OBJECTIVE:To investigate the effectiveness and safety of Jiawei Xiaoyao pill(加味逍遥丸,JXP)in the treatment of symptoms associated with premenstrual syndrome(PMS).METHODS:A total of 144 regularly menstruating women with PMS were recruited at 8 sites in China from August 2017 to December 2018,and randomized to receive either a JXP or a matching placebo(12 g/d,6 g twice a day)for 3 menstrual cycles.The primary indicator was the reduced Daily Record of Severity of Problems(DRSP)scores in the luteal phase after 3 months of treatment.The safety outcomes included clinical adverse events(AEs),adverse reactions(ARs),changes in vital signs,and laboratory tests.RESULTS:JXP surpassed the placebo in reducing DRSP scores(psychological/somatic dysfunction)in the luteal phase over 3 menstrual cycles of treatment(PFAS=0.002,PPPS=0.001).Additionally,there were no significant differences in the incidence of AEs,severe AEs,withdrawal due to AEs and ARs between the two groups(all P>0.05),and no clinically significant adverse medical events related to the test drug observed.CONCLUSIONS:JXP was superior to the placebo in relieving the symptoms associated with PMS,which signified that JXP may be effective,safe,and welltolerated as an alternative therapy.

Genomic imprinting can lead to allele-specific expression(ASE),where one allele is preferentially expressed more than the other.Perturbations in genomic imprinting or ASE genes have been widely observed across various neurological disorders,notably autism spectrum disorder(ASD).In this study,we crossed rhesus cynomolgus monkeys to produce hybrid monkeys and established a framework to evaluate their allele-specific gene expression patterns using the parental genomes as a reference.Our proof-of-concept analysis of the hybrid monkeys identified 353 genes with allele-biased expression in the brain,enabling us to determine the chromosomal locations of ASE clusters.Importantly,we confirmed a significant enrichment of ASE genes associated with neuropsychiatric disorders,including ASD,highlighting the potential of hybrid monkey models in advancing our understanding of genomic imprinting.

Mosaicism occurs in most founder genemodified monkeys generated by virus infection and programmable-nucleases[1–3]. In addition, macaque monkeys have a long preadolescence period, with four or more years usually required for male macaque monkeys to become sexually mature [4,5]. The mosaicism and long time to sexual maturity have largely prevented wide usage of gene-modified monkey models.

Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. Thus, BMAL1 knockout monkeys are potentially useful for studying the physiological consequences of circadian disturbance, and for developing therapies for circadian and psychiatric disorders.

为探究马铃薯的最佳遗传转化体系,以紫色马铃薯‘华颂66号’无菌组培苗茎段为外植体,用农杆菌介导法将含有植物沉默载体质粒pBWA(V)KS-miR8019转化马铃薯外植体,测定不同预培养时间、不同激素组合及抗生素对马铃薯愈伤再生转化体系的影响。结果表明:1)茎段预培养时间为2 d时,愈伤组织诱导率最高;2)最适的外植体愈伤组织诱导的培养基配方为:MS干粉式培养基+2.0 mg/L 6-BA+1.0 mg/L 2,4-D,愈伤组织的诱导率为96.7%。最适愈伤组织分化的培养基为:MS干粉式培养基+4.0 mg/L 6-BA+0.1 mg/L NAA+1.0 mg/L GA3;3)植株再生及生根的抗生素最适筛选浓度为75 mg/L的卡那霉素;4)农杆菌抑制剂的最适浓度为300 mg/L的特美汀,且在愈伤组织诱导、分化及生根阶段都需要加入。综上,通过对农杆菌介导的愈伤再生转化过程中各个影响因素进行试验,初步建立了马铃薯‘华颂66号’的遗传转化体系。

Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer

Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. This study demonstrates that the SCNT approach could be used to generate cloned monkeys from fibroblasts of a young adult monkeys and paves the way for the development of macaque monkey disease models with uniform genetic backgrounds.

