Circulating tumor cells(CTCs)are recognized as the main source of tumor recurrence and metastasis.Eliminating the CTCs in peripheral blood provides a new strategy to reduce the probability of recurrence or metastasis.Here,we proposed a concept to eliminate CTCs by inserting a needle in the superficial blood vessel.Using the property of ZnO and the structure of nanoflowers,we designed a medical needle coated with ZnO nanoflowers(ZNFs),which killed about 90%of captured CTCs in vitro and prevented the injecting CTCs from spreading to lung tissue in BABL/c mouse model.Results in vitro and in vivo demonstrated that the CTCs not only were captured and killed,but also detached from the needle surface after dead,enabling the ZNFs needle continually eliminate CTCs.Furthermore,a theoretical model was presented to explain the penetration mechanism of cells by nanostructures,which indicated that nanoflowers structure can puncture CTCs more easily than vertical nanowire structure.The concept of inserting an intravascular needle provides a potential strategy to lower the concentration of CTCs in blood and reduce the probability of tumor recurrence or metastasis.

Second-generation DNA-encoded multiple display on a constant macrocyclic scaffold enabled by an orthogonal protecting group strategy

DNA-encoded chemical library(DEL)represents an emerging drug discovery technology to construct com-pound libraries with abundant chemical combinations.While drug-like small molecule DELs facilitate the discovery of binders against targets with defined pockets,macrocyclic DELs harboring extended scaffolds enable targeting of the protein–protein interaction(PPI)interface.We previously demonstrated the design of the first-generation DNA-encoded multiple display based on a constant macrocyclic scaffold,which harvested binders against difficult targets such as tumor necrosis factor-α(TNF-α).Here,we developed a novel strategy which utilized four orthogonal amine-protecting groups on DNA,to explore larger chem-ical combinations on the same constant macrocyclic scaffold,following the parallel paradigm to mimic the versatile antibody-like multivalent epitope recognition patterns.We successfully integrated these or-thogonal protecting groups with acylation and made a mock second-generation DNA-encoded display combination.This work illustrates a strategy to produce larger encoded multiple display on a constant macrocyclic scaffold,which could facilitate potential binder discovery with enhanced affinity to clinically significant PPI targets.

Efficacy,safety,and cost-effectiveness of triple therapy in preventing relapse in rheumatoid arthritis:A randomized controlled trial(ESCoRT study)

Background:Biological agents,such as tumor necrosis factor inhibitors(TNFi),have been widely used in rheumatoid arthritis(RA)patients and greatly improved goal achievement.The aim of this study was to investigate whether conventional synthetic diseasemodifying anti-rheumatic drugs(csDMARDs)combination was better in reducing relapse than methotrexate(MTX)monotherapy,and more cost-effective than continuing TNFi plus MTX in RA patients who achieved low disease activity(LDA)with TNFi and MTX therapy.Methods:RA patients who failed to csDMARDs received an induction therapy of MTX plus TNFi for maximally 12 weeks.Those achieving LDA in 12 weeks were randomly assigned at a 1:1:1 ratio into three groups:(A)adding hydroxychloroquine and sulfasalazine for the first 12 weeks and then discontinuing TNFi for the following 48 weeks;(B)maintaining TNFi and MTX for 60 weeks;and(C)maintaining TNFi and MTX for the first 12 weeks and then discontinuing TNFi for the following 48 weeks.The primary outcome was relapse.Results:A total of 117 patients were enrolled for induction therapy and 67 patients who achieved LDA within 12 weeks were randomized,with 24,21,and 22 patients in groups A,B,and C,respectively.The relapse rates of groups A and B during the entire 60 weeks were comparable[10/22(45.5%)vs.7/20(35.0%),χ^(2)=0.475,P=0.491],however,significantly lower than that of group C[10/22(45.5%)vs.17/20(85.0%),χ^(2)=5.517,P=0.019;7/20(35.0%)vs.17/20(85.0%),χ^(2)=11.035,P=0.004,respectively].Taking RMB 100,000 Yuan as the threshold of willingness to pay,compared to MTX monotherapy(group C),both TNFi maintenance and triple csDMARDs therapies were cost-effective,but triple csDMARDs therapy was better.Conclusion:For RA patients who have achieved LDA with TNFi and MTX,csDMARDs triple therapy was a cost-effective option in favor of reducing relapse.Trial registration:ClinicalTrials.gov,NCT02320630.

