure and strong diapycnal mixing which is far greater than that in the open *** analysis and observations reveal that internal tides,internal solitary waves,and strong winds are the sources of the strong mixing in the northern SCS.A major consequence of the strong mixing is an active mid-deep circulation *** system promotes exchange of water between the SCS and adjacent oceans,and also regulates the upper layer of wind-driven circulation,making the 3 dimensional SCS circulation clearly different from that in other tropical and subtropical marginal *** mass transport capacity of the mid-deep circulation has a substantial impact on marine sedimentation,the biogeochemical cycle,and other processes in the *** paper summarizes the recent advances in middeep sea circulation dynamics of the SCS,and discusses the opportunities and challenges in this area.

Inverted perovskite solar cells(perSCs)are a highly promising candidate in the photovoltaic field due to their low-temperature fabrication process,negligible hysteresis,and easy integration with Si-based solar cells.A cathode interlayer(CIL)is necessary in the development of inverted devices to reduce the trap density and energy barrier between the electron transport layer(ETL)and the ***,most CILs are highly thickness-sensitive due to low conductivity and poor *** this study,we report on a self-doping perylene imide-based ionene polymer(pNpDIN)used as CIL material to modify electrode in inverted *** exhibits high conductivity and a good solubility in polar solvent,which results in an improved power conversion efficiency(pCE)from 10.05%(device without a CIL)to 16.97%.When the blend of pNpDIN and Bphen was used as a mixed CIL,the pCE of perSCs can be further increased to 2..2.%owing to the excellent morphology and matched energy *** importantly,the pCE of the device is highly tolerant to the thickness of the mixed CIL,which benefited from the high conductivity of *** development is expected to provide an excellent mixed CIL material for roll-to-roll processing efficient and stable inverted perSCs.

Objective:Mantle cell lymphoma(MCL)is a rare subtype of non-Hodgkin lymphoma(NHL)with high heterogeneity and a high recurrence *** heterogenous cell populations contribute to relapse remains to be ***:We performed single cell rNA sequencing(scrNA-seq)on approximately 4,000 bone marrow cells sampled from one patient with multidrug resistant *** identified 10 subpopulations comprising 4 malignant B cell subtypes,3 T cell subtypes,2.dendritic cell subtypes and 1 natural killer(NK)cell ***,we identified cell markers,including a series of genes associated with immune escape and drug *** addition,we explored the roles of these genes in the mechanism of immune escape and drug resistance,and we verified the expression imbalance and clinical prognostic potential by using GEO datasets including 2.1 MCL ***:The major immune escape mechanisms of MCL included anti-perforin activity,decreased immunogenicity and direct inhibition of apoptosis and cell killing,as mediated by type I and II B *** drug resistance mechanisms of different cell clusters included drug metabolism,DNA damage repair,apoptosis and survival *** III B cells closely communicate with other *** key genes involved in the resistance mechanisms showed dysregulated expression and may have significant clinical prognostic ***:This study investigated potential immune escape and drug resistance mechanisms in *** results may guide individualized treatment and promote the development of therapeutic drugs.

Elaborating Spatiotemporal Associations Between the Built Environment and Urban Vibrancy:A Case of guangzhou City,china

This study applies multi-source datasets(i.e.,Baidu Heat Map data,points of interest(pOIs)data,and floor area and land use data)and geographically and temporally weighted regression(GTWr)models to elaborate the spatiotemporal relationships between the built environment and urban vibrancy on both weekdays and weekends,using guangzhou City as a ***,we verified the spatially and temporally nonstationary nature of the built environment correlates,which have been largely ignored in previous studies based on local regression *** spatially and temporally heterogeneous effects of the built environment on urban vibrancy are then presented and visualized,based on the GTWr *** found that the elasticity of location(i.e.,distance),land use mix(i.e.,diversity),building intensity and numbers of pOIs with various functions(i.e.,density)are different across time(2.h intervals within a day)and space(grids),due to people’s everyday lifestyle,time-space constraints,and geographical context(e.g.,spatial structure).The findings highlight the importance of a better understanding of the local geography on the spatiotemporal relationships for urban planners and local governments so as to put forward decision-making support for fostering and maintaining urban vibrancy.

A Comparative Analysis of performance-Based resilience Metrics via a Quantitative-Qualitative Combined Approach: Are We Measuring the Same Thing?

Since the proposal of the pioneering“resilience triangle”paradigm,various time-series performance-based metrics have been devised for resilience *** numerous choices diversify the toolbox for measuring this compound system concept;however,this multiplicity causes intractable questions for applications,including“Do these metrics measure the same resilience?”and“Which one to pick under what circumstance?”In this study,we attempted to address these two fundamental issues using a comprehensive comparative *** a quantitative-qualitative combined approach,12.popular performance-based resilience metrics are compared using empirical data from china’s aviation system under the disturbance of *** results indicate that only 12.of the 66 metric pairs are strongly positively correlated and with no significant differences in quantification outcomes;qualitative results indicate that the majority of the metrics are based on different definition interpretations,basic components,and expression forms,and thus essentially measure different *** advantages and disadvantages of each metric are comparatively discussed,and a“how to choose”guideline for metric users is *** study is an introspective investigation of resilience quantification studies,aiming to offer a new perspective to scrutinize those benchmarking metrics.

Joint effects of micro-sized polystyrene and chlorpyrifos on zebrafish based on multiple endpoints and gut microbial effects

Microplastics often co-occur with a variety of organic contaminants in aquatic environment and pose combined risks to aquatic wildlife. Here, we investigated joint effects of microsized polystyrene(mpS, 5 μm) and an organophosphate pesticide chlorpyrifos on zebrafish,using multiple endpoints at both fish individual and gut microbiota levels. It was revealed that mpS ingested by zebrafish accumulated in gut and liver, and caused oxidative stress, hyperactive swimming performance and histological damages in fish, and induced disorders and diversity alterations of the gut microbial community. More importantly, mpS exhibited considerable adsorption capacity against chlorpyrifos, and those adsorbing chlorpyrifos presented greater effects on fish individuals but no different effects on gut microbiota compared to single mpS exposure. Together with body residues of chlorpyrifos in zebrafish,it was proposed that the joint effects between mpS and chlorpyrifos were attributed to the chlorpyrifos released from mpS within zebrafish. The present results provided a comprehensive understanding of joint effects of mpS and contaminants co-occurring in the environment and emphasized the importance of considering the adsorbed chemicals in toxicological studies of microplastics.
