Abundance and diversity of ammonia-oxidizing archaea in response to various habitats in Pearl River Delta of China, a subtropical maritime zone

Ammonia-oxidizing archaea （AOA） are widely considered key to ammonia oxidation in various environments. However, little work has been conducted to simultaneously investigate the abundance and diversity of AOA as well as correlations between archaeal amoA genotypes and environmental parameters of different ecosystems at one district. To understand the abundance, diversity, and distribution of AOA in Pearl River Delta of China in response to various habitats, the archaeal amoA genes in soil, marine, river, lake, hot spring and wastewater treatment plant （WWTP） samples were investigated using real-time fluorescent quantitative PCR and clone libraries. Our analyses indicated that the diversity of AOA in various habitats was different and could be clustered into five major clades, i.e., estuary sediment, marine water/sediment, soil, hot spring and Cluster 1. Phylogenetic analyses revealed that the structure of AOA communities in similar ecological habitats exhibited strong relation. The canonical correspondence method indicated that the AOA community structure was strongly correlated to temperature, pH, total organic carbon, total nitrogen and dissolved oxygen variables. Assessing AOA amoA gene copy numbers, ranging from 6.84× 10^6 to 9.45 × 10^7 copies/g in dry soil/sediment, and 6.06× 10^6 to 2.41 ×10^7 copies/L in water samples, were higher than ammonia-oxidizing bacteria （AOB） by 1-2 orders of magnitude. However, AOA amoA copy numbers were much lower than AOB in WWTP activated sludge samples. Overall, these studies suggested that AOA may be a major contributor to ammonia oxidation in natural habitats but play a minor role in highly aerated activated sludge. The result also showed the ratio of AOA to AOB amoA gene abundance was positively correlated with temperature and less correlated with other environmental parameters. 以贺兰山东麓‘赤霞珠’干红葡萄酒为原料,采用不锈钢罐、微氧罐、橡木桶进行陈酿处理,与原酒作为对照,以期探究不同容器陈酿对干红葡萄酒基本理化指标、香气成分和感官特性的影响。采用电子鼻与顶空固相微萃取-气相色谱-质谱联用(HS-SPME-GC-MS)技术对不同容器陈酿的干红葡萄酒挥发性化合物进行分析和比较。结果表明,不同容器陈酿葡萄酒总糖、总酸、色度间存在显著差异(P<0.05),相较于原酒,3种容器陈酿的葡萄酒色度值均有显著提升,但对葡萄酒酒度、挥发酸、pH不会产生显著的影响。电子鼻和HS-SPME-GC-MS结果均表明原酒与不同容器陈酿的干红葡萄酒挥发性成分之间存在差异。HS-SPME-GC-MS在4类酒样中共检测出66种挥发性成分,包括酯类、醇类、酸类、醛酮类、萜烯类及其他类物质,其中酯类物质含量最多,醇类物质次之。主成分分析结合感官评价结果显示,与原酒相比,不同容器陈酿能够明显增强葡萄酒花香和果香特征,降低酒样的脂肪味,并且经橡木桶和微氧罐陈酿的葡萄酒酸甜适宜,口感更加柔顺。因此,相较于不锈钢罐,微氧罐陈酿有利于葡萄酒的熟化,对葡萄酒的感官提升有重要作用。

Ⅳ型胶原蛋白A4在肾透明细胞癌中的表达及其与预后和免疫浸润的相关性分析

目的探究Ⅳ型胶原蛋白A4(collagen typeⅣalpha 4 chain,COL4A4)在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)中的表达及其与临床预后和免疫浸润的关系。方法利用癌症基因组图谱(The Cancer Genome Atlas,TCGA)和高通量基因表达(Gene Expression Omnibus,GEO)数据库分析COL4A4在ccRCC组织和癌旁正常组织中的表达差异。Western blot实验探究COL4A4在人ccRCC细胞株(786-O、OS-RC-2、Caki-1、SW839和RCC4)和人正常肾小管上皮细胞株HK-2中的表达差异。Kaplan-Meier曲线用于分析COL4A4表达与ccRCC患者的临床总生存期、无进展生存期和疾病特异性生存期的关系,并用受试者工作特征(receiver operating characteristic,ROC)曲线评价COL4A4对预测ccRCC发生的诊断价值。利用cBioPortal和COSMIC数据库分析COL4A4在ccRCC中的突变情况。采用STRING数据库构建COL4A4的蛋白互作(protein-protein interaction,PPI)网络。采用LinkedOmics数据库构建COL4A4的共表达基因网络。采用R软件进行基因本体(Gene Ontology,GO)功能富集分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)信号通路富集分析。基于TCGA数据的单样本基因集富集分析(single sample gene set enrichment analysis,ssGSEA)分析ccRCC中COL4A4表达与免疫浸润的相关性。结果TCGA和GEO数据库均显示,COL4A4在ccRCC组织中低表达(均P<0.05)。Western blot实验证实,COL4A4在ccRCC细胞株(786-O、OS-RC-2、Caki-1、SW839和RCC4)中均低表达(均P<0.05)。高表达COL4A4的患者具有更好的总生存期、无进展生存期和疾病特异性生存期(均P<0.01)。COL4A4是ccRCC患者的独立危险因素(HR=0.562,95%CI:0.352~0.898,P<0.05)。COSMIC数据库发现,COL4A4突变频率较低,在ccRCC中稳定表达。PPI网络显示,COL4A4与整合素家族密切相关。功能富集分析发现,COL4A4参与GTP酶活性调节、细胞核分裂、细胞周期转换、自噬和T细胞激活等生物学过程。信号通路富集分析提示,COL4A4主要参与沙门氏菌感染和细胞间的紧密连接。免疫浸润分析发现,COL4A4表达与调节性T细胞浸润呈负相关(r=-0.428,P<0.05)。结论COL4A4是ccRCC的潜在生物预后标志物和治疗靶点。