Designing a photo-assisted Co-C_(3)N_(4) cathode for high performance Li-O_(2) batteries

Li-O_(2) batteries with extremely high specific energy density have been regarded as a kind of promising successor to current Li-ion batteries.However,the high charge overpotential for the decomposition of Li_(2)O_(2) discharge product reduces the energy efficiency and triggers a series of side reactions that cause the Li-O_(2) batteries to have a limited lifetime.Herein,Co-doped C_(3)N_(4)(Co-C_(3)N_(4))photocatalysts were designed by an in situ thermal evaporation method to take advantage of the photo-assisted charging technology to conquer the shortcomings of Li-O_(2) batteries encountered in the charge process.Different from the commonly used photocatalysts,the Co-C_(3)N_(4) photocatalysts perform well no matter with and without illumination,owing to the Co doping induced conductivity and electrocatalytic ability enhancement.This makes the Co-C_(3)N_(4) reduce the charge and discharge overpotentials and improve the cycling performance of Li-O_(2) batteries(from 20 to 106 cycles)without illumination.While introducing illumination,the performance can be further improved:Charge voltage reduces to 3.3 V,and the energy efficiency increases to 84.84%,indicating that the Co-C_(3)N_(4) could behave as a suitable photocathode for Li-O_(2) batteries.Besides,the low charge voltage and the continuous illumination together weaken the corrosion of the Li anode,making the long-term high-efficiency operation of Li-O_(2) batteries no longer just extravagant hope.

Development of a random-forest-copula-factorial analysis(RFCFA)method for predicting propagation between meteorological and hydrological drought

In the context of global climate warming,the propagation of meteorological drought(MD)may aggravate the devastating impact of hydrological drought(HD)on water security and sustainable development.There are challenges in accurately predicting the propagation of drought and effectively quantifying the effects of uncertainty,especially in data-deficient regions.In this study,a novel method called RFCFA is developed through integrating random forest(RF),copula,and factorial analysis(FA)into a general framework as well as applied to the Aral Sea Basin(a typical arid and data-scarce basin in Central Asia)under considering the impact of climate change.Several findings can be summarized:(1)the projected future drought propagation probability of ASB is 39.2%,which is about 8%higher than historical level;(2)drought propagation is mainly affected by mean climate condition,catchment characteristics(i.e.,elevation,LUCC,and slope),and human activities(i.e.,irrigation and reservoir operation);(3)the lower propagation probability in spring is expected under SSP1-2.6 due to increased snow meltwater,and the drought propagation probability in autumn is the highest(reaching 45.4%)under the influence of reservoir operation;(4)the combined effects of meteorological conditions and agricultural irrigation can lead to a higher probability of future propagation in the upper river basin in summer.Findings are valuable for predicting drought propagation risk,revealing main factors and inherent uncertainties,as well as providing support for drought management and disaster prevention.

Characteristics of Boundary Layer Structure during a Persistent Haze Event in the Central Liaoning City Cluster, Northeast China

The characteristics of boundary layer structure during a persistent regional haze event over the central Liaoning city cluster of Northeast China from 16 to 21 December 2016 were investigated based on the measurements of particulate matter（PM） concentration and the meteorological data within the atmospheric boundary layer（ABL）. During the observational period, the maximum hourly mean PM_（2.5） and PM10 concentrations in Shenyang, Anshan, Fushun, and Benxi ranged from 276 to 355 μg m–3 and from 378 to 442 μg m–3, respectively, and the lowest hourly mean atmospheric visibility（VIS） in different cities ranged from 0.14 to 0.64 km. The central Liaoning city cluster was located in the front of a slowly moving high pressure and was mainly controlled by southerly winds. Wind speed（WS） within the ABL（〈 2 km） decreased significantly and WS at 10-m height mostly remained below 2 m s–1 during the hazy episodes, which was favorable for the accumulation of air pollutants. A potential temperature inversion layer existed throughout the entire ABL during the earlier hazy episode [from 0500 Local Time（LT） 18 December to 1100 LT 19 December], and then a potential temperature inversion layer developed with the bottom gradually decreased from 900 m to 300 m. Such a stable atmospheric stratification further weakened pollutant dispersion. The atmospheric boundary layer height（ABLH） estimated based on potential temperature profiles was mostly lower than 400 m and varied oppositely with PM_（2.5） in Shenyang. In summary, weak winds due to calm synoptic conditions, strong thermal inversion layer, and shallow atmospheric boundary layer contributed to the formation and development of this haze event.The backward trajectory analysis revealed the sources of air masses and explained the different characteristics of the haze episodes in the four cities.

