Hepatic ischemia–reperfusion(IR)injury is a serious clinical problem that complicates liver resection and transplantation.Despite recent advances in understanding of the pathophysiology of hepatic IR injury,effective interventions and therapeutics are still lacking.Here,we examined the role of transient receptor potential melastatin 2(TRPM2),a Ca^(2+)-permeable,non-selective cation channel,in mediating hepatic IR injury.Our data showed that TRPM2 deficiency attenuated IR-induced liver dysfunction,inflammation,and cell death in mice.Moreover,RNA sequencing analysis indicated that TRPM2-induced IR injury occurs via ferroptosis-related pathways.Consistently,as a ferroptosis inducer,(1S,3R)-RSL3 treatment induced mitochondrial dysfunction in hepatocytes and a TRPM2 inhibitor suppressed this.Interestingly,TRPM2-mediated calcium influx caused mitochondrial calcium accumulation via the mitochondrial Ca^(2+)-selective uniporter and increased the expression level of arachidonate 12-lipoxygenase(ALOX12),which results in mitochondrial lipid peroxidation during hepatic IR injury.Furthermore,hepatic IR injury-induced ferroptosis was obviously relieved by a TRPM2 inhibitor or calcium depletion,both in vitro and in vivo.Collectively,these findings demonstrate a crucial role for TRPM2-mediated ferroptosis in hepatic IR injury via increased Ca^(2+)-induced ALOX12 expression,indicating that pharmacological inhibition of TRPM2 may provide an effective therapeutic strategy for hepatic IR injury-related diseases,such as during liver resection and transplantation.

A room-temperature electrodeposition method with an organic electrolyte was developed to fabricate a HNO3-pretreated graphene paper Cu(GP'-Cu)composite.To improve the interfacial bonding of GP'-Cu composite,magnetron sputtering technology was used to create a"sandwich"structural gradient GP'-Cu composite.The selection of the intermediate transition layer metal was based on two-dimensional disregistry.Scanning electron microscopy,X-ray photoelectron spectroscopy,and other analytical methods confirmed that the addition of an intermediate transition metal(Cr,Ni)layer reduced the gap distance and enhanced the interfacial bonding of the GP'and Cu deposited layers.The GP'-Ni-Cu composite exhibited the largest increase in tensile strength and conductivity.In addition,it had the highest thermal diffusivity and elongation at break among the GP'-Cu,GP'-Cr-Cu and GP'-Ni-Cu composites.

Computed tomography radiogenomics:A potential tool for prediction of molecular subtypes in gastric stromal tumor

BACKGROUND Preoperative knowledge of mutational status of gastrointestinal stromal tumors(GISTs)is essential to guide the individualized precision therapy.AIM To develop a combined model that integrates clinical and contrast-enhanced computed tomography(CE-CT)features to predict gastric GISTs with specific genetic mutations,namely KIT exon 11 mutations or KIT exon 11 codons 557-558 deletions.METHODS A total of 231 GIST patients with definitive genetic phenotypes were divided into a training dataset and a validation dataset in a 7:3 ratio.The models were constructed using selected clinical features,conventional CT features,and radiomics features extracted from abdominal CE-CT images.Three models were developed:ModelCT sign,modelCT sign+rad,and model CTsign+rad+clinic.The diagnostic performance of these models was evaluated using receiver operating characteristic(ROC)curve analysis and the Delong test.RESULTS The ROC analyses revealed that in the training cohort,the area under the curve(AUC)values for model_(CT sign),model_(CT sign+rad),and modelCT_(sign+rad+clinic)for predicting KIT exon 11 mutation were 0.743,0.818,and 0.915,respectively.In the validation cohort,the AUC values for the same models were 0.670,0.781,and 0.811,respectively.For predicting KIT exon 11 codons 557-558 deletions,the AUC values in the training cohort were 0.667,0.842,and 0.720 for model_(CT sign),model_(CT sign+rad),and modelCT_(sign+rad+clinic),respectively.In the validation cohort,the AUC values for the same models were 0.610,0.782,and 0.795,respectively.Based on the decision curve analysis,it was determined that the model_(CT sign+rad+clinic)had clinical significance and utility.CONCLUSION Our findings demonstrate that the combined modelCT_(sign+rad+clinic)effectively distinguishes GISTs with KIT exon 11 mutation and KIT exon 11 codons 557-558 deletions.This combined model has the potential to be valuable in assessing the genotype of GISTs.