在湖南南县长期紫云英-双季稻轮作试验的基础上,选择单施紫云英(MV)、单施化肥(F100)和紫云英配施全量化肥(MV+F100)处理,研究不同施肥处理下土壤大团聚体(>250μm)、小团聚体(53~250μm)、粉黏粒(2~53μm)及<2μm团聚体的黏粒组分(100~2000、25~100 nm)中矿物的类型、含量和分布特征。结果表明,不同处理各粒级团聚体黏粒组分中的矿物组成相似,主要以伊利石和高岭石为主,包含少量蛭石和1.4nm过渡矿物。MV和MV+F100处理中,相比于<2μm团聚体,大团聚体、小团聚体及粉黏粒中100~2000 nm组分高岭石含量有所降低,蛭石含量增加。与MV相比,MV+F100处理明显增加了<2μm团聚体颗粒中伊利石的相对含量,以及各粒级团聚体颗粒中蛭石的相对含量。随着各团聚体黏粒组分由100~2000nm减小至25~100nm,黏粒矿物的类型减少,伊利石的相对含量显著提高,高岭石含量降低且结晶度变差。与F100相比,MV+F100处理中各粒级团聚体100~2000 nm颗粒的硅铝率明显降低,出现脱硅富铝化现象。

目的评估老年人5年间蛋白质需要量的变化情况。方法在2017年14名老年人蛋白质需要量研究的前期基础上,纳入其中4名老年人(70~80岁)作为研究对象,采用指示剂氨基酸氧化法再次评估老年人蛋白质需要量。设7个蛋白质水平,分别为0.1、0.3、0.6、0.9、1.2、1.5和1.8 g/(kg·d)。每个蛋白质水平前2天均给予维持膳食。第3天进行稳定同位素研究,在氨基酸配比膳食的基础上,采用L-13C-苯丙氨酸作为指示剂,伴随氨基酸配比膳食口服摄入体内,当L-13C-苯丙氨酸的代谢在体内达到稳态时,收集呼出气、尿液样本。通过测定样本中标记氨基酸动力学参数,构建待测蛋白质摄入量和标记氨基酸氧化率的非线性混合效应模型,曲线拐点对应的蛋白质摄入量即为该研究人群的蛋白质平均需要量。结果根据4名老年人不同蛋白水平下呼气中^(13)CO_(2)的产生率可以获得老年人蛋白质平均需要量为1.05(95%CI 0.51~1.60)g/(kg·d)。采用蛋白质平均需要量推算推荐摄入量的变异系数进行推算后获得蛋白质推荐摄入量为1.31(95%CI 0.64~2.00)g/(kg·d)。结论老年人的蛋白质需要量在5年间有所提高,肌少症可能是导致老年人蛋白质需要量增加的主要原因。

Robust Optimal Operation of Active Distribution Network Based on Minimum Confidence Interval of Distributed Energy Beta Distribution

With the gradual increase of distributed energy penetration,the traditional optimization model of distribution network can no longer guarantee the stable and efficient operation of the distribution network.In order to deal with the inevitable uncertainty of distributed energy,a new robust optimal operation method is proposed for active distribution network(ADN)based on the minimum confidence interval of distributed energy Beta distribution in this paper.First,an ADN model is established with second-order cone to include the energy storage device,capacitor bank,static var compensator,on-load tap changer,wind turbine and photovoltaic.Then,the historical data of related distributed energy are analyzed and described by the probability density function,and the minimum confidence interval is obtained by interval searching.Furthermore,via taking this minimum confidence interval as the uncertain interval,a less conservative two-stage robust optimization model is established and solved for ADN.The simulation results for the IEEE33-bus distribution network have verified that the proposed method can realize a more stable and efficient operation of the distribution network compared with the traditional robust optimization method.

腰椎间盘突出症是纤维环破裂后髓核突出压迫神经根造成以腰腿痛为主要表现的疾病,是临床常见病、多发病。针刀治疗严重的、病变范围较广的腰椎间盘突出症尚在探索中,笔者在经筋、筋骨系统理论指导下,充分利用中医整体观念,结合近20年的系统针刀疗法治疗重症腰椎间盘突出症较为成熟的临床经验,形成了上海金山“系统针刀疗法”治疗腰椎间盘突出症的特色诊疗方法。此文从对针刀疗法的认识、常规针刀疗法入手,阐述系统针刀疗法治疗腰椎间盘突出症的理论基础、穴位探究、具体方案,并结合典型案例分析系统针刀疗法治疗腰椎间盘突出症的辨证思路和治疗特点,以期丰富腰椎间盘突出症的临床治疗方案。