Background:Reports on the prevalence of psoriatic arthritis(PsA)among Chinese patients with psoriasis are very limited.This study,conducted by rheumatologists,estimated the prevalence of PsA in a large number of Chinese patients with psoriasis.Methods:Consecutive patients with a confirmed diagnosis of psoriasis attending nine dermatology clinics in five hospitals were recruited.All psoriasis patients were asked to complete a questionnaire comprising 16 questions to identify possible cases of PsA.All patients with one or more positive answers to the questionnaire were evaluated by two experienced rheumatologists.Results:A total of 2434 psoriasis patients,including 1561 males and 873 females,were enrolled.Both the questionnaire and rheumatologists’examinations were completed in the dermatology clinics.The results identified 252 patients with PsA,comprising 168 males and 84 females.The overall prevalence of PsA among psoriasis patients was 10.4%(95%confidence interval[95%CI],9.1%-11.7%).By sex,the prevalence was 10.8%(95%CI,9.2%-12.5%)for males and 9.6%(95%CI,7.7%-11.9%)for females and there was no significant sex difference in the prevalence of PsA(P=0.38).Of the 252 PsA patients,125(49.6%,95%CI,41.3%-59.1%)were newly diagnosed by rheumatologists.Consequently,the prevalence of undiagnosed PsA among psoriasis patients was 5.2%(95%CI,4.4%-6.2%).Conclusion:The prevalence of PsA in the Chinese population with psoriasis is about 10.4%,which is almost double that of previous reports in the Chinese population,but lower than that in Caucasians.

Gel electrolyte via in situ polymerization to promote durable lithium-air batteries

Aprotic lithium-air batteries(LABs)have been known as the holy grail of energy storage systems due to their extremely high energy density.However,their real-world application is still hindered by the great challenges from the Li anode side,like dendrite growth and corrosion reactions,thus a pure oxygen atmosphere is usually adopted to prolong the lifetime of LABs,which is a major obstacle to fully liberate the energy density advantages of LABs.Here,a gel polymer electrolyte has been designed through in-situ polymerization of 1,3-dioxolane(DOL)by utilizing the unique semi-open nature of LABs to protect the Li anode to conquer its shortcomings,enabling the high-performance running of LABs in the ambient air.Unlike common liquid electrolytes,the in-situ formed gel polymer electrolyte could facilitate constructing a gradient SEl film with the gradual decrease of organic components from top to bottom,preventing the Li anode from dendrite growth and air-induced corrosion reactions and thus realizing durable Li repeated plating/stripping(2000h).Benefiting from the anode protection effects of the gradient SEI film,the LABs display a long lifetime of 17o cycles,paving an avenue for practical,long-term,and high-efficiency operation of LABs.

DNA-encoded chemical libraries technology has become a novel approach to finding hit compounds in early drug discovery.The chemical space in a DEL would be expanded to realize its full potential,especially when integrating privileged scaffold dihydroquinazoline that has demonstrated a variety of diverse bioactivities.Driven by the requirement of parallel combinatorial synthesis,we here report a facile synthesis of on-DNA dihydroquinazolinone from aldehyde and anthranilamide.This DNA-compatible reaction was promoted by antimony trichloride,which has been proven to accelerate the reaction and improve conversions.Notably,the broad substrate scope of aldehydes and anthranilamides was explored under the mild reaction condition to achieve moderate-to-excellent conversion yields.We further applied the reaction into on-DNA macrocyclization,obtaining macrocycles embedded dihydroquinazolinone scaffold in synthetically useful conversion yields.