Formation conditions of Sinian–Cambrian large lithologic gas reservoirs in the north slope area of central Sichuan Basin,SW China

Based on analyses of characteristics,hydrocarbon charging history and geological conditions for the formation of Sinian-Cambrian reservoirs in the north slope area of central Sichuan paleo-uplift,the natural gas origin,accumulation evolution,accumulation pattern and formation conditions of large lithologic gas reservoirs have been investigated.Through comprehensive analyses of natural gas composition,carbon and hydrogen isotopic compositions,fluid inclusions,reservoir bitumen,and geological conditions such as lithofacies paleogeography and beach body characterization,it is concluded that:(1)The natural gas in the Sinian-Cambrian of the north slope area is mainly oil cracking gas,and different contribution ratios of multiple sets of source rocks lead to different geochemical characteristics of natural gas in different reservoirs.(2)Although the both Sinian and Cambrian gas reservoirs in this area are lithologic gas reservoirs under monocline background,the former has normal-pressure and the latter has high-pressure.There are three types of source-reservoir-caprock combinations:single source with lower generation and upper reservoir,double sources with lower generation and upper reservoir or with side source and lateral reservoir,double sources with lower generation and upper reservoir or with upper generation and lower reservoir.The Permian-Triassic is the main generation period of oil,Early-Middle Jurassic is the main generation period of oil cracking gas and wet gas,and Late Jurassic-Cretaceous is the main generation period of dry gas.(3)The Sinian-Cambrian system of the north slope area has two favorable conditions for formation of large lithologic gas reservoirs,one is that the large scale beach facies reservoirs are located in the range of ancient oil reservoirs or near the source rocks,which is conducive to the"in-situ"large-scale accumulation of cracked gas in the paleo-oil reservoirs,the other is that the large scale mound-beach complex reservoirs and sealing layers of inter beach tight zones match effecti

According to clinical statistics,the mortality of patients with early brainstem hemorrhage is high.In this study,we established rat models of brainstem hemorrhage by injecting type Ⅶ collagenase into the right basotegmental pontine and investigated the pathological changes of early brainstem hemorrhage using multi-sequence magnetic resonance imaging and histopathological methods.We found that brainstem hematoma gradually formed in the injured rats over the first 3 days and then reduced after 7 days.The edema that occurred was mainly of the vasogenic type.No complete myelin sheath structure was found around the focus of the brainstem hemorrhage.The integrity and continuity of nerve fibers gradually deteriorated over the first 7 days.Neuronal degeneration was mild in the first 3 days and then obviously aggravated on the 7^(th)day.Inflammatory cytokines,interleukin-1β,and tumor necrosis factorαappeared on the 1st day after intracerebral hemorrhage,reached peak levels on the 3^(rd)day,and decreased from the 7^(th)day.Our findings show the characteristics of the progression of early brainstem hemorrhage.

A spawning particle filter for defocused moving target detection in GNSS-based passive radar

Global Navigation Satellite System(GNSS)-based passive radar(GBPR)has been widely used in remote sensing applications.However,for moving target detection(MTD),the quadratic phase error(QPE)introduced by the non-cooperative target motion is usually difficult to be compensated,as the low power level of the GBPR echo signal renders the estimation of the Doppler rate less effective.Consequently,the moving target in GBPR image is usually defocused,which aggravates the difficulty of target detection even further.In this paper,a spawning particle filter(SPF)is proposed for defocused MTD.Firstly,the measurement model and the likelihood ratio function(LRF)of the defocused point-like target image are deduced.Then,a spawning particle set is generated for subsequent target detection,with reference to traditional particles in particle filter(PF)as their parent.After that,based on the PF estimator,the SPF algorithm and its sequential Monte Carlo(SMC)implementation are proposed with a novel amplitude estimation method to decrease the target state dimension.Finally,the effectiveness of the proposed SPF is demonstrated by numerical simulations and pre-liminary experimental results,showing that the target range and Doppler can be estimated accurately.

