BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ***,the positive regulators of ANKRD26 are still *** To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 *** Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family *** to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activi

Experimental Study on a New Dual-Layer Granular Bed Filter for Removing Particulates

A new dual-layer granular bed filter for hot gas cleanup was invented and studied experimentally. Fine sand,0.5-1 mm grain size and about 1350 kg/m3 bulk density,was used as the lower layer of the filter. Expanded perlite par-ticles,2-5 mm grain size and about 70 kg/m3 bulk density,was used for the upper layer of the filter in this study. It was confirmed that the sizes and densities of these two media matched well; the binary media remained in complete segre-gation during regeneration by fluidization. Test results show that the filtration of the expanded perlite particle layer was characterized as "deep bed filtration." Filtration of the fine sand layer was "surface cake filtration." The expanded per-lite particle layer contributed about 90% to the bed dust capacity,but only about 20% to the total bed pressure drop,which increased the bed dust capacity ten fold compared to a single-layer bed of the same sand and the same total bed pressure drop. The dust cake on the surface of the fine sand layer raised the collection efficiencies to over 99.99%.

Sulphur-template method for facile manufacturing porous silicon electrodes with enhanced electrochemical performance

Sulphur(S)-template method based on conventional slurry-casting method has been developed to pro-duce porous silicon(Si)*** facile fabrication technology is suitable for current production line and expected to be widely applied to various electrode materials under large volume change during ***,S particles as template agent are mixed with active material Si,carbon conductor and binder forming uniform *** casting and drying,the electrodes are immersed in carbon disul-fide solution to remove S particles rapidly,generating pores in-situ at the original position of S *** analysis shows that the pores inside electrodes are able to shorten lithium ion diffusion paths,reduce normal expansion rate and decrease formation of cracks in the Si electrode(2 mg_(Si)/cm^(2)),demonstrating a reversible capacity of 951 mAh/g at 0.5 A/g after 100 cycles(with a capacity retention of 99.5%)and a capacity of-826 mAh/g at 2 A/g.

As a promising imaging technology, the low sensitivity of fluorine-19 magnetic resonance imaging(19F MRI) severely hinders its biomedical applications. Herein, we have developed an unprecedented rotaxanebased strategy to improve the sensitivity of19F MRI agents. By threading the fluorinated macrocycle into2-blade pinwheel [2]rotaxanes, the19F longitudinal relaxation rate R1was dramatically increased, resulting in a significant19F MRI signal intensity enhancement of up to 79%. Through comparative molecular dynamics studies using a series of solution and solid-state1H/19F nuclear magnetic resonance(1H/19F NMR) and molecular dynamics simulations, it was found that the formation of mechanical bonds dramatically restricts the motion of the wheel fluorines and thus increasing the R1for higher19F MRI *** a novel strategy for improving19F MRI sensitivity, this study has established19F NMR/MRI as a valuable technology for monitoring the molecular dynamics of rotaxanes, which may shed new light on high-performance19F MRI agents and molecular devices.

Process-driven susceptibility assessment of glacial lake outburst debris flow in the Himalayas under climate change

Global warming is causing glaciers to retreat and glacial lakes to expand in the Himalayas,which amplifies the risk of glacial lake outburst debris flows(GLODFs)and poses a significant threat to downstream lives and ***,the complex interplay between GLODF occurrences and associated indicators,coupled with the lack of a comprehensive susceptibility indicator system that considers the entire GLODF process,presents a substantial challenge in assessing GLODF susceptibility in the *** study proposes a process-driven GLODF susceptibility assessment indicator system responding to climate change that considers the complete process of GLODF formation,incorporating relevant parameters about upstream,themselves,and downstream of glacial ***,to mitigate subjective factors associated with traditional evaluation methods,we developed three novel hybrid machine-learning models by integrating classic machine-learning algorithms with the whale optimization algorithm(WOA)to delineate the distribution of GLODF susceptibility in the *** the hybrid models effectively predicted the GLODFs occurrence,with the WOA-SVC model demonstrating the highest prediction *** 34%of the catchments exhibit high and very high susceptibility levels,primarily concentrated along the north and south sides of the Himalayan ridge,particularly in the eastern and central *** capturing the physical formation process of hazards,such as topographic potential(highest relative importance value of 40%),can precisely identify GLODF.A total of 128 catchments pose potential transboundary threats,with 24 classified as having a very high susceptibility level and 25 as having a high susceptibility ***,the border region between China and Nepal is a prominent hotspot for transboundary threats of *** findings can provide valuable clues for disaster prevention,mitigation,and cross-border coordination in the Himalayas.

Research advances in enhanced coal seam gas extraction by controllable shock wave fracturing

A Novel Foreign Object Detection Method in Transmission Lines Based on Improved YOLOv8n

The rapid pace of urban development has resulted in the widespread presence of construction equipment andincreasingly complex conditions in transmission corridors. These conditions pose a serious threat to the safeoperation of the power *** vision technology, particularly object recognition technology, has beenwidelyemployed to identify foreign objects in transmission line images. Despite its wide application, the technique faceslimitations due to the complex environmental background and other auxiliary factors. To address these challenges,this study introduces an improved YOLOv8n. The traditional stepwise convolution and pooling layers are replacedwith a spatial-depth convolution (SPD-Conv) module, aiming to improve the algorithm’s efficacy in recognizinglow-resolution and small-size objects. The algorithm’s feature extraction network is improved by using a LargeSelective Kernel (LSK) attention mechanism, which enhances the ability to extract relevant features. Additionally,the SIoU Loss function is used instead of the Complete Intersection over Union (CIoU) Loss to facilitate fasterconvergence of the algorithm. Through experimental verification, the improved YOLOv8n model achieves adetection accuracy of 88.8% on the test set. The recognition accuracy of cranes is improved by 2.9%, which isa significant enhancement compared to the unimproved algorithm. This improvement effectively enhances theaccuracy of recognizing foreign objects on transmission lines and proves the effectiveness of the new algorithm.