Prognostic Nomogram for pancreatic cancer with lung metastasis:a SEER database-based study

Bioinspired Tactile Sensation Based on Synergistic Microcrack-Bristle Structure Design toward High Mechanical Sensitivity and Direction-Resolving Capability

Natural tactile sensation is complex,which involves not only contact force intensity detection but also the perception of the force direction,the surface texture,and other mechanical ***,the vast majority of the developed tactile sensors can only detect the normal force,but usually cannot resolve shear force or even distinguish the directions of the ***,we present a new paradigm of bioinspired tactile sensors for resolving both the intensity and the directions of mechanical stimulations via synergistic microcrack-bristle structure design and cross-shaped configuration *** microcrack sensing structure gives high mechanical sensitivity to the tactile sensors,and the synergistic bristle structure further amplifies the sensitivity of the *** cross-shaped configuration engineering of the synergistic microcrack-bristle structure further endows the tactile sensors with good capability to detect and distinguish the directions of the applied mechanical *** as-fabricated tactile sensors exhibit a high sensitivity(25.76 N^(−1)),low detection limit(5.4 mN),desirable stability(over 2,500 cycles),and good capability to resolve both mechanical intensity and directional *** promising application scenarios,surface texture recognition and biomimetic path explorations are successfully demonstrated with these tactile *** newly proposed tactile sensation strategy and technology have great potential applications in ingenious tactile sensation and construction of various robotic and bionic prostheses with high operational dexterity.

The configuration of Au^(δ+)-ZrO_(2)^(δ−)species induced activation enhances electrocatalytic CO_(2)to formate conversion

Electrochemical CO_(2)conversion into value-added chemicals is a promising technology to solve the greenhouse effect and recycle chemical ***,the electrochemical CO_(2)reduction reaction(e-CO_(2)RR)is seriously compromised by weak CO_(2)adsorption and a rough CO_(2)activation process based on the chemical inertness of the CO_(2)molecule and the formed fragile metal–C/O *** this paper,we designed and fabricated Au particles embedded in ZrO_(2).The configuration of Au particles being of positive charge and ZrO_(2)with negative charge is induced and generated by metal–support interactions(MSIs).As a result,Au/ZrO_(2)@C presents a big difference in the CO_(2)conversion compared with the known work,affording a formate yield of 112.5μmol·cm^(−2)·h^(−1)at−1.1 V *** hydrogen electrode(RHE),and a max formate Faradaic efficiency of up to 94.1%at−0.9 V *** superior performance was attributed to the activated Au–ZrO_(2)interface to form the Au^(δ+)*** insitu Fourier transform infrared(FTIR)spectroscopy and theoretical calculations show that the MSIs configuration can be inclined to the*OCO intermediate generation on Au^(δ+)species activating CO_(2)molecules and then accelerate the formation of the*OCHO intermediate in e-CO_(2)RR,thereby favoring the CO_(2)conversion to formate.

The friction coefficient and wear rate of pretreated graphite with liquid nitrogen were obtained by using a ball-on-disk tester,and the wear of GCr15–graphite seal pairs with the low-temperature time-dependent pretreatment was discussed by comparing the wear *** results show that liquid nitrogen pretreatment can affect the hardness and interlayer spacing of *** range of the friction coefficients of pretreated graphite changes from 0.17 to *** the increase of liquid nitrogen pretreatment time,the wear mechanism of graphite would change from dominated three-body wear to adhesion *** experimental results of the mechanical seal with liquid nitrogen pretreatment show that the wear rate of stator is less than 0.00165 mm^(3)·N^(−1)·m^(−1),and the graphite shows a good low-temperature compatibility.