Exploring mechanism of hidden,steep obliquely inclined bedding landslides using a 3DEC model:A case study of the Shanyang landslide in Shaanxi Province,China

Catastrophic geological disasters frequently occur on slopes with obliquely inclined bedding structures(also referred to as obliquely inclined bedding slopes),where the apparent dip sliding is not readily visible.This phenomenon has become a focal point in landslide research.Yet,there is a lack of studies on the failure modes and mechanisms of hidden,steep obliquely inclined bedding slopes.This study investigated the Shanyang landslide in Shaanxi Province,China.Using field investigations,laboratory tests of geotechnical parameters,and the 3DEC software,this study developed a numerical model of the landslide to analyze the failure process of such slopes.The findings indicate that the Shanyang landslide primarily crept along a weak interlayer under the action of gravity.The landslide,initially following a dip angle with the support of a stable inclined rock mass,shifted direction under the influence of argillization in the weak interlayer,moving towards the apparent dip angle.The slide resistance effect of the karstic dissolution zone was increasingly significant during this process,with lateral friction being the primary resistance force.A reduction in the lateral friction due to karstic dissolution made the apparent dip sliding characteristics of the Shanyang landslide more pronounced.Notably,deformations such as bending and uplift at the slope’s foot suggest that the main slide resistance shifts from lateral friction within the karstic dissolution zone to the slope foot’s resistance force,leading to the eventual buckling failure of the landslide.This study unveils a novel failure mode of apparent dip creep-buckling in the Shanyang landslide,highlighting the critical role of lateral friction from the karstic dissolution zone in its failure mechanism.These insights offer a valuable reference for mitigating risks and preventing disasters related to obliquely inclined bedding landslides.

Design and analysis of an untethered micro flapping robot which can glide on the water

Flapping-wing flying insects possess various advantages,such as high agility and efficiency.The design and manufacture of insect-scale flapping-wing micro aerial vehicle(FWMAV)have attracted increasing attention in recent decades.Due to the limitations of size and weight,the FWMAV with an onboard battery which can fully mimic insect flight has not been achieved.In this work,we design and fabricate a highly integrated flapping-wing microrobot named Robomoth.The Robomoth consists of a carbon chassis,customized power and control devices,and two piezoelectric ceramic actuators symmetrically distributed in the thorax and controlled individually.It weighs 2.487 g,spans 5.9 cm in length,possesses 9 cm of wingspan,and carries a 0.355 g rechargeable lithium battery.We demonstrate the mobility of the Robomoth through untethered gliding and making turns on the water surface.A simplified dynamic model of the flapping system is proposed to explain the relationship between the driving frequency and the flapping amplitude.The Robomoth is one new untethered bioinspired flapping-wing robot that can perform stable water surface motion,which holds potential applications such as search and rescue on the water.The robot can also provide insight for designing insect-scale flying vehicles.

目前传统的间歇制备过程由于受制于宏观尺度难以对工艺参数、产物结构及性能实现精准调控,导致产物的均一性差,制备过程存在显著的安全隐患。因此,如何实现含能材料的安全、可控和高效的制备已成为目前研究的热点。微反应器以其微型化、集成化、高度安全性和出色的传质传热效率等诸多优势,高度契合含能材料制备过程的需求。因此,微反应器技术逐渐引起了研究者的广泛关注,在含能材料制备中崭露头角。本文介绍了微反应器在危险化工合成过程及含能材料制备中应用的国内外研究进展,特别关注了其在单质含能材料和复合含能材料制备中的潜在应用,对未来的研究方向进行了展望,强调了未来微反应器技术领域的发展需求包括规模化生产、三废处理及智能化平台建设等。
